RESUMEN

A cationic conjugated polyelectrolyte was designed and synthesized based on poly(fluorene-co-phenylene) containing 5 mol% benzothiadiazole (BT) as a low energy trap and 15-crown-5 as a recognizing group for potassium ions. A potassium ion can form a sandwich-type 2:1 Lewis acid-based complex with 15-crown-5, to cause the intermolecular aggregation of polymers. This facilitates inter-chain fluorescence resonance energy transfer (FRET) to a low-energy BT segment, resulting in fluorescent signal amplification, even at dilute analyte concentrations. Highly sensitive and selective detection of K(+) ions was demonstrated in water. The linear response of ratiometric fluorescent signal as a function of [K(+) ] allows K(+) quantification in a range of nanomolar concentrations with a detection limit of ≈0.7 × 10(-9) M.

RESUMEN

This study concentrates on the potential application of conjugated polyelectrolytes (CPEs) to cell imaging and DNA delivery. Four different types of polyfluorene copolymers, namely, PAHFP-Br, PAEFP-Br, PAHFbT-Br, and PSBFP-Na, which have the same π-conjugated backbone but different side chains, were synthesized. For cytotoxicity testing, L-929 fibroblastic cells were treated with increasing concentrations (0-50 µM) of each CPE and then cell viability was determined by WST-8 assay. Cellular uptake of CPEs into cultured L-929 cells was observed by fluorescence microscopy. To examine DNA delivery by CPEs, the cells were incubated for 1 H with PAHFP-Br/fluorescein (Fl)-labeled single-stranded DNA (ssDNA-Fl) complex and then visualized by fluorescence microscopy. Cytotoxicity of CPEs was increased in a dose-dependent manner but at lower than 10 µM, PAHFP-Br, PAEFP-Br, and PSBFP-Na did not show any cytotoxic effects on the cells. When added to cell cultures at 1 µM, PAHFP-Br/ssDNA-Fl complex was delivered and then dissociated into PAHFP-Br and ssDNA-Fl within the cells. This result implies that PAHFP-Br can enable cell imaging and DNA delivery into fibroblastic cells. Therefore, it is suggested that PAHFP-Br with various advantages such as low cytotoxicity and high fluorescence efficiency can be extensively used as a potential agent for cell imaging and gene delivery.

RESUMEN

Resonance Raman spectroscopy is an efficient tool for multiplex imaging because of the narrow bandwidth of the electronically enhanced vibrational signals. However, Raman signals are often overwhelmed by concurrent fluorescence. In this study, we synthesized a series of truxene-based conjugated Raman probes to show structure-specific Raman fingerprint patterns with a common 532 nm light source. The subsequent polymer dot (Pdot) formation of the Raman probes efficiently suppressed fluorescence via aggregation-induced quenching and improved the dispersion stability of particles without leakage of Raman probes or particle agglomeration for more than 1 year. Additionally, the Raman signal amplified by electronic resonance and increased probe concentration exhibited over 103 times higher relative Raman intensities versus 5-ethynyl-2'-deoxyuridine, enabling successful Raman imaging. Finally, multiplex Raman mapping was demonstrated with a single 532 nm laser using six Raman-active and biocompatible Pdots as barcodes for live cells. Resonant Raman-active Pdots may suggest a simple, robust, and efficient way for multiplex Raman imaging using a standard Raman spectrometer, suggesting the broad applicability of our strategy.

A low-density polyethylene (LDPE) degrading bacterial strain (ATKU1) was isolated (99.86% similar with Bacillus siamensis KCTC 13613T) from a plastic dumping site to study interactions between microplastics (< 5 mm) and microorganisms. The strain was found (by scanning electron microscopy) to form biofilm on the microplastic surface after its interaction with LDPE (avg. Mw~4,000 Da and avg. Mn~1,700 Da) as a sole carbon source. Atomic force microscopy (AFM) showed the biofilm's 3-D developmental patterns and significantly increased Young's modulus of the LDPE surface after microbial treatment. Most of the viable bacteria attached to biofilms rather than media, which suggested their ability to utilize LDPE. Absorption bands of carbonyl, alkenyl, acyl, ester, primary-secondary alcohol, alkene groups and nitric oxides were found on the treated LDPE particles using Fourier-transform infrared spectroscopy. Fourier transform-ion cyclotron resonance mass spectrometry of the media indicated compositional shifts of the compounds after treatment (i.e., increase in the degree of unsaturation and increment in oxygen-to-carbon ratio) and presence of unsaturated hydrocarbons, polyketides, terpenoids, aliphatic/peptides, dicarboxylic acids, lipid-like compounds were hinted. The plastic degrading abilities of Bacillus siamensis ATKU1 suggest its probable application for large scale plastic bioremediation facility.

RESUMEN

The use of novel oral anticoagulants (NOACs) has increased in recent times in an effort to overcome the shortcomings of warfarin. They are being used primarily for the prevention of thrombosis caused by atrial fibrillation and offer the advantages of having fewer drug interactions than warfarin, no dietary restrictions, and no requirement for regular blood tests. Although there is reportedly less postoperative bleeding even if the drug is not discontinued during procedures that can cause local bleeding, such as dental procedures, no well-designed clinical studies have assessed postoperative bleeding associated with the use of these drugs. This article reports a case of a 74-year-old male patient who was taking rivaroxaban. The patient underwent a dental implant procedure after discontinuing rivaroxaban for one day and subsequently suffered delayed bleeding on postoperative day 6. Accordingly, this article also reports that the use of NOACs may also lead to delayed bleeding.

BACKGROUND: Osteoclasts are the only cell type capable of breaking down bone matrix, and its excessive activation is responsible for the development of bone-destructive diseases. Euphorbia lathyris L. (ELL) is an herbal plant that belongs to the Euphorbiaceae family. This study investigated the effects of the methanol extract of the aerial part of ELL on receptor activator of nuclear factor-kappa B ligand (RANKL)-induced osteoclast formation and signaling pathways. METHODS: Osteoclasts were formed by co-culturing mouse bone marrow with osteoblasts or by culturing mouse bone marrow-derived macrophages (BMMs) with macrophage colony-stimulating factor (M-CSF) and RANKL. Bone resorption assays were performed using dentine slices. The expression level of mRNA was analyzed by real-time polymerase chain reaction (PCR) or reverse transcription (RT)-PCR. Western blotting assays were performed to detect the expression or activation level of proteins. RESULTS: ELL inhibited RANKL-induced osteoclast formation without cytotoxicity. Furthermore, the RANKL-stimulated bone resorption was diminished by ELL. Mechanistically, ELL blocked the RANKL-triggered p38 mitogen-activated protein kinase (MAPK) phosphorylation, which resulted in the suppression of the expression of c-Fos and nuclear factor of activated T cells (NFATc1). In osteoblasts, ELL had little effect on the mRNA expression of RANKL and osteoprotegerin (OPG). CONCLUSIONS: The present data suggest that ELL has an inhibitory effect on osteoclast differentiation and function via downregulation of the p38/c-Fos/NFATc1 signaling pathways. Thus, ELL could be useful for the treatment of bone diseases associated with excessive bone resorption.
