RESUMO

The aim of this study was to develop and optimise a saikosaponin a and saikosaponin d compound liposome (SSa-SSd-Lip) formulation with reduced hemolysis and enhanced bioavailability. A screening experiment was done with Plackett-Burman design, and response surface methodology of five factors (EPC/SSa-SSd ratio, EPC/Chol ratio, water temperature, pH of PBS, and ultrasound time) was employed to optimise the mean diameter, entrapment efficiency of SSa and SSd, and the reduction of hemolysis for SSa-SSd-Lip. Under the optimal process conditions (EPC/SSa-SSd ratio, EPC/Chol ratio, water temperature and pH of PBS were 26.71, 4, 50 °C and 7.4, respectively), the mean diameter, the entrapment efficiency of SSa, the entrapment efficiency of SSd and the hemolysis were 203 nm, 79.87%, 86.19%, 25.16% (SSa/SSd 12.5 mg/mL), respectively. The pharmacokinetic studies showed that the SSa-SSd-Lip had increased circulation time, decreased Cl, and increased AUC, MRT and T1/2ß (p < 0.05) for both SSa and SSd after intravenous administration in comparison with solution.

RESUMO

Distribution of lignin in the cell walls of Chinese fir branches emerged in the spring season were first studied by using ultraviolet microscope based on their cell microstructure observation and lignin qualitative measurement by the lightmicroscope and confocal laser scanning microscope. The results showed that the contents of lignin are inhomogeneously distributed in different micro-areas of the cell walls. The order of lignin concentrations is the cell corner>the middle lamellar>the secondary with the absorbance values of ultraviolet wave of 0.489, 0.307 and 0.278, respectively. The result of quantitative analysis consists with that of qualitative analysis. A new measurement method was proposed to study the distribution of lignin content in wood cell walls in CFhina.

Thermotropic phase behavior of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) liposomes containing 5 mol% cholesterol, or 5 mol% stigmasterol, or 5 mol% paeonol have been investigated by differential scanning calorimetry (DSC) and synchrotron X-ray diffraction (XRD) techniques, to investigate the competitive molecular interaction among paeonol-loaded liposomes. The results show that both sterol and paeonol can incorporate into hydrophobic region and interact with acyl chains of DPPC. Both 5 mol% sterols and 5 mol% paeonol can promote the formation of rippled gel phase of DPPC liposomes at room temperature. 5 mol% paeonol can induce the occurrence of phase separation in DPPC liposomes, but 5 mol% cholesterol or 5 mol% stigmasterol cannot induce this phenomenon. Both the repeat distance and the correlation length of paeonol-poor domain are larger than those of coexisted paeonol-rich domain. Both calorimetric data and SAXS patterns show that sterols have more favorable, stabilizing interactions with DPPC than paeonol, implying that high concentrations of sterols will have a negative effect on the loading of paeonol. In addition, calorimetric data show that cholesterol have a little more favorable, stabilizing interactions with DPPC than stigmasterol. The results of this study will play an important role in optimizing the formulation of paeonol-loaded liposomes.

OBJECTIVE: To observe the immunologic changes in patients who have complications after polyacrylamide hydrogel (HPG) injection. METHODS: To compare the patients in the HPG injection group and the control group, the expressions of immunoglobulin IgG, IgM, IgA and complement C3, C4 of serum were measured by immunoturbidimetry. RESULTS: The expressions of IgG and C3 in the injection group were lower than the control group with extremely significant difference (P < 0.01). The expressions of IgM were higher than the control group with significant difference (P < 0.05). The expression of IgA and C4 had no significant differences between the two groups. The immunologic changes had no relation to the injection doses. CONCLUSIONS: Clinical application of HPG as a soft tissue filling material may cause foreign-body rejection reaction. The long-term immunologic effect of HPG needs further observation in more cases.

OBJECTIVE: To explore the reasons for the complications of polyacrylamide hydrogel injection. METHODS: 39 patients were included in this study, who had complications after polyacrylamide hydrogel injection in the breasts, nose, temproal area or depressed locus. The clinical manifestations of the complications were analyzed. RESULTS: Postoperative infection occurred in 5 cases, induration in 23, pain in 25, ulceration of the puncture points in 9, displacement in 6, galactostasis in 1, skin necrosis in 1, breast deformation in 3, bilateral asymmetry in 4, aseptic inflammation in 10, and skin-acne-like changes on the face in 4. CONCLUSION: Some complications were caused mainly by incorrect manipulation and others were relative to the injected material. The applications of polyacrylamide hydrogel as a clinical implant material need further investigations. Polyacrylamide hydrogel injection should be cautiously used.
