BACKGROUND: The mountainous region of Kashmir is a biodiversity hotspot, with diverse local communities and a rich cultural history linked to nature. Mountain ecosystems are highly vulnerable to climate change. This study emphasises the need to record the indigenous ethnoecological knowledge of wild plants used for the treatment of various ailments at higher elevations in remote areas where globalisation poses a threat to this traditional knowledge. METHODS: The field survey was carried out in 2020-2022, to collect data on wild medicinal plants. Informants were selected randomly to collect indigenous medicinal knowledge using semi-structured interviews and group discussions. Various quantitative indices were employed to evaluate ethnomedicinal data. RESULTS: A total of 110 medicinal plants belonging to 49 families were recorded in the study area. These medicinal plants are extensively used by local communities for the treatment of 20 major disease categories. Asteraceae was the dominant family contributing (9.09%) to medicinal plants, followed by Polygonaceae (8.18%), Apiaceae (7.27%), Lamiaceae (5.45%), and Ranunculaceae (5.45%). We observed 166 remedies were used for the treatment of various diseases in humans, and 9 remedies were used for animals. The most frequently used medicinal remedy was tea or decoction (30.91%). Among the medicinal plants, herbs (85.5%) were most frequently used by the local populations of Kashmir, whereas leaves (10.26%) were used for the treatment of various ailments. Out of 110 species, 31 were endemic, 15 of which are endemic to the Kashmir region and 16 to the Western Himalaya. The highest RFC value was reported for Allium humile (0.77), the highest UV value for Fritillaria cirrhosa (1.33), and the highest ICF value for gastro-intestinal/digestive disorders (0.85). CONCLUSIONS: Local communities still rely on wild medicinal plants for primary healthcare. These communities retained valuable indigenous knowledge, which needs to be preserved for the conservation and sustainable utilisation of natural resources. Further field exploration is required to fully explore indigenous knowledge in the mountainous regions of Kashmir, and this knowledge has the potential to support the ongoing ecological transition.

Phaseolus vulgaris L. is the major pulse cultivated and culturally inculcated in the food habits of the locals in the Himalayan mountainous region of Azad Jammu and Kashmir (AJK), Pakistan. The current study was designed to investigate the role of P. vulgaris cultivation in providing livelihood support and to evaluate its production and consumption patterns correlated with the household variables in the state of AJK. The socio-economic data was collected from nine bean cultivated areas in six districts of AJK. The data was acquired by administrating a total of 522 detailed semi structured questionnaires from a diverse array of the respondents following the snowball technique focusing on yield, consumption, revenue generation and livelihood support provided by bean cultivation. The results revealed that common bean cultivation provided significant livelihood support to the local mountainous populations with an average annual income of 50.80 $/family. Subsequently, bean production contributed an average annual per capita income of 6.81 $ in the area, which was attributed to the large family size. Local populations showed an average bean production of 33.93 kg/family, whereas the average annual bean consumption was recorded as 31.99 kg/family in the region. Bean crops were recorded to have an average price of $1.49/kg, with significant variations in the study area correlated with local yield. A data analysis indicated a strong correlation in bean production and consumption patterns. Common bean farmers had a very small farm size, averaging 0.24 ha, where 100% of farmers cultivated common beans as an intercrop with Maize as the primary crop. A Pearson's test (p value < 0.05) revealed significant correlations between land holding and bean production as well as consumption, and bean production with annual per capita income. Small farm size, declining soil fertility, low bean pricing and the unavailability of market mechanisms were identified as the major challenges faced by the common bean farmers. It is recommended to employ an integrated bean farming approach to enhance the economic impact of common bean cultivation in the socioeconomic appraisal of the local populations.

Monitoring of heavy metals in agricultural soils and the crops grown in them is essential to design mitigation strategies to reduce toxic heavy metals in diet and food chains. We determined chromium (Cr), arsenic (As), cadmium (Cd), and lead (Pb) concentrations in the soil-plant system from agricultural fields of Siran Valley, Mansehra, Pakistan, to assess their potential health risk. Although the concentrations of the heavy metals in soils were within the permissible limits for agricultural soils, heavy metal concentrations in many of the vegetables exceeded the recommended safe values. Among the six leafy vegetables tested, all had greater than safe limits for As, four also for Cr and two also for Cd. As level was greater than safe limits in all five fruit and flower vegetables, two had Cr, and one had Pb also at unsafe levels. Among the five tuber, bulb, and root vegetables, As was higher than safe limits in all and Cd in one. The transfer factor in all three categories of vegetables followed the descending order Cd > As > Cr > Pb. Daily intake of metals were within limit set by USEPA for all heavy metals except As. The health risk indices for Cr, As, Cd, and Pb indicated that values greater than 1 for As suggest that the vegetables studied here pose a risk of chronic arsenic poisoning, but other heavy metals do not pose such a risk. Our study reinforces the need for mitigation strategies to reduce unsafe levels of heavy metals in vegetables.

Background: The upper belt of Azad Kashmir is a hilly, mountainous, and remote area where the indigenous communities mainly believe in traditional medicines for the treatment of different ailments. This study aimed to conserve scientifically and culturally important medicinal knowledge of Primula species in Azad Kashmir, Western Himalaya, Pakistan. The additional objective was to evaluate the antibacterial activity of these plants against pathogenic bacteria. Methods: The ethnomedicinal data of Primula species was explored by conducting structured interviews with 40 informants of the study area, especially asking about the medicinal uses of Primula species. The indigenously used Primula species were further analyzed for their antibacterial activity against both gram-positive and gram-negative bacteria by using disc diffusion assay supplemented with a more robust minimum inhibitory concentration assay. Results: Ethnomedicinal data revealed that indigenous communities living in upper regions of Azad Kashmir use 5 Primula species for the treatment of various disorders. The highly cited disease category was ophthalmic disorders. P. denticulata and P. macrophylla were the most cited plant species with higher use reports such as 104 and 93, respectively. One or more extracts of different parts of Primula species showed a noteworthy antibacterial activity against one or more tested bacteria. Conclusion: This study provides novel information regarding several categories of traditional uses and antibacterial activity of Primula species in Azad Kashmir, Western Himalaya. The need for novel and more effective drugs derived from natural products is more important than ever, making future studies on herbal remedies both justified and urgently required.

Background: Different parts of Taraxacum officinale (L.) were used in traditional medicine in various parts of the world for the treatment of health problems, and they possess significant biological activities. The present study aimed to estimate phytochemical and biological activities of T. officinale using different extraction solvents. Methods: Methanolic, acetone, and n-hexane extracts of selected species were prepared, and ten secondary metabolites were examined using standard protocols. The antioxidant activity was performed using three in vitro methods, namely, DPPH assay, total reducing power (TRP) assay, and total antioxidant capacity (TAC). Toxicological analysis was done using the brine shrimp cytotoxic assay and radish seed phytotoxic assay. Results: The T. officinale methanolic extract showed the highest phenolic (178.27 ± 17.17 mg/GAE/g) and flavonoid (18.50 ± 1.64 mg QE/g) contents. Similarly, the methanolic extract also revealed the highest DPPH activity (32.80 ± 9.66 IC50), reducing potential (0.53 ± 0.02 mg/g), and TAC (19.42 ± 0.97 mg/g) as compared to the acetone and n-hexane extracts. The Pearson correlation analysis confirmed a strong positive correlation (r > 0.9) between total phenolic content (TPC), total flavonoid content (TFC), and all antioxidant assays. Furthermore, a heat map displayed the methanolic extract (red color) as a valuable source of phytochemicals and antioxidant agents. Moreover, the T. officinale methanolic extract also showed the highest (7.12 ppm) cytotoxic potential whereas both methanolic and acetone extracts were revealed as moderate phytotoxic agents when compared with the standard. Conclusion: The T. officinale methanolic extract exhibited comparatively notable phytochemicals that are actively involved in antioxidant activities and possess toxicological properties. This upholds the folkloric use of T. officinale as a possible source to develop natural plant-based drugs. Further investigations to isolate bioactive compounds and elements and on their safety need to be conducted.

BACKGROUND: Wild edible mushrooms (WEM) are economically significant and used in traditional medicines worldwide. The region of Jammu and Kashmir (Western Himalayas) is enriched with the diversity of edible mushrooms, collected by the rural people for food and income generation. This is the first detailed study on diversity and ethno-medicinal uses of mushrooms from the State of Jammu and Kashmir. METHODS: Consecutive surveys were conducted to record ethnomycological diversity and socio-economic importance of wild edible mushrooms value chain in rural areas of Azad Jammu and Kashmir during 2015-2019. Ethnomycological data were collected with a semi-structured questionnaire having a set of questions on indigenous mycological knowledge and collection and retailing of wild edible mushrooms. A total of 923 informants from the study area provided the results identifying the gender, type of mushroom species, medicinal uses, and marketing of mushrooms. Diversity of mushrooms was studied by using quadrat and transect methods. Principal component analysis (PCA) and detrended correspondence analysis (DCA) were also applied to the dataset to analyse the relationship between species distribution, the underlying environmental factors, and habitat types. PCA identified the major species-specific to the sites and put them close to the sites of distribution. RESULTS: A total of 131 mushroom species were collected and identified during 2015-2019 from the study area. Ninety-seven species of mushrooms were reported new to the State of Azad Jammu and Kashmir. The dominant mushroom family was Russulaceae with 23 species followed by Agaricaceae, 16 species. Major mushroom species identified and grouped by the PCA were Coprinus comatus, Lactarius sanguifluus, Amanita fulva, Armillaria gallica, Lycoperdon perlatum, Lycoperdon pyriforme, and Russula creminicolor. Sparassis crispa, Pleurotus sp, and Laetiporus sulphureus were recorded most edible and medicinally significant fungi. Morels were also expensive and medicinally important among all harvested macro-fungal species. These were reported to use against common ailments and various health problems. CONCLUSIONS: Collection and retailing of WEM contribute to improving the socio-economic status, providing alternative employment and food security to rural people of the area. These mushrooms are used as a source of food and traditional medicines among the rural informants and could be used as a potential source of antibacterial and anticancer drugs in the future.

Autophagy is an intracellular process in all eukaryotes which is responsible for the degradation of cytoplasmic constituents, recycling of organelles, and recycling of proteins. It is an important cellular process responsible for the effective virulence of several pathogenic plant fungal strains, having critical impacts on important crop plants including potatoes. However, the detailed physiological mechanisms of autophagy involved in the infection biology of soil-borne pathogens in the potato crop needs to be investigated further. In this study, the autophagy-related gene, FoATG12, in potato dry rot fungus Fusarium oxysporum was investigated by means of target gene replacement and overexpression. The deletion mutant ∆FoATG12 showed reduction in conidial formation and exhibited impaired aerial hyphae. The FoATG12 affected the expression of genes involved in pathogenicity and vegetative growth, as well as on morphology features of the colony under stressors. It was found that the disease symptoms were delayed upon being inoculated by the deletion mutant of FoATG12 compared to the wild-type (WT) and overexpression (OE), while the deletion mutant showed the disease symptoms on tomato plants. The results confirmed the significant role of the autophagy-related ATG12 gene in the production of aerial hyphae and the effective virulence of F. oxysporum in the potato crop. The current findings provid an enhanced gene-level understanding of the autophagy-related virulence of F. oxysporum, which could be helpful in pathogen control research and could have vital impacts on the potato crop.

Fusarium oxysporum is the most important pathogen of potatoes which causes post-harvest destructive losses and deteriorates the market value of potato tubers worldwide. Here, F. oxysporum was used as a host pathogen model system and it was revealed that autophagy plays a vital role as a regulator in the morphology, cellular growth, development, as well as the pathogenicity of F. oxysporum. Previous studies based upon identification of the gene responsible for encoding the autophagy pathway components from F. oxysporum have shown putative orthologs of 16 core autophagy related-ATG genes of yeast in the genome database which were autophagy-related and comprised of ubiquitin-like protein atg3. This study elucidates the molecular mechanism of the autophagy-related gene Foatg3 in F. oxysporum. A deletion (∆) mutants of F. oxysporum (Foatg3∆) was generated to evaluate nuclear dynamics. As compared to wild type and Foatg3 overexpression (OE) strains, Foatg3∆ strains failed to show positive MDC (monodansylcadaverine) staining which revealed that Foatg3 is compulsory for autophagy in F. oxysporum. A significant reduction in conidiation and hyphal growth was shown by the Foatg3∆ strains resulting in loss of virulence on potato tubers. The hyphae of Foatg3∆ mutants contained two or more nuclei within one hyphal compartment while wild type hyphae were composed of uninucleate hyphal compartments. Our findings reveal that the vital significance of Foatg3 as a key target in controlling the dry rot disease in root crops and potato tubers at the postharvest stage has immense potential of disease control and yield enhancement.

Autophagy is a universal catabolic process preserved in eukaryotes from yeast to plants and mammals. The main purpose of autophagy is to degrade cytoplasmic materials within the lysosome/vacuole lumen and generate an internal nutrient pool that is recycled back to the cytosol during nutrient stress. Here, Fusarium oxysporum was utilized as a model organism, and we found that autophagy assumes an imperative job in affecting the morphology, development, improvement and pathogenicity of F. oxysporum. The search of autophagy pathway components from the F. oxysporum genome database recognized putative orthologs of 16 core autophagy-related (ATG) genes of yeast, which additionally incorporate the ubiquitin-like protein atg22. Present study elucidates the unreported role of Foatg22 in formation of autophagosomes. The deletion mutant of Foatg22 did not demonstrate positive monodansylcadaverine (MDC) staining, which exposed that Foatg22 is required for autophagy in F. oxysporum. Moreover, the ∆Foatg22 strains exhibited a decrease in hyphal development and conidiation, and reduction in pathogenicity on potato tubers and leaves of potato plant. The hyphae of ∆Foatg22 mutants were less dense when contrasted with wild-type (WT) and overexpression (OE) mutants. Our perceptions demonstrated that Foatg22 might be a key regulator for the control of dry rot disease in tuber and root crops during postharvest stage.

Mushrooms have had a long association with humankind and have great biological and economic effects. This study aimed to record ethnomycological and nutritional analysis of some edible mushrooms from the Western Himalayas (Neelum Valley), Azad Jammu and Kashmir, Pakistan. Mushrooms from different taxonomical and ecological groups were collected and properly identified with the help of the available literature: Morchella esculenta, M. crassipes, M. elata, M. conica, Pleurotus ostreatus, Lycoperdon gemmatum, Helvella crispa, Tricholoma megnivelare, Gyrometra esculenta, Agaricus campestris, Hydnum imbricatum, and Sparassis crispa. The local communities are familiar with the morphological features, habitats, and qualities of these mushrooms. Ethnomycological data were collected through the use of a questionnaire; we found that these species have great medicinal value against different ailments. Four species (A. campestris, H. imbricatum, P. ostreatus, and S. crispa) were selected for nutritional analysis (proteins, fats, fiber, and moisture) on the basis of their frequent use in the study area. The largest amounts of protein (1.627 and 1.493 mg/mL) were found in H. imbricatum and S. crispa, respectively. The largest amount of fiber was found in P. ostreatus (14%); S. crispa contains the largest amount of fat (12.7%).

OBJECTIVE: Himalayan mountain populations have been dependent upon indigenous plant resources for their health care for many years. Tribal women are interested in use of local herbs for cosmetic purposes. The present work is based on the results of research conducted on cosmetic uses of some important plants by the tribal women in District Poonch, Azad Kashmir Pakistan. MATERIALS AND METHODS: An ethno botanical survey was carried out during summer 2012. The data were collected from 310 female informants from 16 villages using questionnaire method and semi structured interviews. RESULTS: A total of 39 plants species belonging to 20 families, being used for various cosmetic purposes were recorded. Indigenous species are traditionally used by the locals for problems including acne (16%), hair growth (11%), bad breath (12%), facial spots (9%), allergy, (9%), fairness (8%), wrinkles (8%), eye and lip care (9%). Seventy different recipes were recorded to be practiced by locals using herbal parts. The major plant parts utilized in herbal recipes included fruit (32.8%), Leaves (25.2%), seeds (13.4%) and roots (8.9%). Women of older (>30 years) age group showed greater (67%) response regarding knowledge and practice of cosmetic herbs. CONCLUSION: This study was the 1(st) ever project focusing on cosmetic perspectives of ethno-botany in the area. Our study contributes to an improved understanding of ignored aspect of cosmetic ethnobotany among the local women. Further detailed investigations are recommended to record and preserve precious ethno-botanical knowledge of the area.

BACKGROUND: Mountain ecosystems all over the world support a high biological diversity and provide home and services to some 12% of the global human population, who use their traditional ecological knowledge to utilise local natural resources. The Himalayas are the world's youngest, highest and largest mountain range and support a high plant biodiversity. In this remote mountainous region of the Himalaya, people depend upon local plant resources to supply a range of goods and services, including grazing for livestock and medicinal supplies for themselves. Due to their remote location, harsh climate, rough terrain and topography, many areas within this region still remain poorly known for its floristic diversity, plant species distribution and vegetation ecosystem service. METHODS: The Naran valley in the north-western Pakistan is among such valleys and occupies a distinctive geographical location on the edge of the Western Himalaya range, close to the Hindu Kush range to the west and the Karakorum Mountains to the north. It is also located on climatic and geological divides, which further add to its botanical interest. In the present project 120 informants were interviewed at 12 main localities along the 60 km long valley. This paper focuses on assessment of medicinal plant species valued by local communities using their traditional knowledge. RESULTS: Results revealed that 101 species belonging to 52 families (51.5% of the total plants) were used for 97 prominent therapeutic purposes. The largest number of ailments cured with medicinal plants were associated with the digestive system (32.76% responses) followed by those associated with the respiratory and urinary systems (13.72% and 9.13% respectively). The ailments associated with the blood circulatory and reproductive systems and the skin were 7.37%, 7.04% and 7.03%, respectively. The results also indicate that whole plants were used in 54% of recipes followed by rhizomes (21%), fruits (9.5%) and roots (5.5%). CONCLUSION: Our findings demonstrate the range of ecosystem services that are provided by the vegetation and assess how utilisation of plants will impact on future resource sustainability. The study not only contributes to an improved understanding of traditional ethno-ecological knowledge amongst the peoples of the Western Himalaya but also identifies priorities at species and habitat level for local and regional plant conservation strategies.
