RESUMO

Investigating the influence of the ambient chemical environment on molecular behaviors in liposomes is crucial for understanding and manipulating cellular vitality as well as the capabilities of lipid drug carriers in various environments. Here, we designed and synthesized a second harmonic generation (SHG) and fluorescence probe molecule called Pyr-Py+-N+ (PPN), which possesses membrane-targeting capability. We employed PPN to investigate the response of lipid vesicles composed of cardiolipin to the presence of exogenous salt. The kinetic behaviors, including the adsorption and embedding of PPN on the surface of small unilamellar vesicles (SUVs) composed of cardiolipin, were analyzed. The response of the SUVs to the addition of NaCl was also monitored. A rapid decrease in vesicle size can be evidenced through the rapid drop in SHG emission originating from PPN located on the vesicle surface.

OBJECTIVE: To investigate the expression of SCD1 in TFE3-rRCC and its effect on the proliferation and migration of TFE3-rRCC cells. METHODS: GEPIA database was used to analyze the expression level of SCD1 in different tumors and its effect on the prognosis of patients. The expression levels of SCD1 in TFE3-rRCC patients and cell lines UOK109 and UOK120 were detected by QPCR and Western blot. Liposomal shRNA was used to knock down SCD1 expression in cell lines. The changes of cell proliferation and migration ability before and after SCD1 knockdown were detected by CCK-8 and Transwell experiments. RESULTS: SCD1 expression levels were higher in all three common renal cancers, and patients with high SCD1 expression had shorter survival and worse prognosis (Logrank P<0.001). The mRNA and protein levels of SCD1 were also significantly increased in renal cancer tissues of patients with high expression of TFE3 and in TFE3-rRCC cell lines UOK109 and UOK120. After SCD1 knockdown, the proliferation and migration ability of UOK109 and UOK120 cells decreased significantly. CONCLUSION: SCD1 is highly expressed in TFE3-rRCC and can promote the proliferation and migration of TFE3-rRCC cell lines, which may be a key molecule in promoting the development of TFE3-rRCC.

Bimetallic alloy nanomaterials are promising anode materials for potassium-ion batteries (KIBs) due to their high electrochemical performance. The most well-adopted fabrication method for bimetallic alloy nanomaterials is tube furnace annealing (TFA) synthesis, which can hardly satisfy the trade-off among granularity, dispersity and grain coarsening due to mutual constraints. Herein, we report a facile, scalable and ultrafast high-temperature radiation (HTR) method for the fabrication of a library of ultrafine bimetallic alloys with narrow size distribution (≈10-20 nm), uniform dispersion and high loading. The metal-anchor containing heteroatoms (i.e., O and N), ultrarapid heating/cooling rate (≈103  K s-1 ) and super-short heating duration (several seconds) synergistically contribute to the successful synthesis of small-sized alloy anodes. As a proof-of-concept demonstration, the as-prepared BiSb-HTR anode shows ultrahigh stability indicated by negligible degradation after 800 cycles. The in situ X-ray diffraction reveals the K+ storage mechanism of BiSb-HTR. This study can shed light on the new, rapid and scalable nanomanufacturing of high-quality bimetallic alloys toward extended applications of energy storage, energy conversion and electrocatalysis.

AIMS: Denture use may potentially increase the risk of cardiometabolic diseases (CMDs), but the casual relevance and strength of the associations are currently unknown. METHODS AND RESULTS: A total of 495 938 participants from the UK Biobank were included in the observational analyses. Linkage disequilibrium score (LDSC) regression and Mendelian randomization analyses were employed to estimate genetic correlation and the associations between the genetic liability for denture use with coronary artery disease, myocardial infarction, heart failure (HF), any stroke (AS), ischaemic stroke, haemorrhagic stroke, type 2 diabetes (T2D), and related clinical risk factors. In observational analysis, denture use was associated with 14-25% higher risks of various CMDs. The LDSC analysis found that denture use showed a positive genetic correlation with CMDs (rg 0.21-0.38). Genetic liability for denture use was associated with an elevated risk of HF [odds ratio: 1.49 (1.20-1.83)] and T2D [1.11 (1.01-1.24)]. By integrating genetic summary data of denture use with the sum of decayed, missing, and filled tooth surfaces (DMFS), a clinical measure of dental caries obtained from an independent source, genetically determined denture use/DMFS was also associated with an elevated risk of AS [1.21 (1.04-1.40)]. Furthermore, genetically predicted denture use/DMFS was significantly associated with established cardiometabolic risk factors, including HDL cholesterol, triglycerides, waist circumference, waist-to-hip ratio, and height. CONCLUSION: Our study supported potential causal associations between the genetic liability for denture use and risks for HF, AS, T2D, and related clinical risk factors. These findings may inform prevention and intervention strategies targeting dental diseases and CMDs.


This study examined the association of denture use with cardiometabolic diseases (CMDs) and related clinical risk factors through Mendelian randomization analyses using data from UK Biobank and published consortia. Genetic liability for denture use was associated with an 11­49% higher risk of heart failure, stroke, and type 2 diabetes.The potential causal relationship between denture use and CMDs was further strengthened by the associations of denture use with HDL cholesterol, triglycerides, waist circumference, waist-to-hip ratio, and height, which are among the major risk factors of CMDs.

Alginate like extracellular polymers (ALE) recovered from flocculent sludge has been identified as a kind of highly valuable biomaterials. However, the extraction protocols limit the production of biopolymers as ALE extracted from flocculent sludge is at a lower level, around 90-190 mg/g VSS. Under this circumstance, the eco-friendly and effective optimizations for the ALE extraction protocols are expected, and thus surfactants have gained an attention to enhancing the ALE extraction. With this study, different surfactants with different structures and chemical characteristics, such as sodium dodecyl sulfate (SDS), cetyltrimethylammonium bromide (CTAB) and octyl phenyl polyoxyethylene ether (Triton X-100), were experimented to improve the ALE extraction, and in turn the optimal conditions and the associated mechanisms were evaluated and figured out. The experimental results indicated that surfactants could enhance the ALE extraction but also improve the alginate purification of ALE. With the optimal dosage of surfactants, the ALE extraction increased from 124.1 mg/g VSS to about 222.8-281.9 mg/g VSS, and the alginate purify was at around 54%-70%, in which the efficiency of the ALE extraction was improved by 79.5%-127.2%. Among others, Triton X-100 had the best performance on improving the ALE extraction, followed by CTAB and SDS. The mechanisms of surfactants on enhancing the ALE extraction and improving the alginate purify can be attributed to: i) surfactants micelles, which can solubilize flocs and extracellular biopolymers; ii) similar structures of surfactants and ALE, which follows the rule of "like dissolves like"; iii) functional groups adsorption, which facilitates the ALE release from matrixes. In a word, the optimized extraction protocol by using surfactants can be effectively applied to extract ALE from flocculent sludge.

Owing to its intricate pathophysiology, impaired wound healing is one of the substantial challenges in the treatment of burn wounds (BWs). Despite the variety of conventional therapies available, morbidities associated with BWs have not subsided. Therefore, aim of the present study was to design an advanced nanotechnology-composited therapy for effectual management of BWs. Hyaluronic acid (HA)-functionalized curcumin (CUR) and quercetin (QUE) co-loaded nanoparticle (HA-CUR-QUE-CSNPs) were fabricated, optimized, characterized and evaluated for successful co-encapsulation of drugs, morphology, stability, drug release, cell proliferation, penetration across the skin, localization in the epidermis and dermis, and in vivo wound healing efficacy. Fabricated HA-functionalized CSNPs exhibited ultra-small size (177 ± 11 nm), good zeta potential (+37.0 ± 3.2 mV), high encapsulation efficiency (EE) (QUE ∼84% and CUR ∼64%) and loading capacity (LC) (QUE ∼38% and CUR ∼43%), and spherical shape with uniformly rough surface. HA-functionalized CSNPs showed a triphasic release pattern with Fickian diffusion kinetics, a time-mannered progression in MC3T3-E1 cells proliferation, improved penetration of CUR (2414 µg/cm2) and QUE (1984 µg/cm2) through stratum corneum, and good localization of drugs in the epidermis and dermis. A superior wound healing efficacy (98% wound closure rate at day 28) with marked histological signs of minimal infiltration of inflammatory cells, re-epithelization, ECM formation, fibroblast infiltration at wound site, granulation tissue formation, angiogenesis, and collagen deposition were also evidenced. This study concludes that HA-functionalization of polymeric NPs could be a promising approach to maximize skin penetration efficiency, localization of drugs in skin tissues, tissue regeneration and BWs healing.

OBJECTIVE: To find the best treatment of penile strangulation and to analyze the sexual psychological factors of the patients. METHODS: We retrospectively analyzed the experiences in removing foreign objects around the penis in 21 patients aged 19 - 61 years with the strangulation time varying from 10 hours to 4 days. The objects were mostly made of metal or plastics. RESULTS: All the objects were successfully removed, 5 of them with the help of lubricant, 4 by aspirating the corpus cavernosum, 8 by shipping with pliers, 2 with the diamond-tipped dental drill, and the other 2, which virtually defied cutting, by aspirating the corpus cavernosum following degloving surgery. CONCLUSION: In removing foreign objects around the penis, simpler methods should be given precedence over more complex ones, and for those that virtually defy cutting, the best option is degloving surgery with particular attention to the survival of the penile skin flap.

Extracellular polymeric substances (EPS) are biopolymers that can be recovered from excess sludge, which could contribute to a more sustainable wastewater treatment plant (WWTP) operation. An example is alginate like extracellular polymers (ALE) contained in the biopolymers could be a potential resource with a highly-added value. EPS extraction for ALE from aerobic granules sludge (AGS) has already been well studied and applied in the Netherlands. On the other hand, there is little attention to the recovery of biopolymers from conventional activated sludge (CAS). In this study, flocculent sludge from eight CAS-WWTPs in China was collected and their EPS/biopolymers were extracted to investigate their recovery potential, chemical & physical properties and limiting factors. The results revealed that the biopolymers extracted and purified from CAS ranged from 90 to 190 mg/g VSS. The compositional characteristics of the biopolymers were observed by FT-IR, 3D-EEM and UV-Visible spectra, demonstrating some differences in the composition and property of the biopolymers from the different WWTPs. The biopolymers had a similarity of about 60% to a commercial alginate with respect to chemical functional groups and the alginate equivalent was >400 mg/g biopolymers. Moreover, the biopolymers consisted of poly (guluronic acid) blocks (20%-30%) and poly (guluronic acid-mannuronic acid) blocks (8%-28%), and the ionic hydrogel formation tests indicated that condensed beads were immediately formed once the drops of the biopolymers came in contact with CaCl2 solution. These results demonstrated that the biopolymers extracted had a relatively high gel-forming capacity and might also have a potential application as commercial biopolymers. Furthermore, the factors influencing the biopolymers' formation such as influent substrate, nutrient content and microbial community and the related mechanisms were investigated. Among them, increasing soluble organics (SCOD) content and low nutrient content (C/N/P) in the influent could promote the biopolymers' formation. Also, different bacteria in BNR processes might have positive or negative effects on the biopolymers' formation. In conclusion, the diversity and abundance of bacteria were identified to be a crucial and decisive factor controlling biopolymers' extraction and composition.

OBJECTIVE: To evaluate the usefulness of the measurement of midnight salivary cortisol (SC) in the initial diagnosis of Cushing's syndrome (CS). METHODS: A total of 18 patients with pathologically confirmed CS were recruited in the study. Thirteen patients with pure obesity and 36 healthy people served as controls. Salivary samples at 8:00, 16:00, 24:00 and 8:00 the next day after the 1 mg dexamethasone suppression test(DST) were collected by a commercially-available saliva collection device, and assayed by Electro Chemiluminescence Immuno Assay (ECLIA). RESULTS: There were highly significant correlations between salivary cortisol and plasma cortisol (PTC) at the 4 time points. The correlation coefficient was 0.73 (0.71, 0.74, 0.78 and 0.61 at the four time points, respectively) (P < 0.05). Midnight salivary cortisol level in Cushing's Syndrome [(10.58 +/- 5.17) nmol/L] was significantly higher than healthy people [(1.55 +/- 0.60) nmol/L]. The receiver operating characteristics curve (ROC) analysis showed a sensitivity of 100.00% and specificity of 84.00% at 8:00 with the cut-off point of 6.21 nmol/L. CONCLUSION: The measurement of midnight salivary cortisol is a convenience and noninvasive screening test for the initial diagnosis of Cushing's Syndrome.

A novel electrochemical sensor has been facilely fabricated by applying composite elements consisting of nitrogen and sulfur co-doped activated graphene (N,S-AGR) and molecularly imprinted polymer (MIP). N,S-AGR was synthesized by one-pot pyrogenation of a mixture of thiourea, KOH and graphene oxide, which was introduced to improve electron transfer capability and surface area of the electrode, while the electro-polymerized MIP layer afforded simultaneous recognition and quantification of cyclophosphamide (CPA) by utilizing Fe(CN)63-/4- as probe to indicate electrical signals. Under the optimal conditions, a calibration curve of current shift versus concentration of CPA was got in the range of 8×10-12-8×10-7molL-1, and the developed sensor gave a remarkably low detection limit (LOD) of 3.4×10-12mol L-1 (S/N=3). Moreover, the as-prepared sensor illustrated other good merits like stability and selectivity, and played a role in real-time therapeutic drug monitoring after CPA administration in rabbit.

Polysaccharide-modified flocculant is a notable material in the field of wastewater treatment. Synthesis of biopolysaccharide derivatives as eco-friendly flocculants is remarkably desired for environmental protection. This work presents an efficient flocculant synthesized through copolymerization of acrylamide, sodium acrylate (AS), and dextran. Physicochemical properties of the flocculant were evaluated. Process parameters of coal-washing wastewater flocculation were tested using Response Surface Method. The application of graft polymers exhibited efficient flocculation performance at low level of flocculant dosage in alkalescent environment. The improved dextran contributes to handle industrial effluent and sanitary sewage.

OBJECTIVE: To construct an eukaryotic expression vector containing human CD40 gene for its efficient, continuous and stable expression in human umbilical vein endothelial ECV-304 cells. METHODS: The recombinant plasmid pUCD40 was digested with endonucleases to obtain human CD40 gene fragment, which was cloned into pCDNA3.1 vector to construct recombinant eukaryotic expression vector pCDNA3.1(+)/CD40. The recombinant vector was identified by enzyme digestion before introduced into ECV-304 cells via liposome, with the positive cell clones selected with G418. The stable transfection and expression of CD40 in ECV-304 cells were identified by reverse transcription (RT)-PCR, Western blotting and flow cytometry, respectively. RESULTS: Enzyme digestion analysis showed that target gene had been cloned into the recombinant vector. The transfected ECV-304 cells successfully expressed human CD40 as determined by RT-PCR and Western-blotting, and 95% of the cells were CD40-positive as shown by flow cytometry. CONCLUSION: The recombinant eukaryotic expression vector pCDNA3.1(+)/CD40 has been successfully constructed, which is capable of stable transfection and expression of CD40 in ECV-304 cells to facilitate further investigation of the roles of CD40 molecule in antiatherosclerotic drug development.

Cherubism is a rare, nonneoplastic, self-limiting fibro-osseous that occurs in children. Affected children usually appear normal at birth. Lesions are characterized by the replacement of bone with fibrovascular tissue containing many multinucleated giant cells. Most studies have reported cherubism to be familial and with bilateral involvement of the mandibles. The authors describe a nonfamilial case of cherubism, involving both the mandible and the maxilla, in a 4-year-old female child with slowly enlarging, painless, symmetrical swelling of both cheeks.Cherubism is a rare disease that is usually limited to the mandible, but the maxilla may be involved. Computed tomography scan and biopsy are helpful for early diagnosis.
