RESUMO

Nitro polycyclic aromatic hydrocarbons (NPAHs) and microplastics (MPs) are emerging contaminants that pose a threat to the aquatic ecosystem. Knowledge of the NPAHs and MPs interaction will help the understanding of their fate and risks in natural environment. Here, the adsorption behavior and mechanism of typical NPAHs on microplastics were investigated. The adsorption kinetic and isotherm data showed that the adsorption of NPAHs was controlled by chemical adsorption and hydrophobic partition, because of excellent fit of kinetic and isothermal equations (R2 > 0.9). The adsorption capacity (587-744 µg g-1) was largely dependent on the hydrophobicity of NPAHs. The experiment of environmental factors confirmed the important role of pollutant hydrophobicity, with 1-Npyr of the highest hydrophobicity having the greatest adsorption on MPs (adsorption rate >90%) and less affected by solution pH and ionic strength (changer <5%). In the mixture system, MPs displayed high adsorption capacity for each compound; Interestingly, because compounds with smaller size were easy to occupy the adsorption sites in the pores of MPs, the adsorption of 2-Nflu (724 µg g-1) was even greater than that of 9-Nant (713 µg g-1) and 1-Npyr (703 µg g-1). The model calculation of adsorption also shows that there is surface adsorption and hydrophobic distribution in the adsorption process. The findings provide new insights into the interactions of MPs with organic pollutants in complex environments.

RESUMO

OBJECTIVE: To assess whether Community Periodontal Index (CPI) teeth and random half-mouth methods are representative of full-mouth method in gingival bleeding examination. METHODS: Data from 1000 untreated adults (age > 18) collected in Beijing from January 2000 to January 2001 were utilized in the analysis. Half of the subjects were examined by Gingival Bleeding Index (GBI) and the other half by Sulcus Bleeding Index (SBI). The data were sorted out and calculated according to CPI teeth and random half-mouth teeth methods. The results of the 2 methods were separately compared with those of full-mouth examination. RESULTS: There was a significant difference between CPI teeth method and full-mouth examination (P < 0.05) in both GBI and SBI. In contrast, compared with full-mouth examination, significant difference was not observed in diagonal or ipsilateral half-mouth results in SBI (P > 0.05), nor in diagonal half-mouth results in GBI (P > 0.05). CONCLUSIONS: The results of CPI teeth method on gingival bleeding could not reflect the full-mouth situation. Ipsilateral and diagonal half-mouth results can serve as a substitute for full-mouth results in SBI, while diagonal half-mouth results in GBI.

RESUMO

Additive Manufactured (AM) Polyether-ether-ketone (PEEK) orthopaedic implants offer new opportunities for bone substitutes. However, owing to its chemical inertness, the integration between PEEK implants and soft tissue represents a major challenge threatening the early success of the PEEK implants. Here we investigated the influence of hydroxyapatite (HA) fillers and porous structure of AM HA/PEEK scaffolds on the integration with soft tissue through in-vitro cellular experiments and in-vivo rabbit experiments. Among the animal experiments, HA/PEEK composite scaffolds with HA contents of 0, 20 wt%, 40 wt% and pore sizes of 0.8 mm, 1.6 mm were manufactured by fused filament fabrication. The results indicated that HA promoted the proliferation and adhesion of myofibroblasts on PEEK-based composites by releasing Ca2+ to active FAK and its downstream proteins, while the surface morphology of the scaffolds was also roughened by the HA particles, both of which led to the tighter adhesion between HA/PEEK scaffolds and soft tissue in-vivo. The macroscopic bonding force between soft tissue and scaffolds was dominated by the pore size of the scaffolds but was hardly affected by neither the HA content and nor the surface morphology. Scaffolds with larger pore size bonded more strongly to the soft tissue, and the maximum bonding force reached to 5.61 ± 2.55 N for 40 wt% HA/PEEK scaffolds with pore size of 1.6 mm, which was higher than that between natural bone and soft tissue of rabbits. Although the larger pore size and higher HA content of the PEEK-based scaffolds facilitated the bonding with the soft tissue, the consequent outcome of reduced mechanical properties has to be compromised in the design of the porous PEEK-based composite implants. The present study provides engineering-accessible synergistic strategies on material components and porous architecture of AM PEEK orthopaedic implants for improving the integration with soft tissue.

RESUMO

Microplastics, as emerging pollutant, are predicted to act as carriers for organic pollutants, but the carrier role and bio-toxic effects with other pollutants in environments are poorly acknowledged. In this study, both the single and combined effects of polyethylene (PE, 10 and 40 mg/L) with the particle size of 100-150 µm and 9-Nitroanthracene (9-NAnt, 5 and 500 µg/L) on zebrafish (Danio rerio) had been investigated. The results illustrated that PE could be as 9-NAnt carrier to enter into zebrafish body, but significantly reduced the bioaccumulation of 9-NAnt, due to the occurrence of adsorption interactions between the simultaneous presence of both PE and 9-NAnt. After 4 days, the enzymes activity of cytochrome P4501A, acetylcholinesterase (AChE), superoxide dismutase (SOD), catalase (CAT), lactate dehydrogenase (LDH), and the abundance of malondialdehyde (MDA), lipid peroxide (LPO) responded strongly to low-dose PE exposure (10 mg/L). After 7 days exposure to PE-9-NAnt (40 mg/L), the P4501A activity increased significantly, but the activities of AChE and LDH were inhibited clearly, causing certain neurotoxicity and disorders of energy metabolism to zebrafish. The analysis of integrated biomarker response index (IBR) suggested that PE had greater bio-toxicity to zebrafish in all exposure groups after short-term exposure, but the PE-9-NAnt complex showed greater bio-toxicity after 7 days, which indicated that complex exposure of PE-9-NAnt had a delayed effect on the bio-toxicity of zebrafish. Furthermore, analysis of the intestinal microbiota exhibited that under the conditions of the exposure group with 9-NAnt, the relative abundance of the five dominant bacterial phyla (Proteobacteria, Firmicutes, Fusobacteriota, Bacteroidota and Verrucomicrobiota) changed greatly. Overall, this study confirmed that PE could carry 9-NAnt into fish causing bioaccumulation, but in the case of coexisting exposures, PE reduced 9-NAnt bioaccumulation, suggesting that microplastics with other emerging pollutants in chronic toxicity are probably next objects in future works.

RESUMO

OBJECTIVE: To investigate the situation of gingival bleeding in patients with untreated gingivitis. METHODS: Totally 444 patients with untreated gingivitis were enrolled in one district located in Beijing. The gingival bleeding, except the third molars, was evaluated using the gingival bleeding index (GBI). RESULTS: The sites of GBI=1 were 8.15+/-7.17 in males and 5.65+/-5.65 in females (P=0.0003). The sites of GBI=2 were 33.48+/-21.65 among subjects aged 35-50 years and 24.31+/-18.15 among subjects aged 18-29 years (P=0.03). The first left mandible molars had the highest GBI, while the right maxillary incisors got the lowest GBI. The posterior teeth, the lingual surface, and the proximal region had more sites than the anterior teeth, the buccal surface, and the central region on GBI=1, GBI=2 and total sites P<0.05. CONCLUSION: The severity of gingival bleeding is associated with age and gender, and tends to be site-specific.

RESUMO

Microplastics and Nitropolycyclic aromatic hydrocarbons (NPAHs) are two types of emerging pollutants that are strong potential threats to aquatic ecosystems and organisms. The adsorption of NPAHs on microplastics may explain the fate and effects of NPAHs in natural environments. In this study, the adsorption behavior of 9-Nitroanthrene (9-NAnt) on polyethylene (PE), polypropylene (PP) and polystyrene (PS) was investigated. Kinetic experiments revealed that 9-NAnt was inclined to be adsorbed onto microplastics, especially PE, which had a large adsorption amount of 734 µg g-1. A linear isothermal model better described the isothermal adsorption process for 9-NAnt, which indicated that a hydrophobic distribution may be the main adsorption mechanism in an aqueous solution. Water environment factors, such as the pH and ionic strength, had negligible effects on the adsorption for PE. In contrast, alkaline and high ionic strength conditions resulted in the inhibition of adsorption of PP and PS. In addition, the particle size of microplastics was negatively correlated with the log Kd of 9-NAnt, and the performance of transient aging treatments on microplastics reduced their affinity for 9-NAnt, due to the addition of oxygen-containing functional groups. Above all, hydrophobic and electrostatic processes were the main adsorption mechanisms between microplastics and 9-NAnt.

RESUMO

In this work, a dummy molecularly imprinted polymers (MIPs) were developed as the selective sorbents for preparation of the high purity gingerols from ginger for the first time. The dummy template molecule with similar structural skeleton to gingerols, N-vanillylnonanamide, has been designed and synthesized. The performance of the MIPs and non-imprinted polymers (NIPs) were evaluated including selective recognition capacity, adsorption isotherm, and adsorption kinetics. Optimization of various parameters affecting dummy molecular imprinted solid phase extraction (MISPE), such as the type and flow rate of the loading solvent, the composition and volume of the eluting solvent, and the composition and volume of the washing solvent were investigated. Gingerols with the percent recovery of 80 and the percent purity of 99.1 were obtained from the extracts of ginger by MISPE. Besides, gingerols obtained by MISPE had more powerful activity of eliminating free radical compared with extracts before extraction with the MISPE column. Application of MIPs with a high affinity toward three gingerols provides a novel method for obtaining a group of compounds which have likely active groups from natural products.

RESUMO

Porphyromonas gingivalis (Pg) is a major etiologic agent for chronic periodontitis. Tissue destruction by Pg results partly from induction of host inflammatory responses through TLR2 signaling. This work examines the role of apoptosis-associated speck-like protein containing a caspase-recruitment domain (ASC), an adaptor molecule important for TLR-mediated caspase-1 activation. Results demonstrate that ASC levels are stable upon infection of human THP1 monocytic cells with Pg but decrease after cytokine induction. Using short hairpin RNA, we demonstrate an essential role for ASC in induction of IL-1beta by TLR2, 4, and 5 agonists, live Escherichia coli, and Pg. Induction of IL-6, IL-8, IL-10, and TNF also requires ASC, but this induction is not inhibited by IL-1 receptor antagonist or caspase-1 inhibitor. Similar results in U937 indicate broad applicability of these findings. Pg-infected ASC knockdown THP1 cells exhibit reduced transcript levels and NF-kappaB activation. These results suggest a role for ASC in cytokine induction by Pg involving both caspase-1-dependent and -independent mechanisms.
