RESUMEN

Multiple and two-way reversible shape memory polymers (M/2W-SMPs) are highly promising for many fields due to large deformation, lightweight, strong recovery stress, and fast response rates. Herein, a semi-crystalline block poly(urethane-urea-amide) elastomers (PUUAs) are prepared by the copolymerization of isocyanate-terminated polyurethane (OPU) and amino-terminated oligomeric polyamide-1212 (OPA). PUUAs, composed of OPA as stationary phase and PTMEG as reversible phase, exhibit excellent rigidity, flexibility, and resilience, and cPUUA-C7 -S25 exhibits the best tensile property with strength of 10.3 MPa and elongation at break of 360.2%. Besides, all the PUUAs possess two crystallization/melting temperatures and a glass transition temperature, which endow PUUAs with multiple and reversible two-way shape memory effect (M/2W-SME). Physically crosslinked PUUA-C0 -S25 exhibits excellent dual and triple shape memory, and micro chemically crosslinked cPUUA-C7 -S25 further shows quadruple shape memory behavior. Additionally, both PUUA-C0 -S25 and cPUUA-C7 -S25 have 2W-SME. Intriguingly, cPUUA-C7 -S25 can achieve a higher temperature (up to 165 °C) SME, which makes it suitable for more complex and changeable applications. Based on the advantages of M/2W-SME, a temperature-responsive application scenario where PUUAs can transform spontaneously among different shapes is designed. These unique M/2W-SME and high-temperature SME will enable the applications of high-temperature sensors, actuators, and aerospace equipment.

AIM: To investigate the pharmacodynamic and pharmacokinetic parameters of pegylated liposomal doxorubicin (PLD) combined with cyclophosphamide, vincristine, and prednisolone in patients with peripheral T-cell lymphomas (PTCL). METHODS: Seven chemonaive patients and four patients with relapsed peripheral T-cell lymphomas were treated with a CCOP regimen consisting of an intravenous administration of cyclophosphamide (750 mg/m(2)), vincristine (1.4 mg/m(2)), and PLD (30 mg/m(2)) on d 1, as well as an oral administration of prednisolone (60 mg/m(2)) on d 1-5. This regimen was repeated every 3 weeks for six cycles, and the clinical response and toxicity of the regimen were monitored. In addition, the plasma concentration of PLD at different time points was determined before and after treatment. The pharmacokinetics (PKs) software was used to estimate the pharmacokinetic parameters of PLD. RESULTS: The 11 PTCL patients received 35 treatment cycles. Three of them achieved complete response (CR), two partial response (PR), four stable disease (SD), and two progressive disease (PD). The overall response rate (ORR) was 45.5%, and the CR rate was 27.3%. In the 7 chemonaive patients, three achieved CR, two PR, one SD, and one PD. The ORR was 71.4%, and CR rate was 42.9%. The median follow-up time was 15 months, but 6 out of 11 patients were dead at the time of data analysis. The 1-year overall survival rate was 45.5%, and the median progression-free survival (PFS) rate was 6.5 [95% confidence interval (95% CI) 3.17-19.02] with a survival rate of 11.5 months (95% CI 6.65-16.36). The main toxicity was myelosuppression. Oral mucositis and hand-foot syndrome seldom occurred. The PLD plasma concentration from nine patients ranged from 1.7036 to 9.2207 mg·L(-1) after administration of the CCOP regimen (0-168 h). The pharmacokinetic parameters AUC(0-∞), CL, t(1/2), and V(d) were 910.76 mg/L·h, 0.043 L·h(-1)·m(-2), 68.40 h, and 3.56 L/m(2), respectively. CONCLUSION: The CCOP regimen was effective and well tolerated in patients with peripheral T-cell lymphomas. The results of the pharmacokinetic parameters showed that PLD had long retention time in blood circulation.

OBJECTIVE: To explore the advantages of acupuncture treatment guided by channel palpation on stroke-sequel patients. METHODS: This research was randomized, traditional acupuncture controlled trial using channel palpation acupuncture to treat stroke-sequel patients. Totally 148 patients who were randomly assigned to two experimental groups. The treatment group, i.e, the channel palpation group was treated with Dr. Wang Juyi's Channel Palpation. Patients in control group received acupuncture according to New Century Acupuncture. Every patient was needled at Renzhong (GV 26), Baihui (GV 20), Neiguan (PC 6), Jiquan (HT 1), Chize (LU 5), Weizhong (BL 40), Sanyinjiao (SP 6), Zusanli (ST 36), and each acupuncture treatment was modified according different syndrome differentiations including liver yang rising [Taichong (LR 3), Taixi (KI 3)], wind-phlegm blocking collaterals [Fenglong (ST 40), Hegu (LI 4)], phlegm-heat occupying in the Fu-organs [Quchi (LI 11), Neiting (ST 44), Fenglong (ST 40)], Qi deficiency with blood stasis [Qihai (CV 6), Xuehai (SP 10)], Yin Deficiency with wind [Taixi (KI 3), Fengchi (GB 20)]; for wry mouth, add Jiache (ST 6), Dicang (ST 4); for paralyzed arms, add Jianyu (LI 15), Quchi (LI 11), Shousanli (LI 10) and Hegu (LI 4), for paralyzed legs, add Huantiao (GB 30), Yinlingquan (SP 9) and Fengshi (GB 31). The duration of each treatment was 6 weeks. Then the Fugl-Meyer score, the Stroke Specific Quality of Life scale (SS-QOL), and the National Institute of Health Stroke Scale (NIHSS) were assessed before treatments, after 6 and 12 weeks of treatments to evaluate the acupuncture effect in each group. The data were collected and analyzed after the completion of treatment by SPSS 17 using paired sample t-test. RESULTS: Totally 148 participants were recruited, and 136 eligible patients were included in this study. The results showed that for FMA motor function and Fugl-Meyer balance function and NIHSS, there is no statistic difference between two groups at the baseline period, after 6-week treatment and after 12-week follow-up (P > 0.05); However, for NIHSS and SS-QOL, there is no statistic difference between two groups at the baseline period and 6 weeks after treatment (P > 0.05). However, statistical difference starts to appear after 12-week (P = 0.028, 0.037 < 0.05). CONCLUSION: We conclude that as for improving the nerve function and the quality of life, Dr. Wang Juyi's Applied Channel Theory presents a better clinical result.

Cancer vaccines mostly aim to induce cytotoxic T lymphocytes (CTLs) against tumors. An appropriate adjuvant is of fundamental importance for inducing cellular immune response. Since the antigen in particulate form is substantially more immunogenic than soluble form antigen, it is beneficial to interact with antigen-presenting cells membrane to induce robust CD8+ T cell activation following vaccination. Based on previous research, we designed an adjuvant formulation by combining Astragalus saponins, cholesterol, and liposome to incorporate antigen into a particulate delivery system, so as to enhance cellular immune response. Meanwhile, angiogenesis contributes to tumor growth and metastasis, and basic fibroblast growth factor (bFGF) is involved in tumor angiogenesis. Therefore, using lipo-saponins adjuvant formulation and a human recombinant bFGF antigen protein, we tried to induce bFGF-specific CTL response to inhibit tumor angiogenesis to achieve antitumor activity. After five immunizations, the lipo-saponins/bFGF complex elicited robust antibody response and markedly higher amount of interferon-γ in BALB/c mice, resulting in superior antitumor activities. Decreased microvessel density in CD31 immunohistochemistry and the lysis of vascular endothelial cells by the T lymphocytes from the immunized mice indicated that the immunity inhibited the angiogenesis of tumors and further led to the inhibition of tumors. Our data suggest that the approach to construct adjuvant formulation between liposome and Astragalus saponins appeared highly desirable, and that Astragalus saponins may be utilized as a valuable additive for enhancing the effectiveness of vaccines and stimulating an appropriate immune response that can benefit tumor therapy.
