Globally, the demands for sustainably sourced functional foods like prebiotic oligosaccharides have been constantly increasing. This study assessed the potential of pineapple leaves (PL) as lignocellulosic feedstock for sustainable production of cellulose and hemicellulose-derived oligosaccharides through its hydrothermal pretreatment (HT) followed by controlled enzymatic hydrolysis. PL was subjected to HT at 160, 175, and 190 °C for 20, 30, 60, and 90 min without any catalyst for xylooligosaccharide (XOS) production, whereas, the resulting solid content after HT was subjected to controlled enzymatic hydrolysis by commercial cellulase using conduritol B epoxide (0.5-5 mM) for glucooligosaccharides (GOS) production. HT at 160 °C for 60 min resulted in maximum yield of XOS and GOS at 23.7 and 18.3 %, respectively, in the liquid phase. Controlled enzymatic hydrolysis of HT treated (160 °C) PL solids for 20 and 30 min yielded âˆ¼ 174 mg cellobiose/g dry biomass within 24 h, indicating overall high oligosaccharide production.

RESUMO

Microplastics (MPs) are plastic particles in a size ranging from 1 mm to 5 mm in diameter, and are formed by the breakdown of plastics from different sources. They are emerging environmental pollutants, and pose a great threat to living organisms. Improper disposal, inadequate recycling, and excessive use of plastic led to the accumulation of MP in the environment. The degradation of MP can be done either biotically or abiotically. In view of that, this article discusses the molecular mechanisms that involve bacteria, fungi, and enzymes to degrade the MP polymers as the primary objective. As per as abiotic degradation is concerned, two different modes of MP degradation were discussed in order to justify the effectiveness of biotic degradation. Finally, this review is concluded with the challenges and future perspectives of MP biodegradation based on the existing research gaps. The main objective of this article is to provide the readers with clear insight, and ideas about the recent advancements in MP biodegradation.

In the recent past, an increasing interest is mostly observed in using microwave and ultrasonic irradiation to aid the biological conversion of waste materials into value-added products. This study is focused on various individual impacts of microwaves and ultrasonic waves for the treatment of biomass before the synthesis of value-added products. Following, a comprehensive review of the mechanisms governing microwaves and ultrasonication as the treatment methods, their effects on biomass disruption, solubilization of organic matter, modification of the crystalline structure, enzymatic hydrolysis and production of reducing sugars was performed. However, based on the lab-scale experiments evaluated, microwaves and ultrasonication were studied to be economically and energetically ineffective despite their beneficial effects on the waste biomass. This article reviews some of the difficulties associated with using microwaves and ultrasonic irradiation for the efficient processing of waste biomasses and identified some potential directions for future study.

Lignocellulosic (LC) biomass is the most abundant renewable resource for mankind gravitating society towards sustainable solution for energy that can reduce the carbon footprint. The economic feasibility of 'biomass biorefinery' depends upon the efficiency cellulolytic enzymes which is the main crux. Its high production cost and low efficiencies are the major limitations, that need to be resolved. As the complexity of the genome increases, so does the complexity of the proteome, further facilitated by protein post-translational modifications (PTMs). Glycosylation is regarded the major PTMs and hardly any recent work is focused on importance of glycosylation in cellulase. By modifying protein side chains and glycans, superior cellulases with improved stability and efficiency can be obtained. Functional proteomics relies heavily on PTMs because they regulate activity, localization, and interactions with protein, lipid, nucleic acid, and cofactor molecules. O- and N- glycosylation in cellulases influences its characteristics adding positive attributes to the enzymes.

A hydrothermal process is a sustainable approach for biorefinery leading to conversion of lignocellulosic (LC) biomass into value-added products. This study is based on the production of xylooligosaccharides (XOS) from pineapple leaves (PL) waste by using mild acid like gluconic acid (GA). GA, when used as catalyst in hydrothermal process to produce XOS the yield improved. The above process can be integrated with bacterial cellulose (BC) production bioprocess via Komagataeibacter europaeus 14,148 where gluconic acid is produced as by-product. Maximum XOS (2-5 degree of polymerisation) yield of 67.79 % in the liquid fraction was obtained via hydrothermal treatment at 160 °C for 60 min with 5% gluconic acid concentration. It is based on the selective solubilization of hemicellulose fraction. Enzymatic hydrolysis of GA hydrothermally pretreated solid fraction of PL biomass gave 14.5 g/L glucose with 5% solid loading and 10 FPU/gds enzyme loading which was employed for Bacterial cellulose production.

Environmental concerns have taken a center stage in our lives driving the society towards biorefinery. Bioprocess development to produce valuable products utilizing waste has its own significance in circular bioeconomy and environmental sustainability. In the present study, production of bacterial cellulose using pineapple waste as carbon source by Komagataeibacter europaeus was undertaken and it was applied for removal of vanadium, a heavy metal which is generated as waste by semiconductors industry in Taiwan. Highest yield of bacterial cellulose (BC) e.i. 5.04 g/L was obtained with pineapple core hydrolysate (HS-PC) replacing glucose in HS medium. The vanadium adsorption capacity by BC produced by HS medium was 5.24 mg/g BC at pH 4 and 2.85 mg/g BC was observed on PCH medium. BC was characterised via SEM, FTIR and XRD.

The grave concerns arisen as a result of environmental pollution and diminishing fossil fuel reserves in the 21st century have shifted the focus on the use of sustainable and environment friendly alternative resources. Lignocellulosic biomass constituted by cellulose, hemicellulose and lignin is an abundantly available natural bioresource. Lignin, a natural biopolymer has over the years gained much importance as a high value material with commercial importance. The present review provides an in-depth knowledge on the journey of lignin from being considered a roadblock to a bridge connecting diverse industries with widescale applications. The successful valorization of lignin for the production of bio-based platform chemicals and fuels has been the subject of intensive investigation. A deeper understanding of lignin characteristics and factors governing the biomass conversion into valuable products can support improved biomass consumption. The components of lignocellulosic biomass might be totally transformed into a variety of value-added products with the improvements in bioprocess techniques that valorize lignin. In this review, the recent advances in the lignin extraction and depolymerization methods that may help in achieving the cost-economics of the bioprocess are summarized and compared. The industrial potential of lignin-derived products such as aromatics, biopolymers, biofuels and agrochemicals are also outlined. Additionally, assessment of the recent research trends in lignin valorization into value-added chemicals has been done and present scenario of technological-industrial applications of lignin with economic perspectives is highlighted.

This study addresses the pressing need for sustainable bioremediation solutions to combat increasing pollution challenges in alignment with sustainability development goals. The research focuses on developing a co-culture approach involving microalgae and Komagataeibacter europaeus BCRC 14148 bacterium to create a biocomposite for efficient ammonia removal. Nanocellulose, produced by the bacterium, serves as a substrate for microalgae attachment. Optimization using specific growth media ratios resulted in biocomposite yields of 4.05 ± 0.16 g/L and 3.83 ± 0.13 g/L in HS medium with fructose and glucose, respectively. The optimal conditions include a 40:60 ratio of HS-F to TAP medium, 25 ℃ incubation, 6000 Lux light intensity, pH 5.5, and a 48-hour incubation period. When applied to wastewater treatment, the biocomposite demonstrated exceptional ammonium removal efficiency at 91.64 ± 1.27 %. This co-culture-derived biocomposite offers an eco-friendly, recyclable, and effective solution for sustainable environmental bioremediation.

The need for alternative sources of energy became increasingly urgent as demand for energy and the use of fossil fuels both soared. When processed into aromatic compounds, lignin can be utilized as an alternative to fossil fuels, however, lignin's complex structure and recalcitrance make depolymerization impractical. This article presented an overview of the most recent advances in lignin conversion, including process technology, catalyst advancement, and case study-based end products. In addition to the three established methods (thermochemical, biochemical, and catalytic depolymerization), a lignin-first strategy was presented. Depolymerizing different forms of lignin into smaller phenolic molecules has been suggested using homogeneous and heterogeneous catalysts for oxidation or reduction. Limitations and future prospects of lignin depolymerization have been discussed which suggests that solar-driven catalytic depolymerization through photocatalysts including quantum dots offers a unique pathway to obtain the highly catalytic conversion of lignin.

Practically, high-yield conversion of biomass into value-added products at low cost is a primary goal for any lignocellulosic refinery. In the industrial context, the limitation in the practical adaptation of the conventional techniques practically involves multiple reactors for the conversion of biomass to bioproducts. Therefore, the present manuscript critically reviewed the advancements in one-pot reaction systems with a major focus on the scientific production of value-added products from lignocellulosic biomass. In view of that, the novelty of one-pot reactions is shown during the fractionation of biomass into their individual constituents. The importance of the direct conversion of cellulose and lignin into a range of valuable products including organic acids and platform chemicals are separately discussed. Finally, the article is concluded with the opportunities, existing troubles, and possible solutions to overcome the challenges in lignocellulosic biorefinery. This article will assist the readers to identify the economic-friendly-one-pot conversion of lignocellulosic biomass.

The bioprocessing of lignocellulosic biomass to produce bio-based products under biorefinery setup is gaining global attention. The economic viability of this biorefinery would be inclined by the efficient bioconversion of all three major constituents of lignocellulosic biomass i.e. cellulose, hemicellulose, and lignin for value-added biochemicals and biofuels production. Although the lignocellulosic biorefinery setup has a clear value proposition, the commercial success at the industrial scale is still inadequate. This can be attributed mainly to irregular biomass supply chain, market uncertainties, and scale-up challenges. Global research efforts are underway by public and private sectors to get deeper market penetration. A comprehensive account of important factors, limitations, and propositions are worth consideration for the commercial success of lignocellulosic biorefineries. In this article, the importance of integration of lignocellulosic biorefineries with existing petrochemical refineries, the technical challenges of industrialization, SWOT analysis, and future directions have been reviewed.

Consolidated bioprocessing (CBP) is characterized by a single-step production of value-added compounds directly from biomass in a single vessel. This strategy has the capacity to revolutionize the whole biorefinery concept as it can significantly reduce the infrastructure input and use of chemicals for various processing steps which can make it economically and environmentally benign. Although the proof of concept has been firmly established in the past, commercialization has been limited due to the low conversion efficiency of the technology. Either a native single microbe, genetically modified microbe or a consortium can be employed. The major challenge in developing a cost-effective and feasible CBP process is the recognition of bifunctional catalysts combining the capability to use the substrates and transform them into value-added products with high efficiency. This article presents an in-depth analysis of the current developments in CBP around the globe and the possibilities of advancements in the future.

Strategic valorization of readily available sugarcane bagasse (SB) is very important for waste management and sustainable biorefinery. Conventional SB pretreatment methods are ineffective to meet the requirement for industrial adaptation. Several past studies have highlighted different pretreatment procedures which are lacking environmentally benign characteristics and effective SB bioconversion. This article provides an in-depth review of a variety of environmentally acceptable thermochemical and biological pretreatment techniques for SB. Advancements in the conversion processes such as pyrolysis, liquefaction, gasification, cogeneration, lignin conversion, and cellulose conversion via fermentation processes are critically reviewed for the formation of an extensive array of industrially relevant products such as biofuels, bioelectricity, bioplastics, bio adsorbents, and organic acids. This article would provide comprehensive insights into several crucial aspects of thermochemical and biological conversion processes, including systematic perceptions and scientific developments for value-added products from SB valorization. Moreover, it would lead to determining efficient pretreatment and/or conversion processes for sustainable development of industrial-scale sugarcane-based biorefinery.

Interest in functional food, such as non-digestible prebiotic oligosaccharides is increasing day by day and their production is shifting toward sustainable manufacturing. Due to the presence of high carbohydrate content, lignocellulosic biomass (LCB) is the most-potential, cost-effective and sustainable substrate for production of many useful products, including lignocellulose-derived prebiotic oligosaccharides (LDOs). These have the same worthwhile properties as other common oligosaccharides, such as short chain carbohydrates digestible to the gut flora but not to humans mainly due to their resistance to the low pH and high temperature and their demand is constantly increasing mainly due to increased awareness about their potential health benefits. Despite several advantages over the thermo-chemical route of synthesis, comprehensive and updated information on the conversion of lignocellulosic biomass to prebiotic oligomers via controlled enzymatic saccharification is not available in the literature. Thus, the main objective of this review is to highlight recent advancements in enzymatic synthesis of LDOs, current challenges, and future prospects of sustainably producing prebiotic oligomers via enzymatic hydrolysis of LCB substrates. Enzyme reaction engineering practices, custom-made enzyme preparations, controlled enzymatic hydrolysis, and protein engineering approaches have been discussed with regard to their applications in sustainable synthesis of lignocellulose-derived oligosaccharide prebiotics. An overview of scale-up aspects and market potential of LDOs has also been provided.

In recent years, the increase of microplastics in the sea exerted a negative impact on coral health. This study has been undertaken to analyze the impact of microplastics on corals. Here, Goniopora columna was exposed to different concentrations of polyethylene microplastics (PE-MP). The daily polyps length and adaptability were recorded. Analysis of the zooxanthellae density and antioxidant activity of coral was done after 1, 3, 5 and 7 days. Further tissue morphology and accumulation of PE-MP were analyzed. The results showed that PE-MP at different concentrations can be adsorbed on the surface of corals and enter inside corals after 7 days. PE-MP at different concentrations reduced polyps length, adaptability and cause the changes in the density of zooxanthellae to be the reason for unbalancing of corals. PE-MP at different concentrations reduced the superoxide dismutase (SOD) activity after exposure to 1 day. PE-MP increased the catalase (CAT) activity at 100 mg/L after exposure; even after reducing the concentration has the same effect. PE-MP at various concentrations increased the glutathione peroxidase (GPx) activity after exposure to 5 and 7 days. It also increased the glutathione transferase (GST) and glutathione (GSH) activity after exposure to 5 and 7 days. PE-MP at different concentrations increased the malondialdehyde (MDA) content after exposure from 1 to 7 days. Analysis of tissue morphology and tissue accumulation shows that different concentrations of PE-MP cause mesenteric atrophy, vacuole, and accumulation in the coral mesenteric. These results indicate that the PE-MP can impact the antioxidant system and hampers the function of enzymes responsible for detoxification of G. columna, increase lipid peroxide content and also cause tissue damage through accumulating in the coral mesenteric.

Bacterial cellulose (BC) represents a novel bio-origin nonomaterial with its unique properties having diverse applications. Increased market demand and low yield are the major reason for its higher cost. Bacteria belonging to Komagataeibacter sp are the most exploited ones for BC production. Development of a cost-effective bioprocess for higher BC production is desirable. Though static fermentation modes have been majorly employed for BC production using tray fermenters, agitated mode has also been employed successfully with air-lift fermenters as well as stirred tank reactors. Bioprocess advances in recent years has led BC production to an upper level; however, challenges of aeration requirement and labor cost towards the higher end is associated with static cultivation at large scale. We have discussed the bioprocess development for BC production in recent years along with the challenges associated and the path forward.

Increasing reliance on non-renewable fuels has shifted research attention to environmentally friendly and sustainable energy sources.The inherently recalcitrant nature of lignocellulosic biomass (LCB) makes downstream processing of the bioprocess challenging. Deep eutectic solvents (DESs) are popular and inexpensive green liquids found effective for LCB valorisation. DESs have negligible vapor-pressure and are non-flammable, recyclable, cost-economic, and thermochemically stable. This review provides a detailed overview on the DESs types, properties and their role in effective delignification and enzymatic digestibility of polysaccharides for cost-effective conversion of LCB into biofuels and bioproducts. The conglomeration of DESs with assistive pretreatment techniques can augment the process of biomass deconstruction. The current challenges in upscaling the DESs-based pretreatment technology up to commercial scale is summarized, with possible solutions and future directions. These insights would fill the knowledge-gaps to towards development of lignocellulosic biorefineries and to address the global energy crisis and environment issues.

The need to develop sustainable alternatives for pretreatment and hydrolysis of lignocellulosic biomass (LCB) is a massive concern in the industrial sector today. Breaking down of LCB yields sugars and fuel in the bulk scale. If explored under nanotechnology, LCB can be refined to yield high-performance fuel sources. The toxicity and cost of conventional methods can be reduced by applying nanoparticles (NPs) in refining LCB. Immobilization of enzymes onto NPs or used in conjugation with nanomaterials would instill specific and eco-friendly options for hydrolyzing LCB. Nanomaterials increase the proficiency, reusability, and stability of enzymes. Notably, magnetic NPs have bagged their place in the downstream processing of LCB effluents due to their efficient separation and cost-effectiveness. The current review highlights the role of nanotechnology and its particles in refining LCB into various commercial precursors and value-added products. The relationship between nanotechnology and LCB refinery is portrayed effectively in the present study.

Bacterial cellulose (BC) is higher in demand due to its excellent properties which is attributed to its purity and nano size. Komagataeibacter xylinum is a model organism where BC production has been studied in detail because of its higher cellulose production capacity. BC production mechanism shows involvement of a series of sequential reactions with enzymes for biosynthesis of cellulose. It is necessary to know the mechanism to understand the involvement of regulatory proteins which could be the probable targets for genetic modification to enhance or regulate yield of BC and to alter BC properties as well. For the industrial production of BC, controlled synthesis is desired so as to save energy, hence genetic manipulation opens up avenues for upregulating or controlling the cellulose synthesis in the bacterium by targeting genes involved in cellulose biosynthesis. In this review article genetic modification has been presented as a tool to introduce desired changes at genetic level resulting in improved yield or properties. There has been a lack of studies on genetic modification for BC production due to limited availability of information on whole genome and genetic toolkits; however, in last few years, the number of studies has been increased on this aspect as whole genome sequencing of several Komagataeibacter strains are being done. In this review article, we have presented the mechanisms and the targets for genetic modifications in order to achieve desired changes in the BC production titer as well as its characteristics.

Biorefining of lignocellulosic biomass is a relatively new concept but it has strong potential to develop and partially replace the fossil derived fuels and myriad of value products to subsequently reduce the greenhouse gas emissions. However, the energy and cost intensive process of releasing the entrapped fermentable sugars is a major challenge for its commercialization. Various factors playing a detrimental role during enzymatic hydrolysis of biomass are inherent recalcitrance of lignocellulosic biomass, expensive enzymes, sub-optimal enzyme composition, lack of synergistic activity and enzyme inhibition caused by various inhibitors. The current study investigated the mechanism of enzyme inhibition during lignocellulosic biomass saccharification especially at high solid loadings. These inhibition factors are categorized into physio-chemical factors, water-soluble and -insoluble enzyme inhibitors, oligomers and enzyme-lignin binding. Furthermore, different approaches are proposed to alleviate the challenges and improve the enzymatic hydrolysis efficiency such as supplementation with surfactants, synergistic catalytic/non-catalytic proteins, and bioprocess modifications.
