RESUMO

Blends of poly(lactic acid) (PLA) with poly(propylene carbonate) (PPC) are currently in the phase of intensive study due to their promising properties and environmentally friendly features. Intensive study and further commercialization of PPC-based polymers or their blends, as usual, will soon face the problem of their waste occurring in the environment, including soil. For this reason, it is worth comprehensively studying the degradation rate of these polymers over a long period of time in soil and, for comparison, in phosphate buffer to understand the difference in this process and evaluate the potential application of such materials toward agrochemical and agricultural purposes. The degradation rate of the samples was generally accompanied by weight loss and a decrease in molecular weight, which was facilitated by the presence of PPC. The incubation of the samples in the aqueous media yielded greater surface erosions compared to the degradation in soil, which was attributed to the leaching of the low molecular degradation species out of the foils. The phytotoxicity study confirmed the no toxic impact of the PPC on tested plants, indicating it as a "green" material, which is crucial information for further, more comprehensive study of this polymer toward any type of sustainable application.

RESUMO

The aim of this study was to fabricate novel microparticles (MPs) for efficient and long-term delivery of amikacin (AMI). The emulsification method proposed for encapsulating AMI employed low-molecular-weight poly(lactic acid) (PLA) and poly(lactic acid-co-polyethylene glycol) (PLA-PEG), both supplemented with poly(vinyl alcohol) (PVA). The diameters of the particles obtained were determined as less than 30 µm. Based on an in-vitro release study, it was proven that the MPs (both PLA/PVA- and PLA-PEG/PVA-based) demonstrated long-term AMI release (2 months), the kinetics of which adhered to the Korsmeyer-Peppas model. The loading efficiencies of AMI in the study were determined at the followings levels: 36.5 ± 1.5 µg/mg for the PLA-based MPs and 106 ± 32 µg/mg for the PLA-PEG-based MPs. These values were relatively high and draw parallels with studies published on the encapsulation of aminoglycosides. The MPs provided antimicrobial action against the Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and Klebsiella pneumoniae bacterial strains. The materials were also comprehensively characterized by the following methods: differential scanning calorimetry; gel permeation chromatography; scanning electron microscopy; Fourier transform infrared spectroscopy-attenuated total reflectance; energy-dispersive X-ray fluorescence; and Brunauer-Emmett-Teller surface area analysis. The findings of this study contribute toward discerning new means for conducting targeted therapy with polar, broad spectrum antibiotics.

This study focuses on the adsorption kinetics of four highly potent sex hormones (estrone (E1), 17ß-estradiol (E2), 17α-ethinylestradiol (EE2), and estriol (E3)), present in water reservoirs, which are considered a major cause of fish feminization, low sperm count in males, breast and ovarian cancer in females induced by hormonal imbalance. Herein, electrospun polymeric nanostructures were produced from cellulose acetate, polyamide, polyethersulfone, polyurethanes (918 and elastollan), and polyacrylonitrile (PAN) to simultaneously adsorbing these estrogenic hormones in a single step process and to compare their performance. These nanofibers possessed an average fiber diameter in the range 174-330 nm and their specific surface area ranged between 10.2 and 20.9 m2g-1. The adsorption-desorption process was investigated in four cycles to determine the effective reusability of the adsorption systems. A one-step high-performance liquid chromatography technique was developed to detect and quantify concurrently each hormone present in the solution. Experimental data were obtained to determine the adsorption kinetics by applying pseudo-first-order, pseudo-second-order and intraparticle diffusion models. Findings showed that E1, E2 and EE2 best fitted pseudo-second-order kinetics, while E3 followed pseudo-first-order kinetics. It was found that polyurethane Elastollan nanofibers had maximum adsorption capacities of 0.801, 0.590, 0.736 and 0.382 mg g-1for E1, E2, EE2 and E3, respectively. In addition, the results revealed that polyurethane Elastollan nanofibers had the highest percentage efficiency of estrogens removal at ∼58.9% due to its strong hydrogen bonding with estrogenic hormones, while the least removal efficiency for PAN at ∼35.1%. Consecutive adsorption-desorption cycles demonstrated that polyurethane maintained the best efficiency, even after being repeatedly used four times compared to the other polymers. Overall, the findings indicate that all the studied nanostructures have the potential to be effective adsorbents for concurrently eradicating such estrogens from the environment.

This study investigated the effect of natural antioxidants inherent to beetroot (Beta vulgaris var. Vulgaris) on the ageing of environmentally friendly plastics. Certain properties were examined in this context, comprising thermal, mechanical, and morphological properties. A visual evaluation of relevant changes in the given polymers (polylactide and polycaprolactone) was conducted during an ageing test in a UV chamber (45 °C, 70% humidity) for 720 h. The films were prepared by a casting process, in which samples with the extract of beetroot were additionally incorporated in a common filler (bentonite), this serving as a carrier for the extract. The results showed the effect of the incorporated antioxidant, which was added to stabilize the biodegradable films. Its efficiency during the ageing test in the polymers tended to exceed or be comparable to that of the reference sample.

This study is focussed on micro-encapsulation of essential oils in polylactic acid (PLA) and a poly(methyl methacrylate) (PMMA) matrix as well as blends of the same. Microspheres were prepared by the solvent evaporation technique and characterised by scanning electron microscopy (SEM), differential scanning calorimetry (DSC) and Fourier transform infra-red spectroscopy (FTIR). The encapsulation efficiencies and release profiles of the essential oils were studied by gas chromatography mass spectrometry (GC-MS) and head-space solid-phase microextraction GC-MS, respectively. Furthermore, the microspheres were tested for antibacterial activity against both Gram-negative and Gram-positive bacterial strains. The results showed that the microspheres compositions (PLA/PMMA ratio) have significant effect on their characteristics. The process adopted for preparing the microspheres promoted formation of spherical particles at the sizes of 1.5-9.5 µm. The highest encapsulation efficiency of the prepared microspheres was observed in systems consisting of linalool (81.10 ± 10.0 wt. % for PLA system and 76.0 ± 3.3 wt. % for PMMA system). Confirmation was also made that the release rate of the microspheres was affected by the size of the same.

This work is focused on preparation of novel porous type of core-shell-structured microparticles based on polylactide (shell) and poly(vinyl alcohol) cross-linked with glutaric acid (GA) (core) prepared by water-in-oil-in-water solvent evaporation technique. The microparticle systems were used as delivery systems for immobilisation of model antibacterial agent - nisin. The effect of cross-linking and the initial amount of nisin on their morphology was investigated using scanning electron microscopy, BET analysis, zeta potential measurement and Fourier transform infra-red spectroscopy. Encapsulation efficiency and release profile of nisin from the microparticles were studied by high performance liquid chromatography. Antibacterial activity of the prepared systems was tested by dilution and spread plate technique. Results showed the microparticles in the size range of 9-16 µm in diameter with spherical multi-hollow core-shell structure. The presence of cross-linking agent GA influences the release profile of the peptide and has synergistic effect on Listeria monocytogenes growth reduction.

This work describes the preparation of a novel biopolymer hydrogel based on acid whey, cellulose derivatives and polyvinyl alcohol (PVA). The hydrogel was prepared and characterized with the aim of producing an environmentally-friendly soil amendment to increase water retention capacity of the soil. The findings showed considerable swelling properties of the hydrogels depending on the PVA content and crosslinking density. The samples with PVA in a concentration 2.5 % and 5 % were more rigid, the gel fraction increased with a subsequently decrease in their swelling capacity. The hydrogels crosslinked with 15 % of citric acid demonstrated a constant swelling ratio (SR) of around 500 % within 10 swelling/drying cycles. The hydrogels crosslinked with 10 % citric acid and supplemented with 1 % of PVA showed SR of 1000-1400 % caused by less crosslinked polymer network and increased pore volume for water uptake. It was found that hydrogel with a higher gel fraction had a stable structure. Supplementing PVA at 5 % extended the period of decomposition of the hydrogel material by almost 60 % in the soil environment and soil humidity was maintained for longer. Applying 2 % of the hydrogel 5PVA to soil increased the water retention capacity by 19 %.

The polymeric cytisine-enriched fibers based on poly(3-hydroxybutyrate) were obtained using electrospinning method. The biocompatibility study, advanced thermal analysis and release of cytisine from the poly(3-hydroxybutyrate) fibers were carried out. The nanofibers' morphology was evaluated by scanning electron microscopy. The formation and description of phases during the thermal processes of fibers by the advanced thermal analysis were examined. The new quantitative thermal analysis of polymeric fibers with cytisine phases based on vibrational, solid and liquid heat capacities was presented. The apparent heat capacity of fibers was measured using the standard differential scanning calorimetry. The quantitative analysis allowed for the study of the glass transition and melting/crystallization process. The mobile amorphous fraction, degree of crystallinity and rigid amorphous fraction were determined depending on the thermal history of semicrystalline polymeric fibers. Furthermore, the cytisine dissolution behaviour was studied. It was observed that the kinetic of the release from polymeric nanofiber is delayed than for the marketed product. The immunosafety of the tested polymeric nanofibers with cytisine was confirmed by the Food and Drug Agency Guidance as well as the European Medicines Agency. The polymeric matrix with cytisine seems to be a promising candidate for the prolonged release formulation.

A set of renewable and biodegradable hydrogels based on acid whey and cellulose derivatives blended with poly(lactic acid) (PLA) were designed as eco-friendly biopolymeric material for sustainable agricultural applications. The physico-chemical properties of the hydrogel were evaluated using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and rheological measurements. The effect of the whey/polysaccharide/PLA hydrogel on soil quality improvement (water retention study, biodegradability, loading capacity and release of the fertilizers) and the growth pattern of Raphanus sativus and Phaseolus vulgaris has been also studied. The addition of PLA has been found to improve mechanical properties of the hydrogel. The introduction of 20% wt PLA extended decomposition time of hydrogels by 25% which makes the material more stable in the environment and maintaining the soil humidity for longer. The increasing the amount of PLA led to a rise in hydrogel viscosity brought about better entrapment efficiency of the fertilizers (86-92% for KNO3 and 87-96% for urea, resp.) compared to control (82% for KNO3 and 85% for urea, resp.). The novel hydrogels with swelling ratio of up to 500% showed potential as a sustainable water reservoir for plants improving water retention capacity of the soil by 30%.

Bio-based and biodegradable packaging combined with chemical sensors and indicators has attracted great attention as they can provide protection combined with information on the actual freshness of foodstuffs. In this study, we present an effective, biodegradable, mostly bio-sourced material ideal for sustainable packaging that can also be used as a smart indicator of ammonia (NH3) vapor and food spoilage. The developed material comprises a blend of poly(lactic acid) (PLA) and poly(propylene carbonate) (PPC) loaded with curcumin (CCM), which is fabricated via the scalable techniques of melt extrusion and compression molding. Due to the structural similarity of PLA and PPC, they exhibited good compatibility and formed hydrogen bonds within their blends, as proven by Fourier transform infrared (FTIR) and X-ray diffraction (XRD). Thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC) analysis confirmed that the blends were thermally stable at the used processing temperature (180 °C) with minimal crystallinity. The rheological and mechanical properties of the PLA/PPC blends were easily tuned by changing the ratio of the biopolymers. Supplementing the PLA/PCC samples with CCM resulted in efficient absorption of UV radiation, yet the transparency of the films was preserved (T700 ∼ 68-84%). The investigation of CCM extract in ethanol with the DPPH• assay demonstrated that the samples could also provide effective antioxidant action, due to the tunable release of the CCM. Analyses for water vapor and oxygen permeability showed that the PPC improved the barrier properties of the PLA/PPC blends, while the presence of CCM did not hinder barrier performance. The capacity for real-time detection of NH3 vapor was quantified using the CIELab color space analysis. A change in color of the sample from a yellowish shade to red was observed by the naked eye. Finally, a film of PLA/PPC/CCM was successfully applied as a sticker indicator to monitor the spoilage of shrimps over time, demonstrating an evident color change from yellow to light orange, particularly for the PPC-containing blend. The developed system, therefore, has the potential to serve as a cost-effective, easy-to-use, nondestructive, smart indicator for food packaging, as well as a means for NH3 gas monitoring in industrial and environmental applications.

In recent years, a great deal of attention has been paid to the combined use of multiple antitumor drugs for better cancer treatment. The aims of the study are to construct a nanoparticle drug delivery system for the co-delivery of irinotecan hydrochloride and curcumin and to develop an analytical method for simultaneously quantifying these molecules, which is essential for further studies of the co-delivered nano system. The irinotecan hydrochloride and curcumin co-delivered nanoparticle (ICN) were prepared by combinatorially entrapping them into polyethylene glycol-poly lactic acid-co-glycolic acid (PEG-PLGA) polymeric nanoparticles. A simple, sensitive and rapid high-performance liquid chromatography method was developed and validated to simultaneously quantify the compounds in the co-delivered nanoparticle system. Acetonitrile and ultrapure water containing sodium dodecyl sulfate (0.08 mol/L), disodium phosphate (Na2HPO4, 0.002 mol/L) and acetic acid (4%, v/v) were used as the mobile phase and their ratio was set at 50:50. The flow rate was set to 1.0 mL/min, and the temperature in the column oven was maintained at 40°C. The analysis was carried out at 256 and 424 nm to assess irinotecan hydrochloride and curcumin, respectively. Detectors with only one channel can also visualize both analytes in one chromatogram at 379 nm and still demonstrate acceptable sensitivity. The retention times for irinotecan hydrochloride and curium were 3.317 and 5.560 min, respectively. The method developed was confirmed to be sensitive, accurate (recovery, 100 ± 2%), precise (relative standard deviation, RSD ≤ 1%), robust and linear (R2 ≥ 0.9996) in the range from 2.05 to 1050 µg/mL. The presented method has been used to quantify irinotecan hydrochloride and curcumin in the co-delivered ICN nano system to assess the drug delivery quality of the nanoparticles and can also be used for routine analysis because of its simplicity and accuracy.

PolyElectrolyte Nanoparticles (PENs) obtained by layer-by-layer self-assembly of polycations/polyanions suffer from a lack of colloidal stability in physiological conditions. We report a simple innovative approach for increasing their stability by multiple ionic cross-linkers. Herein, a chitosan-based core was stabilized by polyanions such as tripolyphosphate and dextran sulfate at pHs of 3 (aPENs) and 8 (bPENs) to improve the quality of electrostatic interactions in the core and manage self-assembly of polyethyleneimine shell onto the core. The physicochemical properties of the particles were characterized by DLS, SEM, TEM, FT-IR, and TGA. TEM micrographs showed visible core/shell structures of bPENs. From particle size and polydispersity indices, the bPENs stability was salt concentration-dependent. The release profiles of PENs using nicotinic acid demonstrated sustained release in a pH-independent manner with a good fit of Korsmeyer-Peppas model. These results suggest that multiple ionic cross-linkers can be an efficient approach to increase the colloidal stability of PENs.

Poly(3-hydroxybutyrate) is a biopolymer used to production of implants in the human body. On the other hand, the physical and mechanical properties of poly(3-hydroxybutyrate) are compared to the properties of isotactic polypropylene what makes poly(3-hydroxybutyrate) possible substitute for polypropylene. Unfortunately, the melting point of poly(3-hydroxybutyrate) is almost equal to its degradation temperature what gives very narrow window of its processing conditions. Therefore, numerous attempts are being made to improve the poly(3-hydroxybutyrate) properties. In the present work, hybrid nanobiocomposites based on poly(3-hydroxybutyrate) as a matrix with the use of organic nanoclay - Cloisite 30B and linear polyurethane as a second filler have been manufactured. The linear polyurethane was based on diphenylmethane 4,4'-diisocyanate and diol with imidazoquinazoline rings. The obtained nanobiocomposites were characterized by X-ray diffraction, scanning and transmission electron microscopies, thermogravimetry, differential scanning calorimetry and their selected mechanical properties were tested. The resulting hybrid nanobiocomposites have intercalated/exfoliated structure. The nanobiocomposites are characterized by a higher thermal stability and a wider range of processing temperatures compared to the unfilled matrix. The plasticizing influence of nanofillers was also observed. In addition, the mechanical properties of the discussed nanobiocomposites were examined and compared to those of the unfilled poly(3-hydroxybutyrate). The new-obtained nanobiocomposites based on poly(3-hydroxybutyrate) containing 1% Cloisite 30B and 5 wt. % of the linear of polyurethane characterized the highest improvement of processing conditions. They have the biggest difference between the temperature of degradation and the onset melting temperature, about 100 °C.

Poly(lactic acid)-block-poly(oxirane)s (PLA-b-POE) of various compositions were prepared using a one-pot approach and then extended in a reaction with l-lysine diethyl ester diisocyanate, thereby forming polyester-ether-urethanes (PEU) with prolonged chains and units with increased degradability. The PEUs are processed by electrospinning to prepare degradable nanofibrous sheet materials with and without encapsulating the antibiotic Vancomycin (VAC). PLA block isomerism and POE blocks oligomeric content (1000 g/mol) affect the thermal properties, processability, nanofibrous sheet morphology, abiotic degradation, cytocompatibility, and encapsulated antibiotic release rate of prepared PEUs. Therefore, our findings provide an effective approach to tuning the functional properties of these advanced biocompatible materials. © 2019 Wiley Periodicals, Inc. RESUMO

An innovative microcarrier based on a carboxy-enriched and branched polylactic acid derivative was developed to enhance the in vitro phototoxicity of the photosensitizer and prodrug 5-aminolevulinic. Microparticles, prepared by double emulsion technique and loaded with the prodrug were carefully characterized and the effect of the polymer structure on the chemical, physical and biological properties of the final product was evaluated. Results showed that microparticles have a spherical shape and ability to allocate up to 30 µg of the photosensitizer per mg of carrier despite their difference in solubility. Release studies performed in various simulated physiological conditions demonstrate the influence of the branched structure and the presence of the additional carboxylic groups on the release rate and the possibility to modulate it. In vitro assays conducted on human epithelial adenocarcinoma cells proved the not cytotoxicity of the carriers in a wide range of concentrations. The hemocompatibility and surface proteins adsorption were evaluated at different microparticles concentrations to evaluate the safety and estimate the possible microparticles residential time in the bloodstream. The advantages, of loading 5-aminolevulinic acid in the prepared carrier has been deeply described in terms of enhanced phototoxicity, compared to the free 5-aminolevulinic acid formulation after irradiation with light at 635 nm. The obtained results demonstrate the advantages of the prepared derivative compared to the linear polylactide for future application in photodynamic therapy based on the photosensitizer 5-aminolevulinic acid.

Three biodegradable plastics materials, namely pure poly(l-lactide) (PLA), PLA with plasticizer triacetine (TAC) and the mixture PLA/polyhydroxybutyrate (PHB) and TAC were investigated concerning changes of physical properties due to biodegradation in compost at 58°C up to 16days. With rising time of degradation in compost, both number and weight molecular masses were decreasing progressively, but only marginal change of the polydispersity index was observed which indicates that biodegradation is not random process. FTIR spectroscopy revealed that in spite of the extensive decrease of molecular weight, no substantial change in chemical composition was found. The most significant modification of the spectra consisted in an appearing of the broad band in region 3100-3300cm-1, which was assigned to a formation of biofilm on the sample surfaces. This effect appeared for all three materials, however, it was much more pronounced for samples containing also triacetine. Measurement of changes in crystalline portion confirmed that amorphous phase degrades substantially faster compared to crystalline part. The plasticizer triacetine is disappearing also rather fast from the sample resulting besides other effect also in a temporary increase of Tg, which at the beginning grows almost to the value typical for PLA without plasticizer but later the Tg is decreasing due to substantial changes in molecular weight. Generally during composting, the samples keep shape for up to 8days, after that time the material disintegrates to rough powder.

In this work, nanocomplexes based on chitosan grafted by carboxy-modified polylactic acid (SPLA) were prepared with the aim of loading simultaneously two anticancer drugs - doxorubicin and 5-fluorouracil, as well as to control their release, reduce the initial burst and boost cytotoxicity. The SPLA was prepared by a polycondensation reaction, using pentetic acid as the core molecule, and linked to the chitosan backbone through a coupling reaction. Nanocomplexes loaded with both drugs were formulated by the polyelectrolyte complexation method. The structure of the SPLA was characterized by 1H NMR, while the product CS-SPLA was analyzed by FTIR-ATR to prove the occurrence of the reaction. Results showed that the diameters and ζ-potential of the nanocomplexes fall in the range 120-200nm and 20-37mV, respectively. SEM and TEM analysis confirmed the spherical shape and dimensions of the nanocomplexes. The presence of hydrophobic side chain SPLA did not influence the encapsulation efficiency of the drugs but strongly reduced the initial burst and prolonged release over time compared to unmodified chitosan. MS analysis showed that no degradation or interactions between the drugs and carrier were exhibited after loading or 24h of release had taken place, confirming the protective role of the nanocomplexes. In vitro tests demonstrated an increase in the cytotoxicity of the drugs when loaded in the prepared carriers.

Polyelectrolytic multilayers (PEMs) with enhanced antibacterial properties were built up onto commercial poly(ethylene terephthalate) (PET) films based on the layer by layer assembling of bacterial contact killing chitosan and bacterial repelling highly hydrated hyaluronic acid. The optimization of the aminolysis modification reaction of PET was carried out by the study of the mechanical properties and the surface characterization of the modified polymers. The layer by layer assembly was successfully monitored by TEM microscopy, surface zeta-potential, contact angle measurements and, after labeling with fluorescein isothiocyanate (FTIC) by absorption spectroscopy and confocal fluorescent microscopy. Beside, the stability of the PEMs was studied at physiological conditions in absence and in the presence of lysozyme and hyaluronidase enzymes. Antibacterial properties of the obtained PEMs against Escherichia coli were compared with original commercial PET.

Chitosan and chitosan-grafted polylactic acid as a matrix for BSA encapsulation in a nanoparticle structure were prepared through a polyelectrolyte complexation method with dextran sulfate. Polylactic acid was synthetized via a polycondensation reaction using the non-metal-based initiator methanesulfonic acid and grafted to the chitosan backbone by a coupling reaction, with 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide as the condensing agent. The effect of concentration of the polymer matrix utilized herein on particle diameter, ζ-potential, encapsulation efficiency, and the release kinetic of the model protein bovine serum albumin at differing pH levels was investigated. The influence of pH and ionic strength on the behavior of the nanoparticles prepared was also researched. Results showed that grafting polylactic acid to chitosan chains reduced the initial burst effect in the kinetics of BSA release from the structure of the nanoparticles. Furthermore, a rise in encapsulation efficiency of the bovine serum albumin and diminishment in nanoparticle diameter were observed due to chitosan modification. The results suggest that both polymers actually show appreciable encapsulation efficiency; and release rate of BSA. CS-g-PLA is more suitable than unmodified CS as a carrier for controlled protein delivery.

The aim of this work was to investigate the potential of an amphiphilic system comprising chitosan-grafted polylactide and carboxyl-functionalized polylactide acid as a carrier for the controlled release and co-release of two DNA alkylating drugs: doxorubicin and temozolomide. Polylactide and carboxyl-functionalized polylactide acid were obtained through direct melt polycondensation reaction, using methanesulfonic acid as a non-toxic initiator, and subsequently these were grafted to the chitosan backbone through a coupling reaction, utilizing 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide as a condensing agent. ATR-FTIR analysis and conductometric titration confirmed that a reaction between CS and PLA, PLACA2% and PLACA5% occurred. Chitosan-grafted-polylactide and polylactide-citric acid nanoparticles were prepared via the polyelectrolyte complex technique, applying dextran sulphate as a polyanion, and loaded with doxorubicin and temozolomide. The diameter of particles, ζ-potential and their relationship to temperature and pH were analysed in all formulations. Encapsulation, co-encapsulation efficiency and release studies were conducted in different physiological simulated environments and human serum. Results showed the continuous release of drugs without an initial burst in different physiological media.
