RESUMEN

The multivariate analyses are useful tools to estimate the genetic variability between accessions. In the breeding programs, the Ward-Modified Location Model (MLM) multivariate method has been a powerful strategy to quantify variability using quantitative and qualitative variables simultaneously. The present study was proposed in view of the dearth of information about popcorn breeding programs under a multivariate approach using the Ward-MLM methodology. The objective of this study was thus to estimate the genetic diversity among 37 genotypes of popcorn aiming to identify divergent groups associated with morpho-agronomic traits and traits related to resistance to Fusarium spp. To this end, 7 qualitative and 17 quantitative variables were analyzed. The experiment was conducted in 2014, at Universidade Estadual do Norte Fluminense, located in Campos dos Goytacazes, RJ, Brazil. The Ward-MLM strategy allowed the identification of four groups as follows: Group I with 10 genotypes, Group II with 11 genotypes, Group III with 9 genotypes, and Group IV with 7 genotypes. Group IV was distant in relation to the other groups, while groups I, II, and III were near. The crosses between genotypes from the other groups with those of group IV allow an exploitation of heterosis. The Ward-MLM strategy provided an appropriate grouping of genotypes; ear weight, ear diameter, and grain yield were the traits that most contributed to the analysis of genetic diversity.

A challenge faced by popcorn breeding programs is the existence of a negative correlation between the two main traits, popping expansion and yield, which hinders simultaneous gains. The objective of this study was to investigate the use of a new variable or super trait, which favors the reliable selection of superior progenies. The super trait 'expanded popcorn volume per hectare' was introduced in the evaluation of 200 full-sib families of the eighth recurrent intrapopulation selection cycle, which were arranged in randomized blocks with three replicates in two environments. Although the inability to obtain simultaneous gains through selection via popping expansion or yield was confirmed, the super trait was positively associated with both yield and popping expansion, allowing simultaneous gains via indirect selection using 'expanded popcorn volume per hectare' as the main trait. This approach is recommended because this super trait can be used in breeding programs to optimize selective gains for the crop.

The successful development of hybrid cultivars depends on the reliability of estimated combining ability of the parent lines. The objectives of this study were to assess the combining ability of partially inbred S3 families of popcorn derived from the open-pollinated variety UENF 14, via top-crosses with four testers, and to compare the testers for their ability to discriminate the S3 progenies. The experiment was conducted in the 2015/2016 crop season, in an incomplete-block (Lattice) design with three replications. The following agronomic traits were evaluated: average plant height, grain yield (GY), popping expansion (PE), and expanded popcorn volume per hectare. The top-cross hybrid, originating from the BRS-Angela vs S3 progeny 10 combination, was indicated as promising, showing high values for specific combining ability for GY and PE. For the S3 progenies that showed high and positive GCA values for GY and PE, the continuity of the breeding program is recommended, with the advance of self-pollination generations. Fasoulas' differentiation index discriminated the BRS-Angela tester as the most suitable for identifying the superior progenies.

The popcorn breeding program of Universidade Estadual do Norte Fluminense Darcy Ribeiro aims to provide farmers a cultivar with desirable agronomic traits, particularly with respect to grain yield (GY) and popping expansion (PE). We evaluated full-sib families from the seventh cycle of recurrent selection and estimated the genetic progress with respect to GY and PE. Eight traits were evaluated in 200 full-sib families that were randomized into blocks with two replicates per set in two contrasting environments, Campos dos Goytacazes and Itaocara, located in north and northwest Rio de Janeiro State, respectively. There were significant differences between sets in families with respect to all traits evaluated, which indicates genetic variability that may be explored in future cycles. Using random economic weights in the selection of superior progenies, the Mulamba and Mock index showed gains for PE and GY of 5.11 and 7.78%, respectively. Significant PE and GY increases were found when comparing the evolution of mean values of these two parameters that were assessed at cycles C0-C6 and predicted for C7. Thus, an advanced-cycle popcorn cultivar with genotypic superiority for the main traits of economic interest can be made available to farmers in Rio de Janeiro State.

Pathogenesis-related proteins (PRs) are among the defense mechanisms of plants that work as an important barrier to the development of pathogens. These proteins are classified into 17 families according to their amino acid sequences, serology, and/or biological or enzyme activity. The present study aimed to identify PRs associated with the pathosystem of Capsicum baccatum var. pendulum: Pepper yellow mosaic virus (PepYMV). Forty-five-day-old plants from accession UENF 1624, previously identified as resistant to PepYMV, were inoculated with the virus. Control and infected leaves were collected for analysis after 24, 48, 72, and 96 h. The inoculated and control plants were grown in cages covered with anti-aphid screens. Proteins were extracted from leaf tissue and the presence of ß-1,3-glucanase, chitinase, peroxidase, and lipid transport protein was verified. No difference was observed between the protein pattern of control and infected plants when ß-1,3-glucanase, chitinase, and lipid transport protein were compared. However, increased peroxidase expression was observed in infected plants at 48 and 72 h after inoculation, indicating that this PR is involved in the response of resistance to PepYMV in C. baccatum var. pendulum.

We estimated the genetic gains of the 12th cycle of reciprocal recurrent selection for maize traits of agronomic interest. We used 23 ISSR molecular markers in an attempt to maximize genetic variability among and within populations based on selection of S(1) progenies. To this end, 138 full-sib families were evaluated in a randomized block design in two environments (the municipalities of Campos dos Goytacazes and Itaocara, in the State of Rio de Janeiro, Brazil), with replications within sets. Direct selection for grain yield was used for the selection of the families. To assess genetic diversity among and within populations, we examined plants produced from part of the S(1s) seeds from the parents that originated the 42 full-sib families that were selected from the agronomic traits. Direct selection for grain yield provided good gains for the traits evaluated, with estimated improvement of -0.87 days for days to flowering, 0.35 plants, 1.79 ears per plot, 0.58 g per 100-grain weight, 308.21 g ear weight per plot, and 261.83 kg/ha grain yield. Application of molecular markers at the stage of superior progeny selection led to increased genetic distance among populations, which is a very important factor for utilization of heterosis and providing greater longevity to the reciprocal recurrent selection program.

As part of the Universidade Estadual do Norte Fluminense recurrent selection program of popcorn, we evaluated full-sib families of the sixth cycle of recurrent selection and estimated genetic progress for grain yield and expansion capacity. We assessed 200 full-sib families for 10 agronomic traits, in a randomized block design, with two replications within sets in two environments: Campos dos Goytacazes and Itaocara, in the State of Rio de Janeiro, Brazil. There were significant differences for families/"sets" for all traits, indicating genetic variability that could be exploited in future cycles. In the selection of superior progenies, the Mulamba and Mock index gave the best gains for popping expansion (PE) and grain yield (GY), with values of 10.97 and 15.30%, respectively, using random economic weights. By comparing the evolution of the means obtained for PE and GY in the cycles C(0), C(1), C(2), C(3), C(4), C(5), and predicted for C(6), a steady increase was observed for both PE and GY, with the addition of 1.71 mL/g (R(2) = 0.93) and 192.87 kg/ha (R(2) = 0.88), respectively, in each cycle. Given the good performance of this popcorn population in successive cycles of intrapopulation recurrent selection, we expect that a productive variety with high expansion capacity will soon be available for producers in the north and northwest regions of Rio de Janeiro State, Brazil.

We examined the effect of incorporation of molecular markers on variability between and within populations in order to maximize heterotic effects and longevity of a maize reciprocal recurrent selection program. Molecular variability was quantified by inter-simple sequence repeat (ISSR) markers between and within the maize populations Cimmyt and Piranão in the 10th cycle of a reciprocal recurrent selection program. Forty-two S(1) progenies of each population were analyzed, these being families of full-sibs selected according to their agronomic traits. Thirteen primers were selected, which produced 140 bands; 114 of them were polymorphic and 26 monomorphic. Based on UPGMA grouping analysis and by genetic distances, it was possible to identify "contaminant" progenies. These progenies belong to the Piranão or Cimmyt groups, but cluster in the opposite heterotic group. Identification of "contaminant" progenies is relevant for selection, because, besides identifying genotypes that should be eliminated at the recombination stage, it allows increased heterosis expression in crosses between more genetically distinct individuals. After the elimination of the "contaminant" progenies and those that were allocated between the heterotic groups, a new statistical analysis was carried out, which demonstrated increased genetic distances between the populations. It was concluded that the application of molecular markers in reciprocal recurrent selection programs allows the optimization of the monitoring of genetic variability within and between populations, favoring recombination between more distant progenies, besides ensuring increased longevity of the reciprocal recurrent selection program.

Studies of genetic effects of early selection of maize based on seed quality traits are rare, especially those that use materials from different heterotic groups. These studies are also useful in tropical environments and for the advancement of sustainable agriculture with cropping during seasons not commonly used for cultivation. We estimated, through diallel crosses, the predominant genetic effects on the expression of agronomic traits and seed quality and on the general combining ability of nine maize lines from commercial hybrids and the specific combining ability of hybrid combinations among them. In the evaluation of seed quality, seven tests were used: first count and final count of seed germination, seedling vigor classification, cold tolerance, seedling emergence rate in a sand seedbed, speed of emergence in a sand seedbed, and speed of emergence index. Plant height, first ear height and grain yield were the estimated agronomic traits. In the diallel analysis, method 3 (model I) proposed by Griffing was used. There was a greater significance of non-additive genetic effects in the genetic control of seed quality of the various lines. The Flash, Dekalb 350 and P 30F80 lines combined high seed quality and high grain yield. For growth during the normal planting season, the combinations CD 3121-1 x P 30F80, Speed x CD 3121-2, Dow 8330 x AG 8080 and Dekalb 350 x CD 3121-2 were the most promising for both seed quality and agronomic traits.

Considering the expected genetic variability of elephant grass (Pennisetum purpureum), due to its cultivation in different continents, we characterized and estimated the genetic divergences between 46 accessions of elephant grass with different edaphoclimatic adaptations, using RAPD and ISSR markers. We evaluated, comparatively, the consistency of the information achieved with these markers. Twenty-six RAPD and 25 ISSR primers were employed. The RAPD markers produced 185 bands, 72% of which were polymorphic, with a mean of 5.11 polymorphic bands per primer. The 25 ISSR starters produced 216 bands; 76% were polymorphic, with a mean of 6.56 polymorphic bands per primer. The correlation between the genetic distances achieved by the RAPD and ISSR markers was 0.76, which is highly significant by the Mantel test. Based on UPGMA grouping, considering the point of sudden change, five and six groups were formed for the data from the RAPD and ISSR markers, respectively. These markers provided partially concordant groups, indicating that these techniques can provide consistent information and consequently could be used in studies of genetic diversity among accessions.

In Brazil, using combining ability of popcorn genotypes to achieve superior hybrids has been unsuccessful because the local genotypes are all members of the same heterotic group. To overcome this constraint, 10 lines (P(1) to P(10)) with different adaptations to tropical or temperate edaphoclimatic environments were used to obtain 45 F(1) hybrids in a complete diallel. Capsicum species are very important in Brazil because of economic, cultural and biological factors, and the country is considered to be a diversity center for this genus. Collection and maintenance of the genetic diversity in Capsicum are important to avoid genetic erosion. Besides the identification of species, the characterization and evaluation of accessions maintained in gene banks are of fundamental importance. For this purpose, multivariate methods have become an important tool in the classification of conserved genotypes. The objectives of this study were: i) to identify and characterize accessions of the Capsicum spp collection and draw conclusions about the potential use of certain accessions in different production sectors; ii) to estimate the genetic divergence among accessions using the Ward-MLM procedure, and iii) to evaluate the efficiency of the analysis of continuous and categorical data using the Ward-MLM procedure. Fifty-six Capsicum spp accessions were evaluated based on 25 descriptors, 14 of which were morphological and 11 agronomic. Based on the qualitative descriptors, it was possible to identify all species and, together with the agronomic descriptors, genotypes could be indicated with potential for use in various production sectors. Five was determined as the ideal number of groups by the criteria pseudo-F and pseudo-t2. The Ward-MLM procedure allowed the differentiation of the species C. annuum, C. frutescens, C. baccatum, and C. chinense in separate groups. The Ward-MLM procedure showed some level of efficiency in clustering Capsicum species analyzing morphological and agronomic data simultaneously.

Accessions in gene banks need to be characterized and evaluated to determine their genetic diversity. We made a joint diversity analysis of the tomato gene bank of the Universidade Estadual do Norte Fluminense Darcy Ribeiro in Rio de Janeiro state, using the Ward-modified location model. Forty Solanum lycopersicum accessions were characterized and evaluated for 22 morphoagronomic descriptors and 131 random amplified polymorphic DNA markers. Based on the pseudo-F and pseudo-t(2) criteria, the optimal number of groups was established as five. Variability within groups was high for both continuous and discrete nominal data. The first two canonical variables explained about 90% of the inter-group variability. Care should be taken in using the Ward-modified location model technique to avoid incorporating excessive and unnecessary markers, which could favor molecular markers when compared with morphoagronomic variables. However, the minimum number of markers is germplasm- dependent and must be recalculated for each new divergence analysis.
