RESUMEN

In recent years, polymers with stimuli-responsive properties have been increasingly reported on due to their diverse applications. However, most of the studies have only focused on the performance of polymers under specific scenarios. The laws of changes in the properties in response to various external stimuli have been less systematically and quantitatively studied. In this paper, we prepared an amphiphilic polymer (PadaMX and PAdaM3QA-X) with temperature-, pH-, ion-, and ß-cyclodextrin (ß-CD)-responsive properties. According to the cloud point tested by the UV-Vis method, the lower critical soluble temperature (LCST) of PAdaM3QA-10% was more sensitive to a change in pH and less sensitive to a change in ions compared with PadaM3 due to quaternized side chains with a stronger intramolecular mutual repulsion. We then fabricated the coatings with responsive properties by immobilizing the adamantyl groups on ß-CD-modified surfaces. The hydrophilicity of the coatings was improved after quaternization, as proven by the water contact angle (WCA) measurement. The antifouling and antibacterial performance was further evaluated via the fluorescence intensity of bovine serum albumin (BSA) adsorbed on the surfaces and the spread plate method. A 78.4% BSA desorption rate and a 96.8% sterilization rate were achieved by the PAdaM3QA-10% coating. In summary, this work prepared a multiple-stimuli-responsive amphiphilic copolymer for antifouling and antibacterial functionality via a "resistance-kill-release" mechanism.

BACKGROUND: Cimicifuga racemosa is one of the herbs used for the treatment of climacteric syndrome, and it has been cited as an alternative therapy to estrogen. Apart from hectic fevers, dyspareunia and so on, dry mouth also increase significantly after menopause. It has not yet been reported whether C. racemosa has any impact on the sublingual gland, which may relate to dry mouth. In an attempt to determine this, we have compared the effects of estrogen and C. racemosa on the sublingual gland of ovariectomized rats. RESULTS: HE staining showed that the acinar cell area had contracted and that the intercellular spaces were broadened in the OVX (ovariectomized rats) group, while treatment with estradiol (E2) and iCR (isopropanolic extract of C. racemosa) improved these lesions. Transmission electron microscopy showed that rough endoplasmic reticulum expansion in mucous and serous acinar epithelial cells and apoptotic cells was more commonly seen in the OVX group than in the SHAM (sham-operated rats) group. Mitochondria and plasma membrane infolding lesions in the striated ducts were also observed. These lesions were alleviated by both treatments. It is of note that, in the OVX + iCR group, the volume of mitochondria in the striated duct was larger than in other groups. Immunohistochemical staining showed that the ratio of caspase-3 positive cells was significantly increased in the acinar cells of the OVX group compared with the SHAM group (p < 0.05); and the MA (mean absorbance) of caspase-3 in the striated ducts also increased (p < 0.05). Estradiol decreased the ratio of caspase-3 positive cells and the MA of caspase-3 in striated ducts significantly (p < 0.05). ICR also reduced the ratio of caspase-3 positive cells and the MA in the striated ducts (p < 0.05), but the reduction of the MA in striated ducts was inferior to that of the OVX + E2 group (p < 0.05). CONCLUSION: Both estradiol and iCR can inhibit subcellular structural damage, and down-regulate the expression of caspase-3 caused by ovariectomy, but their effects were not identical, suggesting that both drugs confer a protective effect on the sublingual gland of ovariectomized rats, but that the specific location and mechanism of action producing these effects were different.

Neurite alignment and elongation play special roles in the treatment of neuron disease, design of tissue engineering implants, and bioelectrodes applications. For instance, the trigeminal neurons (TGNs) free nerve endings are a key component of the pulp-dentin complex. The reinnervation of the pulp canal space requires the recruitment of apically positioned free nerve endings through axonal guidance. Many studies have been carried to develop patterned two-dimensional substrates or three-dimensional scaffolds with aligned topographical structures to guide axonal growth. However, most of the strategies are either complicated/inconvenient in process or time-/cost-sacrifice. One-step dimensionally confined hydrothermal (DCH) technique has been considered an effective and facile approach to fabricate reduced graphene oxide fibers (rGOFs), and the rGOFs have shown significant potential in regulating neural stem cells differentiation toward neurons. Here, inspired by the relationship between the lateral size of GO nanosheets and the electrical conductivity of GO films made from GO sheets as a building block, we fabricated surface conductivity and topography-controlled rGOFs based on the DCH method. Well "self-patterned" directional channel structure of rGOF showed outstanding ability to improve the neurofilament alignment and migration, with the cell deviation angle less than 10° for over 90% of the cells, while a porous surface structure tended to form neuron nets. All of the rGOF possessed excellent cytocompatibility with TGNs. Our results underlined the high degree of alignment of topographical cues in guidance of neurite over high electrical conductivity. The as-prepared rGOFs could be used in many areas including biosensing, electrochemistry, energy, and peripheral or central nerve tissue engineering.
