RESUMO

Berberine (BBR) is currently explored in the oral treatment of many disorders, especially in those involving inflammatory processes. Nanotechnology-based drug delivery systems are emerging as an effective approach for improving the poor oral absorption/bioavailability of BBR. To optimize the BBR immunoregulatory effects on a specific part of the gastrointestinal tract, here we describe a micro- and nanoencapsulated hybrid delivery system (MNEHDS) for colon-targeted oral delivery of BBR and test its therapeutic efficacy in a murine colitis model. The MNEHDS is formed by encapsulation of BBR-loaded poly(lactic-co-glycolic acid) nanoparticles into a pH-sensitive, BBR-pre-entrapped Eudragit FS30D matrix to form a hybrid microparticle composed of the BBR and BBR nanoparticles. Once in the colonic environment, the microencapsulated BBR is almost completely released for immediate action, while BBR nanoparticles can provide sustained release of BBR subsequent to their intestinal absorption. One dose of oral MNEHDS/BBR treatment results in significant attenuation of acute colitis induced by dextran sulfate sodium. The MNEHDS/BBR also proves to be effective during chronically induced colitis with two doses given 1 week apart. The improved efficacy is accompanied by decreased production of colon inflammation. Comparatively, oral treatment with one or two 7-day courses of free BBR has less effect on ameliorating either acute or chronic colitis. Thus, MNEHDS represents a novel delivery system for BBR, and potentially other therapeutic agents, to treat inflammatory bowel disease.

To describe special facial features of children with Williams syndrome in China by using method of three-dimensional craniofacial anthropometry. Using three-dimensional stereo photogrammetric device, 14 craniofacial anthropometric measurements were performed and five indices were calculated in 52 children with Williams syndrome and 208 age and sex matched controls of Han Chinese ethnicity. Except intercanthal width, mouth breadth, morphological face height, nasal height-breadth index, nasal breadth-depth index, morphological ear index, the Williams syndrome group under 3 years old were smaller than the control group in the other 12 variables. Compared with the control group, the Williams syndrome group aged 3-5 years old had smaller biocular breadth, nasal length, nasorostral angle, bitragal breadth, ear width, morphological ear index and face depth. The Williams syndrome group aged above 6 years old had smaller biocular breadth, nasal breadth, bitragal breadth, ear width, ear length and face depth than the control group. The craniofacial variability index of the Williams syndrome group was greater than the control group. Greater variation was found among children with Williams syndrome than normal in China, specifically at eye, nose, ear and face shape, which demonstrate the usefulness of three-dimensional stereo photogrammetric analysis in supporting accurate diagnose of the patient with Williams syndrome.

Microplastics have emerged as an important threat to terrestrial ecosystems. To date, little research has been conducted on investigating the effects of microplastics on ecosystem functions and multifunctionality. In this study, we conducted the pot experiments containing five plant communities consisting of Phragmites australis, Cynanchum chinense, Setaria viridis, Glycine soja, Artemisia capillaris, Suaeda glauca, and Limonium sinense and added polyethylene (PE) and polystyrene (PS) microbeads to the soil (contained a mixture of 1.5 kg loam and 3 kg sand) at two concentrations of 0.15 g/kg (lower concentration, hereinafter referred to as PE-L and PS-L) and 0.5 g/kg (higher concentration, hereinafter referred to as PE-H and PS-H) to explore the effects of microplastics on total plant biomass, microbial activity, nutrient supply, and multifunctionality. The results showed that PS-L significantly decreased the total plant biomass (p = 0.034), primarily by inhibiting the growth of the roots. ß-glucosaminidase decreased with PS-L, PS-H, and PE-L (p < 0.001) while the phosphatase was noticeably augmented (p < 0.001). The observation suggests that the microplastics diminished the nitrogen requirements and increased the phosphorus requirements of the microbes. The decrease in ß-glucosaminidase diminished ammonium content (p < 0.001). Moreover, PS-L, PS-H, and PE-H reduced the soil total nitrogen content (p < 0.001), and only PS-H considerably reduced the soil total phosphorus content (p < 0.001), affecting the ratio of N/P markedly (p = 0.024). Of interest, the impacts of microplastics on total plant biomass, ß-glucosaminidase, phosphatase, and ammonium content did not become larger at the higher concentration, and it is observable that microplastics conspicuously depressed the ecosystem multifunctionality, as microplastics depreciated single functions such as total plant biomass, ß-glucosaminidase, and nutrient supply. In perspective, measures to counteract this new pollutant and eliminate its impact on ecosystem functions and multifunctionality are necessary.

Considering the importance of rapid and effective detection of nitrite in food samples, herein, we report a multistep heterophase synthetic strategy for constructing crystalline-amorphous zinc/cobalt iron porous nanosheets (C-A Zn/Co-Fe PNSs) on carbon cloth. Notably, the C-A Zn/Co-Fe PNSs@CC electrochemical interface consists of interlaced carbon fibers with many uniformly distributed hybrid nanosheets containing crystalline-amorphous interfacial sites. This particular hybrid structure permits local enrichment of nitrite for enhanced nitrite capture efficiency, laying a solid foundation for the ultrasensitive detection of nitrite. In proof-of-concept experiments, we verified that C-A Zn/Co-Fe PNSs@CC could be used to analyze nitrite with good sensitivity and reproducibility. It is worth mentioning that an ultra-low LOD of 0.44 µM was obtained for the detection of nitrite. The designed electrochemical sensor worked well in complex samples such as sausages and tap water under optimal parameters.

Hand, foot, and mouth disease (HFMD) is common among children below 5 years. HFMD has a high incidence in Hubei Province, China. In this study, the Prophet model was used to forecast the incidence of HFMD in comparison with the autoregressive-integrated moving average (ARIMA) model, and HFMD incidence was decomposed into trends, yearly, weekly seasonality and holiday effect. The Prophet model fitted better than the ARIMA model in daily reported incidence of HFMD. The HFMD incidence forecast by the Prophet model showed that two peaks occurred in 2019, with the higher peak in May and the lower peak in December. Periodically changing patterns of HFMD incidence were observed after decomposing the time-series into its major components. In specific, multi-year variability of HFMD incidence was found, and the slow-down increasing point of HFMD incidence was identified. Relatively high HFMD incidences appeared in May and on Mondays. The effect of Spring Festival on HFMD incidence was much stronger than that of other holidays. This study showed the potential of the Prophet model to detect seasonality in HFMD incidence. Our next goal is to incorporate climate variables into the Prophet model to produce an accurate forecast of HFMD incidence.

Acute myocardial infarction (MI) is a common cardiovascular disease with high mortality. In this study, an injectable thermosensitive hydrogel of chitosan (CS)/dextran (DEX)/ß-glycerophosphate (ß-GP) loaded with umbilical cord mesenchymal stem cells (UCMSCs) was prepared for MI treatment. The good biocompatibility of hydrogels was confirmed by 3T3 cells and HUVECs culture study in vitro. HUVECs encapsulated in the hydrogels showed a cell delivery ability. Furthermore, the results indicated that hydrogel could encapsulate most of UCMSCs and the cells exhibited good viability in CS/ 1.0DEX/ß-GP hydrogels. The expression of cardiac markers of cTnI and Cx43 and signaling pathways of p-Akt and p-ERK1/2 were studied, and it showed that UCMSCs differentiate towards myocardium and has a great potential for therapeutic use of cardiac repair. In conclusion, the thermosensitive hydrogel of CS/1.0 DEX/ß-GP loaded UCMSCs was a promising candidate as cell delivery vehicle for cardiac repair to reconstitute damaged myocardium.

Fluoride and arsenic are inorganic contaminants that occur in the natural environment. Chronic fluoride and/or arsenic exposure can induce developmental neurotoxicity and negatively influence intelligence in children, although the underlying molecular mechanisms are poorly understood. This study explored the effects of fluoride and arsenic exposure in drinking water on spatial learning, memory and key protein expression in the ERK/CREB signaling pathway in hippocampal and cerebral cortex tissue in rat offspring. Pregnant rats were divided into four groups. Control rats drank tap water, while rats in the three exposure groups drank water with sodium fluoride (100mg/L), sodium arsenite (75mg/L), and a sodium fluoride (100mg/L) and sodium arsenite (75mg/L) combination during gestation and lactation. After weaning, rat pups drank the same solution as their mothers. Spatial learning and memory ability of pups at postnatal day 21 (PND21) and postnatal day 42 (PND42) were measured using a Morris water maze. ERK, phospho-ERK (p-ERK), CREB and phospho-CREB (p-CREB) protein expression in the hippocampus and cerebral cortex was detected using Western blot. Compared with the control pups, escape latencies increased in PND42 pups exposed to arsenic and co-exposed to fluoride and arsenic, and the short-term and long-term spatial memory ability declined in pups exposed to fluoride and arsenic, both alone and in combination. Compared with controls, ERK and p-ERK levels decreased in the hippocampus and cerebral cortex in pups exposed to combined fluoride and arsenic. CREB protein expression in the cerebral cortex decreased in pups exposed to fluoride, arsenic, and the fluoride and arsenic combination. p-CREB protein expression in both the hippocampus and cerebral cortex was decreased in pups exposed to fluoride and arsenic in combination compared to the control group. There were negative correlation between the proteins expression and escape latency periods in pups. These data indicate that exposure to fluoride and arsenic in early life stage changes ERK, p-ERK, CREB and p-CREB protein expression in the hippocampus and cerebral cortex of rat offspring at PND21 and PND 42, which may contribute to impaired neurodevelopment following exposure.
