RESUMEN

The dark sleeper, Odontobutis potamophila, is a commercially valuable fish that widely distributed in China and Southeast Asia countries. The phenomenon of sexual dimorphism in growth is conspicuous, which the males grow substantially larger and faster than the females. However, the high-quality genome resources for gaining insight into sex-determining mechanisms to develop sex-control breeding are still lacking. Here, a chromosomal-level genome assembly of O. potamophila was generated from a combination of Illumina reads, 10× Genomics sequencing, and Hi-C chromatin interaction sequencing. The assembled genome was 1,134.62 Mb with a contig N50 of 22.25 Mb and a scaffold N50 of 24.85 Mb, representing 94.4% completeness (Benchmarking Universal Single-Copy Orthologs). Using Hi-C data, 96.49% of the total contig bases were anchored to the 22 chromosomes, with a contig N50 of 22.25 Mb and a scaffold N50 of 47.68 Mb. Approximately 54.18% of the genome were identified as repetitive elements, and 23,923 protein-coding genes were annotated in the genome. The assembled genome can be used as a valuable resource for molecular breeding and functional studies of O. potamophila in the future.

RESUMEN

OBJECTIVES: To explore the possibility of using 2-dimensional image assessments in combination with cone beam computed tomography (CBCT) in patients with temporomandibular joint osteoarthritis (TMJOA) to enhance the clinical significance and applicability of the CBCT findings on TMJOA changes. STUDY DESIGN: Fifty-two patients with unilateral TMJOA and 40 healthy adults who had undergone CBCT in preparation for orthodontic treatment were selected for this study. Temporomandibular joint images were acquired with CBCT. All images were classified into 4 phases (slight erosion, widespread erosion, appearance of bone maintenance, and partial loss of condylar head with a remodeled cortical surface) and 4 levels of severity (absent, slight, moderate, and extensive). Quantitative microstructural analysis, including bone value fraction (BV/TV), trabecular number (Tb.N), trabecular thickness, and trabecular separation (Tb.Sp) of each image, was performed by using a customized software. RESULTS: In patients with unilateral TMJOA, the affected side had significantly higher BV/TV and Tb.N and significantly lower Tb.Sp compared with the unaffected side. Compared with healthy individuals, the affected joints in patients with TMJOA had significantly higher BV/TV and trabecular thickness and significantly lower Tb.N and Tb.Sp. No significant difference was found among different phase groups or different severity levels. CONCLUSIONS: CBCT combined with a software investigation protocol for microstructure measurement can be used to provide reliable trabecular parameters in assessing TMJOA.

OBJECTIVE: To detect the ability of Enterococcus faecalis (E. faecalis) biofilm formation and explore the relationship between E. faecalis biofilm formation ability and clinical manifestation. METHODS: 96 well plate with the establishment of 53 E. faecalis in vitro biofilm model, combined with crystal violet staining, was used to test the biofilm formation ability of the clinical isolates E. faecalis and analyze the relationship between biofilm formation capacity and clinical manifestation. RESULTS: In total 53 E. faecalis strains, 40 strains(75.47%) had biofilm forming ability. Statistical analysis revealed that the capacities of biofilm formation between E. faecalis isolated from with fistula and without fistula was significantly different (P<0.05). CONCLUSION: In the retreatment of root canal, the ability of biofilm formation of E. faecalis separated from the teeth without fistula is better than those separated from the teeth with fistula.

RESUMEN

OBJECTIVE: To detect the Enterococcus faecalis (E. faecalis) in post-treatment endodontic disease, and to analyze the relationship between the occurrence of E. faecalis and clinical symptom. METHODS: 108 teeth which need root canal retreatment were collected, and the clinical symptoms and physical signs were recorded. Bacterium samples from root canal were taken, and genome DNA from bacterial samples were extracted. The occurrence of E. faecalis by means of the polymerase chain reaction was investigated. RESULTS: The detection rate of E. faecalis in cases of root canal retreatment was 47.2%, while in cases with symptoms or signs, or cases with both symptoms and signs, the root canal E. faecalis detection rates were 52.6%, 57.9%, 62.5%. The detection rates of E. faecalis between cases with clinical symptom and without clinical symptom demonstrated statistical significance (P < 0.05). The detection rates between cases with both clinical symptom and manifestly aneretic root and cases without clinical symptom and manifestly aneretic root had statistical significance (P < 0.05). In the group of clinical symptom, the detection rate of E. faecalis in cases with biting pain was 66.7%, clearly higher than those without biting pain (P < 0.05). CONCLUSION: The occurrence of E. faecalis in cases of root canal retreatment correlates with clinical symptoms.
