RESUMO

Novel poly(arylene ether ketone) polymers with fluorophenyl pendants and phenoxide-terminated wholly sulfonated poly(arylene ether sulfone) oligomers are prepared via Ni(0)-catalyzed and nucleophilic polymerization, respectively, and subsequently used as starting materials to obtain graft-crosslinked membranes as polymer electrolyte membranes. The phenoxide-terminated sulfonated moieties are introduced as hydrophilic parts as well as crosslinking units. The chemical structure and morphology of the obtained membranes are confirmed by (1) H NMR and tapping-mode AFM. The properties required for fuel cell applications, including water uptake and dimensional change, as well as proton conductivity, are investigated. AFM results show a clear nanoscale phase-separation microstructure of the obtained membranes. The membranes show good dimensional stability and reasonably high proton conductivities under 30-90% relative humidity. The anisotropic proton conductivity ratios (σ(formula see text) ) of the membranes in water are in the range 0.65-0.92, and increase with an increase in hydrophilic block length. The results indicate that the graft-crosslinked membranes are promising candidates for applications as polymer electrolyte membranes.

RESUMO

An anaerobic membrane bioreactor (AnMBR) was used to treat low to high (5-200 mg/L) concentrations of 2-chlorophenol (2-CP) wastewater. The AnMBR achieved high and stable chemical oxygen demand removal and 2-CP removal with an average value of 93.2% and 94.2% under long hydraulic retention times (HRTs, 48-96 h), respectively. 2-CP removal efficiency of 98.6 mg/L/d was achieved with 2-CP concentration of 200 mg/L, which was much higher than that of other anaerobic bioreactors. Furthermore, volatile fatty acids didn't accumulate under high 2-CP loading. Long HRTs significantly reduced the membrane fouling as the fouling rate (0.90 × 109-5.44 × 109 m-1h-1) was low. Spirochaetaceae and Methanosaeta were the dominant microbes responsible for dechlorination, methanogenesis, and shock resistance. All these results demonstrate that this AnMBR operated under long HRTs is good and robust for fluctuating chlorophenols wastewater treatment, which has high potential for treating fluctuating refractory organics wastewater with the low membrane fouling rate.

RESUMO

The objective of this article is to obtain a more detailed insight into poly-beta-hydroxybutyrate (PHB) metabolism through network-based metabolic pathway analysis. We employ extreme pathways to perform this study, because calculating and interpreting extreme pathways is a promising way for pathway analysis and metabolic engineering. After giving an in silico model of butanoate metabolism of Bacillus thuringiensis 97-27 (btk), extreme pathways were calculated and classified. Furthermore, the type I and II extreme pathways were further classified and analyzed in detail based on their structure and functional capabilities. Besides "historical" biochemical pathways, the results also suggest that there are some novel pathways.

Pulcherrimin, a potential biocontrol agent produced by microorganisms, has the promising applications in the agricultural, medical, and food areas, and the low yield of pulcherrimin has hindered its applications. In this study, the red pigment produced by Bacillus licheniformis DW2 was identified as pulcherrimin through the spectrometry analysis and genetic manipulation, and the component of the medium used for pulcherrimin production was optimized. Based on our results, the addition of 1.0 g L-1 Tween 80 could improve the yield of pulcherrimin, and glucose and (NH4)2SO4 were served as the optimal carbon and nitrogen sources for pulcherrimin synthesis, respectively. Furthermore, an orthogonal array design was applied for optimization of the medium. Under optimized condition, the maximum yield of pulcherrimin was 331.17 mg L-1, 5.30-fold higher than that of the initial condition, which was the maximum yield reported for pulcherrimin production. Collectively, this study provided a promising strain and a feasible approach to achieve the high-level production of antimicrobial pulcherrimin.

A new capillary electrophoresis method for determining poly-beta-hydroxybutyric acid (PHB) in Bacillus thuringiensis was established. Poly-beta-hydroxybutyric acid in samples was hydrolyzed by sulphuric acid and neutralized by Ba(OH)2. The content of produced beta-hydroxybutyrate was then determined by capillary zone electrophoresis (CZE) with indirect UV detection at 254 nm. With 5 mM p-hydroxybenzoate and 0.5 mM tetradecyltrimethylammonium bromide (TTAB) at pH 8.0 as carrier electrolyte, beta-hydroxybutyrate can be determined within 6 min. Standard regression equation was made by beta-hydroxybutyrate, and the linear range was 2-1000 microg/ml. The relative standard deviations (RSDs) for migration time and peak area are both less than 1.0%. The detection limit for beta-hydroxybutyrate was 0.2 microg/ml, which is two to three orders of magnitude lower than that of the gas chromatography (GC) method. The capillary electrophoresis method was successfully applied to determine poly-beta-hydroxybutyric acid in fermentation broth and single colony. The added standard recovery was 96%.

The production of alpha-ketoglutarate, adenine, thuringiensin production rate and thuringiensin yield on glucose consumed increased by 22%, 36%, 40% and 40%, respectively, in presence of 2 g citrate/l. However, citrate decreased pyruvate production, poly-beta-hydroxybutyrate (PHB) production rate and PHB yield by 62%, 31% and 45%, respectively. The activities of pyruvate kinase and glucose-6-phosphate dehydrogenase were 36%-45% lower and 50%-120% higher than those of the control, respectively. The results suggest that citrate regulated the carbon flux to synthesis of adenine present in thuringiensin with a higher efficiency of utilization of glucose by decreasing PHB synthesis.
