RESUMEN

Circulating extracellular vesicles (EVs) have emerged as a valuable source of cancer biomarkers. However, the high degree of EV heterogeneity and the complexity of clinical samples pose a challenge in the sensitive identification of tumor-derived EVs. Here we introduce a one-step thermophoretic AND gate operation (Tango) assay that integrates polyethylene glycol (PEG)-enhanced thermophoretic accumulation of EVs and simultaneous AND gate operation on EV membranes by dual-aptamers recognition. By using the Tango assay to detect tumor-derived EVs with co-presence of EpCAM and PSMA directly from serum in a homogeneous, separation-free format, we can discriminate prostate cancer (PCa) patients from benign prostatic hyperplasia (BPH) patients in the diagnostic gray zone with an accuracy of 91 % in 15 min. Our approach streamlines EV enrichment and AND gate operation on EVs in a single assay, providing a rapid, straightforward, and powerful method for precise and non-invasive diagnosis of cancer.

RESUMEN

A liquid plano-convex lens with focal length tuning is proposed, which is formed by sinking an oil droplet onto the bottom of an elastomer. A simple and low-cost fabrication method is presented. The lens aperture and initial focal length can be controlled during the fabrication. Furthermore, focal length tuning is demonstrated. The lens made of a 40 mg oil droplet can achieve the tuning range from 12 to 17 mm. The effective aperture of the lens is about 2.8 mm. In the demonstration of an imaging system, the lens assists in focusing and a clear image can be observed.

Herpetrione(HPE) is an effective compound that has been used in the treatment of liver diseases. To improve its dissolution and absorption, herpetrione nanosuspensions was prepared. Nanosuspensions were proved to achieve intact absorption in vivo. However, the transport mechanisms are not fully understood, especially lack of direct evidence of translocation of particulates. In this study, an environment-responsive dye, P4, was loaded into herpetrione amorphous nanoparticles (HPE-ANPs) to elucidate the absorption and transport mechanism of the nanoparticles. And the amount of HPE and nanoparticles in the samples were quantified using HPLC/LC-MS/MS and IVIS with the model of Caco-2 and Caco-2/HT29-MTX. Results demonstrated that HPE is mainly taken up by passive diffusion in the form of free drugs, while HPE-ANPs are internalized by an energy dependent active transport pathway or intracellular endocytosis. It is speculated that HPE-ANPs may change the original entry pathway of drug molecules. Furthermore, the presence of mucus layer and the use of HPMC E15 may contribute to drug absorption to some extent. Transcellular transport study indicates that HPE-ANPs has a poor absorption. In conclusion, the differences in the absorption behavior trends of HPE-ANPs are caused by the difference in particle properties and the form of existence of the drug.

Recently, electromagnetic radiation is a serious threat to equipment accuracy, military safety and human health. The combination with different materials to fabricate absorber composites with well-designed morphology is expected to ameliorate this issue. In here, CuS/Fe3O4@polypyrrole (CuS/Fe3O4@PPy) flower-like composites are constructed by the combination of hydrothermal method, solvothermal method and in-situ polymerization. CuS with flower-like structure consisting of nanosheets can provide a conductive backbone and large specific surface area. Hollow Fe3O4 microspheres play a key role in deciding magnetic loss, and electromagnetic waves can penetrate their hollow structure, result in multiple reflection and refraction. PPy coating can enhance the combined strength of composite, and effectively consume microwaves by scattering and multiple refraction in the intercalated structure. As expected, the minimum reflection loss (RLmin) of CuS/Fe3O4@PPy composites is -74.12 dB at 8.16 GHz with a thickness of 2.96 mm, and the effective absorption bandwidth (EAB) is 4.6 GHz (13.4-18.0 GHz) at 1.68 mm. The excellent electromagnetic wave absorption performances are attributed to the synergy effect of different components. This work provides a unique strategy for the structural design of flower-like microspheres in the field of electromagnetic wave absorption.

Efficient delivery of chemotherapeutic agents into tumor cells and reversal of chemoresistance are crucially important to enhance cancer therapy. We fabricated pH/redox dual responsive nanocarriers based on cell penetrating peptides (TAT) functionalized TPGS (cTAT-TPGS) and polypeptide (PEG-b-poly(aspartic-lipoic acid), PPAL) to reduce the permanent drug release and overcome multidrug resistance. TAT was used to functionalize TPGS and shielded by pH-responsive fatty acids, and polypeptides with lipoic acid side chains (PPAL) were synthesized. Reversibly crosslinked hybrid micelles (RCMs) were fabricated based on cTAT-TPGS and PPAL. RCMs nanocarriers exhibited acid-responsive charge reversal and redox-responsive drug release. The in vitro results showed that the RCMs could be efficiently internalized by the MCF-7/ADR cells in an acidic microenvironment and inhibited the DOX efflux, causing a higher cytotoxicity than non-crosslinked nanocarriers. Furthermore, the dual-responsive structure effectively prolonged the circulation time of RCM nanocarriers and achieved a high level of accumulation in cancer cells in vivo, leading to much more effective inhibition of tumor growth. The DOX-loaded RCMs also showed excellent biosafety, especially for the myocardium tissue. This novel strategy provided an effective platform for drug target delivery and reversal of MDR.

Lipomas are very common benign adipose tissue tumors that most often develop in subcutaneous tissue. A limited number of studies have described the characteristics of parosteal ossifying lipomas located in the fibula, cervical vertebrae and mandible. However, to the best of our knowledge, parosteal ossifying lipoma of the clavicle has not yet been reported. We herein describe the clinical symptoms, radiological and histological findings in a rare case of parosteal ossifying lipoma of the clavicle in a 40-year-old male patient. The characteristic histological appearance, together with computed tomography and/or magnetic resonance imaging characteristics, should aid in the accurate diagnosis of such cases.
