RESUMO

Oral defects lead to a series of function disorders, severely threatening the patients' health. Although injectable hydrogels are widely studied in tissue regeneration, their mechanical performance is usually stationary after implant, without further self-adaption toward the microenvironment. Herein, an injectable hydrogel with programmed mechanical kinetics of instant gelation and gradual self-strengthening along with outstanding biodegradation ability is developed. The fast gelation is realized through rapid Schiff base reaction between biodegradable chitosan and aldehyde-modified sodium hyaluronate, while self-strengthening is achieved via slow reaction between redundant amino groups on chitosan and epoxy-modified hydroxyapatite. The resultant hydrogel also possesses multiple functions including (1) bio-adhesion, (2) self-healing, (3) bactericidal, (4) hemostasis, and (5) X-ray in situ imaging, which can be effectively used for oral jaw repair. We believe that the strategy illustrated here will provide new insights into dynamic mechanical regulation of injectable hydrogels and promote their application in tissue regeneration.

An acid-base switchable [c2]daisy chain rotaxane terminated with two 2,6-diacetylamino pyridine units has been self-assembled with a bis(uracil) linker. The complementary hydrogen-bond recognition patterns, together with lateral van der Waals aggregations, result in the hierarchical formation of unidimensional supramolecular polymers associated in bundles of muscle-like fibers. Microscopic and scattering techniques reveal that the mesoscopic structure of these bundles depends on the extended or contracted states that the rotaxanes show within individual polymer chains. The observed local dynamics span over several length scales because of a combination of supramolecular and mechanical bonds. This work illustrates the possibility to modify the hierarchical mesoscopic structuring of large polymeric systems by the integrated actuation of individual molecular machines.

This paper describes the first reversible, heat-set organogel based on the supramolecular interactions of beta-cyclodextrin (beta-CD). The gel was prepared by interaction of diphenylamine (DPA) with beta-CD and lithium chloride in N,N-dimethylformamide (DMF). In this gel system, DPA could be gelated in DMF as the temperature increased and then dissolved again as the temperature decreased. In the microscopic structure of gel, beta-CDs play a key role in the formation of nanorods and microfibers. Some important features of the gel were observed. (1) The system is a multicomponent solution, in which each of the four components is required for the organogelation property. (2) The system is a reversible, thermo-responsive organogel composed of small organic molecules. When the temperature is lower than T(gel), the gel transforms back into a solution. The reversible thermo-transition was confirmed by differential scanning calorimetry (DSC). The gel system is responsive to the concentration of LiCl. No gel was formed without LiCl. The stimuli responses of the system with other salts such as KCl and NaCl were weaker than with LiCl. (4) The system is responsive to the addition of guest molecules. The structures and sizes of the guest molecules could influence the gel formation. Generally, T(gel) decreased by adding guest molecules in the gel system, but some guest molecules, whose structures are exactly fitted to the cavity of CDs, could prevent gel formation. This work may provide new avenues in delivery of functional molecules as well as design of intelligent materials and biomaterials.
