RESUMEN

Chinese yew, Taxuschinensisvar.mairei is an endangered shrub native to south-eastern China and is widely known for its medicinal value. The increased cultivation of Chinese yew has increased the incidence of various fungal diseases. In this study, Pestalotioid fungi associated with needle spot of Chinese yew were isolated from Guangxi Province. Based on morphological examinations and multi-locus (ITS, tub2, tef-1α) phylogenies, these isolates were identified to five species, including two new species, Pestalotiopsistaxicola and P.multicolor, two potential novel Neopestalotiopsis species, Neopestalotiopsis sp. 3 and Neopestalotiopsis sp. 4, with a known Pestalotiopsis species (Pestalotiopsistrachycarpicola), firstly recorded from Chinese yew. These two new Pestalotiopsis species were morphologically and phylogenetically distinct from the extant Pestalotioid species in Chinese yew. Pathogenicity and culture characteristic tests of these five Pestalotioid species were also performed in this study. The pathogenicity test results revealed that Neopestalotiopsis sp. 3 can cause diseases in Chinese yew needles. These results have indicated that the diversity of Pestalotioid species associated with Chinese yew was greater than previously determined and provided helpful information for Chinese yew disease diagnosis and management.

Diplodia tip blight, caused by Diplodia sapinea (=Sphaeropsis sapinea), are widely distributed in Honghuaerji, Inner Mongolia, China, causing severe damage on natural Mongolian pine (Pinus sylvestris var. mongolica). D. sapinea is an endophyte that becomes pathogenic under conditions of drought, hail damage, or temperature-associated stress. The role of the endophytic community inhabiting different pine tissues in the expression of disease is still unknown. In this study, the diversity and community structure of endophytic fungi among asymptomatic and symptomatic Mongolian pine were detected using culture-based isolation and high-throughput sequencing (HTS), and the potential antagonistic endophytes against D. sapinea were also screened. The results indicated that 198 and 235 strains of endophytic fungi were isolated from different tissues of symptomatic and asymptomatic Mongolian pine, respectively. D. sapinea was the most common endophyte isolated from the current-year needles and shoots of symptomatic trees, and Diplodia was also the most common in the HTS data. There were no significant differences in the endophytic fungal species richness among asymptomatic and symptomatic trees, but there were differences observed within specific sampled tissues. The ANOSIM analysis confirmed that the endophytic fungi community structure significantly differed between sampling tissues among symptomatic and asymptomatic Mongolian pine. Furthermore, the antagonism study revealed Penicillium fructuariae-cellae with the ability to inhibit the growth of D. sapinea in vitro, and the potential performance of this fungus, acting as biological control agent, was evaluated under greenhouse. Our findings can pave the way to a better understanding of the interactions between D. sapinea, other endophytic fungi and their hosts, and provide helpful information for more efficient disease management strategies.

Chrysomyxa rusts are fungal pathogens widely distributed in the Northern hemisphere, causing spruce needle and cone rust diseases, and they are responsible for significant economic losses in China. Taxonomic delimitation and precise species identification are difficult within this genus because some characters often overlap in several species. Adequate species delimitation, enhanced by the use of DNA-based methodologies, will help to establish well-supported species boundaries and enable the identification of cryptic species. Here, we explore the cryptic species diversity in the rust genus Chrysomyxa from China. Species delimitation analyses are conducted using a distance-based method (ABGD) and three tree-based methods (GMYC, bPTP, and mPTP) based on combined LSU and ITS sequences of over 60 specimens. Although there is some incongruence among species delimitation methods, two new species and three putative cryptic species are identified. The key to 20 Chrysomyxa species distributed in China is presented. These results suggest that a significant level of undiscovered cryptic diversity is likely to be found in Chrysomyxa from China. Future studies should consider multiple analytical methods when dealing with multi-locus datasets.
