RESUMEN

Rapid diagnosis of coronavirus disease 2019 (COVID-19) is key for the long-term control of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) amid renewed threats of mutated SARS-CoV-2 around the world. Here, we report on an electrical label-free detection of SARS-CoV-2 in nasopharyngeal swab samples directly collected from outpatients or in saliva-relevant conditions by using a remote floating-gate field-effect transistor (RFGFET) with a 2-dimensional reduced graphene oxide (rGO) sensing membrane. RFGFET sensors demonstrate rapid detection (<5 min), a 90.6% accuracy from 8 nasal swab samples measured by 4 different devices for each sample, and a coefficient of variation (CV) < 6%. Also, RFGFET sensors display a limit of detection (LOD) of pseudo-SARS-CoV-2 that is 10 000-fold lower than enzyme-linked immunosorbent assays, with a comparable LOD to that of reverse transcription-polymerase chain reaction (RT-PCR) for patient samples. To achieve this, comprehensive systematic studies were performed regarding interactions between SARS-CoV-2 and spike proteins, neutralizing antibodies, and angiotensin-converting enzyme 2, as either a biomarker (detection target) or a sensing probe (receptor) functionalized on the rGO sensing membrane. Taken together, this work may have an immense effect on positioning FET bioelectronics for rapid SARS-CoV-2 diagnostics.

RESUMEN

As emerging contaminants, microplastics (MPs) have attracted global attention. They are a potential risk to organisms, ecosystems and human health. MPs are characterized by small particle sizes, weak photodegradability, and are good environmental carriers. They can physically adsorb or chemically react with organic, inorganic and bio-pollutants to generate complex binary pollutants or change the environmental behaviors of these pollutants. We systematically reviewed the following aspects of MPs: (i) Adsorption of heavy metals and organic pollutants by MPs and the key environmental factors affecting adsorption behaviors; (ii) Enrichment and release of antibiotic resistance genes (ARGs) on MPs and the effects of MPs on ARG migration in the environment; (iii) Formation of "plastisphere" and interactions between MPs and microorganisms; (iv) Ecotoxicological effects of MPs and their co-exposures with other pollutants. Finally, scientific knowledge gaps and future research areas on MPs are summarized, including standardization of study methodologies, ecological effects and human health risks of MPs and their combination with other pollutants.

The geochemical characteristics of heavy metals (Cd, Co, Cr, Cu, Ti, Zn) in 61 surface sediments from the Yangtze River Estuary and its adjacent East China Sea were studied. The high values of Cu, Co and Zn are distributed near the mouth of Hangzhou Bay. The high values of Cr appear near Zhoushan Archipelago. The high values of Cd appear in the Yangtze River Estuary, while Ti and Cd have high values in the open sea eastward. The order of pollution degree is Cr > Cd > Co > Zn > Ti > Cu. The enrichment degree is Cd > Cr > Zn > Co > Ti > Cu. Except for some sites, Cd is moderately enriched, but there is no severe enrichment. The main sources of human activities of the six metals include industry and shipping. Agricultural production may be another important source of human activities for Cd.

Many Trichoderma species are cosmopolitan and widespread free-living fungi in various ecological environment, and many are economically important in the fields of agriculture and industry. However, carbon metabolism profiles of Trichoderma species have not been characterized in detail. In this study, Biolog FF MicroPlates were used to contrast carbon utilization and the differences among five Trichoderma species, each representing a unique phenotype. Their metabolic abilities varied greatly. Species producing cellulases and chitinases and with phosphate-solubilizing activities exhibited high efficiency of substrate utilization, whereas low efficiency was shown by saline-alkaline-tolerant species that metabolized simple carbon sources. Species producing cellulases at high levels may be specified to decompose and assimilate cellulose and hemicellulose in woody substrates, and those producing chitinases may have mycoparasitic roles. Species with plant growth-promoting traits are good at uptake of exudates from plant roots. Overall, metabolic models reflect nutritional adaptation of Trichoderma to diverse niches in nature.

The distribution characteristics, sources and ecological risk of thallium (Tl) in the surface sediments of Yangtze Estuary and its adjacent sea were studied. Tl concentrations ranged from 0.369 to 1.197 µg g-1 with an average of 0.674 µg g-1, which was slightly higher than the corresponding background values. Tl concentrations were relatively high in sediments of the south bank of Chongming Island and the Hangzhou Bay mouth, and gradually decreased from inner shelf to outer seas. The variation trend of Tl concentrations was controlled by sediment characteristics, hydrodynamic conditions and sources together. The sediment flux of Tl in the study area was 428.6 t/yr. The Yangtze River, the Yellow River and atmospheric inputs of Tl accounted for 52.7%, 10.5%, and 0.15% of the total sediment flux, respectively. The result of potential ecological index indicated that Tl in surface sediments of the study area had no threat to the ecological environment.

The distribution characteristics, sources and ecological risk of antimony (Sb) in the surface sediments of Changjiang Estuary and the adjacent sea were studied. Sb concentrations ranged from 0.320 to 0.968 µg g-1 with mean value of 0.577 µg g-1. Sb concentrations were relatively high in sediments of the south Yellow Sea, the Hangzhou Bay mouth and the inner Changjiang Estuary. The variation trend of Sb concentrations was controlled by hydrodynamics, Al/Fe/Mn oxides. Sb also showed strong chalcophile property. Correlation analysis and enrichment factor showed Sb came mainly from natural sources. Total Sb sediment flux in the study area was 446.3 t/yr. The Changjiang River, the Yellow River and atmospheric inputs accounted for 85.7%, 13.9%, and 0.4% of the total sediment Sb flux, respectively. The result of potential ecological index indicated the very low Sb concentrations could hardly threat the ecological environment of the study area.

AIM: To describe the clinical characteristics with genetic lesions in a Chinese family with Crouzon syndrome. METHODS: All five patients from this family were included and received comprehensive ophthalmic and systemic examinations. Direct sequencing of the FGFR2 gene was employed for mutation identification. Crystal structure analysis was applied to analyze the structural changes associated with the substitution. RESULTS: All patients presented typical Crouzon features, including short stature, craniosynostosis, mandibular prognathism, shallow orbits with proptosis, and exotropia. Intrafamilial phenotypic diversities were observed. Atrophic optic nerves were exclusively detected in the proband and her son. Cranial magnetic resonance imaging (MRI) implied a cystic lesion in her sellar and third ventricular regions. A missense mutation, FGFR2 p.Cys342Trp, was found as disease causative. This substitution would generate conformational changes in the extracellular Ig-III domain of the FGFR-2 protein, thus altering its physical and biological properties. CONCLUSION: We describe the clinical presentations and genotypic lesions in a Chinese family with Crouzon syndrome. The intrafamilial phenotypic varieties in this family suggest that other genetic modifiers may also play a role in the pathogenesis of Crouzon syndrome.

OBJECTIVE: This study was designed to establish guinea pigs as an animal model for uterine artery embolization (UAE) with tris-acryl gelatin microspheres (TAGM). METHODS: Twenty-five female adult guinea pigs were randomly divided into two groups, including a uterine artery casting mould group (n=10) and a UAE group (n=15). Pelvic angiography and vascular casting mould were performed in the first group. The anatomical characters of the pelvic cavity in guinea pigs were described. In the second group, the technical feasibility of performing UAE with TAGM in guinea pigs was investigated. The histopathological slides of the uterus of guinea pigs after UAE were examined to inspect the outcomes of UAE. RESULTS: The uterine artery springs from the internal iliac artery, ascends tortuously along the cervix, and gives off vertically 8-10 branches to the cervix uteri and uterine horns. The diameters of the trunk of the uterine artery and its first branch were 0.32±0.027 mm and 0.14±0.01 mm, respectively. For UAE animals, the dosages of 40-120 and 100-300 µm TAGM were 0.033±0.003 ml and 0.015±0.002 ml, respectively. On histopathological slides, embosphere particles were found in the first branches of the uterine artery, the subserous arteries, and the intramural arteries. Inflammatory reactions in the uterus were common in guinea pigs after UAE. Local or dispersed areas of necrosis in uterus also were observed in a few guinea pigs. CONCLUSIONS: Guinea pigs are an appropriate and feasible model for UAE with TAGM.

PURPOSE: To evaluate the efficacy of a dentifrice containing 5% potassium nitrate (test) on dental hypersensitivity compared with the fluoride dentifrice (control) over a 4-week period. METHODS: The study design was a randomized ,double-blinded ,controlled clinical trial. 63 subjects were randomly divided into 2 groups, with 31 were given a 5% potassium nitrate dentifrice (experimental group) and the other 32 given a fluoride dentifrice (control group). Both groups were instructed to brush their teeth twice a day. An air blast stimulus was used to measure the degree of pain using a VAS scale. The data were analysed by the nonparametric Mann-Whitney U test and Wilcoxon-test with SPSS13.0 software package. RESULTS: The VAS (baseline=68.29±8.263, 4-week=31.71±6.378) was significantly reduced over the 4 weeks period in the experimental group (P<0.05). After 4 weeks, VAS in the experimental group was significantly lower than that in the control group(P<0.05). CONCLUSION: The study demonstrates that the dentifrice containing 5% potassium nitrate effectively reduces dental hypersensitivity.

PURPOSE: To study the influence of heat treatment on retention force of magnetic attachments. METHODS: Three groups of magnetic attachments (including 10 Magfit EX 400W, 10 Magfit EX 600W, 10 Magfit EX 800W) were fixed on universal test machine respectively. The retention force of each attachment was measured. After heat treatment, their retention force was measured again. The difference of retention force before and after heat treatment was compared using SPSS11.0 software package. RESULTS: The average retention force of magnetic attachments (Magfit EX 400W) was (1.58±0.12)N before heat treatment and (1.64±0.11)N after heat treatment. The average retention force of magnetic attachments (Magfit EX 600W) was (2.67±0.19)N before heat treatment and (2.65±0.14)N after heat treatment.The average retention force of magnetic attachments (Magfit EX 800W) was (3.02±0.25)N before heat treatment and (3.02±0.24)N after heat treatment. The retention force of magnetic attachments had no significant change after heat treatment (P>0.05). CONCLUSION: The magnetic attachments can be treated by waterbath heart treatment in the clinic without significant change of their retention force.
