RESUMEN

Chinese bayberry leaf proanthocyanidins (BLPs) are Epigallocatechin gallate (EGCG) oligomers or polymers, which have a lot of health-promoting activity. The activity is closely related to their behavior during in vitro digestion, which remains unknown and hinders further investigations. To clarify the changes of BLPs during gastrointestinal digestion, further research is required. For in vitro digestion, including gastric-intestinal digestion, colon fermentation was applied. Caco-2 monolayer transportation was also applied to investigate the behavior of different BLPs with different degrees of polymerization. The trimers and the tetramers were significantly decreased during in vitro gastric-intestinal digestion resulting in a significant increase in the content of dimers. The dimers and trimers were the main compounds utilized by gut microbiota and they were assumed not to degrade through cleavage of the inflavan bond. The monomers and dimers were able to transport through the Caco-2 monolayer at a rate of 10.45% and 6.4%, respectively.

Objective: To separate the resting-state network of patients with dental pain using independent component analysis (ICA) and analyze abnormal changes in functional connectivity within as well as between the networks. Patients and Methods: Twenty-three patients with dental pain and 30 healthy controls participated in this study. We extracted the resting-state functional network components of both using ICA. Functional connectivity differences within 14 resting-state brain networks were analyzed at the voxel level. Directional interactions between networks were analyzed using Granger causality analysis. Subsequently, functional connectivity values and causal coefficients were assessed for correlations with clinical parameters. Results: Compared to healthy controls, we found enhanced functional connectivity in the left superior temporal gyrus of anterior protrusion network and the right Rolandic operculum of auditory network in patients with dental pain (p<0.01 and cluster-level p<0.05, Gaussian random field corrected). In contrast, functional connectivity of the right precuneus in the precuneus network was reduced, and were significantly as well as negatively correlated to those of the Visual Analogue Scale (r=-4.93, p=0.017), Hamilton Anxiety Scale (r=-0.46, p=0.027), and Hamilton Depression Scale (r=-0.563, p<0.01), using the Spearman correlation analysis. Regarding the causal relationship between resting-state brain networks, we found increased connectivity from the language network to the precuneus in patients with dental pain (p<0.05, false discovery rate corrected). However, the increase in causal coefficients from the verbal network to the precuneus network was independent of clinical parameters. Conclusion: Patients with toothache exhibited abnormal functional changes in cognitive-emotion-related brain networks, such as the salience, auditory, and precuneus networks, thereby offering a new imaging basis for understanding central neural mechanisms in dental pain patients.

Background: Acute pericoronitis (AP) is a common cause of odontogenic toothache. Pain significantly affects the structure and function of the brain, but alterations in spontaneous brain activity in patients with AP are unclear. Purpose: To apply the amplitude of low-frequency fluctuations (ALFF) method in resting-state functional magnetic resonance imaging to investigate altered spontaneous brain activity characteristics in patients with AP in different frequency bands (typical, slow-4, and slow-5 bands) and assess their correlation with clinical scores. Patients and Methods: Thirty-four right-handed patients with AP and 31 healthy controls (HC), matched for age, sex, education, and right-handedness, were enrolled. All subjects underwent resting-state functional magnetic resonance imaging. DPABI software was used for data preprocessing and extracting the ALFF values in different frequency bands. Subsequently, differences in ALFF values in the three bands were compared between the two groups. Correlation between ALFF values in the differential brain regions and clinical scores was assessed. Results: In the typical band, ALFF values were higher in the left insula, left superior occipital gyrus, left inferior parietal lobule, left posterior cerebellar lobule, and right postcentral gyrus in the AP than in the HC group. In the slow-4 band, ALFF values in the left superior occipital gyrus, right superior occipital gyrus, and right middle occipital gyrus were higher, and those in the right cingulate gyrus and right superior temporal gyrus were lower in the AP than in the HC group. In the slow-5 band, the ALFF values in the left insula and left superior occipital gyrus were higher in the AP than in the HC group. The ALFF values of the typical bands in the left insula, left inferior parietal lobule, and right postcentral gyrus correlated negatively, those of the slow-4 band in the right middle occipital gyrus correlated positively, and those of the slow-5 band in the left insula correlated negatively with the visual analogue scale score in the AP group. Conclusion: Our results suggested that the intrinsic brain activity of AP patients was abnormal and frequency-dependent. This provides new insights to explore the neurophysiological mechanisms of AP.

Curcumin solid dispersions (Cur SDs) were prepared using hydroxypropyl methyl cellulose (HPMC) and sodium carboxymethyl cellulose (CMC) at different dosages. The results of Fourier transform infrared spectroscopy and Raman spectroscopy showed that the characteristic peak of curcumin shifted, and the addition of CMC enhanced this phenomenon. The addition of CMC reduced the contact angle, increased the surface free energy, and improved the solubility of Cur SDs. These changes were positively correlated with the amount of CMC. The surface morphology of Cur SDs changed from needle-like to sheet-like as observed by scanning electron microscopy. Cur SDs prepared by CMC and HPMC retained good biological activity. HT-29 human colon cancer cell analysis showed that the addition of CMC significantly improved the anti-proliferation effect of Cur SDs, thus enhancing the bioavailability of curcumin. Solid dispersions made with CMC and HPMC will be a promising hydrocolloid carrier to improve oral bioavailability and efficacy of curcumin.

Objective: To study the dynamic changes of local metrics in patients with toothache (TA, Toothache) in the resting state, in order to further understand the changes of central neural mechanism in patients with dental pain and its effect on cognition and emotion. Methods: Thirty patients with TA and thirty matched healthy (HC) control volunteers were recruited, and resting-state functional magnetic resonance (rs-MRI) scans were performed on all subjects, and data were analyzed to compare group differences in three dynamic local indices: dynamic regional homogeneity (dReHO), dynamic low-frequency fluctuation amplitude (dALFF) and dynamic fractional low-frequency fluctuation amplitude (dfALFF). In addition, the association between dynamic local metrics in different brain regions of TA patients and scores on the Visual Analog Scale (VAS) and the Hospital Anxiety and Depression Scale (HADS) was investigated by Pearson correlation analysis. Results: In this study, we found that The local metrics of TA patients changed with time Compared with the HC group, TA patients showed increased dReHo values in the left superior temporal gyrus, middle frontal gyrus, precentral gyrus, precuneus, angular gyrus, right superior frontal gyrus, middle temporal gyrus, postcentral gyrus and middle frontal gyrus, increased dALFF values in the right superior frontal gyrus, and increased dfALFF values in the right middle temporal gyrus, middle frontal gyrus and right superior occipital gyrus (p < 0.01, cluster level P < 0.05). Pearson correlation analysis showed that dReHo values of left precuneus and left angular gyrus were positively correlated with VAS scores in TA group. dReHo value of right posterior central gyrus was positively correlated with HADS score (P < 0.05). Conclusion: There are differences in the patterns of neural activity changes in resting-state brain areas of TA patients, and the brain areas that undergo abnormal changes are mainly pain processing brain areas, emotion processing brain areas and pain cognitive modulation brain areas, which help to reveal their underlying neuropathological mechanisms. In the hope of further understanding its effects on cognition and emotion.

OBJECTIVE: The objective of this study was to investigate the antibacterial effects on a cariogenic biofilm of a bioactive glass (BAG) combined with either sodium fluoride (NaF) or triclosan (TCS). DESIGN: According to minimal bactericidal concentrations, 37.5mg/ml of BAG, 4.69 mg/ml of NaF, and 15.53 µg/ml of TCS solutions were prepared. When used alone, the three antimicrobial solutions were increased to double-dosage strength (2 MBC). The study contained the following experimental groups: group 1, BAG (2 MBC); group 2, NaF (2 MBC); group 3, TCS (2 MBC); group 4, BAG+NaF; group 5, BAG+TCS; group 6, control (saline). Streptococcus mutans biofilm was cultured with 0.1% sucrose anaerobically on 66 sterilized coverslips (1 × 1 cm(2)) for 24h uninterrupted. After 10 min of exposure to the experimental groups, the microbial kinetics, morphology, and viability of the S. mutans biofilms were assessed by evaluation of colony-forming units (CFUs), scanning electron microscopy (SEM), and confocal laser scanning microscopy (CLSM). RESULTS: BAG (2 MBC) used alone showed significantly stronger antibacterial effects than the other two antimicrobials used alone. The combination groups also displayed the same or greater biofilm inactivation effects as BAG (2 MBC) in the plate count test. SEM showed smaller stacks (towers) and fewer surrounding bacteria in groups BAG (2 MBC), BAG+NaF, and BAG+TCS. Confocal microscopy also determined higher live/dead ratios in groups NaF (2 MBC), TCS (2 MBC), and control than in groups BAG (2 MBC), BAG+NaF, and BAG+TCS. CONCLUSIONS: The combinations of BAG with either NaF or TCS enhanced the inactivation effects of BAG (2 MBC) on S. mutans biofilm, and these findings should be further investigated clinically for the control of dental biofilms.
