BACKGROUND: The pregnant women with intrahepatic cholestasis were at high risk of fetal distress, preterm birth and unexpected stillbirth. Intrahepatic cholestasis of pregnancy (ICP) was mainly caused by disorder of bile acid metabolism, whereas the specific mechanism was obscure. METHODS: We performed proteomics analysis of 10 ICP specimens and 10 placenta specimens from patients without ICP through data-independent acquisition (DIA) technique to disclose differentially expressed proteins. We executed metabolomic analysis of 30 ICP specimens and 30 placenta specimens from patients without ICP through UPLC-MS/MS to identify differentially expressed metabolites. Enrichment and correlation analysis was used to obtain the direct molecular insights of ICP development. The ICP rat models were constructed to validate pathological features. RESULTS: The heatmap of proteomics analysis showed the top 30 up-regulated and 30 down-regulated proteins. The metabolomic analysis revealed 20 richer and 4 less abundant metabolites in ICP samples compared with placenta specimens from patients without ICP, and enrichment pathways by these metabolites included primary bile acid biosynthesis, cholesterol metabolism, bile secretion, nicotinate and nicotinamide metabolism, purine metabolism and metabolic pathways. Combined analysis of multiple omics results demonstrated that bile acids such as Glycohyocholic acid, Glycine deoxycholic acid, beta-Muricholic acid, Noncholic acid, cholic acid, Gamma-Mercholic Acid, alpha-Muricholic acid and Glycochenodeoxycholic Aicd were significantly associated with the expression of GLRX3, MYL1, MYH7, PGGT1B, ACTG1, SP3, LACTB2, C2CD5, APBB2, IPO9, MYH2, PPP3CC, PIN1, BLOC1S1, DNAJC7, RASAL2 and ATCN3 etc. The core protein ACAT2 was involved in lipid metabolic process and animal model showed that ACAT2 was up-regulated in placenta and liver of pregnant rats and fetal rats. The neonates had low birth weight and Safranin O-Fast green FCF staining of animal models showed that poor osteogenic and chondrogenic differentiation of fetal rats. CONCLUSION: Multiple metabolites-alpha-Muricholic acid, beta-Muricholic acid, Glycine deoxycholic acid and Glycochenodeoxycholic Acid etc. were perfect biomarkers to predict occurrence of ICP. Bile acids were significantly associated with varieties of protein expression and these proteins were differentially expressed in ICP samples. Our study provided several biomarkers for ICP detection and potential therapeutic targets for ICP development.

Supercontinuum generation (SCG) from silica-based photonic crystal fibers (PCFs) is of highly technological significance from microscopy to metrology, but has been hindered by silica's relatively low intrinsic optical nonlinearity. The prevailing approaches of filling PCF with nonlinear gases or liquids can endow fibre with enhanced optical nonlinearity and boosted SCG efficiency, yet these hybrids are easily plagued by fusion complexity, environmental incompatibility or transmission mode instability. Here this work presents a strategy of embedding solid-state 2D MoS2 atomic layers into the air-holes of PCF to efficiently enhance SCG. This work demonstrates a 4.8 times enhancement of the nonlinear coefficient and a 70% reduction of the threshold power for SCG with one octave spanning in the MoS2-PCF hybrid. Furthermore, this work finds that the SCG enhancement is highly layer-dependent, which only manifests for a real 2D regime within the thickness of five atomic layers. Theoretical calculations reveal that the critical thickness arises from the trade-off among the layer-dependent enhancement of the nonlinear coefficient, leakage of fundamental mode and redshift of zero-dispersion wavelength. This work provides significant advances toward efficient SCG, and highlights the importance of matching an appropriate atomic layer number in the design of functional 2D material optical fibers.

Rabbits are an attractive meat livestock species that can efficiently convert human-indigestible plant biomass, and have been commonly used in biological and medical researches. Yet, transcriptomic landscape in muscle tissue and association between gene expression level and growth traits have not been specially studied in meat rabbits. In this study Oxford Nanopore Technologies (ONT) long-read sequencing technology was used for comprehensively exploring transcriptomic landscape in Longissimus dorsi for 115 rabbits at 84 days of age, and transcriptome-wide association studies (TWAS) were performed for growth traits, including body weight at 84 days of age and average daily gain during three growth periods. The statistical analysis of TWAS was performed using a mixed linear model, in which polygenic effect was fitted as a random effect according to gene expression level-based relationships. A total of 18,842 genes and 42,010 transcripts were detected, among which 35% of genes and 47% of transcripts were novel in comparison with the reference genome annotation. Furthermore, 45% of genes were widely expressed among more than 90% of individuals. The proportions (±SE) of phenotype variance explained by genome-wide gene expression level ranged from 0.501 ± 0.216 to 0.956 ± 0.209, and the similar results were obtained when explained by transcript expression level. In contrast, neither gene nor transcript was detected by TWAS to be statistically significantly associated with these growth traits. In conclusion, these novel genes and transcripts that have been extensively profiled in a single muscle tissue using long-read sequencing technology will greatly improve our understanding on transcriptional diversity in rabbits. Our results with a relatively small sample size further revealed the important contribution of global gene expression to phenotypic variation on growth performance, but it seemed that no single gene has an outstanding effect; this knowledge is helpful to include intermediate omics data for implementing genetic evaluation of growth traits in meat rabbits.

Atmospheric drag is an important influencing factor in precise orbit determination and the prediction of low-orbit space debris. It has received widespread attention. Currently, calculating atmospheric drag mainly relies on different atmospheric density models. This experiment was designed to explore the impact of different atmospheric density models on the orbit prediction of space debris. In the experiment, satellite laser ranging data published by the ILRS (International Laser Ranging Service) were used as the basis for the precise orbit determination for space debris. The prediction error of space debris orbits at different orbital heights using different atmospheric density models was used as a criterion to evaluate the impact of atmospheric density models on the determination of space-target orbits. Eight atmospheric density models, DTM78, DTM94, DTM2000, J71, RJ71, JB2006, MSIS86, and NRLMSISE00, were compared in the experiment. The experimental results indicated that the DTM2000 atmospheric density model is best for determining and predicting the orbits of LEO (low-Earth-orbit) targets.

Reversible thermochromism presents depth at low temperatures and brightness at high temperatures, which is not conducive to the application of the passive energy-saving and shading effect. Here, we propose an innovative strategy for unconventional reversible thermochromic energy-storage flexible wood films (FT-PCMs-2) by the contribution of polyvinyl alcohol/propylene glycol (PVA/PG). Upon heating, the FT-PCMs-2 changes from light red/green to dark red/green, in contrast to the color change of the non-flexible composites without PVA/PG. The FT-PCMs-2 has good flexibility, mechanical strength and thermal stability. Among them, MSR-DEW-6PVA and MSG-DEW-6PVA have better comprehensive properties, including suitable phase change temperatures (38.6 °C and 37.2 °C), high latent heat values (59.79 J g-1 and 73.02 J g-1), and low supercooling degrees (2 °C and 0.6 °C). FTIR, XRD and SEM analysis show that the binary fatty acid eutectics were successfully loaded into the eucalyptus fibre skeleton with physical interaction with PVA/PG. In addition, MSR-DEW-6PVA and MSG-DEW-6PVA have photothermal conversion rates of 48.3% and 36%, good cycling stability and anti-leakage performance after 50 cycles, and have promising applications in the fields of building energy saving, intelligent windows and decoration.

Atmospheric drag is an important factor affecting orbit determination and prediction of low-orbit space debris. To obtain accurate ballistic coefficients of space debris, we propose a calculation method based on measured optical angles. Angle measurements of space debris with a perigee height below 1400 km acquired from a photoelectric array were used for orbit determination. Perturbation equations of atmospheric drag were used to calculate the semi-major-axis variation. The ballistic coefficients of space debris were estimated and compared with those published by the North American Aerospace Defense Command in terms of orbit prediction error. The 48 h orbit prediction error of the ballistic coefficients obtained from the proposed method is reduced by 18.65% compared with the published error. Hence, our method seems suitable for calculating space debris ballistic coefficients and supporting related practical applications.

Obesity has become a major health problem worldwide, and increasing evidence supports the importance of microRNAs (miRNAs) in its pathogenesis. Recently, we found that miR-383-5p_1 is highly expressed in the perirenal fat of high-fat-fed rabbits, but it is not yet known whether miR-383-5p is involved in lipid metabolism. Here, we used transcriptome sequencing technology to screen 1642 known differentially expressed genes between miR-383-5p mimic groups and miR-383-5p negative control groups. Gene Ontology Resource (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) were enriched in the pathway related to lipid metabolism, and glycine biosynthesis, the NOD receptor signal pathway and nonalcoholic fatty liver were significantly enriched. Afterwards, our research results indicated that miR-383-5p can promote the proliferation and differentiation of rabbit preadipocytes, and there is a direct targeting relationship with RAD51AP1. Mechanistically, miR-383-5p directly interacts with the lipid metabolism and participates in adipogenesis and lipid accumulation by targeting RAD51AP1. In conclusion, our data highlight a physiological role for miRNA in lipid metabolism and suggest the miR-383-5p/RAD51AP1 axis may represent a potential mechanism for controlling lipid accumulation in obesity.

BACKGROUND: The co-occurrence of Anti-phospholipase A2 receptor-associated membranous nephropathy (anti-PLA2R-MN) and human immunodeficiency virus (HIV) infection is a rare clinical scenario, presenting significant challenges in terms of management and treatment. CASE SUMMARY: A 32-year-old Chinese male diagnosed with HIV infection presented with a clinical history of proteinuria persisting for over two years. A kidney biopsy demonstrated subepithelial immune complex deposition and a thickened glomerular basement membrane, indicative of stage I-II membranous nephropathy. Immunofluorescence staining revealed granular deposition of PLA2R (3+) along the glomerular capillary loops, corroborated by a strongly positive anti-PLA2R antibody test (1:320). Initial treatment involving losartan potassium, rivaroxaban, tacrolimus, and rituximab was discontinued due to either poor effectiveness or the occurrence of adverse events. Following a regimen of weekly subcutaneous injections of telitacicept (160 mg), a marked decline in the 24 h urine protein was observed within a three-month period, accompanied by a rise in serum albumin level. No significant reductions in peripheral blood CD3+CD4+T and CD3+CD8+T cell counts were detected. The patient's physical and psychological conditions showed significant improvements, with no adverse events reported during the treatment course. CONCLUSION: Telitacicept might offer a potential therapeutic avenue for patients diagnosed with anti-PLA2R-MN concomitant with HIV infection.

Passive daytime radiative cooling (PDRC) as a zero-energy consumption cooling method has broad application potential. Common commercial crystalline silicon (c-Si) solar cell arrays suffer working efficiency loss due to the incident light loss and overheating. In this work, a radiative cooler with PDMS (polydimethylsiloxane) film and embedded SiO2 microparticles was proposed to use in silicon solar cells. Both anti-reflection and radiative cooling performance can be improved through numerical parametric study. For the best performing of PDMS/SiO2 radiative cooler, the thickness of PDMS layer, volume fraction and radius of the embedded SiO2 particles have been determined as 55 µm, 8% and 500 nm, respectively. 94% of emissivity in first atmospheric window band (8-13 µm) for radiative cooling and 93.4% of solar transmittance at the crystalline silicon absorption band (0.3-1.1 µm) were achieved. We estimated that the PDMS/SiO2 radiative cooler can lower the temperature of a bare c-Si solar cell by 9.5°C, which can avoid 4.28% of efficiency loss. More incident light can enter and be utilized by silicon layer to enhance the efficiency of the solar cells. The proposed difunctional radiative cooling coating may become guidance for next generation encapsulation of crystalline silicon solar cells.

BACKGROUND: Choosing the appropriate antipsychotic drug (APD) treatment for patients with schizophrenia (SCZ) can be challenging, as the treatment response to APD is highly variable and difficult to predict due to the lack of effective biomarkers. Previous studies have indicated the association between treatment response and genetic and epigenetic factors, but no effective biomarkers have been identified. Hence, further research is imperative to enhance precision medicine in SCZ treatment. METHODS: Participants with SCZ were recruited from two randomized trials. The discovery cohort was recruited from the CAPOC trial (n = 2307) involved 6 weeks of treatment and equally randomized the participants to the Olanzapine, Risperidone, Quetiapine, Aripiprazole, Ziprasidone, and Haloperidol/Perphenazine (subsequently equally assigned to one or the other) groups. The external validation cohort was recruited from the CAPEC trial (n = 1379), which involved 8 weeks of treatment and equally randomized the participants to the Olanzapine, Risperidone, and Aripiprazole groups. Additionally, healthy controls (n = 275) from the local community were utilized as a genetic/epigenetic reference. The genetic and epigenetic (DNA methylation) risks of SCZ were assessed using the polygenic risk score (PRS) and polymethylation score, respectively. The study also examined the genetic-epigenetic interactions with treatment response through differential methylation analysis, methylation quantitative trait loci, colocalization, and promoter-anchored chromatin interaction. Machine learning was used to develop a prediction model for treatment response, which was evaluated for accuracy and clinical benefit using the area under curve (AUC) for classification, R2 for regression, and decision curve analysis. RESULTS: Six risk genes for SCZ (LINC01795, DDHD2, SBNO1, KCNG2, SEMA7A, and RUFY1) involved in cortical morphology were identified as having a genetic-epigenetic interaction associated with treatment response. The developed and externally validated prediction model, which incorporated clinical information, PRS, genetic risk score (GRS), and proxy methylation level (proxyDNAm), demonstrated positive benefits for a wide range of patients receiving different APDs, regardless of sex [discovery cohort: AUC = 0.874 (95% CI 0.867-0.881), R2 = 0.478; external validation cohort: AUC = 0.851 (95% CI 0.841-0.861), R2 = 0.507]. CONCLUSIONS: This study presents a promising precision medicine approach to evaluate treatment response, which has the potential to aid clinicians in making informed decisions about APD treatment for patients with SCZ. In 2020, Changchun Observatory developed a 280 mm wide-field optical telescope array to improve surveillance of space debris in the geosynchronous belt. There are many advantages including a wide field of view, the ability to observe a large area of sky and high reliability. However, the wide field of view causes a significant number of background stars to appear in the image when photographing space objects, making it difficult to detect them. This research focuses on the precise detection of GEO space objects from images taken by this telescope array in order to position them in large quantities. Our work further investigates the motion feature of an object, namely that the object can be seen as being in a uniform linear motion for a brief length of time. Based on this feature, the belt can be divided into a number of smaller areas and the telescope array scans each smaller area one at a time from east to west. To detect objects in the subarea, a combination of image differencing with trajectory association is used. The image differencing algorithm is used to remove most stars and screen out suspected objects in the image. Next, the trajectory association algorithm is employed to further filter out the real objects among the suspected ones, and the trajectories attributed to the same object are linked. The feasibility and accuracy of the approach were verified by the experiment results. The accuracy rate of trajectory association exceeds 90% and on average, more than 580 space objects can be detected per observation night. Since the J2000.0 equatorial system can accurately describe the apparent position of an object, the object can be detected by using this coordinate system as opposed to the pixel coordinate system.

Imbalances in the gut microbiota composition may lead to several reproductive disorders and diseases during pregnancy. This study investigates the fecal microbiome composition between primiparous and multiparous cows during non-pregnancy and pregnancy to analyze the host-microbial balance at different stages. The fecal samples obtained from six cows before their first pregnancy (BG), six cows during their first pregnancy (FT), six open cows with more than three lactations (DCNP), and six pregnant cows with more than three lactations (DCP) were subjected to 16S rRNA sequencing, and a differential analysis of the fecal microbiota composition was performed. The three most abundant phyla in fecal microbiota were Firmicutes (48.68%), Bacteroidetes (34.45%), and Euryarchaeota (15.42%). There are 11 genera with more than 1.0% abundance at the genus level. Both alpha diversity and beta diversity showed significant differences among the four groups (p < 0.05). Further, primiparous women were associated with a profound alteration of the fecal microbiota. The most representative taxa included Rikenellaceae_RC9_gut_group, Prevotellaceae_UCG_003, Christensenellaceae_R_7_group, Ruminococcaceae UCG-005, Ruminococcaceae UCG-013, Ruminococcaceae UCG-014, Methanobrevibacter, and [Eubacterium] coprostanoligenes group, which were associated with energy metabolism and inflammation. The findings indicate that host-microbial interactions promote adaptation to pregnancy and will benefit the development of probiotics or fecal transplantation for treating dysbiosis and preventing disease development during pregnancy.

Microcombs generated in photonic integrated circuits can provide broadband and coherent optical frequency combs with a high repetition rate from microwave to terahertz. Coherent microcombs formed in normal group velocity dispersion microresonators usually have a flat-top temporal profile, called platicon. Here, we propose a novel scheme to generate platicon in Si3N4 microresonator with the assistance of third-harmonic generation. The nonlinear coupling between the fundamental and the third-harmonic waves that draws support from third-order sum/difference frequency generation provides a new mechanism to achieve the phase matching of four-wave mixing in normal dispersion microresonators. We show that single or multiple platicons can be obtained by changing the third-harmonic nonlinear coupling strength and phase matching condition for third-order sum/difference frequency generation. Our work provides a promising solution to facilitate coherent and visible microcomb generation in a pure χ(3) microresonator, which is potential for self-referencing combs and optical clock stabilization.

In the past few years, the increasing amount of space debris has triggered the demand for distributed surveillance systems. Long exposure time can effectively improve the target detection capability of the wide-area surveillance system. Problems that also cause difficulties in space-target detection include large amounts of data, countless star points, and discontinuous or nonlinear targets. In response to these problems, this paper proposes a high-precision space-target detection and tracking pipeline that aims to automatically detect debris data in space. First, a guided filter is used to effectively remove the stars and noise, then Hough transform is used to detect space debris, and finally Kalman filter is applied to track the space debris target. All experimental images are from Jilin Observatory, and the telescope is in star-tracking mode. Our method is practical and effective. The results show that the proposed automatic extraction channel of space debris can accurately detect and track space targets in a complex background.

With the comprehensive prohibition of antibiotics in the feed industry in China, the incidence of diarrhea in rabbits increased, such as loss of appetite, vomiting, and excretion of atheromatous feces. In order to explore the pathological and the molecular mechanisms of the diarrhea in the rabbitry fed with antibiotic-free diet, we used microbial metagenomics, transcriptome, and non-targeted metabolomics sequencing. The results showed that the Firmicutes level was significantly decreased (p < 0.001) and the Proteobacteria level was significantly increased (p < 0.05). The functional enrichment of cecum revealed that most differentially expressed genes (DEGs) were expressed in immune, inflammatory, and metabolic processes. The enrichment of the cecal fecal metabolites focused on the bile secretion, antifolate resistance, and tryptophan metabolism pathways, which are mainly associated with inflammation. The results of correlation analysis showed that Fournierella was positively correlated with myricetin, ursolic acid, and furtherly might cause bile secretion and tryptophan metabolism disorder, aggravate intestinal inflammation, change intestinal permeability, and reduce host immunity, leading to diarrhea in rabbits. This study provides a theoretical basis for illustrating the reason for diarrhea and developing new feeds for the health of rabbits.

In this article, we study how the choice of parameters of a slow saturable absorber (SSA) affects the stable operation of a soliton fiber comb laser. We show that a shorter recovery time for the SSA does not always lead to shorter modelocked pulses. Instead, increasing the cavity gain plays a critical role in generating stable modelocked pulses with higher energy and shorter durations. We find that more stable, shorter, and more energetic output pulses can be achieved with lower saturation energies of the SSA and/or higher anomalous dispersion within the cavity.

We demonstrate high quality (Q) factor microring resonators in high index-contrast GeSbSe chalcogenide glass waveguides using electron-beam lithography followed by plasma dry etching. A microring resonator with a radius of 90 µm shows an intrinsic Q factor of 4.1 × 105 in the telecom band. Thanks to the submicron waveguide dimension, the effective nonlinear coefficient was determined to be up to ∼110 W-1m-1 at 1550 nm, yielding a larger figure-of-merit compared with previously reported submicron chalcogenide waveguides. Such a high Q factor, combined with the large nonlinear coefficient and high confinement, shows the great potential of the GeSbSe microring resonator as a competitive platform in integrated nonlinear photonics.

RESUMEN

In this study, two kinds of form-stable multifunctional materials with thermal and electrical response (FPCMs: DP-E7U3-CNT, DP-E7T3-CNT) are composed of wood-based honeycomb-like celluloses micro-framework (DP), carbon nanotubes (CNT), erythritol-urea (E7U3) or erythritol-thiourea (E7T3). In FPCMs, DP acts as a skeleton structure to seal E7U3 and E7T3 and provide more pathways for heat conduction. The CNT acts as an extended surface to further improve thermal conductivity. FE-SEM showed that the honeycomb-like pore structure of DP was completely filled with E7U3, E7T3 and CNT. FTIR and XRD analysis show that there is only a combination of physical interactions between the components of FPCMs. DSC curves and thermal conductivity analysis results show that DP-E7U3-1.5CNT and DP-E7T3-1.5CNT with the mass fraction of carbon nanotubes (1.5 wt%) have the highest latent heat values (230.3 J/g, 272.2 J/g) and thermal conductivity (0.9832 W/(m·K), 0.9363 W/(m·K)). Both DP-E7U3-1.5CNT and DP-E7T3-1.5CNT exhibit high latent heat retention and thermal stability after 100 heating-cooling cycles. In addition, DP-E7U3-1.5CNT and DP-E7T3-1.5CNT show excellent performance in light-heat energy conversion-storage, actual latent heat storage and release, thermal and electrical response performance, which make it has great potential to be multifunctional materials with thermal storage sand electrical response.
