RESUMEN

Tissue elasticity is universally recognized as a diagnostic and prognostic biomarker for prostate cancer. As the first diagnostic test, the digital rectal examination is used since malignancy changes the prostate morphology and affects its mechanical properties. Currently, this examination is performed manually by the physician, with an unsatisfactory positive predictive value of 42%. A more objective and spatially selective technique is expected to provide a better prediction degree and understanding of the disease. To this aim, here we propose a miniaturized probe, based on optical fiber sensor technology, for mechanical characterization of the prostate with sub-millimeter resolution. Specifically, the optical system incorporates a customized Fiber Bragg Grating, judiciously integrated in a metallic cannula and moved by a robotic arm. The probe enables the local measurement of the force upon tissue indentation with a resolution of 0.97 mN. The system has been developed in such a way to be potentially used directly in vivo. Measurements performed on phantom tissues mimicking different stages of the prostatic carcinoma demonstrated the capability of our device to distinguish healthy from diseased zones of the prostate. The study on phantoms has been complemented with preliminary ex vivo experiments on real organs obtained from radical surgeries. Our findings lay the foundation for the development of advanced optical probes that, when integrated inside biopsy needle, are able to perform in vivo direct mechanical measurements with high sensitivity and spatial resolution, opening to new scenarios for early diagnosis and enhanced diagnostic accuracy of prostate cancer.

Nowadays, epidural space identification is made by using subjective and manual techniques characterized by failure rates up to 7%. In this work, we propose a fiber optic sensor technology based needle guidance system, that is directly inspired by the most common technique currently used for epidurals; through real-time strain measurements, the fiber Bragg grating integrated inside the needle lumen is able to effectively perceive the typical force drop occurring when the needle enters the epidural space. An in vivo swine study demonstrates the validity of our approach, paving the way for the development of lab-in-a-needle systems.

In this work, we report on the first demonstration of Lab on Fiber (LOF) dosimeter for ionizing radiation monitoring at ultra-high doses. The new dosimeter consists in a metallo-dielectric resonator at sub-wavelength scale supporting localized surface plasmon resonances realized on the optical fiber (OF) tip. The resonating structure involves two gold gratings separated by a templated dielectric layer of poly(methyl methacrylate) (PMMA). Two LOF prototypes have been manufactured and exposed at the IRRAD Proton Facility at CERN in Geneva to 23 GeV protons for a total fluence of 0.67 × 1016 protons/cm2, corresponding to an absorbed dose of 1.8 MGy. Experimental data demonstrated the "radiation resistance" feature of the LOF devices and a clear dependence of the reflected spectrum versus the total dose, expressed by a cumulative blue-shift of ~1.4 nm of the resonance combined with a slight increase of 0.16 dBm in the reflected spectrum. The numerical analysis carried out to correlate the experimental results with the dimensional and physical properties of the resonator, expected to be tightly connected to the absorbed dose, suggests that the main phenomenon induced by exposure to proton beam and able to explain the measured spectral behavior is the reduction of the PMMA thickness, which is also consistent with past literature in the field. Preliminary results demonstrated the potentiality of the proposed platform as dosimeter at MGy dose levels for high energy physics experiments.

RESUMEN

Placing the needle inside the epidural space for locoregional anesthesia is a challenging procedure, which even today is left to the expertise of the operator. Recently, we have demonstrated that the use of optically sensorized needles significantly improves the effectiveness of this procedure. Here, we propose an optimized configuration, where the optical fiber strain sensor is directly integrated inside the epidural catheter. The new design allows the solving of the biocompatibility issues and increases the versatility of the former configuration. Through an in vivo study carried out on a porcine model, we confirm the reliability of our approach, which also opens the way to catheter monitoring during insertion inside biological spaces.

Integrating multi-responsive polymers such as microgels onto optical fiber tips, in a controlled fashion, enables unprecedented functionalities to Lab-on-fiber optrodes. The creation of a uniform microgel monolayer with a specific coverage factor is crucial for enhancing the probes responsivity to a pre-defined target parameter. Here we report a reliable fabrication strategy, based on the dip coating technique, for the controlled realization of microgel monolayer onto unconventional substrates, such as the optical fiber tip. The latter was previously covered by a plasmonic nanostructure to make it sensitive to superficial environment changes. Microgels have been prepared using specific Poly(N-isopropylacrylamide)-based monomers that enable bulky size changes in response to both temperature and pH variations. The formation of the microgel monolayer is efficiently controlled through the selection of suitable operating pH, temperature and concentration of particle dispersions used during the dipping procedure. The effect of each parameter has been evaluated, and the validity of our procedure is confirmed by means of both morphological and optical characterizations. We demonstrate that when the coverage factor exceeds 90%, the probe responsivity to microgels swelling/collapsing is significantly improved. Our study opens new paradigms for the development of engineered microgels assisted Lab-on-Fiber probes for biochemical applications.

The integration of microfluidics and photonic biosensors has allowed achievement of several laboratory functions in a single chip, leading to the development of photonic lab-on-a-chip technology. Although a lot of progress has been made to implement such sensors in small and easy-to-use systems, many applications such as point-of-care diagnostics and in vivo biosensing still require a sensor probe able to perform measurements at precise locations that are often hard to reach. The intrinsic property of optical fibers to conduct light to a remote location makes them an ideal platform to meet this demand. The motivation to combine the good performance of photonic biosensors on chips with the unique advantages of optical fibers has thus led to the development of the so-called lab-on-fiber technology. This emerging technology envisages the integration of functionalized materials on micro- and nano-scales (i.e. the labs) with optical fibers to realize miniaturized and advanced all-in-fiber probes, especially useful for (but not limited to) label-free chemical and biological applications. This review presents a broad overview of lab-on-fiber biosensors, with particular reference to lab-on-tip platforms, where the labs are integrated on the optical fiber facet. Light-matter interaction on the fiber tip is achieved through the integration of thin layers of nanoparticles or nanostructures supporting resonant modes, both plasmonic and photonic, highly sensitive to local modifications of the surrounding environment. According to the physical principle that is exploited, different configurations - such as localized plasmon resonance probes, surface enhanced Raman scattering probes and photonic probes - are classified, while various applications are presented in context throughout. For each device, the surface chemistry and the related functionalization protocols are reviewed. Moreover, the implementation strategies and fabrication processes, either based on bottom-up or top-down approaches, are discussed. In conclusion we highlight some of the further development opportunities, including lab-in-a-needle technology, which could have a direct and disruptive impact in localized cancer treatment applications.

Sensing schemes based on Rayleigh anomalies (RAs) in metal nanogratings exhibit an impressive bulk refractive-index sensitivity determined solely by the grating period. However, the surface sensitivity (which is a key figure of merit for label-free chemical and biological sensing) needs to be carefully investigated to assess the actual applicability of this technological platform. In this paper, we explore the sensitivity of RAs in metal nanogratings when local refractive-index changes are considered. Our studies reveal that the surface sensitivity deteriorates up to two orders of magnitude by comparison with the corresponding bulk value; interestingly, this residual sensitivity is not attributable to the wavelength shift of the RAs, which are completely insensitive to local refractive-index changes, but rather to a strictly connected plasmonic effect. Our analysis for increasing overlay thickness reveals an ultimate surface sensitivity that approaches the RA bulk value, which turns out to be the upper-limit of grating-assisted surface-plasmon-polariton sensitivities.

We demonstrate the operation of a flexible optical filter based on guided mode resonances that operates in the visible regime. The filter is fabricated on a free standing polymeric membrane of 1.3 µm thickness and we show how the geometrical design parameters of the filter determine its optical properties, and how various types of filter can be made with this scheme. To highlight the versatility and robustness of the approach, we mount a filter onto a collimated fibre output and demonstrate successful wavelength filtering.

En el presente trabajo se examinó la interacción de las amanitinas de Amanita phalloides (Basidiomycetes) con los venenos de las serpientes Bothrops neuwiedi diporus ("yarará pequeña"), B. alternatus ("yarará grande"), Crotalus durissus terrificus ("serpiente de cascabel") y de la abeja mielera Apis mellifera. Se aplicaron las técnicas de Ouchterlony, inmunotransferencia, electroforesis rocket y electroforesis en gel de poliacrilamida a los anti-venenos y anti-toxinas obtenidos por inmunización en caballos y/o en conejos. Los anti­sueros de serpientes y las amanitinas reaccionaron en forma cruzada, así como el veneno de abeja y las amanitinas. Cuando los venenos de Bothrops neuwiedii diporus y Crotalus durissus terrificus se preincubaron con las amanitinas y se analizaron por electroforesis en gel de poliacrilamida-dodecilsulfato de sodio (dodecylsulfate-polyacrylamide gel electrophoresis: SDS-PAGE), algunas bandas de proteínas desaparecieron y otras se redujeron notablemente. Estos resultados revelan por primera vez la interacción y la degradación de las proteínas de los venenos de serpientes por las amanitinas. Por otra parte, la modificación del tiempo de coagulación de la sangre humana, debida a los venenos, se corrigió con los ciclopéptidos de Amanita. Estos resultados también se informan por primera vez en este trabajo. La presencia de polipéptidos tóxicos en los venenos de serpientes y abejas, así como en A. phalloides y la reactividad cruzada demostradas en este trabajo, sugieren la existencia de epítopos comunes a todos ellos. Teniendo en cuenta estas reacciones, el uso de anti-venenos heterólogos parece ser de utilidad en el tratamiento del envenenamiento.


In the present work, the interaction of the amanitins of Amanita phalloides (Basidiomycetes) with the venoms of Bothrops neuwiedi diporus ("small yarará snake"), B. alternatus ("big yarará"), Crotalus durissus terrificus ("rattlesnake"), and honey bee Apis mellifera was examined. Ouchterlony, immunotransfer, rocket-electrophoresis, and polyacrylamide gel electrophoresis techniques were applied to anti-venoms and anti-toxins obtained by immunization in horses and/or in rabbits. Snake antisera and amanitins cross-reacted as well as bee venom and amanitins. When venoms of Bothrops neuwiedii diporus and Crotalus durissus terrificus were preincubated with amanitins and analysed by sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE), some protein bands disappeared and others were significantly reduced. These results reveal for the first time the interaction and degradation of proteins in snake venoms by amanitins. Moreover, the modification of the human blood clotting time due to snake venoms was corrected by the Amanita cyclopeptides. These results are also reported for the first time in this work. The occurrence of toxic polypeptides in the snake and bee venoms as well as in A. phalloides, and the cross-reactivity demostrated herein, suggest the occurrence of epitopes common to all of them. Taking into account these reactions,the use of heterologous anti-venoms seems to be of value in envenomation treatment.

Foram aplicadas as técnicas de Ouchterlony, imunotransferência, eletroforese rocket e eletroforese em gel de poliacrilamida aos anti-venenos e anti-toxinas obtidos por imunização em cavalos e/ou em coelhos. Os anti-soros de serpentes e as amanitinas reagiram em forma cruzada, bem como o veneno de abelha e as amanitinas. Quando os venenos de Bothrops neuwiedii diporus e Crotalus durissus terrificus foram incubados previamente com as amanitinas e foram analisados por eletroforese em gel de poliacrilamida-dodecilsulfato de sódio (dodecylsulfate-polyacrylamide gel electrophoresis: SDS-PAGE), algumas faixas de proteínas desapareceram e outras se reduziram notavelmente. Estes resultados revelam por primeira vez a interação e a degradação das proteínas dos venenos de serpentes pelas amanitinas. Por outra parte, a modificação do tempo de coagulação do sangue humano, devido aos venenos, se corrigiu com os ciclopeptídeos de Amanita. Estes resultados também se informam por primeira vez neste trabalho. A presença de polipeptídeos tóxicos nos venenos de serpentes e abelhas, bem como em A. phalloides e a reatividade cruzada demonstradas neste trabalho, sugerem a existência de epítopos comuns a todos eles. We propose a reliable fabrication process enabling the integration of dielectric and metallic nanostructures on the tip of optical fibers, thus representing a further step in the "lab-on-fiber" technology roadmap. The proposed fabrication procedure involves conventional deposition and nanopatterning techniques, typically used for planar devices, but here adapted to directly operate on optical fiber tip. Following this approach, we demonstrate a first technological platform based on the integration onto the optical fiber tip of two-dimensional hybrid metallo-dielectric nanostructures supporting localized surface plasmon resonances. By means of experimental measurements and full-wave numerical simulations, we characterize these resonant phenomena and investigate the underlying physics. We show that resonances can be easily tuned by acting on the physical and geometrical parameters of the structure. Moreover, with a view toward possible applications, we present some preliminary results demonstrating how the proposed device can work effectively as an optical probe for label-free chemical and biological sensing as well as a microphone for acoustic wave detection.

Leaves extracts and essential oil of Nectandra angustifolia were explored for the first time for neutralization of Bothrops neuwiedi diporus snake venom. The ethanol extract was the most active and inhibited both venom activities (hemolytic and coagulant), while the oil was only active on the coagulant activity. These observations confirmed that certain medicinal plants from Corrientes and Chaco Provinces possess significant snake venom neutralizing capacity and need further examination for their active constituents. Analysis by GC and GC-MS of the essential oil and the enantiomeric excess found for alpha-pinene, beta-pinene and limonene allowed a better characterization of this species.

Lemon verbena (Aloysia citriodora Palau) is indigenous to South America and was introduced into Europe. It is cultivated mainly due to the lemon-like aroma emitted from its leaves, which are utilized for the preparation of herbal tea reputed to have antispasmodic, antipyretic, sedative and digestive properties. In this work we introduce the enantiomeric distribution of sabinene and limonene by bidimensional gas chromatography (chiral GC-GC) as a genuine quantitative parameter in order to improve the knowledge so far available on A. citriodora oil. Multivariate analysis afforded information on the similarities and differences of wild and cultivated A. citriodora populations during different seasons in the same environmental conditions. The results indicated that it was possible to discard the environmental and seasonal effect on the chemical composition of A. citriodora for wild and cultivated materials belonging to the same genetic origin.

We report on the first experimental evidence of guided resonances (GRs) in photonic crystal slabs based on aperiodically ordered supercells. Using Ammann-Beenker (quasiperiodic, eightfold symmetric) tiling geometry, we present our study on the fabrication, experimental characterization, and full-wave numerical simulation of two representative structures (with different filling parameters) operating at near-IR wavelengths (1300-1600 nm). Our results show a fairly good agreement between measurements and numerical predictions and pave the way for the development of new strategies (based on, e.g., the lattice symmetry breaking) for GR engineering.

In this paper, we present a comparative study of the tuning efficiency and sensitivity of guided resonances (GRs) in photonic crystal (PC) holed slabs based on periodic and aperiodically-ordered unit cells, aimed at assessing the applicability of these important technology platforms to ultra-compact optical sensors and active devices. In particular, with specific reference to square-lattice periodic PCs and aperiodically-ordered Ammann-Beenker photonic quasi-crystals, we study the effects of the hole radius, slab thickness, and refractive index on the GR sensitivity and tunability with respect to variation in the hole refractive index. Finally, we carry out a theoretical and numerical analysis in order to correlate the GR shift with the field distribution of the unperturbed (air holes) structures. Our results indicate that the spatial arrangement of the holes may strongly influence the tuning and sensitivity efficiency, and may provide new degrees of freedom and tools for the design and optimization of novel photonic devices for both sensing and telecommunication applications.

In this paper, we study the excitation of guided resonances (GRs) in photonic-crystal slabs based on point-defected aperiodically-ordered supercells. With specific reference to perforated-slab structures and the Ammann-Beenker octagonal lattice geometry, we carry out full-wave numerical studies of the plane-wave responses and of the underlying modal structures, which illustrate the representative effects induced by the introduction of symmetry-preserving and symmetry-breaking defects. Our results demonstrate that breaking the supercell mirror symmetries via the judicious introduction of point-defects enables for the excitation of otherwise uncoupled GRs, with control on the symmetry properties of their field distributions, thereby constituting an attractive alternative to those GR-engineering approaches based on the asymmetrization of the hole shape. In this framework, aperiodically-ordered supercells seem to be inherently suited, in view of the variety of inequivalent defect sites that they can offer.

Lippia alba (Verbenaceae) is a shrub whose essential oil has important biological, pharmacological, and aromatizing properties. The species has a natural range from Central America to Argentina, being cultivated for its commercial value in Brazil, Argentina and Guatemala, and has been introduced into India and Spain. To reach the economic potential of the plant, the present study was aimed at evaluating L. alba for different chemotypes. The composition of the essential oil from two native populations of L. alba, collected from Argentina and two accessions from Costa Rica, were screened by GC and GC-MS. The results obtained led us to adopt the concept of a biodistribution map, as was proposed previously for the species, representing an approach to the natural biological distribution of the species in America based on the chemotypes described and their geographical distribution. Moreover, the biodiversity reported for the species (seven of eight chemotypes described for L. alba are present in Argentina) suggests the southern region of South America as the centre of distribution for L. alba.

In this paper, we report on the first evidence of guided resonances (GRs) in aperiodically-ordered photonic crystals, tied to the concept of "quasicrystals" in solid-state physics. Via a full-wave numerical study of the transmittance response and the modal structure of a photonic quasicrystal (PQC) slab based on a representative aperiodic geometry (Ammann-Beenker octagonal tiling), we demonstrate the possibility of exciting GR modes, and highlight similarities and differences with the periodic case. In particular, we show that, as for the periodic case, GRs arise from the coupling of the incident plane-wave with degenerate modes of the PQC slab that exhibit a matching symmetry in the spatial distribution, and can still be parameterized via a Fano-like model. Besides the phenomenological implications, our results may provide new degrees of freedom in the engineering of GRs, and pave the way for new developments and applications.

In this paper, slow and fast light in optical fiber through tilted fiber Bragg grating (TFBG) are reported. The experimental results show the capability of TFBGs to enable group velocity control of an optical pulse in optical fiber, due to the anomalous dispersion features induced by the coupling between the propagating core mode and each counter-propagating coupling cladding mode. In particular, superluminal propagation of a pulse train has been observed at optical communication wavelengths with time advancements in the picoseconds time scale in 1cm long TFBG and group velocity as large as about two times the speed of light in optical fiber (approximately 1.3 x c0). Very good agreement has been obtained comparing the measured group delay of the TFBG with the one retrieved from the amplitude response through Hilbert transform. Finally, tunable slow and fast light has also been reported, demonstrating the possibility to control the group velocity at single wavelength through both fluidic and thermal actuation.

En base a una encuesta nuestra anterior que abarcó a más de mil quinientas personas, respecto al empleo de plantas de la flora autóctona del chaco argentino en la medicina popular, hemos examinado la presencia de compuestos cianogénicos en las especies más utilizadas, con el objeto de validar su acción farmacológica o alertar sobre sus efectos tóxicos. Las especies analizadas son empleadas bajo diversas formas, tales como macerados, masticadas directamente, con el mate, como agua de beber, en infusiones o decocciones. En trabajos anteriores hemos informado sobre la presencia de saponinas y sapogeninas, antraquinonas y alcaloides en las mismas especies

RESUMEN

En anteriores trabajos hemos revelado la presencia de compuestos cianogenéticos y alcaloides en plantas autóctonas usadas en medicina popular en el Chaco argentino. En la presente comunicación exponemos sobre el reconocimiento de la presencia de saponinas o sapogeninas en los diversos órganos de diecisiete especies vernáculas con tradición de empleo en etnomedicina, cuidando la sanidad de las muestras, e identificando la procedencia de cada población vegetal, muestreo que se ha extendido durante tres años consecutivos y a períodos vegetativos diversos a efectos de diferenciar variaciones ecológicas, fenológicas o estacionales. Las especies han sido seleccionadas en base a la tradición indígena como también de acuerdo a una encuesta que ha abarcado a aproximadamente mil quinientos pobladores actuales
