RESUMO

Despite significant advances in therapy, the 5-year survival rates for patients with advanced stage oral cancers still remains poor as an appropriate treatment has not been found yet, due to side effects of chemo/radiotherapy. Verbascoside (VB), a major bioactive constituent of the Tsoong herb, displays pharmacological properties by exhibiting anti-oxidative, anti-inflammatory and anti-cancer activities. However, the underlining function and mechanism of VB in human oral squamous cell carcinoma (OSCC) remains unclear. In this study, we show that VB significantly decreased the viability and metastasis of HN4 and HN6 tumor cells, while promoting apoptosis. A xenograft OSCC mouse model further showed that intraperitoneal injection of VB strongly inhibited growth and lung metastasis of implanted tumor cells. Immunoblot analysis confirmed that VB effectively suppressed nuclear factor (NF)-κB activation and downstream Bcl-2/Bcl-XL expression, resulting in increased OSCC cell apoptosis. In addition, VB suppressed mRNA and protein expression of matrix metalloproteinase-9 via suppression of NF-κB activation, thereby inhibiting tumor cell metastasis. Inspiringly, compared to cisplatin-treated group, VB is a biocompatible agent without signficant side effects in vivo. Collectively, our results demonstrate that VB effectively inhibits OSCC tumor cell growth and metastasis via suppression of IκB kinase complex (IKK)/NF-κB-related signaling activation, suggesting that VB has potential use as a potent anticancer agent in OSCC therapeutic strategies.

The eruption of primary teeth is a basic event during physical development of children, which is affected by heredity and environment. This study aimed to analyze the changes in primary teeth eruption among Chinese children with social development. A total of 249,264 healthy children under 2 years were extracted from the 1995, 2005, and 2015 National Survey on the Physical Growth and Development of Children in Nine Cities of China. Their primary teeth were examined and percentiles of primary teeth eruption age were calculated by probit analysis. The median primary teeth eruption age were 6.8 months, 6.7 months, 6.6 months in 1995, 2005 and 2015. Primary teeth eruption age of boys was 0.2 months, 0.3 months, 0.3 months earlier than that of girls in 1995, 2005 and 2015. Primary teeth eruption age was the earliest in children from northern region and was the latest in children from southern region, and this regional difference did not change over time. These findings suggest that primary teeth eruption age slightly advanced with social development, and their gender difference and regional difference have always existed, which supplied some data for understanding the secular trend of primary teeth development in stomatology, pediatrics, anthropology, and other related fields.

OBJECTIVE: To explore the extraction and purification technology of total saponins from the effective parts of Liriope spicata. METHODS: Orthogonal design was used. Macroporous resin was selected to separate and purify total saponin from the effective parts of Liriope spicata. The process validation was conducted. The total saponins was determined by Ultraviolet Spectrophotometry. RESULTS: The optimal extraction conditions were as follows: 10 times the amount of ethanol (70%) for each occasion and hot reflux (3 x 2 h). Total saponins was purified by D101 macroporous resin. The concentration of eluting ethanol was 50% - 70%. Ethanol (70%) was selected as the best eluent. The result of process validation was consistent with the study. CONCLUSION: The process is simple and stable enough to significantly improve the extraction rate of the effective parts. The study can provide reference for the research and production of effective parts of traditional Chinese medicine such as Liriope spicata.

In this paper, a portable fluorescence-based functional hydrogel loaded with ß-d-glucose pentaacetate (ß-D-GP) is designed for high-sensitive quantification of amine vapor and visual monitoring of freshness of shrimp. We found for the first time that amine vapor can mediate ß-D-GP to generate photoluminescent polymer-carbon nanodots (PCNDs) with good optical properties. On this basis, a functional hydrogel sensing platform is simply formed by solidifying ß-D-GP in agarose hydrogels. When exposure to the volatile amines released from the spoilage of shrimp, ß-D-GP in hydrogel is immediately mediated by amines to generate PCNDs, resulting in obvious fluorescence-based color variation of functional hydrogel. Notably, a smartphone is used to obtain digital photographs and RGB (Red/Green/Blue) information of hydrogels for on-site quantitative analysis. The gray value of G/(R + B) of hydrogel shows good linearity with trimethylamine (TMA) vapor concentration in the range of 0-59.49 × 10-9 mol dm-3. More importantly, the G/(R + B) value of functional hydrogel is successfully used to assess the freshness of shrimp. Consequently, this strategy provides a low-cost, portable fluorescence analysis device with promising applications in achieving high-sensitive, nondestructive, and on-site food safety evaluation of animal-derived aquatic products.

Context: Long-acting recombinant human growth hormone (rhGH) has transformed growth hormone deficiency (GHD) treatment. However, the possibility and rationality for flexible time regimen are pending. Objective: We studied the efficacy of biweekly versus weekly PEGylated rhGH (PEG-rhGH) therapy in GHD children. Design Setting and Patients: This multicenter, phase IV trial with a non-inferiority threshold ≥20% enrolled 585 Tanner stage I GHD children. Intervention: Subjects randomly received 0.20 mg/kg once-weekly or biweekly PEG-rhGH, or 0.25 mg/kg.w rhGH once daily for 26 weeks. Main Outcome Measure: The primary outcome was height SD scores for chronological age (HtSDSCA) at week 26 and safety measurements including adverse events (AEs), IGF-2, and IGFBP-2 changes. Results: At week 26, the median HtSDSCA changed from -2.75, -2.82, and -2.78 to -2.31, -2.43, and -2.28 with weekly and biweekly PEG-rhGH, and daily rhGH, respectively. The difference in HtSDSCA was 0.17 ± 0.28 between weekly and biweekly PEG-rhGH, and 0.17 ± 0.27 between daily rhGH and biweekly PEG-rhGH, failing the non-inferiority threshold. Nevertheless, the height velocity of children receiving biweekly PEG-rhGH reached 76.42%-90.34% and 76.08%-90.60% that of children receiving weekly PEG-rhGH and daily rhGH, respectively. The rate of AEs was comparable among the groups. No statistical difference was observed in IGF-2 and IGFBP-2 levels among the groups. IGFBP-2 levels decreased over time in all groups, with no notable difference in IGF-2 and IGFBP-2 changes among the three treatment groups. Conclusions: Although notably promoted height velocity, biweekly PEG-rhGH failed the non-inferiority threshold as compared with either weekly PEG-rhGH or daily rhGH. Compared with short-term rhGH, long-acting PEG-rhGH did not significantly increase tumor-associated IGF-2 and IGFBP-2 expressions. In this work, a new flow injection analysis (FIA) for the determination of Pb(2+) in Chinese medicinal herbs was developed. In the buffer solution of borax-NaOH (pH 10.5), Pb(2+) reacted with 2-[(5-bromo-2-pyridyl)-azo]-5-(diethyl-amino)phenol (5-Br-PADAP) to form a complex. The experimental results showed that the sensitivity was enhanced in the presence of polyethyleneglycol-800 (PG-800). The main factors affecting the determination were investigated in detail. Under the optimum conditions, the linear range and detection limit is 0.0-0.3microg/mL and 1.5ng/mL (correlation coefficient r=0.9996), respectively. The linear regression equation is A=-0.005+0.60c (microg/mL). The sample throughout is 10h(-1). Foreign substrates effects were also investigated. The proposed method has been successfully applied to the determination of lead in reference material, goldthread and lepidium seed.

OBJECTIVE: To investigate the relationship between the levels of plasma adrenaline and norepinephrine and gene polymorphism of ß1 adrenergic receptor G1165C in children with enterovirus 71 (EV71) infection in hand foot and mouth disease (HFMD). METHODS: The polymerase chain reaction (PCR) was used to detect the expression of gene polymorphism of ß1 adrenergic receptor G1165C in vitro. The levels of plasma adrenaline and norepinephrine were measured by enzyme-linked immunosorbent assay (ELISA). RESULTS: The plasma norepinephrine level of severe group was significantly higher than the mild group in children with EV71 infection in HFMD (P < 0.05); however, the levels of plasma adrenaline in two groups had no statistical differences (P > 0.05); There was no significant difference in the distribution of ß1 adrenergic receptor G1165C genotype and allele between EV71 infection group and healthy control group (P > 0.05). Further analysis of EV71 infection group by dividing it into mild and severe groups showed that there was no significant difference in the distribution of genotype and allele between these two groups as well (P > 0.05). There was no significant difference in the levels of epinephrine and norepinephrine in different genotypes of EV71 infection group (P > 0.05), and in the levels of plasma epinephrine and norepinephrine in the mild and severe groups (P > 0.05). CONCLUSIONS: As the disease gets worse, the plasma norepinephrine level has a rising trend in children with EV71 infection in HFMD, which is an important indicator to evaluate the progress of the disease. However, the gene polymorphism of ß1 adrenergic receptor G1165C have no significant correlation, not only with the susceptibility and severity of EV71 infection in hand, foot and mouth disease, but also with the levels of catecholamine.

Low sugar concentration and the presence of various inhibitors are the major challenges associated with lignocellulosic hydrolyzates as a fermentation broth. Vacuum membrane distillation (VMD) process can be used to concentrate sugars and remove inhibitors (furans) efficiently, but it's not desirable for the removal of less volatile inhibitors such as acetic acid. In this study, a VMD-adsorption process was proposed to improve the removal of acetic acid, achieving simultaneous concentration and detoxification of lignocellulosic hydrolyzates by one step process. Results showed that sugars were concentrated with high rejections (>98%) and little sugar loss (<2%), with the significant reduction in nearly total furans (99.7%) and acetic acid (83.5%) under optimal operation conditions. Fermentation results showed the ethanol production of hydrolyzates concentrated and detoxified using the VMD-adsorption method were approximately 10-fold greater than from untreated hydrolyzates.

Cell-derived microvesicles (MVs) have been recently shown as an efficient carrier to deliver small RNAs into the target cells. In the present study, we characterized the inhibitory effect of TGF-ß1 siRNA delivered by mouse fibroblast L929 cell-derived MVs (L929 MVs) on the growth and metastasis of murine sarcomas 180 cells both in vitro and in vivo. We found that, comparing to the same concentration of free TGF-ß1 siRNA, TGF-ß1 siRNA delivered by L929 MVs much more efficiently decreased the level of TGF-ß1 in the recipient tumor cells. Functionally, MVs containing TGF-ß1 siRNA significantly decreased the viability and migration of sarcomas 180 cells and promoted the apoptosis of tumor cells. Co-immunoprecipitation with Argonaute 2 (AGO2) via anti-AGO2 antibody indicated that the majority of TGF-ß1 siRNA in the MVs were associated with AGO2 complex. A tumor implantation mouse model further showed that intravenous injection of TGF-ß1 siRNA-containing MVs strongly suppressed TGF-ß1 expression and TGF-ß1 signaling downstream in the implanted tumor cells, and thus inhibited the growth and lung metastases of tumor cells. In conclusion, our results collectively demonstrate that the delivery of therapeutic TGF-ß1 siRNA by cell-derived MVs provides an effective strategy to control tumor cell growth and metastasis.

In this study, vacuum membrane distillation (VMD) was used to remove two prototypical fermentation inhibitors (acetic acid and furfural) from lignocellulose hydrolyzates. The effect of operating parameters, such as feed temperature and feed velocity, on the removal efficiencies of inhibitors was investigated. Under optimal conditions, more than 98% of furfural could be removed by VMD. However, the removal efficiency of acetic acid was considerably lower. After furfural and acetic acid were selectively removed from hydrolyzates by VMD, ethanol production efficiency increased by 17.8% compared to original hydrolyzates.
