RESUMEN

Significance: The initiation of goal-directed actions is a complex process involving the medial prefrontal cortex and dopaminergic inputs through the mesocortical pathway. However, it is unclear what information the mesocortical pathway conveys and how it impacts action initiation. In this study, we unveiled the indispensable role of mesocortical axon terminals in encoding the execution of movements in self-initiated actions. Aim: To investigate the role of mesocortical axon terminals in encoding the execution of movements in self-initiated actions. Approach: We designed a lever-press task in which mice internally determine the timing of the press, receiving a larger reward for longer waiting periods. Results: Our study revealed that self-initiated actions depend on dopaminergic signaling mediated by D2 receptors, whereas sensory-triggered lever-press actions do not involve D2 signaling. Microprism-mediated two-photon calcium imaging further demonstrated ramping activity in mesocortical axon terminals approximately 0.5 s before the self-initiated lever press. Remarkably, the ramping patterns remained consistent whether the mice responded to cues immediately for a smaller reward or held their response for a larger reward. Conclusions: We conclude that mesocortical dopamine axon terminals encode the timing of self-initiated actions, shedding light on a crucial aspect of the intricate neural mechanisms governing goal-directed behavior.

BACKGROUND: Instillation of highly soluble nanoparticles (NPs) into the lungs of rodents can cause acute eosinophilia without any previous sensitizations by the role of dissolved ions. However, whether gradually dissolving NPs can cause the same type of eosinophilia remains to be elucidated. We selected nickel oxide (NiO) as a gradually dissolving NP and evaluated the time course pulmonary inflammation pattern as well as its mechanisms. METHODS: NiO NPs were intratracheally instilled into female Wistar rats at various concentrations (50, 100, and 200 cm(2)/rat) and the lung inflammation was evaluated at various time-points (1, 2, 3, and 4 days). As positive controls, NiCl2 and the ovalbumin-induced allergic airway inflammation model was applied. NiCl2 was instilled at 171.1 µg Ni/rat, which is equivalent nickel concentration of 200 cm(2)/rat of NiO NPs. Cytological analysis and biochemical analysis including lactate dehydrogenase (LDH), total protein, and pro-inflammatory cytokines were measured in bronchoalveolar lavage fluid (BALF). The levels of total immunoglobulin E (IgE) and anaphylatoxins (C3a and C5a) were measured in BALF and serum. The levels of eotaxin were measured in the alveolar macrophages and normal lung tissue before and after addition of cell lysis buffer to evaluate whether the direct lysis of cells can release intracellular eotaxin. RESULTS: NiO NPs produced acute neutrophilic inflammation throughout the study. However, eosinophils were recruited at 3 and 4 days post-instillation of NiO NPs and the magnitude and pattern of inflammation was similar with NiCl2 at 24 h post-instillation. The eosinophil recruitment by NiO NPs was not related with either the levels of total IgE or anaphylatoxins. The lysis of alveolar macrophages and normal lung tissue showed high levels of intracellular eotaxin and the levels of LDH showed positive correlation with the levels of eotaxin. CONCLUSIONS: Instillation of NiO NPs produced neutrophilia at 1 and 2 days after instillation, while the mixed type of neutrophilic and eosinophilic inflammation was produced at 3 and 4 days post-instillation, which was consistent with NiCl2. The mechanism of the eosinophilia involves the direct release of intracellular eotaxin due to the rupture of cells by the accumulated solubilized nickel ions in the phagolysosome.

This study presents the inductively coupled plasma mass spectrometry (ICP-MS) as a method for tracing the regional origin of ginseng. The results of the analysis of 15 Korean ginsengs from three different regions and of 15 Chinese ginsengs from three different regions reveal that the Sr isotope ratios (87)Sr/(86)Sr of the ginsengs differed according to their origin. For pretreatment, the ginseng samples were dried, and were dissolved through microwave digestion, then were each made to amount to 6ml with 11.9M HCl. Rb was then separated from Sr to enable an interference-free measurement through cation exchange chromatography. Six millilitres of the ginseng sample were injected in the column, and 60ml of 11.9M HCl was passed through the column at a 1mlmin(-1) flow rate to separate Rb from Sr. After Rb was eluted completely, 60ml of 5.0M HCl was passed at a 1mlmin(-1) flow rate to collect Sr. In the Sr collection step, the first 10ml portion of 30ml eluate was discarded, and the next 10ml portion was taken and was diluted with de-ionized water at a ratio of 1:3, for analysis purposes. The results of the analysis of 30 ginseng samples revealed that the Chinese ginsengs have an (87)Sr/(86)Sr ratio range of 0.672-0.701, and the Korean ginsengs 0.705-0.714. The Korean ginsengs, therefore, have a higher (87)Sr/(86)Sr ratio range than the Chinese ginsengs. Of the Korean ginsengs, (87)Sr/(86)Sr ratio range of ginsengs from Punggi, Geumsan and Hongcheon are about 0.706-0.709, 0.705-0.706, and 0.710-0.714, respectively.
