RESUMO

In this paper, both arginine-glycine-aspartic acid (RGD)-containing peptide and transferrin (Tf) were conjugated to the thermosensitive poly(N-isopropylacrylamide-co-propyl acrylic acid) (poly(NIPAAm-co-PAAc)) nanogel to prepare a dual-targeting drug carrier. The obtained nanogel was characterized in terms of fluorescence spectroscopy, UV-vis spectroscopy, dynamic light scattering (DLS) and transmission electron microscopy (TEM). In order to track the dual-ligand conjugated nanogel, fluorescein isothiocyanate (FITC) was further conjugated to the nanogel. A cell internalization experiment showed that the dual-ligand conjugated nanogel exhibited obviously enhanced endocytosis by HeLa cells as compared with non-tumorous cells (COS-7 cells). The drug-loaded dual-ligand conjugated nanogel could be transported efficiently into the target tumor cells and the anti-tumor effect was enhanced significantly, suggesting that the dual-ligand conjugated nanogel has great potential as a tumor targeting drug carrier.

Hydrogels with pH-sensitive poly(acrylic acid) (PAAc) chains and biodegradable acryloyl-poly(-caprolactone)-2-hydroxylethyl methacrylate (AC-PCL-HEMA) chains were designed and synthesized. The morphology of hydrogel was observed by scanning electron microscopy. The degradation of the hydrogel in the presence of Pseudomonas lipase was studied. The in vitro release of bovine serum albumin from the hydrogel was investigated. Cytotoxicity study shows that the AC-PCL-HEMA/AAc copolymer exhibits good biocompatibility. Cell adhesion and migration into the hydrogel networks were evaluated by using different cell lines. The hydrogel with a lower cross-linking density and a larger pore size exhibited a better performance for cells migration.

To develop chitosan-based efficient gene vectors, chitosans with different molecular weights were chemically modified with low molecular weight polyethylenimine. The molecular weight and composition of polyethylenimine grafted N-maleated chitosan (NMC-g-PEI) copolymers were characterized using gel permeation chromatography (GPC) and (1)H NMR, respectively. Agarose gel electrophoresis assay showed that NMC-g-PEI had good binding ability with DNA, and the particle size of the NMC-g-PEI/DNA complexes was 200-400 nm, as determined by a Zeta sizer. The nanosized complexes observed by scanning electron microscopy (SEM) exhibited a compact and spherical morphology. The NMC-g-PEI copolymers showed low cytotoxicity and good transfection activity, comparable to PEI (25 KDa) in both 293T and HeLa cell lines, except for NMC 50K-g-PEI. The results indicated that the molecular weight of NMC-g-PEI has an important effect on cytotoxicity and transfection activity, and low molecular weight NMC-g-PEI has a good potential as efficient nonviral gene vectors.

In order to investigate the influence of the continuous alkylamide sequence having pH sensitive unit on the temperature sensitivity of poly(N-isopropylacrylamide) (PNIPAAm)-based hydrogel, a monomer, N-(2-(dimethylamino) ethyl)-methacrylamide (DMAEMA), having an ethylamide group as well as an aliphatic tertiary amino group, was designed and synthesized. Hydrogels based on NIPAAm and DMAEMA were prepared via free radical polymerization. The resulted P(NIPAAm-co-DMAEMA) hydrogels were characterized in terms of maximum swelling ratio, swelling kinetics, temperature response kinetics, and effect of pH. The data obtained show that the novel hydrogels have the strong desire to respond to external temperature and pH stimuli. Importantly, because the P(NIPAAm-co-DMAEMA) hydrogels have the continuous alkylamide sequence containing isopropylamide pendant groups from PNIPAAm and ethylamide pendant groups from PDMAEMA, the incorporation of DMAEMA moiety not only provides the pH sensitivity, but also maintains the thermal properties of P(NIPAAm-co-DMAEMA) hydrogels, even as the molar percentage of DMAEMA moiety reaches 14 mol%.

Novel composite poly(N-isopropylacrylamide) (PNIPAAm) hydrogels, containing poly(N-isopropylacrylamide)-block-poly(methyl methacrylate) (PNIPAAm-b-PMMA) micelles for sustained drug delivery were prepared and characterized. Various amounts of thermal sensitive PNIPAAm-b-PMMA micelles were incorporated physically into thermosensitive PNIPAAm bulk hydrogel to form composite PNIPAAm hydrogels. The resultant composite hydrogels were characterized by scanning electron microscopy (SEM) and differential scanning calorimetry (DSC) for morphological and thermal properties, respectively. The temperature dependence of swelling ratio and response kinetics upon heating or cooling were also investigated to understand the smart properties, that is, temperature-sensitive properties of the composite PNIPAAm hydrogels. These composite PNIPAAm hydrogels exhibited a faster shrinking kinetics than the one of pure PNIPAAm hydrogels. In addition, prednisone acetate, used as model drug, was loaded into the micelles incorporated to the composite PNIPAAm hydrogels. The controlled release behavior of the novel composite PNIPAAm hydrogels at different temperatures (22 and 37 degrees C) was examined.

To investigate the temperature sensitive mechanism and develop temperature sensitive materials, a novel poly(N-isopropyl-3-butenamide) (PNIPBAm) hydrogel was designed and synthesized. The maximum swelling ratio, temperature dependence of swelling ratio and deswelling/reswelling kinetics of the resulted PNIPBAm hydrogel were characterized. The data obtained exhibited that the temperature sensitivity of the PNIPBAm hydrogel depended on the amount of crosslinker, i.e. the temperature sensitivity decreased with the increasing content of crosslinker. This novel temperature sensitive hydrogel would have the potential applications in bioengineering and biotechnology fields.

In this study, two types of BolA-like amphiphilic peptides with dual ligands comprising a tumor-targeting moiety of RGD sequence and a cell-penetrating moiety of R8 sequence are designed and synthesized as gene vectors. The BolA-structural peptide carriers can self-assemble into spherical nanoparticles with a hydrophilic core and shell, which are similar to the viral capsid and can bind plasmid DNA in an aqueous medium to form viral-mimetic complexes. It is found that the BolA-like dual ligands system exhibits significantly enhanced gene expression in both HeLa and 293T cell lines, as compared with poly(ethylenimine) PEI. These BolA-like amphiphilic peptides are promising in clinical trials of gene therapy.

The aim of this study was to evaluate the biocompatibility and biodegradability of RGD peptide hydrogel in the posterior segment of the eye as a biomaterial potentially useful for sustained drug delivery systems. RGD peptide hydrogel was injected into the vitreous cavity and suprachoroidal space of rabbit eyes. Clinical follow-up and histological observation were performed up to four weeks. The biodegradability was also evaluated by the lifetime of the hydrogel which was defined by ophthalmoscopic observation or ultrasonography. The results showed that RGD peptide hydrogel was well tolerated in the vitreous cavity and suprachoroidal space, and disappeared from the injection sites progressively. As for suprachoroidal injection, the hydrogel was clearly identified by ultrasound echography and was confirmed innoxious to the retinal vessels by fluorescein angiography. Histological observations showed that the structures of retina, choroid and other tissues around the injection site remained normal after the injection. The lifetime of the hydrogel was 25.7 ± 2.65 days and 14.3 ± 3.3 days in the vitreous cavity and suprachoroidal space, respectively. The results obtained demonstrated that RGD peptide hydrogel, which showed excellent biocompatibility and favorable biodegradability in the posterior segment of rabbit eyes, appears to be a promising biomaterial to deliver drugs focally to the choroid and the retina.

To investigate the possibility of in situ thermosensitive hydrogel formation via Michael-type addition reaction, we designed and prepared thiol- and vinyl-modified poly(N-isopropylacrylamide) (PNIPAAm)-based copolymers. When the solutions of these two kinds of PNIPAAm-based copolymers were mixed at physiological temperature (37 degrees C), a physical gelation resulting from the hydrophobic aggregation of PNIPAAm based copolymers and chemical cross-linking between thiol and vinyl functional groups or so-called chemical gelation occurred, resulting in the formation of a three-dimensional hydrogel. Because all the gelations were performed at a high temperature (above LCSTs of the PNIPAAm based copolymers), these in situ formed hydrogels presented heterogeneous network structures, resulting in an improved thermosensitivity in comparison with the conventional one.

An effective strategy was developed and demonstrated to improve the properties of thermosensitive poly(N-isopropylacrylamide) (PNIPAAm) hydrogel by using diethyl ether as a precipitation agent during the polymerization/crosslinking. Results reveal that the modified PNIPAAm hydrogels have the heterogeneous network structures and decreased LCSTs in a comparison with the normal PNIPAAm hydrogel. The modified PNIPAAm hydrogels also exhibit significantly improved sensitive properties, including fast response and stable, rapid, large magnitude oscillatory shrinking-swelling upon temperature cycles around lower critical solution temperature. In addition, fish DNA, used as a model drug, is loaded into the modified PNIPAAm hydrogels, the controlled release behaviors of the drug loaded hydrogels at different temperatures (22 and 37 degrees C) are further examined.

A series of novel hydrogels based on poly(N-isopropylacrylamide) (PNIPAAm) with pendant poly(N-(2-(dimethylamino) ethyl)-methacrylamide) (PDMAEMA) grafts were designed and synthesized. The influence of the pendant PDMAEMA grafts on the properties of the resulted hydrogels was examined in terms of morphology observed by scanning electron microscopy (SEM), thermal property characterized by differential scanning calorimetry (DSC) and shrinking/swelling kinetics upon external temperature changes. In comparison with the conventional PNIPAAm hydrogels, resulting hydrogels presented favorable pH sensitivity as well as improved thermosensitive properties, including enlarged water containing capability at room temperature and faster shrinking/swelling rate upon heating. In addition, fish DNA, used as a model drug, was loaded into the hydrogels, and the controlled release behavior of the drug-loaded hydrogels at different temperatures (22 and 37 degrees C) was further studied.

Supramolecular hydrogels self-assembled by alpha-cyclodextrin and methoxypolyethylene glycol-poly(caprolactone)-(dodecanedioic acid)-poly(caprolactone)-methoxypolyethylene glycol (MPEG-PCL-MPEG) triblock polymers were prepared and characterized in vitro and in vivo. The sustained release of dextran-fluorescein isothiocyanate (FITC) from the hydrogels lasted for more than 1 month, which indicated that the hydrogels were promising for controlled drug delivery. ECV304 cells and marrow mesenchymal stem cells (MSC) were encapsulated and cultured in the hydrogels, during which the morphologies of the cells could be kept. The in vitro cell viability studies and the in vivo histological studies demonstrated that the hydrogels were non-cytotoxic and biocompatible, which indicated that the hydrogels prepared were promising candidates as injectable scaffolds for tissue engineering applications.

A novel class of functional poly(N-isopropylacrylamide) (PNIPAAm) hydrogels with pendent micellar structure resulting from the pending amphiphilic polymers was designed and prepared. The influence of the pendent micellar structure on the properties of the resulted PNIPAAm hydrogels was examined in terms of morphology observed by scanning electron microscopy, thermal response through differential scanning calorimetry, and deswelling/reswelling kinetics upon external temperature changes. In comparison with the conventional ones, the novel PNIPAAm hydrogels with pendent micellar structure presented improved temperature-sensitive properties, i.e., enlarged water containing capability at room temperature, as well as improved deswelling rate upon heating.
