RESUMO

The aim of the present study was to explore the risk factors of postoperative airway complications in children with oral floor mass. The first choice of auxiliary examination method for children with oral floor mass is also proposed. This retrospective study included 50 children with floor-of-mouth (FOM) masses. Medical records were reviewed, and information on age of onset, functional impacts present, age at consultation, imaging findings, history of preoperative aspiration, pathology findings, properties of biopsied fluid, treatment modality, postoperative outcomes, and operation were recorded. A total of 20 patients exhibited functional impacts such as difficulty in breathing and feeding. Ultrasound examination was performed in 28 cases; and magnetic resonance imaging, in 38 cases. The diagnosis was lymphatic malformation in 12 cases, developmental cyst in 29 cases, and solid mass in 7 cases. There were 28 cases of surgical resection, 9 cases underwent multiple puncture volume reduction followed by surgery, 11 cases treated using sclerotherapy injection, and 1 case treated using sclerotherapy injection and surgical resection. Young age, functional impact, and high grade of lymphatic duct malformation increased the risk of surgical treatment. B-scan ultrasound is the first choice for the diagnosis of FOM masses in children.

Objective.Reliable and user-friendly electrodes can continuously and real-time capture the electroencephalography (EEG) signals, which is essential for real-life brain-computer interfaces (BCIs). This study develops a flexible, durable, and low-contact-impedance polyvinyl alcohol/polyacrylamide double-network hydrogel (PVA/PAM DNH)-based semi-dry electrode for robust EEG recording at hairy scalp.Approach.The PVA/PAM DNHs are developed using a cyclic freeze-thaw strategy and used as a saline reservoir for semi-dry electrodes. The PVA/PAM DNHs steadily deliver trace amounts of saline onto the scalp, enabling low and stable electrode-scalp impedance. The hydrogel also conforms well to the wet scalp, stabilizing the electrode-scalp interface. The feasibility of the real-life BCIs is validated by conducting four classic BCI paradigms on 16 participants.Main results.The results show that the PVA/PAM DNHs with 7.5 wt% PVA achieve a satisfactory trade-off between the saline load-unloading capacity and the compressive strength. The proposed semi-dry electrode exhibits a low contact impedance (18 ± 8.9 kΩ at 10 Hz), a small offset potential (0.46 mV), and negligible potential drift (1.5 ± 0.4µV min-1). The temporal cross-correlation between the semi-dry and wet electrodes is 0.91, and the spectral coherence is higher than 0.90 at frequencies below 45 Hz. Furthermore, no significant differences are present in BCI classification accuracy between these two typical electrodes.Significance.Based on the durability, rapid setup, wear-comfort, and robust signals of the developed hydrogel, PVA/PAM DNH-based semi-dry electrodes are a promising alternative to wet electrodes in real-life BCIs.

A large amount of wastewater is generated in the processing of coffee from fruit to cup. Thermophilic high-solids co-digestion of coffee processing wastewater (CPW) and waste activated sludge (WAS) has been succeeded by anaerobic membrane bioreactor (AnMBR). Chemical oxygen demand (COD) removal efficiencies of 92 ±â€¯3% with an average methane yield of 0.28 LCH4/gCODremoved were achieved at a high solids content of 50 g/L in the AnMBR. The optimal digestion performance of 82.4% removal COD conversion to CH4 was achieved at hydraulic retention time (HRT) 10 d. Energy balance analysis revealed AnMBR has succeeded in energy positive at all the HRTs. The net energy potential (NEP) was determined to average 2.12-2.82 kJ/gCOD, amongst which the maximum NEP was achieved at HRT 15 d. These results indicated the high-solids co-digestion by AnMBR is a promising approach to maximize the bioenergy conversion from the co-substrate of CPW and WAS.

The present study aims to investigate the effect of recombinant human bone morphogenetic protein-2 (rhBMP-2) on the osteogenesis of periodontal ligament (PDL) cells. The expression vector of rhBMP-2 (pcDNA3.1-rhBMP-2) was established. PDL cells were obtained through the enzymatic digestion and tissue explant methods and verified by immunohistochemistry. Cells were classified into experimental (cells transfected with pcDNA3.1/rhBMP-2-EGFP), blank (cells with no transfection) and control group (cells transfected with empty plasmid). rhBMP-2 expression was assessed via Western blotting analysis. The mineralization ability, alkaline phosphatase (ALP) activity and level of related osteogenic biomarkers were detected to evaluate the osteogenic characteristics of PDL cells. The rhBMP-2 expression vector (pcDNA3.1-rhBMP-2) was successfully established. Primary PDL cells displayed a star or long, spindle shape. The cultured cells were long, spindle-shaped, had a plump cell body and homogeneous cytoplasm and the ellipse nucleus contained two or three nucleoli. Cells displayed a radial, sheaf-like or eddy-like arrangement after adherence growth. Immunohistochemical staining confirmed that cells originated from mesenchymal opposed to epithelium. The experimental group exhibited an enhanced mineralization ability, higher ALP activity and increased expression of rhBMP-2 and osteogenic biomarkers (Runx2, collagen type I and osteocalcin) than the blank and control group. The present study demonstrated that rhBMP-2 transfection enhances the osteogenesis of PDL cells and provides a possibility for the application of rhBMP-2 expression products in dental disease treatment.

Periodontal ligament stem cells (PDLSCs) are tissue-specific mesenchymal stem cells (MSCs), having an important role in regenerative therapy for teeth loss. Interleukin-7 (IL-7) is a key cytokine produced by stromal cells including MSCs, and exhibits specific roles for B and T cell development and osteoblasts differentiation of multiple myeloma. However, the effect of IL-7 on osteogenic differentiation of PDLSCs remains unclear. Therefore, in the present study we determined whether IL-7 affects the proliferation and osteogenic differentiation of PDLSCs in vitro and explored the associated signaling pathways for IL-7-mediated cell differentiation. The results demonstrated that the isolated human PDLSCs possessed MSCs features, highly expressing CD90, CD44, CD105, CD29 and CD73, and almost did not expressed CD34, CD45, CD11b, CD14 and CD117. IL-7 could not significantly affect the proliferation of PDLSCs, but it decreased their osteogenic differentiation and inhibited alkaline phosphatase (ALP) activity. The results of quantitative reverse transcription-polymerase chain reaction (qRT-PCR) and Western blotting exhibited that the expression levels of Runx-2, SP7 and osteocalcin (OCN) were significantly reduced by IL-7. Further studies indicated that IL-7 did not significantly change JNK, ERK1/2 and p38 protein production, but markedly suppressed their phosphorylation levels. These data suggest that IL-7 inhibits the osteogenic differentiation of PDLSCs probably via inactivation of mitogen-activated protein kinase (MAPK) signaling pathway.

OBJECTIVE: Periodontal disease is one of the most prevalent oral diseases, which is associated with inflammation of the tooth-supporting tissues. Tormentic acid (TA), a triterpene isolated from Rosa rugosa, has been reported to exert anti-inflammatory effects. The aim of this study was to investigate the anti-inflammatory effects of TA on lipopolysaccharide (LPS)-stimulated human gingival fibroblasts (HGFs). METHODS: The levels of inflammatory cytokines such as interleukin (IL)-6 and chemokines such as IL-8 were detected by enzyme-linked immunosorbent assay (ELISA). The expression of Toll-like receptor 4 (TLR4), nuclear factor kappa B (NF-κB), IκBα, p38, extracellular signal-regulated kinase (ERK), and c-Jun N-terminal kinase (JNK) was determined by Western blotting. RESULTS: The results showed that Porphyromonas gingivalis LPS significantly upregulated the expression of IL-6 and IL-8. TA inhibited the LPS-induced production of IL-6 and IL-8 in a dose-dependent manner. Furthermore, TA inhibited LPS-induced TLR4 expression; NF-κB activation; IκBα degradation; and phosphorylation of ERK, JNK, and P38. CONCLUSION: TA inhibits the LPS-induced inflammatory response in HGFs by suppressing the TLR4-mediated NF-κB and mitogen-activated protein kinase (MAPK) signalling pathway.

OBJECTIVE: To study the effect of different surface treatments and different bonding agents on the bonding strength between self-made metal stub and Vita Mark II ceramic block. METHODS: Sandblasting and grinding were used to treat the surface of metal stub; epoxy resin and alpha-cyanoacrylate ester were selected; shear bonding strength was tested and the water-resist test was carried out. RESULTS: At normal temperature, the maximum bonding strength between the surface of metal stub (treated by sandblasting) and alpha-cyanoacrylate ester (as bonding agent) was determined to be (842.60 +/- 56.06) N, but it decreased to 0 after 15-day water-resist test. The maximum bonding strength between the surface of stub (treated by grinding) and epoxy resin (as bonding agent) was determined to be (762.44 +/- 214.82) N, which was not significantly different from the maximum bonding strength of the sandblasting-treatment group (P > 0.05), and no significant change of the bonding strength was seen after 15-day water-resist test. CONCLUSION: Epoxy resin has good properties; it is the suitable bonding agent of choice for forming a firm bond between metal stub and ceramic block.

To investigate the mechanism of cartilage degradation induced by overloading in the temporomandibular joint (TMJ), the effect of cyclic mechanical compressive stress on the activity of plasminogen activator (PA) and the expression of the predominant components of the PA system were analyzed in cultured mandibular condylar chondrocytes (MCCs) in rats. MCCs were exposed to cyclic mechanical compressive stress (2000, 4000 and 6000 µ strain) at 0.5 Hz by a four­point bending system. The activity of PA was determined by hydrolysis of the chromogenic substrate H­D-Val-Leu-Lys­pNA (S­2251). The mRNA and protein expression levels of urokinase­type PA (uPA), tissue­type PA (tPA), uPA receptor (uPAR) and PA inhibitor 1 (PAI­1) were detected by qPCR and western blot analysis, respectively. Cyclic mechanical stress at 4000 and 6000 µ strain induced the expression of uPA, tPA and uPAR, and increased the activity of PA. Furthermore, cyclic mechanical stress at 6000 µ strain also inhibited the expression of PAI­1. Analysis of pericellular proteolytic activity demonstrated that PA functioned as the active enzyme in excessive mechanical stress responsiveness (e.g., 4000 and 6000 µ strain) largely via uPAR, not PAI­1. Cyclic mechanical stress at 2000 µ strain induced the expression of tPA and PAI­1; however, it did not change the activity of PA. These results suggested that the mechanical induction of uPA, tPA and uPAR upregulated PA activity, which may provide a proteolytic environment of extracellular matrix components and subsequently contribute to the cartilage degradation in TMJ osteoarthritis.

OBJECTIVE: To evaluate the accuracy of Cercon CAD/CAM system and the influence of different spacer thickness and spacer coverage area on the internal and marginal fit of the copings. METHODS: A stainless steel master model of first molar was replicated. After scanning, designs of copings were made with different spacer thickness and spacer coverage area. There were six groups: S10C70, S10C90, S30C70, S30C90, S60C70, S60C90. Measurements of the adaptation were performed with silicone impression material and an image analysis system. RESULTS: Univariate ANOVA showed that variation in the internal fit of copings was related to measuring location, spacer thickness and spacer coverage area. There was significant difference between different spacer coverage area groups, and significant difference between spacer thickness 10 microm and 30 microm groups, 10 microm and 60 microm groups, no significant difference between spacer thickness 30 microm and 60 microm groups. Mean marginal fit was (27.7+/-7.6) microm. There was statistically significant difference between both mesial, distal marginal gaps and both buccal, lingual marginal gaps. No difference between mesial and distal marginal gaps, and between buccal and lingual marginal gaps. CONCLUSION: The accuracy by Cercon CAD/CAM system for the marginal and internal fit of zirconium dioxide copings is well within the range of clinical acceptability. Spacer thickness and spacer coverage area have statistically significant influence on the internal fit.
