RESUMO

Polymeric vehicles often exhibit batch-to-batch variations due to polydispersity, limiting their reproducibility for biomedical applications. In contrast, polyhedral oligomeric silsesquioxane (POSS) has emerged as an attractive candidate for drug delivery due to its precise chemical structure and rigid molecular shape. A promising strategy to enhance drug efficacy while reducing systemic toxicity is the development of multi-stimuli-responsive delivery systems capable of targeted drug release at a disease site. Herein, we developed a drug delivery platform based on POSS-polymer conjugates. By functionalizing the POSS with amino groups and establishing B-N coordination with boronic acids, the nanoparticles (NPs) exhibit responsive behavior to stimuli, including adenosine-5'-triphosphate (ATP), acidic pH, and nucleophilic reagents. We successfully encapsulated two boronic acid-containing molecules: tetraphenylethylene (TPE), serving as a fluorescent probe, and bortezomib (BTZ), an anticancer drug. The TPE@NPs were employed to visualize the cellular uptake of NPs by tumor cells, while the BTZ@NPs exhibited increased cytotoxicity in tumor cells compared with normal cells. This POSS-PEG conjugate offers a nanoparticle platform for encapsulating versatile boronic acid-containing molecules, thereby enhancing drug efficacy while minimizing systemic toxicity. Given the wide-ranging applications of boronic acid-containing molecules in biomedicine, our platform holds significant promise for the development of intelligent drug delivery systems for diagnostics and therapeutics.

The effects of Ca2+ on membrane fouling and trace organic compounds (TrOCs) removal in an electric field-assisted microfiltration system were investigated in the presence of Na+ alone for comparison. In the electric field, negatively charged bovine serum albumin (BSA) migrated towards the anode far away from the membrane surface, resulting in a 42.9% transmembrane pressure (TMP) reduction in the presence of Na+ at 1.5 V. In contrast, because of the stronger charge shielding of Ca2+, the electrophoretic migration of BSA was limited and led to a neglectable effect of the electric field (1.5 V) on membrane fouling. However, under 3 V applied voltage, the synergistic effects of electrochemical oxidation and bridging interaction between Ca2+ and BSA promoted the formation of denser settleable flocs and a thinner porous cake layer, which alleviated membrane fouling with a 64.5% decrease in TMP and nearly 100% BSA removal. The TrOCs elimination increased with voltage and reached 29.4%-80.4% at 3 V. The electric field could prolong the contact between TrOCs and strong oxidants generated on the anode, which enhanced the TrOCs removal. However, a stronger charge shielding ability of Ca2+ weakened the electric field force and thus lowered the TrOCs removal.

The affected infraorbital nerve (IFBN) and inferior alveolar nerve (IFAN) status in patients with jaw fibrous dysplasia has not been definitely depicted. In this study, the authors try to explore the status of affected IFBN and IFAN in patients with jaw fibrous dysplasia. Ten patients with jaw fibrous dysplasia were included in this study. The complaints of numbness in the IFBA and IFAN innervated area were asked and recorded, and careful clinical examination was performed to evaluate the touch sense, pain sense, pressure sense, and temperature sense in the IFBA and IFAN innervated areas. Computed tomography scans also were performed to evaluate the imaging characteristics of affected IFBA and IFAN. The results showed that 1 patient with maxillary lesion showed complaints of slight numbness, and clinical examination showed that the patient exhibited slight insensitive in pain sense. In addition, 1 patient with mandibular lesion showed relative obvious complaints of numbness, and clinical examination showed that the patient exhibited slight insensitive in pain sense and temperature sense, but not serious. All other patients exhibited no numbness in the IFBA and IFAN innervated area. Although the position and morphology changed in some patients, all neural canal of affected IFBA or IFAN existed and showed no invasion of lesion. Taking these findings together, it further confirmed that evaluation of the function of IFBAN and IFAN is necessary for patients with jaw fibrous dysplasia, and the affected IFBAN and IFAN may should be reserved in most patients with jaw fibrous dysplasia when resecting or recontouring the lesion.

Applying an electric field in the membrane filtration was an effective method to alleviate membrane fouling and enhance the trace organic compounds (TrOCs) removal. The secondary effluent of a municipal wastewater treatment plant was used as feed water to evaluate the performance of the electric field coupled microfiltration system. Applying a 1.25 V voltage reduced 22.9% membrane fouling by electrophoretic force, and the membrane fouling was alleviated by 70.8% at 3 V by electrochemical oxidation and electric field force. At 3 V, active chlorine and hydroperoxide generated on the electrodes and the acidic environment formed around the anode significantly inhibited the growth of microorganisms and their attachment on the membrane surface, and thus reduced the membrane fouling formed by microorganisms. Electrochemical oxidation also removed the protein in wastewater and changed the main organic components of membrane fouling from microorganisms, protein, and polysaccharide to humic substances and polysaccharide. Furthermore, the electrophoretic force and acidic environment reduced the electrostatic repulsion of humic substances and made them tend to aggregate and form hydrophilic porous fouling structures, which obviously lowered filtration resistance and showed significant membrane fouling mitigation. Also, the electric field effectively enhanced the removal of target TrOCs through electrochemical oxidation and electric field force improving the elimination of TrOCs from 8.5% ~ 26.1% at 0 V to 35.9% ~ 84.8% at 3 V.

Human dental pulp stem cells (hDPSCs) have been recognized as a candidate cell source for tissue engineering. Long non-coding RNAs (lncRNAs) are differentially expressed in inflamed human dental pulp tissues. The present study is aimed at investigating the role of lncRNA H19 in the differentiation potential of hDPSCs. hDPSCs were successfully isolated and cultured, followed by conducting gain and loss-of-function experiments on lncRNA H19 and large tumor suppressor 1 (LATS1) to elucidate their respective biological functions in hDPSCs. lncRNA H19 was able to promote, whereas LATS1 was found to inhibit the differentiation, proliferation, and migration capabilities of hDPSCs. LATS1 was found to activate the Hippo-Yes-associated protein (YAP) signaling pathway by decreasing levels of YAP and Tafazzin (TAZ). The effects of lncRNA H19 on hDPSCs were achieved by repressing LATS1 through enhancer of zeste homolog 2-induced trimethylation of histone 3 at lysine 27. Finally, hDPSCs overexpressing lncRNA H19 and/or LATS1 were transplanted into nude mice. It was shown that lncRNA H19 inhibited LATS1 to promote the production of odontoblasts in vivo. Taken together, lncRNA H19 serves as a contributor to the differentiation potential of hDPSCs via the inhibition of LATS1, therefore highlighting novel therapeutic targets for dental pulp repair.

In orthodontics treatment, scholars have tried to introduce nano antibacterial materials into the materials used in orthodontics to reduce the occurrence of enamel demineralization, caries and periodontitis. The experiment investigated the effect of adding titanium dioxide nanoparticles in different proportions on the flexural strength, compressive strength, surface hardness, tribological properties, adhesion properties, fluorine release properties and antibacterial properties of glass ionomer cement. The effect of water cement on orthodontics, the experiment added different proportions of titanium dioxide nanoparticles to traditional glass ion cement, which greatly improved the mechanical strength of glass ion cement, and significantly improved the antibacterial effect of glass ion cement, bending strength, compressive strength and surface hardness. The addition of titanium dioxide nanoparticles produced a significant antibacterial effect on Streptococcus mutants. Nano- TiO2 glass ionomer cement has a significant effect on orthodontics, and the damage to patients' teeth is significantly lower, which is worthy of clinical popularization.

Coupling electric field, coagulation, and powdered activated carbon (PAC) adsorption in the microfiltration (MF) process was an effective strategy for membrane fouling alleviation and trace organic compounds (TrOCs) elimination. In the electric field, the surface charges of bovine serum albumin (BSA) molecules and kaolin particles distributed along the direction of the electric field and formed electric dipoles, which lowered electrostatic repulsion between BSA-BSA, BSA-kaolin, and kaolin-kaolin, resulting in enhanced particle aggregation and turbidity reduction. Electrophoretic migration also strengthened the interaction between particles and polyaluminum chloride (PACl). Protein removal showed a significant linear correlation with the transmembrane pressure (TMP), and the slope variation of the fitting curves reflected the role of the electric field in the BSA removal and membrane fouling control. Under the synergistic function of electric field, coagulation, and PAC adsorption, the MF system achieved an 80.7% reduction on membrane fouling, an average BSA removal of 76.4%, and TrOCs elimination of 65.3%-81.7%. Electrochemical oxidation was the main contributor to the TrOCs removal when 2.5 V voltage was applied, and could also prolong the service life of PAC.

By modulating the metal centers to adjust the coordination surroundings of the products, two mixed-linker coordination polymers [Cu2(L)(biz)(OH)]·H2O (1) and [Zn(HL)(biz)] (2) (H3L = 5-(4-carboxybenzyloxy)isophthalic acid, biz = benzimidazole), have been produced under mild hydrothermal conditions. To develop new candidates for the acute oral mucositis during orthodontic process, the acute oral mucositis rat model was constructed and the enzyme linked immunosorbent assay (ELISA) was used to find out the release levels of inflammatory cytokines Tumor Necrosis Factor-α (TNF-α) and Interleukin-1ß (IL-1ß). Next, the activation of the AKT signaling pathway was estimated through judging the relative expression of the inflammatory genes in the oral mucosa cells via reverse transcription-polymerase chain reaction (RT-PCR). After compounds treatment, the expression level of the AKT signaling pathway was evaluated by a western blot. Finally, the quantity of the reactive oxygen species (ROS) in the oral mucosa cells was gauged with ROS detection kit. All the results in this research indicated the much more excellent treatment activity of compound 1 than 2 on the acute oral mucositis.

Although previous studies have demonstrated that dental pulp stem cells (DPSCs) from mature and immature teeth exhibit potential for multi-directional differentiation, the molecular and biological difference between the DPSCs from mature and immature permanent teeth has not been fully investigated. In the present study, 500 differentially expressed genes from dental pulp cells (DPCs) in mature and immature permanent teeth were obtained from the Gene Expression Omnibus online database. Based on bioinformatics analysis using the Database for Annotation, Visualization and Integrated Discovery, these genes were divided into a number of subgroups associated with immunity, inflammation and cell signaling. The results of the present study suggest that immune features, response to infection and cell signaling may be different in DPCs from mature and immature permanent teeth; furthermore, DPCs from immature permanent teeth may be more suitable for use in tissue engineering or stem cell therapy. The Online Mendelian Inheritance in Man database stated that Sonic Hedgehog (SHH), a differentially expressed gene in DPCs from mature and immature permanent teeth, serves a crucial role in the development of craniofacial tissues, including teeth, which further confirmed that SHH may cause DPCs from mature and immature permanent teeth to exhibit different biological characteristics. The Search Tool for the Retrieval of Interacting Genes/Proteins database revealed that SHH has functional protein associations with a number of other proteins, including Glioma-associated oncogene (GLI)1, GLI2, growth arrest-specific protein 1, bone morphogenetic protein (BMP)2 and BMP4, in mice and humans. It was also demonstrated that SHH may interact with other genes to regulate the biological characteristics of DPCs. The results of the present study may provide a useful reference basis for selecting suitable DPSCs and molecules for the treatment of these cells to optimize features for tissue engineering or stem cell therapy. Quantitative polymerase chain reaction should be performed to confirm the differential expression of these genes prior to the beginning of a functional study.

A pot experiment including five treatments, i.e., CK (neither fertilizer nor super absorbent polymers), U (urea alone), S [super absorbent polymers (SAP) alone], SUM (SAP mixed with urea) and SUG (gel made of SAP and urea) was conducted to evaluate their effects on fine root morphological characteristics, fine root absorption area, fine root nitrogen metabolism and nitrogen use efficiency of Platycladus orientalis bareroot seedlings. Results showed that compared with U treatment, the SUG treatment significantly increased the biomass, length, specific root length, surface area and volume of fine root. The total absorption area and active absorption area of fine root were also increased remarkably under the SUG treatment. The specific root length in the SUG treatment was increased by 34.7%, 37.9%, 41.1% and 12.4% compared with the treatments of CK, U, S and SUM, respectively. Compared with the U treatment, the activities of nitrate reductase, glutamine synthase, glutamate synthase and glutamate dehydrogenase in fine root of the SUG treatment was improved by 41.2%, 76.6%, 30.7% and 125.8%, respectively, while the ratio of GS to GDH decreased. Not only the ground diameter and plant height, but also the dry matter accumulation of aboveground and underground parts was remarkably enhanced under the SUG treatment. The nitrogen use efficiency was 40.7% in the SUG treatment, being 118.8% and 44.5% higher than the U and SUM treatments, respectively. Gel made of SAP and urea was able to significantly increase the nitrogen use efficiency, promote the growth and enhance the ability of drought resistance by improving the morphological characteristics, enhancing the absorption area and key enzymes activities of nitrogen metabolism in the fine root of P. orientalis bareroot seedlings.

By using polyethylenimine molecules as the linker, l-cysteine was immobilized onto graphene nanosheets, endowing the biocompatible l-cysteine-functionalized graphene film with the ability for catalytic decomposition of exogenous or endogenous donors to generate nitric oxide, and thus inhibiting the platelet activation and aggregation and reducing platelet adhesion.

Recently, a combination of chemotherapy with photothermal therapy (PTT) has received great attention for the construction of a near infrared (NIR)-controlled drug-delivery system for synergistic treatment of cancer, ultimately resulting in the enhancement of the therapeutic efficacy of anticancer drugs. Here, we developed a novel system for synergistic cancer therapy based on bismuth sulfide (Bi2S3) nanoparticle-decorated graphene functionalized with polyvinylpyrrolidone (PVP) (named PVP-rGO/Bi2S3). The as-prepared PVP-rGO/Bi2S3 nanocomposite has a high storage capacity for anticancer drugs (∼500% for doxorubicin (DOX)) and simultaneously has perfect photothermal conversion efficiency in the NIR region. The results of the in vitro accumulative drug release test manifests that the PVP-rGO/Bi2S3 nanocomposite could be applied as a dual pH- and NIR-responsive nanotherapeutic carrier for the controlled release of DOX from DOX-loaded PVP-rGO/Bi2S3 (PVP-rGO/Bi2S3@DOX). Moreover, the treatment of both cancer cells (including Hela, MCF-7, HepG2 and BEL-7402 cells) and BEL-7402 tumor-bearing mice with the PVP-rGO/Bi2S3@DOX complex followed by NIR laser irradiation produces significantly greater inhibition of cancer cell growth than the treatment with NIR irradiation alone or DOX alone, exhibiting a synergistic antitumor effect. Furthermore, due to the obvious NIR and X-ray absorption ability, the PVP-rGO/Bi2S3 nanocomposite could be employed as a dual-modal contrast agent for both photoacoustic tomography and X-ray computed tomography imaging. In addition to the good biocompatibility, the PVP-rGO/Bi2S3 nanocomposite paves a potential way for the fabrication of theranostic agents for dual-modal imaging-guided chemo-photothermal combined cancer therapy.

Spray-freeze-drying (SFD) of oleanolic acid (OA), a BCS Class IV compound, with polyvinylpyrrolidone-40 (PVP-40) as stabilizer and sodium caprate (SC) as wetting agent and penetration enhancer produced kinetically stable, amorphous solid dispersion systems with superior in vitro dissolution performance, and better and more uniform absorption in comparison with commercial OA tablet. Relative to the SC-free formulation, the presence of SC in the formulation resulted in a significant increase in the in vivo absorption rate of OA while exerting no apparent impact on the extent of OA absorption. The SFD-processed OA formulations and commercial OA tablet generally exhibited large inter-animal variability in oral bioavailability, consistent with the absorption characteristics of BCS Class IV compounds. Inclusion of SC coupled with the replacement of OA with its sodium salt (OA-Na) in the formulation was shown to substantially decrease the observed absorption variability. Above results suggested that increases in both dissolution rate and intestinal permeability of BCS Class IV compounds, as exemplified by the SFD-processed dispersion system containing both OA-Na and SC, are critical to reducing the large inter-individual absorption variability commonly observed with this class of drugs.
