BACKGROUND: The ability of a high level of dietary Arthrospira platensis, individually or in combination with two exogenous carbohydrate-degrading enzymes (lysozyme and Rovabio®), to improve systemic antioxidant potential and hepatic lipid metabolism was tested in piglets. Forty male post-weaned piglets, sons of Large White × Landrace sows crossed with Pietrain boars, were allocated into 4 groups (n = 10) and fed during 28 days one of the following diets: 1) a control basal diet (cereal and soybean meal); 2) a basal diet with 10% of A. platensis (AP); 3) the AP diet supplemented with 0.005% of Rovabio® (AP + R); 4) the AP diet supplemented with 0.01% of lysozyme (AP + L). RESULTS: Arthrospira platensis decreased BW gain of piglets, regardless the addition of feed enzymes. The majority of plasma metabolites were affected by diets. A. platensis increased total lipids, total cholesterol and LDL-cholesterol, without changing hepatic fatty acid content or modulating, in an expressive manner, the transcriptional profile of lipid sensitive mediators. The antioxidant potential in general, and total carotenoids in particular, were improved by the microalga, regardless lysozyme or Rovabio®. CONCLUSIONS: Summing up, A. platensis, individually and combined with feed enzymes, impacts negatively on piglets' growth but improves the systemic antioxidant potential and changes plasma lipids with a minor modulation on related hepatic metabolic pathways.

Indigestible fiber-protein-phytate complexes reduce the feeding value of soy products. We investigated the effects of multienzyme supplement (MES, Victus) on standardized ileal digestibility (SID) of amino acids (AA) and apparent total tract digestibility (ATTD) of energy and minerals in roasted full-fat soybean (FFSB) seeds and expelled-extruded soybean meal (SBM) fed to growing pigs. The crude protein (CP) was 33.4% and 42.8% dry matter (DM) in FFSB seeds and SBM, respectively and corresponding values for crude fat were 17.4% and 11.8% DM. Semi-purified diets with 50% of either FFSB seeds or SBM as the sole source of AA were prepared without or with MES supplying phytase, protease, xylanase, and ß-glucanase at 2,200, 8,300, 400, and 100 U/kg of feed, respectively. Diets had TiO2 as an indigestible marker and the ratio of cornstarch to sucrose and corn oil was identical to calculate DE by the difference method. Eight ileal-cannulated barrows (22.1 ± 0.61 kg) were fed diets in a replicated 4 × 4 Latin square design to give eight replicates per diet. The period lasted for 9 d: 5 d for acclimation, 2 d for fecal, and 2 d for ileal digesta samples. There was no (P > 0.05) interaction between soy type and MES or MES effect on SID of AA; SBM had higher (P < 0.05) SID of CP, His, Leu, and Lys. There was no (P > 0.05) interaction between soy type and MES on energy digestibility. The FFSB seeds had higher ATTD of gross energy (GE, 80.2% vs. 76.6%; P < 0.01) than SBM. Pigs fed MES had higher (P < 0.05) ATTD of DM (91.3% vs. 87.7 %), GE (87.5% vs. 82.4%), CP (86.4% vs. 82.9%), crude fat (70.6% vs. 54.9%), Ca (63.2% vs. 60.2%), and P (67.5% vs. 63.2%). In conclusions, differences on AA and energy digestibility in soy products could be linked to processing and compositional differences. Although MES had no effect on SID of AA, the effects on the utilization of minerals and energy demonstrated the value of fiber-degrading enzymes, protease, and phytase in improving the nutritive value of soy products independent of processing.

The effect of a low-ME diet with a multienzyme (Kemzyme Plus, Kemin, Des Moines, IA) blend on performance, meat quality, and carcass traits was evaluated in Hubbard broiler chicks. A total of 120 Hubbard broiler chicks were allocated to the following 4 experimental groups and every group was separated into 6 replicates, with 5 birds per replicate: control (3,180 kcal/kg of ME), control + 0.50 g/kg diet of enzyme (Cont-Enz), low-ME diet (3,080 kcal/kg), and low-ME + 0.50 g/kg diet of enzyme (low-ME-Enz). The trail lasted for 16 D (32 to 48 D of age). No significant differences in growth parameters or carcass traits were observed among treatments. However, liver weight increased with the low-ME-Enz diet (P = 0.038). The low-ME diet recorded the highest weight for the bursa (P = 0.043) and thymus (P = 0.019). Dietary treatments had significant impacts on the length of duodenum, ileum, and cecum, as well as the weight of duodenum. The length of duodenum, ileum, and cecum increased with enzyme supplementation. The myofibril fragmentation index was lower with the Cont-Enz, low-ME, and low-ME-Enz diets than with the control diet (P = 0.043). The shear force increased with the low-ME-Enz diet (P = 0.022) than the control diet. Dietary treatments influenced breast meat yellowness (P = 0.019), whereas the low-ME diet had the lowest yellowness at the slaughtering age. The dietary treatments affected the breast meat pH (P = 0.001), with the control diet having the highest pH value after 24 hours. Thus, there was no effect of low-ME or enzyme supplementation to the control or low-ME diet on growth performance or carcass yield. However, feeding a low-ME diet or Cont-Enz preparation influenced organ and small intestine weights and meat characteristics.

The present study was carried out to explore the impacts of dietary supplementation of enzyme mixture with sodium butyrate on the growth performance, carcass traits, blood profile and economic benefit in two breeds of weanling rabbits adapted to survive in Egypt (New Zealand White and Rex). One-hundred and twenty weaned male rabbits (New Zealand White and Rex) of 6 weeks of age and 770.5 ± 20 g body weight were allotted randomly into four groups in a factorial arrangement. The obtained results indicated that there were non-significant differences in all growth performance traits, blood profile and economic parameters due to the breed effect. However, there were significant differences in most of carcass traits due to the breed effect except total giblets and New Zealand White breed showed the highest value of these parameters including dressing % (p < .01), forequarter and loin % (p < .001) and hindquarter % (p < .003) compared with Rex breed counterparts. The effect of the treatment and its interaction with the breed significantly (p < .05) improved body weight gain, feed consumption and carcass traits (percentage of dressing, forequarter, hind quarter and lion). However, final body weight and feed conversion ratio were not significantly influenced. Supplementing a diet with treatment significantly decreased blood triglycerides, cholesterol and the ratio between albumin and globulin (A/G ratio), while increased blood total protein and globulin. Although higher feed cost and total costs in treated groups than control ones in each breed, they showed higher total return and net return. Rex non-treated rabbit breed showed the lowest profitability measures compared with other groups. In conclusion, dietary supplementation of multi-enzyme with sodium butyrate is highly recommended in growing rabbits due to their beneficial effects on the growth performance and profitability.

The purpose of this study was to investigate the effects of dietary supplementation of non-starch polysaccharide multi-enzyme (NME) in early laying phase of hens on production performance, egg quality, nutrient digestibility, and excreta noxious gas emission. In total, 432 Hy-line brown laying hens at 18 wk of age were used in a 10-wk feeding trail. Hens were randomly assigned to 1 of 3 treatments with 24 replication and 6 hens per replication (1 hen per cage). Dietary treatments were corn-soybean meal-DDGS-based diets supplemented with 0 (based diet, CON), 0.05% (NME1), and 0.1% (NME2) of NME. No significant (P > 0.05) response to increasing NME supplementation was observed for damaged egg rate shown throughout the experiment. Significant (P < 0.05) linear increase was observed for egg production at week 4, 6, and 8; moreover, egg production at week 8 also showed quadratic (P = 0.0344) increase. No significant effects were found on yolk color, eggshell strength, and eggshell thickness during the experiment (P > 0.05) with the increase in NME supplemental levels. Albumin height, haugh unit, and egg color values were linearly (P < 0.05) increased at week 2 and 6 following the increasing NME supplementation, respectively. Additionally, quadratic (P = 0.0013) effect was observed on egg weight at week 6 with the increasing level of NME. Moreover, apparent total tract digestibility of nitrogen and excreta ammonia emission was linearly (P < 0.05) affected increasing NME supplementation. In summary, inclusion of NME containing xylanase, ß-glucanase, galactosidase, and galactomannanase activities in corn-soybean meal-DDGS-based diets increased nitrogen digestibility, decreased excreta ammonia emission, and had no negative effects on production performance and egg quality parameters.

Background: Enzymes are crucial for all aspects of metabolic function. Digestive enzymes from natural sources have been credited with beneficial effects in the digestion and absorption of food. N-SORB is a novel KD120 multienzyme complex (MEC) of metabolically activated enzymes composed of proteases, amylases, lipases, alpha-galactosidase, and glucoamylase from natural sources. These enzymes are encapsulated in a SK713 SLP (non-GMO soy lecithin phospholipid) absorption technology (Prodosome®). Objective: This randomized, double-blind placebo-controlled investigation assessed the safety and efficacy of N-SORB KD120 MEC in healthy male and female volunteers on various parameters of the blood, immunity, body composition, physical health, and quality of life following a 90-day intervention. Methods: Forty-six male and female (mean age: 25.8 ± 12.1 years) healthy volunteers were randomly assigned to receive either N-SORB (1 mL, twice daily) or placebo for 90 consecutive days. Complete blood count, as well as blood glucose, liver enzymes, and lipid profile were assessed pre- and post-intervention. Serum cytokine levels were determined by using a Bio-Plex Pro Human Cytokine 8-plex assay and enzyme linked immunosorbent assay (ELISA). Whole body composition analysis was performed by dual-energy x-ray absorptiometry (DEXA) to determine body fat mass, lean mass, and android and gynoid fat. Body weight, blood pressure, and physical health were assessed. Changes in quality of life were examined using the World Health Organization Quality of Life-abbreviated version and sleep quality was assessed using the 24-item Pittsburgh Sleep Quality Index (PSQI) questionnaire. Adverse events were monitored before, during, and after completion of the study. Results: Of the 46 subjects enrolled, a total of 40 subjects successfully completed the study. Compared to placebo, changes in blood cell counts including hematocrit, hemoglobin, mean corpuscular volume, platelets, and lymphocytes provide evidence of some improvement. Quality of life (QOL) parameters showed a small but significant improvement in the N-SORB group. A significant increase was observed in aspartate aminotransferase level in the placebo group at the end of 90 days of treatment; however, no increase was observed in the N-SORB group. No significant changes in blood urea nitrogen, serum creatinine, alkaline phosphatase, alanine aminotransferase, and lipid profile were observed between the placebo and treatment groups before and following intervention. No adverse effects were reported. Conclusions: This randomized, double blind, placebo-controlled clinical study demonstrates that short-term intervention with N-SORB improves the QOL and PSQI in healthy volunteers and did not significantly alter cardiometabolic parameters, lipid profile, or body composition.

Pasture intake can be a major challenge for free-ranging hens. This study was conducted to examine pasture digestion and to manage its negative effects. A total of 300 ISA Brown laying hens were used to investigate the effect of time on range (T) in short-term (6 wk) and long-term (12 wk) of 2 range types (R) (gravel vs. pasture) and dietary supplements (F) (T1 = xylanase; T2 = xylanase/beta-glucanase/pectinase/protease; T3 = xylanase/benzoic acid/essential oils) on crude protein, crude fiber, acid detergent fiber, neutral detergent fiber, non-starch polysaccharides (NSP), calcium and phosphorus digestibility, pH of the crop, and ileum digesta viscosity and morphology. Hens exposed to the range for 12 wk had lower (P < 0.05) digestibility of crude protein, insoluble rhamnose, ribose, and lower ileal pH compared to hens that ranged for 6 wk. Hens ranging on pasture had lower digestibility (P < 0.05) of crude protein, acid detergent fiber, neutral detergent fiber, insoluble arabinose, and insoluble xylose, but higher digestibility (P < 0.05) of insoluble mannose and glucose compared to hens that ranged on gravel. Hens fed T2 and T3 had higher digestibility (P < 0.05) of CP, acid detergent fiber, and neutral detergent fiber compared to hens fed T1. Hens fed T2 had higher digestibility (P < 0.05) of free oligosaccharide arabinose and xylose than those fed T1 or T3 diets. A significant interaction between T × R was detected for crude fiber digestibility and villus height. Digestibility of crude fiber was reduced and villus height was increased in hens ranged on pasture for 12 wk compared to 6 wk. An interaction between R × F was observed on phosphorus and soluble NSP digestibility (P < 0.05). Hens fed T2 and T3 diets had lower digestibility of phosphorus and NSP on gravel than on pasture. In conclusion, pasture consumption impaired the digestibility of nutrients. Supplementing free-range diets with a multi-enzyme or xylanase/benzoic acid/essential oil product reduced these negative effects and increased the ileal nutrient digestibility.

Lumazine synthase (LS) is a family of enzyme involved in the penultimate step in the biosynthesis of riboflavin. Its enzymatic mechanism has been well defined, and many LS structures have been solved using X-ray crystallography or cryoelectron microscopy. LS is composed of homooligomers, which vary in size and subunit number, including pentamers, decamers, and icosahedral sixty-mers, depending on its species of origin. Research on LS has expanded beyond the initial focus on its enzymatic function to properties related to its oligomeric structure and exceptional conformational stability. These attributes of LS systems have now been repurposed for use in various biomedical fields. This review primarily focuses on the applications of LS as a flexible vaccine presentation system. Presentation of antigens on the surface of LS results in a high local concentration of antigens displayed in an ordered array. Such repetitive structures enable the cross-linking of B-cell receptors and result in strong immune responses through an avidity effect. Potential issues with the use of this system and corresponding solutions are also discussed with the objective of improved utilization of the LS system in vaccine development.

A meta-analysis was performed on eight trials, which included a total of 992 parity 1 to 8 lactating sows, to evaluate the effects of feeding xylanase which is the main enzyme activity present in the enzymatic complex (Rovabio Excel, Adisseo, France) supplement throughout lactation on the following sow performance factors: BW loss, feed intake, backfat depth, and piglet growth. Even a short period of enzyme supplementation during lactation led to a reduction in BW loss of approximately 3 kg per sow (P = 0.003). This reduction represented 1-2% of the BW of sows. This effect could be explained by an increase in feed energy intake and enhanced feed digestibility. Sows fed enzyme-supplemented diets exhibited greater DM, OM, and GE digestibilities (3.4, 3.9, and 4.2% increases, respectively; P < 0.001) than sows fed control diets. During lactation, sows lost from 19 to 25 kg of BW (i.e., approximately 10% of their BW), with a difference between parity groups (P < 0.001). Body reserve mobilization was decreased in sows fed enzyme-supplemented diets (-2.9 kg, P = 0.003), with a more pronounced effect in primiparous than multiparous sows when BW loss is expressed relative to total BW (-2.27 vs. -0.59%, respectively; P = 0.058). Enzyme supplementation also increased litter weight gain up to weaning, with a greater effect in litters from multiparous sows than those from primiparous sows (5.4 vs. 0.6 kg, respectively; P = 0.009). These results could be explained in part by the relationship between their NE intake and either variations in BW or litter weight gain (R2 = 0.51 and 0.49, respectively; P < 0.001). Finally, the meta-analysis suggests that there are differences in the partitioning of the NE intake between growth and milk production and in relation to the sow's parity or physiological status. Extra energy released by enzyme is used for one of these functions (i.e., body mobilization reduction or greater milk export for litter gain).

1. The objective of this study was to investigate the effect of range type, multi-enzyme applications, and a combination of benzoic acid (BA) and essential oils (EO) on the productive performance, organ weight and egg quality of free-range laying hens. 2. Three hundred laying hens were evaluated for the short-term (6 weeks) and long-term (12 weeks) effects of range type (G = no pasture, P = pasture) and feed additives (T1 = control; T2 = betaglucanase/pectinase/protease; T3 = BA/EO). Body weight, feed intake (FI), feed conversion ratio (FCR), egg production (EP), digestive organ weight, and egg quality (EQ) were evaluated. Data were analysed using SPSS 2.2 in a 2×2×3 factorial arrangement. 3. Hens that ranged on pasture were significantly heavier (2043 g vs. 1996 g; p < 0.001), laid heavier eggs (61.9 g vs. 60.3 g; p < 0.001) and produced darker yolk colour (4.3 vs. 7.0; p < 0.001) compared to hens ranged on gravel. Hens fed T2 were significantly heavier (2050 g) compared to hens fed T1 (2005 g) or T3 (2008 g). Organ weights (gizzard, liver and pancreas) were significantly heavier in hens ranged on pasture (16.8 g/kg BW, 22.3 g/kg BW and 1.89 g/kg BW, respectively) compared to hens ranged on gravel (14.2 g/kg BW, 21.7 g/kg BW and 1.83 g/kg BW, respectively). Over time, body weight (1970-2070 g; p < 0.001) and egg weight (59.5-62.8 g; p < 0.001) increased, FI (123-120 g; p = 0.024) was reduced and FCR (2.36-2.10; p = 0.002) improved 4. In conclusion, hens housed on pasture and fed multi-enzyme supplemented diets had significantly heavier body weight and produced heavier eggs with darker yolk colour. Pasture intake and enzyme supplementation increased digestive organ weight significantly.

The aim of this study was to evaluate the effects of two species of poultry (cockerel and Japanese quail), two levels of enzyme (0 and 0.5 g/kg), and 6 processing methods (control (C), gamma irradiation 25 (GI25), and 50 kGy (GI50), autoclaving (AU), fermentation without lactobacillus (F), and fermentation with lactobacillus (FL)) on the nitrogen-corrected apparent metabolizable energy (AMEn) of barley. In this experiment, each processing method was tested at two levels of enzyme (0 and 0.5 g/kg) in six replications with two cockerels or four quails per replication. The results showed that the AMEn values by barley processing methods were: C-11.45 and 12.27; GI25-11.50 and 11.30; GI50-11.49 and 11.29; AU-11.70 and 11.46; F-12.90 and 12.30; FL-1295 and 12.51 MJ/kg in cockerels and quails, respectively. The AMEn (11.49 MJ/kg) of enzyme 0 was significantly lower than that of enzyme 0.5 (12.35 MJ/kg). There was a significant difference in the AMEn of processed barley between cockerels and quails. In conclusion, the most effective processing method of barley for AMEn is fermentation with lactobacillus and enzyme.

Even though erythrocytes transport both oxygen and carbon dioxide, research on blood substitutes has concentrated on the transport of oxygen and its vasoactivity and oxidative effects. Recent study in a hemorrhagic shock animal model shows that the degree of tissue PCO(2) elevation is directly related to mortality rates. We therefore prepared a novel nanobiotechnological carrier for both O(2) and CO(2) with enhanced antioxidant properties. This is based on the use of glutaraldehyde to crosslink stroma free hemoglobin (SFHb), superoxide dismutase (SOD), catalase (CAT) and carbonic anhydrase (CA) to form a soluble PolySFHb-SOD-CAT-CA. It was compared to blood and different resuscitation fluids on the ability to lower elevated tissue PCO(2) in a 2/3 blood volume loss rat hemorrhagic shock model. Sixty minutes of sustained hemorrhagic shock at 30 mm Hg resulted in the increase of tissue PCO(2) to 95 mm ± 3 mmHg from the control level of 55 mm Hg. Reinfusion of whole blood (Hb 15 g/dL with its RBC enzymes) lowered the tissue PCO2 to 72 ± 4.5 mmHg 60 minutes after reinfusion. PolySFHb-SOD-CAT-CA (SFHb 10 g/dL plus additional enzymes) was more effective than whole blood in lowering PCO(2) lowering this to 66.2 ± 3.5 mmHg. Ringer's Lactated solution or polyhemoglobin lowered the elevated PCO2 only slightly to 87 ± 4.5 mmHg and 84.8 ± 1.5 mmHg, respectively. Moreover, ST-elevation for whole blood (Hb 15 g/dL) and PolySFHb-SOD-CAT-CA (Hb 10 g/dL) was respectively 12.8% ± 4% and 13.0% ± 2% of the control 60 minutes after reinfusion. Both are significantly better than those in the Ringer's lactated group and the PolyHb group. In conclusion, this novel approach for blood substitute design has resulted in a novel nanobiotechnological carrier for both O(2) and CO(2) with enhanced antioxidant properties.

A total of 200 (Large White × Landrace) sows were used in a 39-day study to evaluate the effects of feeding a non-starch polysaccharide (NSP)-hydrolysing enzyme multicomplex (Rovabio(®) Excel) in conjunction with a high- or reduced nutrient-density diet during lactation on sow body condition, feed intake and progeny performance. Eight sows were selected each week for 25 weeks, blocked by parity and BW into groups of four, and within the block randomly assigned to one of the four treatments (n = 50/treatment). Treatments were: (1) LND: low energy (13.14 MJ of DE/kg), low CP (15%) diet; (2) LND + RE: LND with 50 mg/kg NSP-hydrolysing enzyme; (3) HND: high energy (14.5 MJ of DE/kg), high CP (16.5%) diet; and (4) HND + RE: HND with 50 mg/kg NSP-hydrolysing enzyme. Sows were fed treatment diets from day 109 of gestation until the day of subsequent service. Between weaning and re-service, Rovabio(®) Excel addition to LND diets resulted in an increase in energy intake; however, a reduction was observed when supplemented to the HND diet (P < 0.05). The inclusion of Rovabio(®) Excel increased feed and energy intake during week 3 (days 15 to 21) of lactation (P < 0.05). Sows fed diets supplemented with Rovabio(®) Excel had greater back-fat depth at weaning and service (P < 0.05); however, the magnitude of change in back-fat depth during lactation and from farrowing to service was not different between treatments. Feeding the HND diet increased energy intake before farrowing, throughout lactation and during the weaning to service interval (P < 0.01); however, overall, average daily feed intake tended to be reduced (P < 0.10). At service, sows fed the HND diet were heavier than sows fed the LND diet (P < 0.05); however, the magnitude of change in BW between treatments was not different. Feeding the HND diet to sows resulted in a tendency for heavier piglets at birth (P = 0.10) that tended to grow at a faster rate and be heavier at weaning than piglets from sows fed the LND diet (P = 0.06). These results indicate that NSP-degrading enzymes offer minimal benefit to sows and their progeny when fed before and during lactation; however, increasing energy intake of sows during lactation may beneficially affect progeny.

Currently available rapid diagnostic tests (RDTs) for malaria show large variation in sensitivity and specificity, and there are concerns about their stability under field conditions. To improve current RDTs, monoclonal antibodies (mAbs) for novel malaria antigens have been developed and screened for their possible use in new diagnostic tests. Three antigens, glutamate rich protein (GLURP), dihydrofolate reductase-thymidylate synthase (DHFR-TS) and heme detoxification protein (HDP), were selected based on literature searches. Recombinant antigens were produced and used to immunize mice. Antibody-producing cell lines were subsequently selected and the resulting antibodies were screened for specificity against Plasmodium falciparum and Plasmodium vivax. The most optimal antibody couples were selected based on antibody affinity (expressed as dissociation constants, KD) and detection limit of crude antigen extract from P. falciparum 3D7 culture. The highest affinity antibodies have KD values of 0.10 nM ± 0.014 (D5) and 0.068 ± 0.015 nM (D6) for DHFR-TS mAbs, 0.10 ± 0.022 nM (H16) and 0.21 ± 0.022 nM (H18) for HDP mAbs and 0.11 ± 0.028 nM (G23) and 0.33 ± 0.093 nM (G22) for GLURP mAbs. The newly developed antibodies performed at least as well as commercially available histidine rich protein antibodies (KD of 0.16 ± 0.13 nM for PTL3 and 1.0 ± 0.049 nM for C1-13), making them promising reagents for further test development.

The DE values and digestible nutrients content of 6 diets were measured in 60-kg male growing pigs fed restricted amount of feed. Diets were prepared from 5 ingredients [wheat (Triticum aestivum), corn (Zea mays), barley (Hordeum vulgare), wheat bran, and soybean (Glycine max) meal; inclusion levels of ingredients were not correlated] with or without carbohydrose enzyme (Rovabio Excel AP; 3300 endo-ß-1,4-xylanase visco units and 300 endo-1,3(4)-ß-glucanase units/kg of feed; 150 g/t of feed) according to a 6 × 2 factorial arrangement; dietary NDF ranged from 10.6 to 20.1% of DM. Pigs (5 per treatment) were placed in metabolism cages that allowed total collections of feces and urine for 10 d after a 11-d adaptation. Samples of feed, urine, and feces were analyzed for GE, ash, and N. Digestibility of GE, N, and OM were calculated. The effects of diet and enzyme (Enz) were evaluated by ANOVA. In addition, the DE and digestible nutrient contents of ingredients were calculated by regression of nutritive values of diets on level of ingredient inclusions. Apparent total tract digestibility of OM, N, and GE of diets were associated with dietary NDF content (r = -0.97; P < 0.001) and were increased (P < 0.05) by Enz addition by 0.4, 1.6, and 0.5%-units (a difference between two percentage values) for OM, N, and GE digestibility, respectively. Improvement in DE value due to Enz averaged 0.09 MJ/kg DM (15.11 vs. 15.02 MJ/kg DM; P < 0.05). The ADG (891 vs. 850 g/d; P < 0.05) was also increased by Enz addition. The calculated DE content without Enz addition averaged 16.3, 16.4, 14.9, 10.5, and 17.2 MJ/kg DM for wheat, corn, barley, wheat bran, and soybean meal, respectively. The Enz addition increased the DE value of ingredients similarly, but the best response was observed for wheat (0.33 MJ/kg DM).

RESUMEN

This report describes a technique of inserting an implantable venous access port (portacath) through a thrombosed and occluded vein employing a pre-existing peripherally inserted central catheter (PICC) as the route of access. The PICC was used as a conduit for venous access in a way that has not been described previously in the literature. This procedure was performed in a young patient with cystic fibrosis in an effort to prevent the use of his virgin contralateral veins, which might be used in the future.

Distal myopathy with rimmed vacuoles (DMRV), also called hereditary inclusion body myopathy, is an autosomal recessive disorder caused by homozygous or compound heterozygous missense mutations in GNE which encodes a protein with two enzymatic activities in sialic acid biosynthesis: UDP-GlcNAc 2-epimerase and ManNAc kinase. The disease starts from 1540 years and is slowly progressive. DMRV preferentially affects tibialis anterior and hamstrings muscles, and has characteristic findings in muscle pathology which include rimmed vacuoles, tubulofilamentous inclusions, deposition of amyloid, and phosphorylated tau. We generated DMRV mice (Gne -/- hGNE D176V-Tg) by crossmating Gne knock-out heterozygous mouse and human GNE p.D176V transgenic mouse. This model mouse recapitulates DMRV clinically, pathologically, and biochemically by developing muscle weakness and atrophy from 21 weeks, amyloid deposition from 31 weeks, and rimmed vacuoles and phosphorylated tau from 41 weeks while having lifelong hyposialylation. We gave three types of GNE metabolites, ManNAc, NeuAc and sialyllactose, to DMRV mice orally from 15 weeks until 55 weeks of age. Sialic acid supplementation almost completely precluded the disease and virtually no sign of DMRV was seen even at 55 weeks of age, indicating that decreased sialic acid is the cause of myopathic phenotype and sialic acid supplementation can prevent the disease process.

Autotaxin, also known as NPP2 (nucleotide pyrophosphatase/phosphodiesterase 2), is a secreted lysophospholipase-D that generates lysophosphatidic acid and thereby promotes the metastatic and invasive properties of tumor cell as well as angiogenesis. We show here that, in mice, NPP2 is cleared from the circulation within minutes and is retained by the liver sinusoidal endothelial cells (LSECs). The binding of NPP2 to isolated LSECs resulted in its degradation and could be competed for with ligands of the scavenger receptor family. Our finding that circulating NPP2 has a rapid turnover has important implications for its development as an anti-cancer target.

1. Exogenous microbial beta-1,3-1,4-glucanases and hemicellulases contribute to improving the nutritive value of cereals rich in soluble non-starch polysaccharides for poultry. 2. In general, plant cell wall hydrolases display a modular structure comprising a catalytic module linked to one or more non-catalytic carbohydrate-binding modules (CBMs). Based on primary structure similarity, CBMs have been classified in 50 different families. CBMs anchor cellulases and hemicellulases into their target substrates, therefore eliciting efficient hydrolysis of recalcitrant polysaccharides. 3. A study was undertaken to investigate the effects of a family 11 beta-glucan-binding domain in the function of recombinant derivatives of cellulase CtLic26A-Cel5E of Clostridium thermocellum that were used to supplement a barley-based diet at lower dosage rates. 4. The results showed that birds fed on diets supplemented with the recombinant CtLic26A-Cel5E modular derivative containing the family 11 CBM or the commercial enzyme mixture Rovabio Excel AP tended to display improved performance when compared to birds fed diets not supplemented with exogenous enzymes. 5. It is suggested that at lower than previously reported enzyme dosage (10 U/kg vs 30 U/kg of basal diet), the beta-glucan-binding domain also elicits the function of the recombinant CtLic26A-Cel5E derivatives. 6. Finally, the data suggest that exogenous enzymes added to barley-based diets act primarily in the proximal section of the gastrointestinal tract.

The objective of this study was to determine the functional location and disappearance of activity of a supplemental Escherichia coli AppA2 phytase and its impact on digesta P and Ca concentrations in the gastrointestinal tract of pigs. In Exp. 1, 18 pigs (8.3 +/- 0.2 kg of BW) were allotted to 3 groups (n = 6 each) and fed a low-P (0.4%) corn-soybean meal, basal diet (BD), BD + phytase [500 units (U)/kg of feed], or BD + inorganic P (iP, 0.1%) for 4 wk. In Exp. 2, 30 pigs (14.5 +/- 0.2 kg of BW) were allotted to 3 groups (n = 10 each) and fed BD, BD + 500 U of phytase/kg of feed, or BD + 2,000 U of phytase/kg of feed for 2 wk. Five or six pigs from each treatment group were killed at the end of both experiments to assay for digesta phytase activity and soluble P concentration in 6 segments of the digestive tract and digesta total P and Ca concentrations in stomach and colon. Compared with pigs fed BD, pigs fed BD + 500 U of phytase/kg of feed in Exp. 1 and BD + 2,000 U of phytase/kg of feed in Exp. 2 had greater (P < 0.05) phytase activities in the digesta of the stomach and upper jejunum (2 m aborally from the duodenum). No phytase activity was detected in the digesta of the lower jejunum (2.12 m cranial to the ileocecal junction) or ileum from any of the treatment groups in either trial. Concentrations of digesta-soluble P peaked in the upper jejunum of pigs fed BD in Exp. 1 and 2, but showed gradual decreases between the stomach and the upper jejunum of pigs fed BD + phytase or BD + iP. In both experiments, pigs fed only BD had greater (P < 0.05) colonic digesta phytase activity and soluble P concentrations than those fed phytase. In Exp. 2, total colonic digesta P or Ca concentrations, or both, of pigs displayed a phytase-dose-dependent reduction (P < 0.05). In conclusion, supplemental dietary AppA2 mainly functioned in the stomach and was associated with a reduced phytase activity in colonic digesta of weanling pigs.
