RESUMEN

PETase exhibits a high degradation activity for polyethylene terephthalate (PET) plastic under moderate temperatures. However, the effect of non-active site residues in the second shell of PETase on the catalytic performance remains unclear. Herein, we proposed a crystal structure- and sequence-based strategy to identify the key non-active site residue. D186 in the second shell of PETase was found to be capable of modulating the enzyme activity and stability. The most active PETaseD186N improved both the activity and thermostability with an increase in Tm by 8.89 °C. The PET degradation product concentrations were 1.86 and 3.69 times higher than those obtained with PETaseWT at 30 and 40 °C, respectively. The most stable PETaseD186V showed an increase in Tm of 12.91 °C over PETaseWT. Molecular dynamics (MD) simulations revealed that the D186 mutations could elevate the substrate binding free energy and change substrate binding mode, and/or rigidify the flexible Loop 10, and lock Loop 10 and Helix 6 by hydrogen bonding, leading to the enhanced activity and/or thermostability of PETase variants. This work unraveled the contribution of the key second-shell residue in PETase in influencing the enzyme activity and stability, which would benefit in the rational design of efficient and thermostable PETase.

Polyethylene terephthalate (PET) is one of the most widely used plastics, and the accumulation of PET poses a great threat to the environment. IsPETase can degrade PET rapidly at moderate temperatures, but its application is greatly limited by the low stability. Herein, molecular dynamics (MD) simulations combined with a sequence alignment strategy were adopted to introduce salt bridges into the flexible region of IsPETase to improve its thermal stability. In the designed variants, the Tm values of IsPETaseI168R/S188D and IsPETaseI168R/S188E were 7.4 and 8.7 °C higher than that of the wild type, respectively. The release of products degraded by IsPETaseI168R/S188E was 4.3 times that of the wild type. Tertiary structure characterization demonstrated that the structure of the variants IsPETaseI168R/S188D and IsPETaseI168R/S188E became more compact. Extensive MD simulations verified that a stable salt bridge was formed between the residue R168 and D186 in IsPETaseI168R/S188D , while in IsPETaseI168R/S188E an R168-D186-E188 salt bridge network was observed. These results confirmed that the proposed computation-based salt bridge design strategy could efficiently generate variants with enhanced thermal stability for the long-term degradation of PET, which would be helpful for the design of enzymes with improved stability.

Microplastic (MP) is ubiquitous in the environment which appeared as an immense intimidation to human and animal health. The plastic fragments significantly polluted the ocean, fresh water, food chain, and other food items. Inadequate maintenance, less knowledge of adverse influence along with inappropriate usage in addition throwing away of plastics items revolves present planet in to plastics planet. The present study aims to focus on the recognition and advance detection technologies for MPs and the adverse effects of micro- and nanoplastics on human health. MPs have rigorous adverse effect on human health that leads to condensed growth rates, lessened reproductive capability, ulcer, scrape, and oxidative nervous anxiety, in addition, also disturb circulatory and respiratory mechanism. The detection of MP particles has also placed emphasis on identification technologies such as scanning electron microscopy, Raman spectroscopy, optical detection, Fourier transform infrared spectroscopy, thermo-analytical techniques, flow cytometry, holography, and hyperspectral imaging. It suggests that further research should be explored to understand the source, distribution, and health impacts and evaluate numerous detection methodologies for the MPs along with purification techniques.

Besides the many advantages of oral drug administration, challenges like premature drug degradation and limited bioavailability in the gastro-intestinal tract (GIT) remain. A prolonged residence time in the GIT is beneficial for enhancing the therapeutic outcome when treating diseases associated with an increased intestinal clearance rate, like inflammatory bowel disease (IBD). In this study, we synthesized rod-shaped mesoporous silica nanoparticles (MSNs) functionalized with polyethylene glycol (PEG) or hyaluronic acid (HA) and investigated their bio-distribution upon oral administration in vivo. The negatively charged, non-toxic particles showed different accumulation behavior over time in healthy mice and in mice with dextran sulfate sodium (DSS)-induced intestinal inflammation. PEGylated particles were shown to accumulate in the lower intestinal tract of healthy animals, whereas inflammation promoted retention of HA-functionalized particles in this area. Overall systemic absorption was low. However, some particles were detected in organs of mice with DSS-induced colitis, especially in the case of MSN-PEG. The in vivo findings were connected to surface chemistry-related differences in particle adhesion on Caco-2/Raji and mucus-producing Caco-2/Raji/HT29 cell co-culture epithelial models in vitro. While the particle adhesion behavior in vivo was mirrored in the in vitro results, this was not the case for the resorption results, suggesting that the in vitro model does not fully reflect the erosion of the inflamed epithelial tissue. Overall, our study demonstrates the possibility to modulate accumulation and retention of MSNs in the GIT of mice with and without inflammation through surface functionalization, which has important implications for the formulation of nanoparticle-based delivery systems for oral delivery applications.

Micro/nano-plastics (MNPs) are considered a heterogeneous class of environmental contaminants that cause multiple toxic effects on biological species. As the commonly used mammalian models to study the effects of MNPs with regard to their toxic effects, the mouse and rat models are making a great contribution to the disciplines of environmental toxicology and medical health. However, the toxic effects of MNPs have not been systematically summarized. Therefore, a systematic review and a meta-analysis of the toxic effects of MNPs on mouse/rat models were conducted. A total of seven main categories were established in this systematic review, and 24 subcategories were further divided according to the specific physiological significance of the endpoint or the classification of the physiological system, which covered all the selected pieces of literature. A total of 1,762 biological endpoints were found, and 52.78% of them were significantly affected. This fact indicates that there are relative factors, including the size, polymer type, concentration, and exposure time of MNPs and different sexes of mouse/rat models that could significantly affect the biological endpoints. These biological endpoints can be classified into various factors, such as the dose-response relationships between MNP concentration and physiological categories of the nervous system, growth, reproduction, digestive tract histopathology, and inflammatory cytokine level, among others. MNPs negatively affected the blood glucose metabolism, lipid metabolism, and reproductive function in mice. The reproductive function in male mice is more sensitive to the toxic effects of MNPs. These findings also provide insights into and directions for exploring the evidence and mechanisms of the toxic effects of MNPs on human health. It is clear that more research is required on the pathological mechanisms at the molecular level and the long-term effects of tissue accumulation.

Volatile organic compounds (VOCs) in urine are valuable biomarkers for noninvasive disease diagnosis. Herein, a facile coordination-driven modular assembly strategy is used for developing a library of gas-sensing materials based on porous MXene frameworks (MFs). Taking advantage of modules with diverse composition and tunable structure, our MFs-based library can provide more choices to satisfy gas-sensing demands. Meanwhile, the laser-induced graphene interdigital electrodes array and microchamber are laser-engraved for the assembly of a microchamber-hosted MF (MHMF) e-nose. Our MHMF e-nose possesses high-discriminative pattern recognition for simultaneous sensing and distinguishing of complex VOCs. Furthermore, with the MHMF e-nose being a plug-and-play module, a point-of-care testing (POCT) platform is modularly assembled for wireless and real-time monitoring of urinary volatiles from clinical samples. By virtue of machine learning, our POCT platform achieves noninvasive diagnosis of multiple diseases with a high accuracy of 91.7%, providing a favorable opportunity for early disease diagnosis, disease course monitoring, and relevant research.

Development and utilization of oxygen absorbing materials for food and beverage packaging are important to protect the oxygen-sensitive foods. In this study, we developed a kind of ionic polymer with excellent oxygen absorbing ability based on reacting ethylene-acrylic acid copolymer (EAA) and sodium sulfite (Na2 SO3 ). By virtue of the hydrophilicity of the ionic polymer, the sulfite in Na2 SO3 is easily hydrolyzed and ionized. The oxygen inhalation reaction was gradually initiated and started, whereas achieved a slow and controllable oxygen absorption process. The oxygen absorbing agent can be directly added in the form of an auxiliary agent, which greatly simplifies the preparation process of the oxygen absorbing material. Furthermore, the ionic structure of the EAA/Na2 SO3 composites were destroyed and mechanical properties of the material did not decrease after oxygen absorption. More importantly, it can effectively prevent the entry of external oxygen because the ionic polymer itself has better barrier properties. PRACTICAL APPLICATION: Typical oxygen-sensitive objects include: beer, coffee, canned food, meat products, dairy products, and so on. The presence of oxygen in the package can cause microbial growth, odor generation, color change and nutrient loss, resulting in a significant reduction in food shelf life. Therefore, controlling the oxygen content of the food package is important to limit the rate of these spoilage and spoilage reactions in the food. Materials with active oxygen scavenging and good barrier properties and packaging performance are highly economically viable in terms of improving product quality and reducing packaging costs.

OBJECTIVES: Dental caries occur through a multifactorial process that may influence all tooth surfaces throughout an individual's life. The application of decision analytical modeling (DAM) has gained an increasing level of attention in long-term outcome assessment and economic evaluation of interventions on caries in recent years. The objective of this study was to systematically review the application of DAM and assess their methodological quality in the context of dental caries. METHODS: A systematic review of the literature published to 31st December 2018 was conducted in Medline, EMBASE, NHSEED, and Web of Science electronic databases. The main information and model characteristics of studies was extracted with the methodological quality of included studies reviewed and assessed using the Philips' checklist. RESULTS: Twenty five studies from different settings were included. Modeling techniques mainly comprised main type of modeling including Markov models (n = 12), Markov/microsimulation mixed model (n = 7), systematic dynamic models (n = 3), microsimulation models (n = 2) and decision tree (n = 1). The mean number of criteria fulfilled was 25.1 out of 60 items, which varied between year of study and research groups. The percentage of criteria fulfilled for data dimension was lower than for the structure and consistency dimension. Critical issues were data quality, incorporation of utility values, and uncertainty analysis in modeling. CONCLUSION: The current review revealed that the methodological quality of DAM in dental caries economic evaluations is unsatisfied. Future modeling studies should adhere more closely to good practice guidelines, especially with respect to data quality evaluation, utility values incorporation, and uncertainty analysis in DAM based studies.

We have previously studied the biocompatibility and mechanical properties of porous zein scaffolds. We based the study on the concept that composite scaffold materials, especially when combined with inorganic materials, are more suited to the mechanical and physiological demands of the host tissue than individual scaffold materials. We investigated the effect of zein/inorganic composite on the physical and biological properties of porous zein scaffolds, which were fabricated by salt-leaching. The composite was prepared by immersion in simulated body fluid. The hydroxyapatite (HA)-coated zein scaffold had the same porosity as the zein scaffold (over 75%). Using scanning electron microscopy, it was established that the morphology of pores located on the surface and within the porous scaffolds showed equally good pore interconnectivity with zein. However, the compressive Young's modulus decreased from 240.1+/-96.8 to 34.4+/-12.6MPa, and compressive strength decreased from 7.8+/-1.2 to 4.2+/-0.8MPa. From the in vitro test with human bone marrow stroma cells, the osteoblastic differentiation on the surface of the HA-coated zein scaffold was increased, as expressed by the alkaline phosphatase activity and reverse transcription-polymerase chain reaction analysis for marker genes. From both the mechanical and biological evaluations, the HA-coated zein scaffold was found to be the optimal biomaterial for bone tissue engineering.

BACKGROUND: Currently, cationic liposome has become the commonly used vehicles for gene transfection. Furthermore, one of the most significant steps in microRNAs expression studies is transferring microRNAs into cell cultures successfully. In this study we aim to approach the feasibility of transfection of cervical cancer cell lines mediated by liposome and to obtain the optimized transfection condition for cervical cancer cell lines. MATERIALS AND METHODS: Lipofectamine(TM)2000 as the carrier, miR-101 mimic was transfected into Hela cells and Siha cells. Using green fluorescent protein as reporter gene, to set different groups according to cell seeding density, the amount of miRNA , miRNA and the proportion of Liposomes, Whether to add serum into medium to study their impact on the liposomal transfection efficiency. Finally, MTT assay was used to analyze the relative minimal cell toxicity of liposome reagents. RESULTS: The seeding density of Hela cell line and Siha are 1.5 x 10(4) (per well of 24 well plates), miRNA amount is 1ul of both, the ratio of miRNA and liposome is 1:0.5 of Hela cell line; 1:0.7 of Siha cell line respectively, after 24 hours we can get the highest transfection efficiency. Compared with serum medium, only Siha cells cultured with serum-free medium obtained higher transfection efficiency before transfection (P<0.01).MTT assay showed that according to the above conditions which has the lowest cytotoxicity. CONCLUSIONS: The method of Liposome to transfected is a suitable way and it can be an efficient reagent for miRNA delivery for Hela cells and Siha cells in vitro. It may serve as a reference for the further research or application.

RESUMEN

A novel lanthanide coordination polymer nanoparticle (LCPN)-based ternary complex was synthesized via the self-assembly of a terbium ion (Tb(3+)) with a nucleotide (GMP) and a mercury ion (Hg(2+)) in aqueous solution. The as-prepared LCPN-based ternary complex (Tb-GMP-Hg) can be applied to the development of time-resolved luminescence assays and oxidase-based biosensors.

A series of zinc complexes based on asymmetrical N,N,O-tridentate ligands were prepared via binaphthyl diamine derivatives. These complexes were characterized and employed as catalysts in lactide polymerization. The X-ray diffraction analyses revealed that molecular structures of 1b and 2b were mononuclear complexes with zinc atoms in distorted octahedral geometries. Upon co-catalysis with isopropanol, complex 2a showed the highest activity among these zinc complexes for the ring-opening polymerization of L-lactide, and complex 3a exhibited the highest stereoselectivity for the ring-opening polymerization of rac-lactide affording substantially isotactic polylactide (PLA) with a P(m) of 0.62. The polymerization kinetics using 2a as a catalyst was studied in detail. The kinetics of the polymerization results revealed that the rates of polymerization were first-order both in the monomer and the catalyst, and there was a linear relationship between the L-LA conversion and the number-average molecular weight of PLA with a narrow molecular distribution (1.07-1.17). The activation energy (31.49 kJ mol(-1)) was deduced according to the Arrhenius equation.

Discomfort of right eye and right maxilla region occurred after a dental implant placement in a patient who surfered stabilized osteofibrous dysplasia (OFD) of sphenoid sinus. In this article, the case was discussed and the relevant literature was reviewed. The possible causes of eye and maxilla discomfort may be associated with OFD, hyper allergic response to implant material and surgical trauma, and psychical factors as well.

Tetrazole compounds have been studied for more than one hundred years and applied in various areas. Several years ago Sharpless and his co-workers reported an environmentally friendly process for the preparation of 5-substituted 1H-tetrazoles in water with zinc salt as catalysts. To reveal the exact role of the zinc salt in this reaction, a series of hydrothermal reactions aimed at trapping and characterizing the solid intermediates were investigated. This study allowed us to obtain a myriad interesting metal-organic coordination polymers that not only partially showed the role of the metal species in the synthesis of tetrazole compounds but also provided a class of complexes displaying interesting chemical and physical properties such as second harmonic generation (SHG), fluorescence, ferroelectric and dielectric behaviors. In this tutorial review, we will mainly focus on tetrazole coordination compounds synthesized by in situ hydrothermal methods. First, we will discuss the synthesis and crystal structures of these compounds. Their various properties will be mentioned and we will show the applications of tetrazole coordination compounds in organic synthesis. Finally, we will outline some expectations in this area of chemistry. The direct coordination chemistry of tetrazoles to metal ions and in situ synthesis of tetrazole through cycloaddition between organotin azide and organic cyano group will be not discussed in this review.

The syntheses and luminescent properties of three novel zinc coordination polymers containing tetrazolyl ligands are described. In situ [2+3] cycloaddition reactions of acetonitrile or p-tolylnitrile with sodium azide in the presence of Zn(ClO(4))(2) as a Lewis acid (Demko-Sharpless tetrazole synthesis method) under hydrothermal (solvothermal) reaction conditions gave [Zn(CH(3)CN(4))(2)](3)(H(2)O) (1) and [Zn(4-MPTZ)(2)] (3) [4-MPTZ = 5-(4-methylphenyl)tetrazole], respectively. On the other hand, [Zn(HCN(4))(2)] (2) was obtained by directly reacting tetrazole with Zn(OAc)(2) under hydrothermal reaction conditions. The structure of 1 shows a super-diamond-like topological network with a diamond subunit as a connecting node. For 2, a diamond-like topological network is also found, but it is 2-fold interpenetrated. The structure of 3 reveals a 2D layered network with a hexagonal net, with the adjacent layers in the network stacked in an ABAB sequence. Photoluminescence studies revealed coordination polymers 1, 2, and 3 exhibit strong blue fluorescent emissions at lambda(max) = 396, 418, and 397 nm, respectively, in the solid state at room temperature.

The reactions of (S)-3-cyanophenylalanine with NaN(3) in the presence of H(2)O and Lewis acids (ZnCl(2) and CdCl(2)) afford two unprecedented 3D homochiral networks, mono[(S)-5-(3-tetrazoyl)-phenylalaninato]zinc(II) (1) and mono[(S)-5-(3-tetrazoyl)-phenylalaninato]cadmium(II) monoaqua(II) (2), respectively. The two compounds are isostructural with noninterpenetrated SrAl(2) topology. The structure of these coordination polymers gives new insight into Sharpless' reaction of chiral 5-substituted 1H-tetrazole and homochiral supramolecular array constructions.
