Objectives: This study explores the significant impact of expressive ties (EI) between DanceSport couples on their competitive performance (CP). Utilizing a dyadic approach, we examined the performance achievement processes of DanceSport couples in relation to their EI. Methods: Participants comprised 67 dyads of Chinese elite dancers aged between 16 and 30 years. The dyadic analysis was carried out using a structural equation model based on the actor-partner interdependence mediation model. Results: With regard to actor effects, both male (ß = 0.292, p = 0.012) and female (ß = 0.443, p < 0.001) dancers' perceived quality of EI had a positive correlation with CP. The males' athlete engagement (AE) partially mediated the impact of EI on CP [indirect effect = 0.144, SE = 0.072, 95% confidence intervals (CI) = 0.020, 0.283]. Regarding partner effects, females' perceived EI quality positively influenced the male's CP (ß = 0.26, p = 0.023) and mediated this association through the male's AE [indirect effect = 0.086, SE = 0.041, 95% confidence intervals (CI) = 0.003, 0.149]. Similarly, the females' AE mediated the effect of males' perceived EI quality on the females' CP [indirect effect = 0.152, SE = 0.074, 95% confidence intervals (CI) =0.002, 0.256]. Conclusion: We not only validated the propositions of the self-determination theory but also provided valuable insights to further enrich it. Our findings underscore that self-determination theory must account for individual gender characteristics.

Objectives: Although the positive association of partnership between DanceSport couples with competitive performance (CP) is documented, less is known about the mediating factors of this relationship. According to the related literature and self-determination theory (SDT), the present study finds and verifies that athlete engagement (AE) mediates the association between partnership and competitive performance. Methods: A total of 242 Chinese sports dancers were recruited using the purposive sampling method. The Partnership Scale-DanceSport Couples (PS-DSC), the Athlete Engagement Questionnaire (AEQ), and the Competitive Performance Questionnaire (CPQ) were adopted to collect data. Results: The obligatory instrumental ties, expressive ties, and interpersonal perception scores are all positively correlated with both athlete engagement and competitive performance, and athlete engagement scores are positively correlated with competitive performance. Athlete engagement completely mediates the association between obligatory instrumental ties and competitive performance, and it partially mediates the association between expressive ties, interpersonal perception, and competitive performance, with the mediating effect accounting for 25.29 and 24.40% of the total effect, respectively. Conclusion: Athlete engagement mediates the association between DanceSport couples' partnership and competitive performance. High levels of athlete engagement are needed to improve the chance of promoting obligatory instrumental ties, expressive ties, and interpersonal perception between DanceSport couples toward excellent competitive performance. Overall, the results represent an attempt to extend our understanding of the mechanisms by which the three partnership stereotype factors individually influence dancers' cognitive and psychological states.

We present electrical, physiological and molecular evidence for substantial electrical coupling of epithelial cells in Malpighian tubules via gap junctions. Current was injected into one principal cell of the isolated Malpighian tubule and membrane voltage deflections were measured in that cell and in two neighboring principal cells. By short-circuiting the transepithelial voltage with the diuretic peptide leucokinin-VIII we largely eliminated electrical coupling of principal cells through the tubule lumen, thereby allowing coupling through gap junctions to be analyzed. The analysis of an equivalent electrical circuit of the tubule yielded an average gap-junction resistance (R(gj)) of 431 kOmega between two cells. This resistance would stem from 6190 open gap-junctional channels, assuming the high single gap-junction conductance of 375 pS found in vertebrate tissues. The addition of the calcium ionophore A23187 (2 micromol l(-1)) to the peritubular Ringer bath containing 1.7 mmol l(-1) Ca(2+) did not affect the gap-junction resistance, but metabolic inhibition of the tubule with dinitrophenol (0.5 mmol l(-1)) increased the gap-junction resistance 66-fold, suggesting the regulation of gap junctions by ATP. Lucifer Yellow injected into a principal cell did not appear in neighboring principal cells. Thus, gap junctions allow the passage of current but not Lucifer Yellow. Using RT-PCR we found evidence for the expression of innexins 1, 2, 3 and 7 (named after their homologues in Drosophila) in Malpighian tubules. The physiological demonstration of gap junctions and the molecular evidence for innexin in Malpighian tubules of Aedes aegypti call for the double cable model of the tubule, which will improve the measurement and the interpretation of electrophysiological data collected from Malpighian tubules.

The development of a culture of the normal mammalian jejunum motivated this work. Isolated crypt cells of the dog jejunum were induced to form primary cultures on Snapwell filters. Up to seven subcultures were studied under the electron microscope and in Ussing chambers. Epithelial markers were identified by RT-PCR, Western blot, and immunofluorescent staining. Confluent monolayers exhibit a dense apical brush border, basolateral membrane infoldings, desmosomes, and tight junctions expressing zonula occludens-1, occludin-1, and claudin-3 and -4. In OptiMEM medium fortified with epidermal growth factor, hydrocortisone, and insulin, monolayer transepithelial voltage was -6.8 mV (apical side), transepithelial resistance was 1,050 Omega.cm(2), and short-circuit current (I(sc)) was 8.1 microA/cm(2). Transcellular and paracellular resistances were estimated as 14.8 and 1.1 kOmega.cm(2), respectively. Serosal ouabain reduced voltage and current toward zero, as did apical amiloride. The presence of mRNA of alpha-epithelial Na(+) channel (ENaC) was confirmed. Na-d-glucose cotransport was identified with an antibody to Na(+)-glucose cotransporter (SGLT) 1. The unidirectional mucosa-to-serosa Na(+) flux (19 nmol.min(-1).cm(-2)) was two times as large as the reverse flux, and net transepithelial Na(+) flux was nearly double the amiloride-sensitive I(sc). In plain Ringer solution, the amiloride-sensitive I(sc) went toward zero. Under these conditions plus mucosal amiloride, serosal dibutyryl-cAMP elicited a Cl(-)-dependent I(sc) consistent with the stimulation of transepithelial Cl(-) secretion. In conclusion, primary cultures and subcultures of the normal mammalian jejunum form polarized epithelial monolayers with 1) the properties of a leaky epithelium, 2) claudins specific to the jejunal tight junction, 3) transepithelial Na(+) absorption mediated in part by SGLT1 and ENaC, and 4) electrogenic Cl(-) secretion activated by cAMP.

The present study investigated the role of calcitonin gene-related peptide (CGRP) in the modulation of nociception in periaqueductal grey (PAG) of rats. Hindpaw withdrawal latencies (HWLs) were tested by hot-plate and Randall Selitto tests. The HWLs to thermal and mechanical stimulation increased significantly after intra-PAG administration of 0.26 or 0.13 nmol of CGRP, but not 0.026 nmol of CGRP. The anti-nociceptive effects induced by CGRP were significantly blocked by intra-PAG administration of 0.026 or 0.26 nmol of the CGRP1 receptor antagonist CGRP8-37. Furthermore, administration of CGRP into the decussation of superior cerebellar peduncle, out of PAG, did not elicit anti-nociceptive effects during 60 min after the injection. The results demonstrated that CGRP plays an important role in anti-nociception in PAG of rats, and CGRP1 receptor is involved in the CGRP-induced anti-nociception.

The V-type H(+)-ATPase is thought to provide the driving force for transepithelial electrolyte and fluid secretion in Malpighian tubules. To confirm the presence of this proton pump in Malpighian tubules of the yellow fever mosquito Aedes aegypti, we used several antibodies raised against the V-type H(+)-ATPase of Manduca sexta. Western blot analysis confirmed the presence of the V-type H(+)-ATPase in Malpighian tubules of Aedes aegypti. In situ immunostaining identified the V-type H(+)-ATPase at the apical membrane of the mitochondrion-rich brush border of principal cells. The V-type H(+)-ATPase was not found in stellate cells. Measurements of ATPase activity revealed that bafilomycin-sensitive and NO(3)(-)-sensitive ATPase activity accounted for 50-60% of total ATPase activity in crude extracts of Malpighian tubules. No significant ouabain- or vanadate-sensitive Na(+)/K(+)-ATPase activity was detected. These results support the conclusion reached previously in electrophysiological studies that the mechanisms for transepithelial electrolyte secretion in the Aedes Malpighian tubules rely on the V-type H(+)-ATPase as the principal energizer of epithelial transport. Measures of transepithelial Na(+) and K(+) secretion and estimates of the H(+) flux mediated by the V-type H(+)-ATPase suggest a 1:1 stoichiometry for Na(+)/H(+) and K(+)/H(+) exchange transport across the apical membrane.
