RESUMEN

To investigate the function and possible mechanism of miR-92a in malignant behaviors such as paclitaxel resistance in ovarian cancer (OC) cells. The miR-92a and PTEN expression were detected by real-time PCR (RT-PCR). The cell viability and apoptosis were detected by MTT, colony formation and flow cytometry assay, respectively. Dual-luciferase reporter assay was adopted to verify the targeting relationship between miR-92a and PTEN. Besides, we measured the relative protein levels of PTEN and p-AKT/AKT by Western blot. MiR-92a was significantly highly expressed in OC cells, and its high expression could notably enhance paclitaxel resistance, cell proliferation and colony formation, as well as inhibit apoptosis in SKOV3-Tax cells. Further luciferase reporter assay and expression detection showed that miR-92a could target and regulate PTEN and that there was a targeted relationship between them. In addition, further exploration of the mechanism revealed that miR-92a regulated PTEN/Akt signaling pathway. MiR-92a not only promotes the proliferation, colony formation and paclitaxel resistance of SKOV3-Tax cells in OC, but also inhibits apoptosis, and it may be related to the regulation of the PTEN/Akt signaling pathway. MiR-92a serves as a potential biomarker for the malignant biological behavior of OC cells.

A highly sensitive silica-alumina (Si-Al)-modified capacitive non-Faradaic glucose biosensor was introduced to monitor gestational diabetes. Glucose oxidase (GOx) was attached to the Si-Al electrode surface as the probe through amine-modification followed by glutaraldehyde premixed GOx as aldehyde-amine chemistry. This Si-Al (∼50 nm) modified electrode surface has increased the current flow upon binding of GOx with glucose. Capacitance values were increased by increasing the glucose concentrations. A mean capacitance value was plotted and the detection limit was found as 0.03 mg/mL with the regression coefficient value, R² = 0.9782 [y = 0.8391x + 1.338] on the linear range between 0.03 and 1 mg/mL. Further, a biofouling experiment with fructose and galactose did not increase the capacitance, indicating the specific glucose detection. This Si-Al-modified capacitance sensor detects a lower level of glucose presence and helps in monitoring gestational diabetes.

OBJECTIVE: The purpose of this study was to investigate the role of cell morphometric factor (FC) and the average density of light in squamous cell carcinomas at the floor of mouth by immunohistochemical method (SABC). METHODS: The slices made from the paraffin specimens of 28 patients with squamous cell carcinomas at the floor of mouth were stained by the immunohistochemical technique with monoclonal antibody CK10 and CK14. In these stained slices the FC parameters and the average density of light were measured by using an image analysis system. RESULTS: In primitive tumors, the FC parameters and the average density of light were significantly different between positive and negative lymph nodes metastasis or between different pathological grades (P < 0.05 or < 0.001). The significance was not existed among clinical stages (P > 0.05). The difference of FC expressed in positive and negative lymph nodes metastasis were significant (P < 0.05 or < 0.001). CONCLUSION: The cell morphometric factor (FC) parameters and the average density of light of CK10 and CK14 could be used as a biological guideline to evaluate the clinically infiltrative and metastasic potential of squamous cell carcinoma at the floor of mouth.
