RESUMEN

Microfluidic channels fabricated over fabrics or papers have the potential to find substantial application in the next generation of wearable healthcare monitoring systems. The present work focuses on the fabrication procedures that can be used to obtain practically realizable fabric-based microfluidic channels (µFADs) utilizing patterning masks and wax, unlike conventional printing techniques. In this study, comparative analysis was used to differentiate channels obtained using different masking tools for channel patterning as well as different wax materials as hydrophobic barriers. Drawbacks of the conventional tape and candle wax technique were noted and a novel approach was used to create microfluidic channels through a facile and simple masking technique using PVC clear sheets as channel stencils and beeswax as the channel barriers. The resulting fabric based microfluidic channels with varying widths as well as complex microchannel, microwell, and micromixer designs were investigated and a minimum channel width resolution of 500 µm was successfully obtained over cotton based fabrics. Thereafter, the PVC clear sheet-beeswax based microwells were successfully tested to confine various organic and inorganic samples indicating vivid applicability of the technique. Finally, the microwells were used to make a simple and facile colorimetric assay for glucose detection and demonstrated effective detection of glucose levels from 10 mM to 50 mM with significant color variation using potassium iodide as the coloring agent. The above findings clearly suggest the potential of this alternative technique for making low-cost and practically realizable fabric based diagnostic devices (µFADs) in contrast to the other approaches that are currently in use.

RESUMEN

Photoacoustic tomographic imaging is a non-invasive medical diagnostic technology for visualizing biological tissue. However, the inverse problem and noise in photoacoustic signals often cause blurred images. Existing regularization methods struggle with staircasing artifacts and edge preservation. To overcome this, an objective function incorporating total generalized variation (TGV) is proposed. However, it failed with high-density Gaussian noise. To address this, an extended version called edge-guided second-order TGV (ESTGV) is introduced. For sparsification, wavelet transform and discrete cosine transform are introduced, while the fast-composite-splitting algorithm is employed for the inverse problem solution. Experimental validation demonstrates the potential of these approaches.

BACKGROUND: Gold nanorods, by virtue of surface plasmon resonance, convert incident light energy (NIR) into heat energy which induces hyperthermia. We designed unique, multifunctional, gold nanorod embedded block copolymer micelle loaded with GW627368X for targeted drug delivery and photothermal therapy. METHODS: Glutathione responsive diblock co-polymer was synthesized by RAFT process forming self-assembled micelle on gold nanorods prepared by seed mediated method and GW627368X was loaded on to the reduction responsive gold nanorod embedded micelle. Photothermal therapy was administered using cwNIR laser (808nm; 4W/cm2). Efficacy of nanoformulated GW627368X, photothermal therapy and combination of both were evaluated in vitro and in vivo. RESULTS: In response to photothermal treatment, cells undergo regulated, patterned cell death by necroptosis. Combining GW627368X with photothermal treatment using single nanoparticle enhanced therapeutic outcome. In addition, these nanoparticles are effective X-ray CT contrast agents, thus, can help in monitoring treatment. CONCLUSION: Reduction responsive nanorod embedded micelle containing folic acid and lipoic acid when treated on cervical cancer cells or tumour bearing mice, aggregate in and around cancer cells. Due to high glutathione concentration, micelles degrade releasing drug which binds surface receptors inducing apoptosis. When incident with 808nm cwNIR lasers, gold nanorods bring about photothermal effect leading to hyperthermic cell death by necroptosis. Combination of the two modalities enhances therapeutic efficacy by inducing both forms of cell death. GENERAL SIGNIFICANCE: Our proposed treatment strategy achieves photothermal therapy and targeted drug delivery simultaneously. It can prove useful in overcoming general toxicities associated with chemotherapeutics and intrinsic/acquired resistance to chemo and radiotherapy.

RESUMEN

Reconstruction of the absorption coefficient of tissue with good contrast is of key importance in functional diffuse optical imaging. A hybrid approach using model-based iterative image reconstruction and a genetic algorithm is proposed to enhance the contrast of the reconstructed image. The proposed method yields an observed contrast of 98.4%, mean square error of 0.638×10⁻³, and object centroid error of (0.001 to 0.22) mm. Experimental validation of the proposed method has also been provided with tissue-like phantoms which shows a significant improvement in image quality and thus establishes the potential of the method for functional diffuse optical tomography reconstruction with continuous wave setup. A case study of finger joint imaging is illustrated as well to show the prospect of the proposed method in clinical diagnosis. The method can also be applied to the concentration measurement of a region of interest in a turbid medium.

RESUMEN

Image quality and photon measurement with good SNR (signal to noise ratio) in continuous wave diffuse optical tomography depend on the source detector density and sensitivity of photo detector. For large volume objects, it is difficult to obtain detectable light intensity with good SNR over the whole boundary. As an alternative, instead of the full boundary, the measurements are taken over a semi circle as in reflection geometry and a partial reconstruction scheme for the same is proposed in this paper. The cross-sectional optical parameters are reconstructed for different half of the sample with modified boundary conditions and finally the average of all the reconstructions are considered as the final reconstructed image. Simulation and experimental results have been illustrated to validate the proposed method. The main advantage of this scheme is to improve signal to noise ratio which controls the quality of reconstruction in actual phantoms. The use of continuous wave measurement makes the system cost effective as well.

Direct noninvasive visualization of wound bed with depth information is important to understand the tissue repair. We correlate skin swept-source-optical coherence tomography (OCT) with histopathological and immunohistochemical evaluation on traumatic lower limb wounds under honey dressing to compare and assess the tissue repair features acquired noninvasively and invasively. Analysis of optical biopsy identifies an uppermost brighter band for stratum corneum with region specific thickness (p < 0.0001) and gray-level intensity (p < 0.0001) variation. Below the stratum corneum, variation in optical intensities is remarkable in different regions of the wound bed. Correlation between OCT and microscopic observations are explored especially in respect to progressive growth and maturation of the epithelial and subepithelial components. Characteristic transition of uniform hypolucid band in OCT image for depigmented zone to wavy highly lucid band in the pigmented zone could be directly correlated with the microscopic findings. The transformation of prematured epithelium of depigmented area, with low expression of E-cadherin, to matured epithelium with higher E-cadherin expression in pigmented zone, implicated plausible change in their optical properties as depicted in OCT. This correlated evaluation of multimodal images demonstrates applicability of swept-source-OCT in wound research and importance of integrated approach in validation of new technology.
