OBJECTS: Changes in condylar position and morphology after mandibular reconstruction are important to aesthetic and functional rehabilitation. We evaluated changes in condylar position and morphology at different stages after mandibular reconstruction using vascularized fibular free flap with condyle preservation. MATERIALS AND METHODS: A total of 23 patients who underwent mandibular reconstruction with fibular flap were included in this retrospective study. CT data of all patients were recorded before surgery (T0), 7 to 14 days after surgery (T1), and at least 6 months after surgery (T2). Five parameters describing the condylar position and 4 parameters describing the morphology were measured in sagittal and coronal views of CT images. The association between clinical characteristics and changes in condylar position and morphology was analyzed. A finite element model was established to investigate the stress distribution and to predict the spatial movement tendency of the condyle after reconstruction surgery. RESULTS: The condylar position changed over time after mandibular reconstruction. The ipsilateral condyles moved inferiorly after surgery (T0 to T1) and continually move anteriorly, inferiorly, and laterally during long-term follow-up (T1 to T2). Contrary changes were noted in the contralateral condyles with no statistical significance. No morphological changes were detected. The relationship between clinical characteristics and changes in condylar position and morphology was not statistically significant. A consistent result was observed in the finite element analysis. CONCLUSION: Condylar positions showed obvious changes over time after mandibular reconstruction with condylar preservation. Nevertheless, further studies should be conducted to evaluate the clinical function outcomes and condylar position. CLINICAL RELEVANCE: These findings can form the basis for the evaluation of short-term and long-term changes in condylar position and morphology among patients who have previously undergone mandibular reconstruction by FFF with condyle preservation.

Tandem semi-stable complementary domains play an important role in life, while the role of these domains in the folding process of nucleic acid molecules has not been systematically studied. Here, we designed a clean model system by synthesizing sequence-defined DNA-OEG copolymers composed of ssDNA fragments with palindromic sequences and orthogonal oligo(tetraethylene glycol) (OEG) linkers. By altering the lengths of DNA units (6-12 nt) and OEG linkers (Xn = 0-4) separately, we systematically studied how stabilities of tandem complementary domains and connecting flexibilities affect the assembly topology. Combining experimental methods and coarse-grained molecular simulation analysis, distributions of multiple assembled conformations (mainly monomers, dimers, and clusters) were characterized. Both results indicated that tandem semi-stable complementary domains tend to form homogeneous closed circular dimers instead of larger clusters due to the synergistic enhancement effect, and the distributions of each conformation highly depend on flexibilities.

An injectable biomaterial has been prepared through co-assembly of lipopeptides C4-Bhc-Glu-Glu-NH2 and C14-Phe-Lys-Lys-NH2. This biomaterial contained a large number of nanofibre bundles (nano-bundles, NBs) of lipopeptide co-assemblies and performed like hydrogels. The morphologies of the NBs were observed by transmission electron microscopy (TEM) and atomic force microscopy (AFM). The rheological properties were investigated with a rheometer. Excitingly, the NB biomaterials exhibited shear thinning and self-healing properties, and could be used as injectable biomaterials. The coumarin group in the lipopeptides endowed the NB biomaterials with both ultraviolet (UV, a one photon process) and near-infrared (NIR) light (a two photon process) responsiveness. A small molecule (Doxorubicin, DOX) and a large molecule (bovine serum albumin, BSA) were used as model drugs, and both of them could be encapsulated in the NB biomaterials and could also be released sustainably or explosively under different conditions (with or without one- and two-photon irradiation). DOX and BSA have different release behaviors because of the NBs. Cell assays showed that the co-assembled NB biomaterials exhibited low cytotoxicity to normal cells. However, when DOX was loaded, the NB biomaterials could kill HeLa cells sustainably. Under UV and NIR irradiation, HeLa cells could be killed rapidly because of the burst release of DOX. The co-assembled supramolecular NB biomaterials with dual-responsiveness, tunable rheological properties and multi-drug encapsulating ability might have potential in biomedical engineering.

Two series of nucleolipids have been designed and synthesized, one with a varying chain length (the dT-Cn series) and the other incorporating an aromatic photo-responsive moiety at the molecular hydrophobic and hydrophilic interface (the P-dT-Cn series). Surface tension measurements revealed the variations of critical micelle concentrations (CMCs) with the alkyl chain length and the incorporation of the photo-responsive segment. The P-dT-Cn series showed broadly lower CMCs and the minimum area per molecule (Amin) values because the π-π stacking between the additional aromatic rings favours more tight packing in the micelle formation. Both series showed similar surface tensions at the CMCs to conventional surfactants with equivalent molecular structures. Their micellar aggregates were used for encapsulation of hydrophobic Nile Red (NR). For the P-dT-Cn series, the encapsulated NR was released upon light irradiation and the controlled release was readily realized by controlling irradiation intensities or switching on and off irradiation. The integration of biocompatibility, complementary base recognition and photo-responsiveness makes the amphiphilic nucleolipids promising in biomedical and biotechnological applications.

Over the past 20 years, the Seneca Valley virus (SVV) has emerged in various countries and regions around the world. Infected pigs display symptoms similar to foot-and-mouth disease and other vesicular diseases, causing severe economic losses to affected countries. In recent years, the number of SVV infections has been increasing in Brazil, China, and the United States. In this study, we comprehensively analyzed SVV genomic sequence data from the perspectives of evolutionary dynamics, phylogeography, and codon usage bias. We aimed to gain further insights into SVV's genetic diversity, spatiotemporal distribution patterns, and evolutionary adaptations. Phylogenetic analysis revealed that SVV has evolved into eight distinct lineages. Based on the results of phylogeographic analysis, it is speculated that the United States might have been the source of SVV, from where it subsequently spread to different countries and regions. Moreover, our analysis of positive selection sites in SVV capsid proteins suggests their potential importance in the process of receptor recognition. Finally, codon preference analysis indicates that natural selection has been a primary evolutionary driver influencing SVV codon usage bias. In conclusion, our in-depth investigation into SVV's origin, dissemination, evolution, and adaptation emphasizes the significance of SVV surveillance and control measures.
