RESUMEN

Salt extraction studies showed that keratohyalin (KH) could be solubilized and extracted from fresh bovine hoof epidermis. The solubility of KH varied in relation to the molarity of the salt solution used for extraction. Using this information, the extracted KH was aggregated in vitro by dialyzing the high salt extract against distilled water. Histochemical, ultrastructural, and immunologic studies of the resultant particles or macroaggregates showed that the latter had the same properties and immunogenicity as the KH granule in situ and produced antibodies against it. Fractionation of the macroaggregates by polyacrylamide gel electrophoresis demonstrated that the macroaggregates were compsed of sets of 20 polymers whose subunits or monomers had a molecular weight of 16,900. Amino acid analyses showed that the macroaggregates and the various fractionated polymers were similar and that the protein ahd 116 amino acid residues. Serine, arginine, glycine, glutamic acid, and histidine constituted 78% of all residues, and serine alone represented 27%. The molecular weight by amino acid analyses was 16,150 after correction for the 8% ribonucleic acid which appears to be complexed to the protein.

X-ray microanalysis has revealed qualitative differences in the elemental composition between so-called hyaline bodies occurring in the connective tissue wall of an odontogenic cyst and hyaline bodies occurring within the epithelial lining of these cysts.

RESUMEN

Microprobe analysis of Araldite embedded sections of hyalin bodies revealed the presence of sulphur, chlorine, calcium and in some cases iron. The results have been compared with those obtained from control tissue sections of human heel skin, a thrombus occluded vessel and mineralized spherules in a dental follicle. Microradiographic studies showed that the density of hyalin bodies increased progressively towards the core. Isodensitracing confirms the laminar configuration of hyalin bodies as seen electron microscopically. The findings conform with the hypothesis that hyalin bodies originate as an epithelial secretion.

The odontogenic cysts contain the hyaline deposits in 69.2%. The hyaline deposits appear as a zone that was continuous with basament membrane or as an irregular deposit within the connective tissue.

Hyaline bodies of odontogenic cyst were examined by histological, histochemical and electron microscopic observations. They were seen only in odontogenic cysts and their prevalence was 6.9% in 576 cases of the cysts. They occurred most frequently within the epithelial lining, but a few bodies were observed in the connective tissue walls. Histochemical reactions of hyaline bodies were similar to those of dental cuticles, but differed from keratin. Ultrastructurally, hyaline bodies were typed into two forms: lamellated and homogeneous. The lamellated bodies were composed of alternate electron-dense and electron-lucent layers. The homogeneous bodies were occasionally formed around granular material, mineralized masses and cholesterol clefts. Hyaline bodies adjoined epithelial cells via a basal lamina-like structure and half-desmosomes. Epithelial cells containing well-developed, rough surfaced, endoplasmic reticulum were seen adjacent to hyaline bodies. Ferritin-like granules were found in the cytoplasm of epithelial cells near hyaline bodies. By electron probe X-ray microanalysis, inorganic elements within the ferritin-like granules were the same in constitution as those in the outermost electron-dense layer of the lamellated bodies. By scanning electron microscopy, hyaline bodies displayed ellipsoidal and stick-like shapes. The present study suggested that hyaline bodies are particular products formed by odontogenic epithelium.

RESUMEN

Fibrinogen and IgG were demonstrated in the hyaline zone with direct immunofluorescent staining after orthodontic tooth movement in man. It was suggested that presence of plasma proteins in the hyaline zone contributed to its hyaline appearance. Fibrin and IgG may, furthermore, attract phagocytes and form a network for the reorganization of the necrotic tissue in the hyaline zone.
