RESUMEN

An Escherichia coli strain producing transposase of a repeated sequence of Bordetella pertussis chromosome (RSBP) was constructed. A defective MGE-helper plasmid method, which allowed the determination of transposase functional activity was developed. It was shown that transposase synthesized in E. coli cells ensures transposition of "defective" RSBP into the host chromosome. Overexpression of transposase was shown to markedly decrease the vital activity of E. coli cells under selective cultivation conditions. Reasons for a decrease in viability transposase-producing cells are discussed. Results showing the impact of transposase on replication of recombinant plasmids and E. coli cell division were obtained.

A method of monitoring the sequential events of IS481 transposition into the ctag site of bvg operon of Bordetella pertussis has been developed. Reproduction of virulent B. pertussis cells in vitro is accompanied by intrachromosomal site-specific IS481 transposition, which, in turn, results in inactivation of bvg operon of the causative agent and cell avirulent state. Avirulent bvg mutants of B. pertussis are incapable of intramolecular IS481 transposition. The frequency of the transposition increases when MgSO4 and nicotinic acid are present the culture medium. In the absence of these modulating factors. IS481 transposition along B. pertussis chromosome is inhibited but not arrested completely. Negative regulation of the bvg-repressed genes of B. pertussis seems to be a mechanism that controls bvg-dependent IS481 transposition.

A computer-aided analysis of the repeating sequence of Bordetella pertussis chromosome (RSBP3) revealed 3 open reading frames, one of whose (ORF1) can code a protein whose structure and properties are similar to those of transposasas, i.e. enzymes in charges for the traveling of migrating genetic elements of pro- and eukaryote. Mutants of the RSBP3 insertion sequence with the affected and unaffected ORF1 sequence were constructed in order to substantiate the above assumption. Two independent experimental models (formation of inter-plasmid co-integrates and of co-integrates between plasmid and E. coli chromosome) were used to show that the RSBP3-stimulated formation of co-integrates is only true for plasmids containing RSBP3 with the unaffected ORF1 sequence. An activity of the Hpr protein (a component of the phosphoenolpyruvate-dependent phosphotransferase) was proven to influence the formation process of inter-plasmid co-integrates.

Long-standing results were studied in 587 patients who sustained reconstructive vascular surgeries when aged under 16 years (the average age was 6 years and 4 months). Out of them 141 patients faced the necessity of repeated surgeries: in 115 (81.6%) patients the extremity function was preserved, in 19 (13.5%) a repeated amputation was performed, 7 patients (4.9%) expired. All those operated on had been actively followed up with the use of periodical instrumental investigation which included dopplerography and ultrasonic angiography. High significance of ultrasonic technique was proved as the use of it permitted one to detect late complications on the early stages of their development and perform surgical corrections before the development of life-threatening complications. Therefore repeated preventive surgeries were performed in 53 patients, 52 of them were a success. Repeated amputations were performed in 1 patient. All those operated on survived.

RESUMEN

During in vitro and in situ experiments ethacrynic acid was shown to reduce the gastric secretion in rats, motility of the small intestine in rats and mice, contractions of the ileum of guinea pigs in response to histamine and acetylcholine. The intestinal water and electrolyte absorption in rats decreased too. All these effects of ethacrynic acid are extrarenal.

In examination of 50 patients with impotence, pathological venous drainage (PVD) was revealed in 11 of them. Perfusion graded cavernosometry/cavernosography is the only method by which PVD can be detected and studied thoroughly. Analysis of the cavernosograms revealed a sphincter of the deep dorsal vein of the penis, the dysfunction of which in 10 of the 11 patients was the cause of PVD; in 5 of these patients the dysfunction was combined with excessive development of the subcutaneous veins of the penis. In one patient PVD was caused by a large venous trunk which emptied into the ostium of the left major subcutaneous vein. The 11 patients underwent operation after which they returned to a normal sex life.

Most complete information on vascular components of erection can be obtained by using three methods: a pharmacological test for creation of artificial passive erection, roentgen-contrast arterio-cavernography on both sides, perfusion graded cavernometry and cavernography. Of 50 patients examined, 5 had no disorders of vascular components of erection. In 34 patients pathology of the arterial duct was revealed, 11 patients had a pathological venous drain. 46 operations were performed on the vessels. Satisfactory and good results were observed in 31 patients (68.8%) within 3 years after operation.

Examination of 50 patients suffering from impotence of vascular genesis included selective angiography of the penis conducted against the background of artificially induced erection, Doppler sonography of the penis and angiography and measurement of the corpora cavernosa. The following causes of impairment of erection were revealed: abnormal venous drainage of corpora cavernosa, derangement of arterial inflow due to occlusion of the aorta, iliac arteries, internal iliac artery, internal pudendal and penile arteries. Surgical correction was aimed at elimination of abnormal venous drainage, inclusion in the major blood flow of the internal iliac arteries and shunting of the penile arteries. Direct favourable results were obtained in 37 (74 per cent) of the patients operated on.

RESUMEN

Chemodectoma of the neck is one of the rare tumors occurring in man. The paper presents an observation of cervical chemodectoma found in a patient aged 31 years. For preoperative differential diagnosis, carotid angiography, ultrasound and radioisotopic examinations were used, which allowed the tumor located at the bifurcation of the left carotid artery to be diagnosed. The material taken during the surgery was histologically and electron microscopically examined to verify benign chemodectoma (an alveolar variant).

The protective effect of triamterene was demonstrated on aconitine (rats) and strophanthin (cats) models of arrhythmias. Furosemide potentiates the antiarrhythmic effects of calcium chloride (rats) and strophanthin, ethacrynic acid potentiates the arrhythmogenic action of all agents used.

RESUMEN

Diuretic and natriuretic action of chloracizine in rats was not associated with its direct influence on the kidney and was accompanied by increase in the plasma volume, by a fall of the hematocrite index and of the total plasma proteins. A study of endogenous creatinine and of osmotic-free water clearance showed the diuretic action of chloracizine to be mainly due to the increase of glomerular filtration and to reduction of the relative proximal reabsorption.

RESUMEN

Chlorathizin and Chlormorphathizin are shown to exercise in 24-hours long tests a substantial diuretic and saluretic action on rats and dogs. Fluorathizin, as well as imizine inhibited the urinary flow. Diprazine augmented the urinary output in dogs, but depressed it in rats. The new antidepressive drug azaphen did not have any marked effect on the urinary flow.
