Limited background data are available on the Mishmi takin (Budorcas taxicolor taxicolor) and Bhutan takin (Budorcas taxicolor whitei) subspecies in the Eastern Himalayas of China because of the lack of systematic field investigations and research. Therefore, mature-animal ecological methods were used to evaluate these takin subspecies' phenotypic characteristics, distribution range, activity rhythm, and population size. From 2013 to 2022, 214 camera traps were installed for wild ungulate monitoring and investigation in all human-accessible areas of the Eastern Himalayas, resulting in 4837 distinguishable takin photographs. The external morphological characteristics were described and compared using visual data. Artificial image correction and related technologies were used to establish physical image models based on the differences between subspecies. MaxEnt niche and random encounter models obtained distribution ranges and population densities. Mishmi takins have a distribution area of 17,314 km2, population density of 0.1729 ± 0.0134 takins/km2, and population size of 2995 ± 232. Bhutan takins have a distribution area of 25,006 km2, population density of 0.1359 ± 0.0264 takins/km2, and population size of 3398 ± 660. Long-term monitoring data confirmed that the vertical migration within the mountain ecosystems is influenced by climate. Mishmi takins are active at 500-4500 m, whereas Bhutan takins are active at 1500-4500 m. The two subspecies were active at >3500 m from May to October yearly (rainy season). In addition, surveying combined with model simulation shows that the Yarlung Zangbo River is not an obstacle to migration. This study provides basic data that contribute to animal diversity knowledge in biodiversity hotspots of the Eastern Himalayas and detailed information and references for species identification, distribution range, and population characteristics.

The Sichuan takin (Budorcas tibetanus) and the golden takin (Budorcas bedfordi) are endemic to China. Studies of their habitat ecology have attracted attention, but dedicated reviews of the topic have not been published. To fill this gap, research advances on the habitat ecology of these takin species were reviewed based on published journal papers from 2001-2022, covering habitat selection and use, as well as habitat assessment and protection. Habitat selection and use by the takins received the most frequent reports in a few protected areas, such as the Tangjiahe Nature Reserve in Sichuan and the Foping Nature Reserve in Shaanxi. Higher herb cover characterized some of the Sichuan takin habitats, while some of the golden takin population exhibited seasonal altitude migration. The focus on habitat assessment and protection differed between the two species. Studying the habitat of the Sichuan takin involves the relationship between its habitat and the protected areas and habitat status in various ranges while studying the habitat of the golden takin involves habitat distribution and change, protection effects, and corridor planning. Some issues worthy of consideration in future research are proposed, including basic habitat information, impacts of human disturbances, habitat restoration, and theoretical, independent, and comprehensive studies on both takins. Habitat conservation frameworks should be formulated for both takins to improve protection and restoration effects. This review provides convenient information to help understand the habitat characteristics of the two takins and is a reference for future research.

This study investigates the non-invasive monitoring of the endocrine ovarian activities of captive female golden takins (Budorcas taxicolor bedfordi) based on long-term fecal sex steroid hormone metabolite dynamics. Fecal progesterone (P4) metabolite dynamics were monitored in nine females for 0.5-15 years between 2004 and 2022. Fecal estradiol-17ß (E2) and estrone (E1) metabolites were measured during certain estrous cycles, and fecal E1 metabolite concentrations were measured during all gestation periods. The breeding season of the captive animals was mainly between May and December, and they were polyestrous animals whose breeding season begins during the long-day period. The onset of the breeding season occurred slightly earlier as age increased. The mean age (±SD) at puberty based on fecal P4 metabolite dynamics was 4.1 ± 2.9 years. The first conception ages ranged from 2.3-10.2 years. The mean estrous cycle period (±SEM) was 25.4 ± 1.1 days, and mounting and mating occurred in periods of low fecal P4 metabolite levels during the breeding season. The mean gestation period (±SD) from the estimated mating date to the calving date was 253.9 ± 5.7 days, and the fecal P4 metabolite distribution during pregnancy was bimodal. Fecal estrone metabolite levels gradually increased 21 weeks before delivery, peaked during the week of delivery, and then markedly decreased in the first week after delivery. Estrus resumed in the first April-August period after delivery (mean ± SD; 103.5 ± 40.9 days) or in May of the year after delivery (421.0 ± 16.5 days). This study revealed that the estrous cycle and pregnancy of female golden takins can be determined by fecal progesterone metabolite dynamics and that fecal estrone metabolite dynamics increases toward parturition and are useful for predicting the date of delivery. This endocrinological information is important for planned breeding efforts for the golden takins.

In this study, we investigated the change in microbiome composition of wild Sichuan takin (Budorcas tibetanus) during winter and spring and analyzed the physiological implications for such changes. Diversity analyses of the microbiome (average 15,091 high-quality reads per sample) in 24 fecal samples (15 from winter, 9 from spring) revealed that spring samples had higher species diversity and were compositionally different from winter samples (P < 0.05). Taxonomic composition analysis showed that the relative abundance increased in spring for Patescibacteria (2.7% vs. 0.9% in winter, P < 0.001) and Tenericutes (1.9% vs. 1% in winter, P < 0.05). Substantial increases in relative abundance of Ruminococcaceae and Micrococcaceae were identified in spring and winter, respectively. Mann-Whitney U and ANCOM identified seven differentially abundant genera: Enterococcus, Acetitomaculum, Blautia, Coprococcus 1, Lachnospiraceae UCG 008, Ruminococcus 2 and Ralstonia. All seven genera were significantly more abundant in spring (average 0.016-1.2%) than winter (average 0-0.16%), with the largest difference found in Ruminococcus (1.21% in spring vs. 0.16% in winter). The other six genera were undetectable in winter. Functional prediction and pathway analysis revealed that biosynthesis of cofactors (ko01240) had the highest gene count ratios in the winter, followed by the two-component system (ko02020). Seasonal variation affects the gut microbiomes in wild Sichuan takins, with winter associated with lower species diversity and spring with enrichment of cellulose-degrading genera and phytopathogens. Such changes were crucial in their adaptation to the environment, particularly the difference in food abundance.

The takin (Budorcas taxicolor) is one of the largest bovid herbivores in the subfamily Caprinae. The takin is at high risk of extinction, but its taxonomic status and genetic diversity remain unclear. In this study, we constructed the first reference genome of Bu. taxicolor using PacBio long High-Fidelity reads and Hi-C technology. The assembled genome is ~2.95 Gb with a contig N50 of 68.05 Mb, which were anchored onto 25+XY chromosomes. We found that the takin was more closely related to muskox than to other Caprinae species. Compared to the common ancestral karyotype of bovidae (2n = 60), we found the takin (2n = 52) experienced four chromosome fusions and one large translocation. Furthermore, we resequenced nine golden takins from the main distribution area, the Qinling Mountains, and identified 3.3 million single nucleotide polymorphisms. The genetic diversity of takin was very low (θπ = 0.00028 and heterozygosity =0.00038), among the lowest detected in domestic and wild mammals. Takin genomes showed a high inbreeding coefficient (FROH =0.217), suggesting severe inbreeding depression. The demographic history showed that the effective population size of takins declined significantly from ~100,000 years ago. Our results provide valuable information for protection of takins and insights into their evolution.

Introduction: The intestinal tract of animals is a complex and dynamic microecosystem that is inextricably linked to the health of the host organism. Takin (Budorcas taxicolor) is a threatened species, and its gut microbiome is poorly understood. Therefore, this study aimed to analyze the microbial community structure and potential pathogens of takin. Methods: Takin fecal samples were collected from five sites in a nature reserve to ensure the uniformity of sample collection, determine the effects of different geographical locations on gut microbes, and analyze the differences in microbial communities between sites. Subsequently, high-throughput 16S rDNA gene sequencing was performed to analyze the microbial diversity and potential pathogens in the gut; the findings were verified by isolating and culturing bacteria and metagenomic sequencing. Results and discussion: The takin gut microflora consisted mainly of four phyla: Firmicutes (69.72%), Bacteroidota (13.55%), Proteobacteria (9.02%), and Verrucomicrobiota (3.77%), representing 96.07% of all microorganisms. The main genera were UCG-005 (20.25%), UCG-010_unclassified (12.35%), Firmicus_unclassified (4.03%), and Rumino coccsea_unclassified (3.49%), while the main species were assigned to Bacteria_unclassified. Potential pathogens were also detected, which could be used as a reference for the protection of takin. Pseudomonas presented the highest abundance at Shuichiping and may represent the main pathogen responsible for the death of takin at the site. This study provides an important reference for investigating the composition of the bacterial community in the intestine of takin.

RESUMEN Objetivo Proponer una herramienta para identificar sectores de población que requieren mayor atención por parte de autoridades locales o gubernamentales en situaciones de crisis biológica, considerando los factores que influyen en la adherencia a las normas de minimización de riesgos. Metodología Se implementó un algoritmo de ordenamiento, tomando como referencia las restricciones de julio del 2021 en Ecuador. El contexto del estudio se resume en siete sectores urbanos de la ciudad de Guayaquil, con una población caracterizada por un nivel de educación promedio por debajo de la educación secundaria superior (70%) y más del 50% entre 20-34 años, con alguna ocupación en el medio de una economía popular debilitada. Siete factores de riesgo fueron identificados después de un análisis estructural de la hipótesis de adherencia (x2/gl=3,6; CFI≥0,91; TLI≥0,90; RMSEA≤0,05), basado en una muestra aleatoria de n=515 adultos viviendo en las áreas afectadas. Resultados El seguimiento de las normas está influenciado por la percepción del clima de seguridad, el riesgo percibido y el entendimiento del riesgo. El umbral de ordenamiento (h) permite establecer relaciones unidireccionales entre variables. Conclusiones Los resultados muestran que Vergeles, Norte y Fertisa representan los sectores con mayor prioridad de atención en materia de salud pública {A4,A5,A6}>{A2}>{A3}>{A1}>{A7}. Se requiere identificar más factores para garantizar una diferenciación óptima.


ABSTRACT Objective To propose a tool to identify local communities that require public health work priority, taking into account factors that influence adherence to risk minimization guidelines, especially in lock-down environments and unconventional workplaces. Methodology An ordering algorithm, based on the theory of uncertainty, was applied to classify population zones exhibiting high levels of infection and non-compliance with regulations in Guayaquil, during the last 'weekend' lockdown episode in July 2021. Seven urban sectors showed the highest number of infections (more than 70% of the local population): Vergeles (A1), Samanes (A2), Socio Vivienda (A3), Guasmo Norte (A4), Fertisa (A5), Alborada (A6), Urdesa (A7). Seven risk factors were identified after a path analysis of compliance hypothesis (x2/gl=3,6; CFI≥0,91; TLI≥0,90, RMSEA≤0,05), based on a random sample of n=515 adults living in the affected areas. Results Adherence to norms is influenced by the safety climate, perceived risk and risk understanding. The ordering threshold (h) leaded unidirectional relationships between variables. Conclusions: Adding more factors are believed to increases the differentiation path. The results showed that Vergeles, Norte and Fertisa were the areas with the highest priority for public health care {A4,A5,A6}>{A2}>{A3}>{A1}>{A7}.

The takin (Budorcas taxicolor) is an endemic ruminant species belonging to the bovine family. The International Union for Conservation of Nature (IUCN) has listed it as an endangered and vulnerable species. However, little is known about its molecular characterization since it lacks a reference genome. This study used RNA sequencing followed by de novo assembly, annotation and simple sequence repeats (SSRs) prediction to assess the transcriptome of Qinling takin (Budorcas taxicolor bedfordi) muscles. In total, 21,648 unigenes with an N50 and mean length of 1388 bp and 817 bp, respectively, were successfully detected and annotated against the public databases (NR, GO, KEGG, and EggNOG). Furthermore, 6222 SSRs were identified using the MIcroSAtellite (MISA) identification tool software. Taken together, these findings will provide valuable information for genetic, genomic, and evolutionary studies on takin.

For many species in zoos, particularly megafauna vulnerable to heat stress, shade is a key environmental resource. However, shade availability has received comparatively less attention than other aspects of the zoo environment. In this study, we share a simple low-cost approach that we applied to document shade availability across 33 zoo enclosures. We then combined these assessments with behavioral observations of enclosure use and shade-seeking behavior during summer months in a case study focused on Sichuan takin (Budorcas taxicolor tibetana) (n = 3), a large cold-adapted bovid. Behavioral observations were conducted before and after installation of a shade sail for the takin. Results indicated that shade availability varied widely across zoo enclosures, with the percent of shaded space ranging from 85 % to 22 % across enclosures during summer months. Shade was a dynamic resource and increased throughout the year and fluctuated across the day, with the least shade available midday. Takin showed general preferences for shaded areas near the walls of their enclosure but were observed using newly available shade from the shade sail after its installation. These accessible methods can be easily applied to assess shade within existing enclosures, evaluate enclosure modifications, and provide guidance for the design of new enclosures.

Infanticide by unrelated individuals is widely reported in the animal kingdom; however, little is known about cases perpetrated by a parent, particularly the mother. This article reports on three cases of mother-initiated infanticide in Qinling golden takins ( Budorcas taxicolor bedfordi) recorded from video and camera images. Based on previous reports in other animals, we propose that the infanticide events observed in golden takins were related to the parental manipulation mechanism - i.e., killing an unhealthy infant to allow the mother to invest more care in potentially healthy offspring, and gain more fruitful reproductive opportunities. This appears to be an evolutionary-based selection strategy, whereby a species can prosper and succeed under the challenges of natural selection. However, further studies on both captive and wild populations are required to answer the various questions raised from our observations.

The flagship species conservation strategy is widely practiced in nature reserves. However, few studies have quantitatively explored whether the current functional zoning adequately conserves the habitats of other species. To examine this problem, we selected the Tangjiahe, Xuebaoding, and Wanglang national nature reserves in southwest China and tested the protective efficiency of the giant panda (Ailuropoda melanoleuca) reserves on a local herbivore-Sichuan takin (Budorcas taxicolor tibetana) in functional zoning. We predicted that the habitat distribution of the Sichuan takin with Maxent and habitat suitability would be classified as very low, low, moderately, or highly suitable. We calculated the percentage of each type of habitat class across the core, buffer, and experimental zones in these reserves and introduced an economic index present value index to further assess the efficacy of the functional zoning in conserving the habitat of Sichuan takin. Our findings indicate that suitable habitat distributions of Sichuan takin are multicore and influenced by diverse variables in different nature reserves. The percentage of moderately suitable and highly suitable habitat in the core zone is 69% and 54% in Tangjiahe, 76% and 75% in Wanglang, and 47% and 28% in Xuebaoding, respectively. The present value index of moderately and highly suitable habitat is 1.01 and 0.79 (Tangjiahe), 0.87 and 0.86 (Wanglang), 0.76 and 0.44 (Xuebaoding), respectively. The efficiency of functional zoning on Sichuan takin seems lower than imagined in giant panda reserves, and species that have important impacts on local ecosystem need further estimation and greater focus.

Hypertrophic osteopathy, a syndrome characterized by painful distal limb swelling and proliferative periosteal reaction, primarily involves the metacarpal and metatarsal bones and phalanges. Lesions are often bilaterally symmetric and typically affect all four limbs. Hypertrophic osteopathy is frequently associated with primary intrathoracic disease, though this condition has also been reported secondary to intra-abdominal and intrapelvic disease and associated with pregnancy in both people and horses. Over a 20-yr period, five adult female Sichuan takin (Budorcas taxicolor tibetana), with 13 total pregnancies, were evaluated because of lameness and distal limb swelling. These clinical signs were observed between 2 and 32 days (mean = 19 days) prior to parturition (gestation period in takin approximately 200-240 days) and resolved in all animals following parturition. Lameness and limb swelling resolved between 8 and 168 days (median = 15 days) after parturition. Sixteen radiographic examinations, from four of the individuals, documented proliferative periosteal reaction, primarily of the metacarpal bones, metatarsal bones, and phalanges. The clinical progression, resolution of signs, and radiographic features in these cases are consistent with hypertrophic osteopathy, secondary to pregnancy. This is the first report describing presumptive hypertrophic osteopathy in takin.

Mishmi takin (Budorcas taxicolor taxicolor) is an endangered and least studies species. We sequenced and annotated a 16,584 bp long complete mitogenome of Mishmi takin (B. t. taxicolor). It was encoded by 37 genes: 13 Protein-Coding Genes (PCGs), two ribosomal RNA genes, 22 transfer RNA, and non-coding control region. All tRNA genes show a typical secondary cloverleaf structure, except tRNAser(AGY) in which dihydrouridine arm did not form a stable structure. We observed 82 bp nucleotide deletions in the control region of Mishmi takin. The overall analysis of 13 PCGs, two rRNA and 22 tRNA of Mishmi takin indicates its close relationship with the B. t. tibetana than the B. t. bedfordi. The overall genetic similarity indicated that all takin species is closely related to the P. nayaur than the C. ibex and Ovibos moschatus. This study provides a useful resource for further studies on conservation and population genetics of Budorcas.

Mycobacterium avium subspecies paratuberculosis (MAP) is a cause of contagious and typically fatal enteric disease, primarily affecting ruminant and pseudoruminant species. During a MAP outbreak in a captive collection, six of nine adult Mishmi takin ( Budorcas taxicolor taxicolor) showed marked weight loss over 1-3 mo, followed by an acute deterioration. Fecal culture and microscopy failed to identify MAP shedding. Necropsy findings included grossly normal intestines and marked enlargement of mesenteric lymph nodes. Histological findings included multibacillary granulomatous enteritis, mesenteric lymphadenitis, and periportal hepatitis. MAP was confirmed by culture of intestinal and lymph node tissues from the index case. Results of antemortem serological testing using an indirect ELISA (ID SCREEN® Paratuberculosis Indirect) were corroborated by findings at necropsy or survival of the outbreak. Mishmi takin appear to show high MAP susceptibility and a rapid disease course compared with domestic ruminant species.

The Sichuan takin inhabits the bamboo forests in the Eastern Himalayas and is considered as a national treasure of China with the highest legal protection and conservation status considered as vulnerable according to The IUCN Red List of Threatened Species. In this study, fecal samples of 71 Sichuan takins were pooled and deep sequenced. Among the 103,553 viral sequences, 21,961 were assigned to mammalian viruses. De novo assembly revealed genomes of an enterovirus and an astrovirus and contigs of circoviruses and genogroup I picobirnaviruses. Complete genome sequencing and phylogenetic analysis showed that Sichuan takin enterovirus is a novel serotype/genotype of the species Enterovirus G, with evidence of recombination. Sichuan takin astrovirus is a new subtype of bovine astrovirus, probably belonging to a new genogroup in the genus Mamastrovirus. Further studies will reveal whether these viruses can also be found in Mishmi takin and Shaanxi takin and their pathogenic potentials.

We investigated the habitat use characteristics of takin in Wanglang and Xiaohegou Nature Reserves to detect whether takin has the different habitat use patterns in different environments. We found that takin has similarities and differences in habitat use in these two nature reserves. Takin at Wanglang usually used the habitats with greater bamboo coverage and bamboo density, whereas takin at Xiaohegou usually used the habitats with a lower canopy. The results of a logistic regression analysis showed that bamboo density, herb-cover proportion, and tree size contributed significantly to the difference between takin plots at Wanglang and Xiaohegou, with an overall correct prediction rate of 92.3%, indicating that these are the main factors discriminating habitat use characteristics of takin in these two nature reserves. Considering takin has different habitat use characteristics in different areas, different protection measures should be taken to protect different takin populations. Based on the habitat use characteristics of takin at Wanglang and Xiaohegou, we summarized adaptation of takin to environments as general adaptation, regional adaptation, and special adaptation.

Chemical elements exposure of endangered golden takins (Budorcas taxicolor bedfordi) living in the Qinling Mountains and in a captive breeding center was assessed by analyzing fecal samples. Concentrations of As, Co, Cr, Cu, Ni and Se were significantly higher in the feces of captive golden takins than the wild. There was no significant difference in the fecal concentrations of Cd, Mn, Hg, Pb or Zn for wild and captive animals. The element concentration of fecal samples collected from captive animals varied seasonally, with concentrations being lowest in spring and highest in winter and/or autumn. The food provided to captive animals varied both in the composition and the concentration of element present. Consumptions of feedstuff and additional foods such as D. sanguinalis and A. mangostanus for the captive golden takins were identified as the possible sources of chemical element exposure. The estimations of dietary intake of most elements by captive takins were below the oral reference dose, except for As and Pb, indicating that As and Pb were the key components which contributed to the potential non-carcinogenic risk for captive golden takins. In conclusion, captive golden takins were exposed to higher concentrations of chemical elements compared with the wild, which were likely due to their dietary difference. Conservation efforts of captive golden takin are potentially compromised by the elevated chemical element exposure and effort should focus on providing uncontaminated food for captive animals.

The gut microbiota of mammals is a complex ecosystem, which is essential for maintaining gut homeostasis and the host's health. The high throughput sequencing allowed us to gain a deeper insight into the bacterial structure and diversity. In order to improve the health status of the endangered golden takins, we first characterized the fecal microbiota of healthy golden takins using high throughput sequencing of the 16S rRNA genes V3-V4 hypervariable regions. Our results showed that, Firstly, the gut microbiota community comprised 21 phyla, 40 classes, 62 orders, 96 families, and 216 genera. Firmicutes (67.59%) was the most abundant phylum, followed by Bacteroidetes (23.57%) and Proteobacteria (2.37%). Secondly, the golden takin maintained higher richness in spring than in the winter while community diversity and evenness was not significantly different. Thirdly, four female golden takins demonstrated highly similar microbiota and the five golden takin males had relatively highly similar microbiota. All of our results might indicate that the fecal microbiota of golden takins were influenced by the season and the animal's sex. The findings provided theoretical basis regarding the gut microbiota of golden takins and may offer new insights to protect this endangered species.

Being able to imagine another person's experience and perspective of the world is a crucial human ability and recent reports suggest that humans "embody" another's viewpoint by mentally rotating their own body representation into the other's orientation. Our recent Magnetoencephalography (MEG) data further confirmed this notion of embodied perspective transformations and pinpointed the right posterior temporo-parietal junction (pTPJ) as the crucial hub in a distributed network oscillating at theta frequency (3-7 Hz). In a subsequent transcranial magnetic stimulation (TMS) experiment we interfered with right pTPJ processing and observed a modulation of the embodied aspects of perspective transformations. While these results corroborated the role of right pTPJ, the notion of theta oscillations being the crucial neural code remained a correlational observation based on our MEG data. In the current study we therefore set out to confirm the importance of theta oscillations directly by means of TMS entrainment. We compared entrainment of right pTPJ at 6 Hz vs. 10 Hz and confirmed that only 6 Hz entrainment facilitated embodied perspective transformations (at 160° angular disparity) while 10 Hz slowed it down. The reverse was true at low angular disparity (60° between egocentric and target perspective) where a perspective transformation was not strictly necessary. Our results further corroborate right pTPJ involvement in embodied perspective transformations and highlight theta oscillations as a crucial neural code.

RESUMEN

A 10-year record review from a zoological institution in the western USA identified four cases of severe laminitis resulting in rotation and protrusion of the third phalanx through the sole. Laminitis is reported in a Masai giraffe (Giraffa camelopardalis tippelskirchi), a Sichuan takin (Budorcas taxicolor tibetana), a greater Malayan chevrotain (Tragulus napu) and a giant eland (Taurotragus derbianus). This is the first report of severe laminitis with pedal bone rotation and protrusion in multiple species of non-domestic hoofstock, and the first report of this disease in three of these species (takin, chevrotain, and giant eland).
