RESUMEN

Reading comprehension deficits are common for students with Autism Spectrum Disorder (ASD) but there are few studies that have examined specific strategies for teaching reading comprehension to this population. The current study investigated the effect of embedding the restricted interests (RI) of two high school students with ASD in text on reading comprehension performance using a single-subject, multi-element research design. Neither participant showed an increase in the number of relevant words shared during oral retell and only one participant showed an increase in the percent of correctly answered reading comprehension questions. Embedding the RI in text more frequently did not impact reading comprehension performance. The results indicate there are potential variables that may limit the effect of embedding the RI of students with ASD in text on reading comprehension.

In this study, we have prepared a series of 4- and 6-arm star-shaped polymers with varying molecular weight and hydrophobicity in order to provide insight into the role and relationship that shape and composition have on the binding and protecting of oral relevant surfaces (hydroxyapatite, HAP) from bacteria colonization. Star-shaped acrylic acid polymers were prepared by free-radical polymerization in the presence of chain transfer agents with thiol groups, and their binding to the HAP surfaces and subsequent bacteria repulsion was measured. We observed that binding was dependent on both polymer shape and hydrophobicity (star vs. linear), but their relative efficacy to reduce oral bacteria attachment from surfaces was dependent on their hydrophobicity only. We further measured the macroscopic effects of these materials to modify the mucin-coated HAP surfaces through contact angle experiments; the degree of angle change was dependent on the relative hydrophobicity of the materials suggesting future in vivo efficacy. The results from this study highlight that star-shaped polymers represent a new material platform for the development of dental applications to control bacterial adhesion which can lead to tooth decay, with various compositional and structural aspects of materials being vital to effectively design oral care products.

BACKGROUND: Stannous fluoride (SnF2) is a compound present in many commercially available dentifrices; however, oxidative decomposition negatively impacts its efficacy. Stannous oxidation is often mitigated through the addition of complexing agents or sources of sacrificial stannous compounds. The authors have found that the addition of zinc phosphate significantly improved stannous stability more effectively than other stabilization methods. The authors evaluated the chemical speciation of stannous compounds within a variety of formulations using x-ray absorption near edge spectroscopy (XANES), a technique never used before in this manner. These data were compared and correlated with several antimicrobial experiments. METHODS: XANES data of various commercially available compounds and Colgate TotalSF were performed and analyzed against a library of reference compounds to determine the tin chemical speciation. The antibacterial assays used were salivary adenosine triphosphate, short-interval kill test, plaque glycolysis, and anaerobic biofilm models. RESULTS: XANES spectra showed a diverse distribution of tin species and varying degrees of SnF2 oxidation. In vitro antimicrobial assessment indicated significant differences in performance, which may be correlated to the differences in tin speciation and oxidation state. CONCLUSIONS: Driven by the excipient ingredients, SnF2 dentifrices contain a distribution of tin species in either the SnF2 or Sn(IV) oxidation state. The addition of zinc phosphate provided significant robustness against oxidation, which directly translated to greater efficacy against bacteria. PRACTICAL IMPLICATIONS: The choice of inactive ingredients in a dentifrice with active SnF2 can dramatically impact product stability.

Water-soluble, linear polymers with high-acid functionality are commonly used in oral care formulations to provide benefits such as bioactive complexation and delivery, as well as inhibition of the bacteria deposition and colonization, commonly referred to as 'anti-attachment'. Unfortunately, structure-activity relationship (SAR) studies of these polymers are scarce, thus, a systematic approach to design polymers with a desired property (e.g. anti-attachment) is limited. Multifunctional anti-attachment amphiphilic molecules (AMs) featuring a sugar backbone, hydrophobic arms, a poly(ethylene glycol) tail, and a chemical anchor effectively deposited on soft ceramic surfaces and reduced bacterial adhesion. The chemical compositions of the AMs were fine-tuned to better coordinate with dental enamel surfaces and prevent bacterial colonization. A graft-to approach was used to investigate the effect of the chemical anchor on AM deposition and retention. The chemical composition, absorption/desorption, and wettability properties of the bioactives and bioactive-coated surfaces were investigated using nuclear magnetic resonance, X-ray photon spectroscopy, quartz crystal microbalance, and contact angle. In addition, the ability of the AMs to provide anti-bacterial attachment on a simulated enamel surface was evaluated in vitro using bacterial repulsion assays. The SAR between surface retention and anti-attachment properties of the AMs demonstrates the feasibility and tunability of using these polymers as bioactive agents that provide anti-attachment benefits on dental enamel surfaces.

A current effort in preventive dentistry is to inhibit surface attachment of bacteria using antibacterial polymer coatings on the tooth surface. For the antibacterial coatings, the physisorption of anionic and cationic polymers directly onto hydroxyapatite (HA) and saliva-treated HA surfaces was studied using quartz crystal microbalance, force spectroscopy, and atomic force microscopy. First, single species adsorption is shown to be stronger on HA surfaces than on silicon oxide surfaces for all polymers (i.e., Gantrez, sodium hyaluronate (NaHa), and poly(allylamine-co-allylguanidinium) (PAA-G75)). It is observed through pH dependence of Gantrez, NaHa, and PAA-G75 adsorption on HA surfaces that anionic polymers swell at high pH and collapse at low pH, whereas cationic polymers behave in the opposite fashion. Thicknesses of Gantrez, NaHa, and PAA-G75 are 52 nm (46 nm), 35 nm (11 nm), and 6 nm (54 nm) at pH 7 (3.5), respectively. Second, absorption of charged polymer is followed by absorption of the oppositely charged polymer. Upon exposure of the anionic polymer layers, Gantrez and NaHa, to the cationic polymer, PAA-G75, films collapse from 52 to 8 nm and 35 to 11 nm, respectively. This decrease in film thickness is attributed to the electrostatic cross-linking between anionic and cationic polymers. Third, for HA surfaces pretreated with artificial saliva (AS), the total thickness decreases from 25 to 16 nm upon exposure to PAA-G75. Force spectroscopy is used to further investigate the PAA-G75/AS coating. The results show that the interaction between a negatively charged colloidal bead and the AS surface is strongly repulsive, whereas PAA-G75/AS is attractive but varies across the surface. Additionally, AFM studies show that AS/HA is smooth with a RMS roughness of 1.7 nm, and PAA-G75-treated AS/HA is rough (RMS roughness of 5.4 nm) with patches of polymer distributed across the surface with an underlying coating. The high roughness of PAA-G75 treated AS/HA is attributed to the strong adsorption of the relatively small PAA-G75 onto the heterogeneously distributed negatively charged AS surface. In addition, uptake of PAA-G75 by pellicle layer (saliva-treated HA surface) is observed, and the adsorbed amount of PAA-G75 on/into pellicle layer is ∼2 times more than that on/into AS layer. These studies show that polymer adsorption onto HA and saliva-coated HA depends strongly on the polymer type and size and that there is an electrostatic interaction between polymer and saliva and/or oppositely charged polymers that stabilizes the coatings on HA. Lastly, assessing the viability of the adherent bacteria collected from the PAA-G75-coated surfaces showed a significant reduction (∼93%) in bacterial viability when compared to bacteria collected from untreated and Gantrez-coated HA. These results suggest the potential antimicrobial activity of PAA-G75.

CASE DESCRIPTION Within a 2-week period, 4 southern cassowaries (Casuarius casuarius) at an exhibit at a Virginia zoo died acutely subsequent to eastern equine encephalitis virus (EEEV) infection. This prompted a search for other EEEV outbreaks in cassowaries, which resulted in the identification of 2 additional cassowaries that died of EEEV infection at a conservation center in Florida. CLINICAL FINDINGS Both juvenile and adult birds were affected. Three of the 6 birds died acutely with no premonitory signs. Clinical disease in the other 3 birds was characterized by lethargy and ataxia. Clinicopathologic findings typically included leukocytosis, hyperuricemia, abnormally high liver enzyme activities, and hyper-ß globulinemia, which was indicative of acute inflammation. TREATMENT AND OUTCOME The 3 birds with clinical disease died despite supportive treatment. Gross abnormalities commonly observed during necropsy included coelomitis and evidence of diarrhea. Frequently observed histologic abnormalities were encephalitis, vasculitis, hepatitis, nephritis, and splenitis. The diagnosis of EEEV infection was confirmed by detection of serum anti-EEEV antibodies or detection of viral RNA in brain tissue by use of a reverse-transcriptase PCR assay. CLINICAL RELEVANCE Findings suggested that EEEV can cause high morbidity and mortality rates in southern cassowaries. Clinical disease might be reduced or prevented by vaccination, isolation of ill birds, and mosquito control strategies.

In order to start providing important early intervention services to preschoolers and toddlers with autism, those children first need to be identified. Despite the availability of specialized autism assessment instruments, there is a need for effective screeners at the early childhood level. Three broadband behavior rating scales were evaluated in this study to determine if any of the scales on the instruments could adequately distinguish between children with autism from other clinically referred children. There were four scales from two instruments that resulted in mean scores outside the average range and had statistically significant differences. However, the small mean score differences and analyses of sensitivity and specificity suggest those scales have limited practical usefulness when used by clinicians.

An 8-yr-old, captive, spayed, female maned wolf (Chrysocyon brachyurus) developed progressive lethargy and weakness over a 24-hr period. Clinical signs included vomiting, recumbency, horizontal nystagmus, possible blindness, pale icteric mucus membranes, and port-wine colored urine. A complete blood cell count revealed severe anemia (packed cell volume [PCV], 6%) and intraerythrocytic piroplasms consistent with a Babesia species. Polymerase chain reaction testing later confirmed babesiosis. The wolf was treated with imidocarb dipropionate, antibiotics, and fluid therapy. A whole-blood transfusion from a sibling maned wolf also was performed. Despite aggressive treatment, the wolf failed to improve and was euthanized. To the authors' knowledge, this is the first documented case of babesiosis in a captive maned wolf in North America. Surveillance of infectious diseases in captive and wild maned wolf populations should be expanded to include screening for Babesia species. Tick control also should be implemented to prevent and decrease transmission of the disease to this endangered species.

Two 12-year-old cats were diagnosed with chronic kidney disease (CKD) based on physical examination, clinicopathologic data and, in one case, abdominal ultrasound findings. Approximately 1 year after the initial diagnosis of CKD both cats developed renal transitional cell carcinoma (TCC)--bilateral in one cat. Based on post-mortem examination, one cat had no evidence of metastasis and the other had metastasis to the large intestine, heart and lungs. This is the first report of de novo bilateral renal TCC in a cat, as well as the first report of renal TCC developing in cats with previous history of confirmed CKD.

Gene-directed enzyme prodrug therapy (GDEPT) has been investigated as a means of cancer treatment without affecting normal tissues. This system is based on the delivery of a suicide gene, a gene encoding an enzyme which is able to convert its substrate from non-toxic prodrug to cytotoxin. In this experiment, we have developed a targeted suicide gene therapeutic system that is completely contained within tumor-tropic cells and have tested this system for melanoma therapy in a preclinical model. First, we established double stable RAW264.7 monocyte/macrophage-like cells (Mo/Ma) containing a Tet-On® Advanced system for intracellular carboxylesterase (InCE) expression. Second, we loaded a prodrug into the delivery cells, double stable Mo/Ma. Third, we activated the enzyme system to convert the prodrug, irinotecan, to the cytotoxin, SN-38. Our double stable Mo/Ma homed to the lung melanomas after 1 day and successfully delivered the prodrug-activating enzyme/prodrug package to the tumors. We observed that our system significantly reduced tumor weights and numbers as targeted tumor therapy after activation of the InCE. Therefore, we propose that this system may be a useful targeted melanoma therapy system for pulmonary metastatic tumors with minimal side effects, particularly if it is combined with other treatments.

Localized magnetic hyperthermia as a treatment modality for cancer has generated renewed interest, particularly if it can be targeted to the tumor site. We examined whether tumor-tropic neural progenitor cells (NPCs) could be utilized as cell delivery vehicles for achieving preferential accumulation of core/shell iron/iron oxide magnetic nanoparticles (MNPs) within a mouse model of melanoma. We developed aminosiloxane-porphyrin functionalized MNPs, evaluated cell viability and loading efficiency, and transplanted neural progenitor cells loaded with this cargo into mice with melanoma. NPCs were efficiently loaded with core/shell Fe/Fe(3)O(4) MNPs with minimal cytotoxicity; the MNPs accumulated as aggregates in the cytosol. The NPCs loaded with MNPs could travel to subcutaneous melanomas, and after A/C (alternating current) magnetic field (AMF) exposure, the targeted delivery of MNPs by the cells resulted in a measurable regression of the tumors. The tumor attenuation was significant (p < 0.05) a short time (24 h) after the last of three AMF exposures.

Two artificial dipeptides containing both a pendant monodentate (pyridine (py)) and tridentate (terpyridine (tpy) or phenyl terpyridine (varphi-tpy)) ligand on an aminoethylglycine (aeg) backbone have been synthesized. These oligopeptides are fully characterized by one and two-dimensional NMR spectroscopy, mass spectrometry, and elemental analysis. The ligands were chosen because they coordinate Cu(2+) to form [Cu(py)(tpy)](2+) complexes; when bound to the dipeptide scaffold, Cu(2+) chelation cross-links the strands to form double-stranded duplex structures with an antiparallel arrangement. Using spectrophotometric titrations, we observe coordination of one Cu(2+) metal per dipeptide strand. Mass spectrometry, NMR spectroscopy, vapor pressure osmometry, and HPLC confirm that the resulting structures are the dipeptide duplex cross-linked by two metal centers.

Multifunctional aminoethylglycine (aeg) derivatized [Ru(bpy)(3)](2+) complexes with pendant bipyridine (bpy) ligands coordinate Cu(2+) to form coordinative chain cross-links in a "hairpin loop" motif. In this paper, we report the synthesis and characterization of a series of Ru aeg hairpins in which the relative aeg chain length and number of pendant bpy ligands is varied. Reaction of each of these with Cu(2+) is monitored using spectrophotometric emission titrations to determine the binding stoichiometry. Coordination of Cu(2+) causes quenching of the emissive excited state Ru species; the degree of quenching efficiency depends on the location and number of coordinated Cu ions. The heterometallic structures are fully characterized, and using the quantum yields and time-resolved emission following excitation of the Ru complexes in deoxygenated solutions, the radiative (k(r)) and nonradiative (k(nr)) relaxation rates are compared. These data reveal only a shallow decrease in k(nr) with increasing distance between the Ru and Cu complexes. Activation energies, determined from temperature dependent studies of the time-resolved emission, also increase as the Ru-Cu separation increases, resulting in the smaller nonradiative rates. Together, these data are suggestive of excited state electron transfer as the quenching mechanism and demonstrate that metal coordination self-assembles structures made from modular artificial amino acids can provide controlled arrangements of chromophores, electron donors, and electron acceptors to shuttle electrons in a new approach for mimicking photosynthesis.

Directed self-assembly using inorganic coordination chemistry is an attractive approach for making functional supramolecular structures. In this article, the synthesis and characterization of Ru(bpy) 3 (2+) compounds derivatized with aminoethylglycine (aeg) substituents containing pendant bipyridine (bpy) ligands is presented. The free bpy ligands in these complexes are available for metal chelation to form coordinative cross-links; addition of Cu (2+) or Zn (2+) assembles heterometallic structures containing two or three transition-metal complexes. Control over relative placement of metal complexes is accomplished using two strategies: two bipyridine-containing aeg strands tethered to Ru(bpy) 3 (2+) allow intramolecular coordination and result in a dimetallic hairpin motif. Ru(bpy) 3 (2+) modified with a single strand forms intermolecular cross-links forming the trimetallic complex. Each of these is characterized by a range of methods, and their photophysical properties are compared. These data, and comparison to an acetyl aeg- modified Ru(bpy) 3 (2+) complex, confirm that the metal ions cross-link bpy-containing aeg strands. Heterometallic complexes containing bound Cu (2+) cause a dramatic reduction in the Ru(bpy) 3 (2+) quantum yields and lifetimes. In contrast, the Ru(bpy) 3 (2+) hairpin with coordinated Zn (2+) has only a slight decrease in quantum yield but no change in lifetime, which could be a result of steric impacts on structure in the dimetallic species. Analogous effects are not observed in the trimetallic Ru-Zn-Ru structures in which this constraint is absent. Each of these heterometallic structures represents a facile and reconfigurable means to construct multimetallic structures by metal-coordination-based self-assembly of modular artificial peptide units.
