RESUMO

Enhancing stalk strength is a crucial strategy to reduce lodging. We identified a maize inbred line, QY1, with superior stalk mechanical strength. Comprehensive analyses of the microstructure, cell wall composition, and transcriptome of QY1 were performed to elucidate the underlying factors contributing to its increased strength. Notably, both the vascular bundle area and the thickness of the sclerenchyma cell walls in QY1 were significantly increased. Furthermore, analyses of cell wall components revealed a significant increase in cellulose content and a notable reduction in lignin content. RNA sequencing (RNA-seq) revealed changes in the expression of numerous genes involved in cell wall synthesis and modification, especially those encoding pectin methylesterase (PME). Variations in PME activity and the degree of methylesterification were noted. Additionally, glycolytic efficiency in QY1 was significantly enhanced. These findings indicate that QY1 could be a valuable resource for the development of maize varieties with enhanced stalk mechanical strength and for biofuel production.

RESUMO

We report an integrated ratiometric lysosomal nitric oxide (NO) nanoprobe based on engineered semiconducting polymer dots (Pdots), LyNO-Pdots, which consist of a newly designed NO-responsive dye, a fluorescent conjugated polymer and two functional polymers. The developed probe LyNO-Pdots exhibit high specificity and stability, good photostability and favorable blood-brain barrier (BBB) penetration ability. The LyNO-Pdots are successfully applied to ratiometric imaging of lysosomal NO variations in brain-derived endothelial cells, brain tissues and mice brains with Alzheimer's disease (AD). The results demonstrate that the NO content in the brains of AD mice is considerably higher than that in normal mice.

INTRODUCTION: Exaggerated neutrophil recruitment and activation are the major features of pathological alterations in periodontitis, in which neutrophil extracellular traps (NETs) are considered to be responsible for inflammatory periodontal lesions. Despite the critical role of NETs in the development and progression of periodontitis, their specific functions and mechanisms remain unclear. OBJECTIVES: To demonstrate the important functions and specific mechanisms of NETs involved in periodontal immunopathology. METHODS: We performed single-cell RNA sequencing on gingival tissues from both healthy individuals and patients diagnosed with periodontitis. High-dimensional weighted gene co-expression network analysis and pseudotime analysis were then applied to characterize the heterogeneity of neutrophils. Animal models of periodontitis were treated with NETs inhibitors to investigate the effects of NETs in severe periodontitis. Additionally, we established a periodontitis prediction model based on NETs-related genes using six types of machine learning methods. Cell-cell communication analysis was used to identify ligand-receptor pairs among the major cell groups within the immune microenvironment. RESULTS: We constructed a single-cell atlas of the periodontal microenvironment and obtained nine major cell populations. We further identified a NETs-related subgroup (NrNeu) in neutrophils. An in vivo inhibition experiment confirmed the involvement of NETs in gingival inflammatory infiltration and alveolar bone absorption in severe periodontitis. We further screened three key NETs-related genes (PTGS2, MME and SLC2A3) and verified that they have the potential to predict periodontitis. Moreover, our findings revealed that gingival fibroblasts had the most interactions with NrNeu and that they might facilitate the production of NETs through the MIF-CD74/CXCR4 axis in periodontitis. CONCLUSION: This study highlights the pathogenic role of NETs in periodontal immunity and elucidates the specific regulatory relationship by which gingival fibroblasts activate NETs, which provides new insights into the clinical diagnosis and treatment of periodontitis.

Oily sewage has made serious impact on environment and people's life, and its treatment has become a global problem to be urgently solved. Oil-water separation has been considered to be an effective method to treat oily sewage at present. In this work, an underwater super-oleophobic/super-hydrophilic membrane with oil-water separation and self-cleaning properties was fabricated by electrochemical oxidation of sodium lignosulfonate doped polypyrrole. The membrane showed super-hydrophilicity for water-removal in air and super-hydrophilicity for oil-removal underwater in both oxidation and reduction states. The oil-water separation efficiency of the membranes for different organics exceeded 98.44%, no matter in oxidation or reduction state. Moreover, the membrane still exhibited excellent performance in terms of the oil-water separation efficiency and flux after 70 cycles, which were greater than 97.18% and 70.14 L·m-2·h-1, respectively. Simultaneously, through exploration of the mechanism, it was found that the larger anion kept intact in the membrane during the redox process, which made the stability of composition and performance. Thus, the membrane with advantageous properties, including underwater super-oleophobic/super-hydrophilicity, high oil-water separation efficiency, high circulating rate and stability, has significant potential in separation and collection of oily sewage.

In order to improve the utilization of industrial lignin as an effective component for ultraviolet (UV) shielding, organic solvent (methanol, ethanol, and acetone) fractionation was applied to improve its UV absorption performance and reduce its apparent color. Physicochemical properties of lignin and lignin-based sunscreens, such as molar mass fraction, functional group content, color change and UV shielding properties, were characterized in detail by GPC, UV spectroscopy, 31P NMR and HSQC-NMR spectroscopy. The results showed that the color and UV-shielding properties of the soluble fraction were significantly superior to those of the original and insoluble fractions. Different lignin fractions were acted as the only active substance in the pure cream and its UV-shielding properties were compared. Among them, the composite sunscreen by adding 5 wt% acetone fractionated lignin had highest sun protection factor (SPF) value of 6.6, approximately 4.5 times higher than those sunscreens mixed with pristine lignin. Overall, this work offers the potential of industrial lignin in value-added applications such as UV protection and cosmetics.

Tinuvin 622, an oligomeric light stabilizer, is widely used in plastics to reduce light and heat induced degradation and extend their service life, therefore its detection is of great importance for quality control of plastic products. However, the classical analytical methods of Tinuvin 622, such as chromatography and mass spectrometry, are difficult to achieve direct qualitative and quantitative analysis, and simultaneously to obtain the molecular weight and molecular weight distribution information. Herein, we propose for the first time the combination of gel permeation chromatography with multi-angle laser light scattering as a simple and direct method to detect Tinuvin 622 in polymers and simultaneously to obtain its molecular weight distribution information. The linearity of the method was good in the concentration range of 0.1-5.0 mg/mL Tinuvin 622 with correlation coefficient (R2 = 0.9999), and the recoveries of Tinuvin 622 at three addition levels ranged from 94.0% to 98.7%, with relative standard deviations of no more than 1.73%. The proposed method has been successfully used to detect Tinuvin 622 in actual samples of polymer additives. Compared with existing analytical methods, Tinuvin 622 has a single peak shape in our method, which is easy to identify and quantify accurately; more importantly, our method can simultaneously characterize the molecular weight and molecular weight distribution of Tinuvin 622, which makes up for the shortcomings of other approaches and provides a new tool for quality monitoring of polymer additives.

Hydrogels have attracted much attention in cancer therapy and tissue engineering due to their sustained gene delivery ability. To obtain an injectable and high-efficiency gene delivery hydrogel, methoxypolyethylene glycol (MPEG) was used to conjugate with the arginine-functionalized poly(l-lysine) dendron (PLLD-Arg) by click reaction, and then the synthesized MPEG-PLLD-Arg interacted with α-cyclodextrin (α-CD) to form the supramolecular hydrogel by the host-guest interaction. The gelation dynamics, hydrogel strength and shear viscosity could be modulated by α-CD content in the hydrogel. MPEG-PLLD-Arg was confirmed to bind and deliver gene effectively, and its gene transfection efficiency was significantly higher than PEI-25k under its optimized condition. After gelation, MMP-9 shRNA plasmid (pMMP-9) could be encapsulated into the hydrogel matrix in situ and be released from the hydrogels sustainedly, as the release rate was dependent on α-CD content. The released MPEG-PLLD-Arg/pMMP-9 complex still showed better transfection efficiency than PEI-25k and induced sustained tumor cell apoptosis. Also, in vivo assays indicated that this pMMP-9-loaded supramolecular hydrogel could result in the sustained tumor growth inhibition meanwhile showed good biocompatibility. As an injectable, sustained and high-efficiency gene delivery system, this supramolecular hydrogel is a promising candidate for long-term gene therapy. STATEMENT OF SIGNIFICANCE: To realize the sustained gene delivery for gene therapy, a supramolecular hydrogel with high-efficiency gene delivery ability was prepared through the host-guest interaction between α-cyclodextrin and PEGylated arginine-functionalized poly(l-lysine) dendron. The obtained hydrogel was injectable and biocompatible with adjustable physicochemical property. More importantly, the hydrogel showed the high-efficiency and sustained gene transfection to our used cells, better than PEI-25k. The supramolecular hydrogel resulted in the sustained tumor growth inhibition meanwhile keep good biocompatibility. As an injectable, sustained and high-efficiency gene delivery system, this supramolecular hydrogel is a promising candidate in long-term gene therapy and tissue engineering.
