RESUMEN

The aim of this study is to evaluate the effect of chitosan or nanodiamond incorporation on the color stability and surface roughness of a bisacrylic resin subjected to artificial aging. Four bisacrylic resins were evaluated, namely, control, chitosan-modified material, nanodiamond-modified material, and chitosan-nanodiamond-modified material. Twenty-four specimens were prepared for each material. The surface roughness was determined using a profilometer with a cut-off of 0.25 mm. The baseline color was measured according to the CIE L*a*b* system using a reflectance spectrophotometer. After these tests, the specimens were individually immersed in cola soft drink, red wine, or distilled water (n = 8) for 28 days. After the aging, the surface roughness and final color were re-evaluated. The color stability was determined using the difference between the coordinates obtained before and after the aging process. The data on roughness and color change were evaluated using ANOVA and the Tukey test (α = 0.05). The results show that the incorporation of nanodiamonds and chitosan into a bisacrylic resin provided a better color stability to the materials (p = 0.007). The storage in red wine resulted in a higher variation in the surface roughness values, especially when only the nanodiamond was incorporated to the material (p < 0.05). The incorporation of both chitosan and nanodiamonds are promising in providing an improvement in the properties of the bisacrylic resin when they are simultaneously incorporated in the product.

OBJECTIVE: The purpose of the study was to design and evaluate novel functional chitosan hydrogels (chitosan-H-propolis, chitosan-H-propolis-nystatin and chitosan-H-nystatin) by using the chitosan-H polymer as "dual function restorative materials". MATERIALS AND METHODS: The nystatin/antioxidant carrier gel was prepared by dispersion of the corresponding component in glycerol and 3% acetic acid with 5% chitosan gelling agent was then added to the dispersion with continuous mixing. The natural bio-adhesive functionalized chitosan hydrogels were combined with built in drug delivery system and bio-actives such as propolis in order to increase the dentin bond strength capacity and maintain therapeutic properties of the alternative drug delivery system. The surface morphology, release behaviors (physiological pH and also in acidic conditions), stability of nystatin:antioxidant:chitosan and the effect of the hydrogels on the shear bond strength of dentin were also evaluated. STATISTICAL ANALYSIS USED: Non-parametric ANOVA test was used to asses significance of higher shear bond values than dentine treated or not treated with phosphoric acid. RESULTS: The release of both nystatin and propolis confer the added benefit of dual action of a functional therapeutic delivery when comparing the newly designed chitosan-based hydrogel restorative materials to commercially available nystatin alone. Neither the release of nystatin nor the antioxidant stability was affected by storage. Chitosan-H, chitosan-propolis, chitosan-nystatin and chitosan-nystatin-propolis treated dentine gives significantly (P < 0.05) higher shear bond values (P < 0.05) than dentine treated or not treated with phosphoric acid. CONCLUSION: The added benefits of their unique functionality involve increased dentin adhesive bond strengths (after 24 h and after 6 months) and positive influence on the nystatin release. Nystatin was a model therapeutic agent, evaluating the concept of using functional materials as carriers for pro-drugs as well as displaying a certain degree of defence mechanism for free radical damage of the novel functional drug delivery. Overall, there was an insignificant relapse in the shear bond strength after 6 months.

RESUMEN

A novel approach in target specific molecular prototype drug delivery system concerns the attempt to employ radical affording substances (RAS) or radical quenching substances (RQS) as prodrugs able to produce irreversible damage on the desired target and therefore to stimulate cellular apoptosis. However, radical species generated can react quickly within the chemical environment prior to reaching its proper site of action. In this short communication, we report our investigations towards developing two alternative novel, simple, flexible and effective drug delivery systems that provide optimal dosage of drugs precisely where and when needed and therefore achieve and sustain a complex delivery profile. We have demonstrated the application of two effective molecular prototype delivery systems able to harness free radical reactivity within the laboratory where biological processes can be studied and controlled, leading to the prevention of disease and the development of new treatments for disease states mediated by free radicals.
