RESUMO

Nanoparticles (NPs) show great advantages in cancer treatment by enabling controlled and targeted delivery of payloads to tumor sites through the enhanced permeability and retention (EPR) effect. In this study, highly effective pH-responsive and biodegradable calcium orthophosphate@liposomes (CaP@Lip) NPs with a diameter of 110 ± 20 nm were designed and fabricated. CaP@Lip NPs loaded with hydrophobic paclitaxel and hydrophilic doxorubicin hydrochloride achieved excellent drug loading efficiencies of 70 and 90%, respectively. Under physiological conditions, the obtained NPs are negatively charged. However, they switched to positively charged when exposed to weak acidic environments by which internalization can be promoted. Furthermore, the CaP@Lip NPs exhibit an obvious structural collapse under acid conditions (pH 5.5), which confirms their excellent biodegradability. The "proton expansion" effect in endosomes and the pH-responsiveness of the NPs facilitate the release of encapsulated drugs from individual channels. The effectiveness and safety of the drug delivery systems were demonstrated through in vitro and in vivo experiments, with a 76% inhibition of tumor growth. These findings highlight the high targeting ability of the drug-loaded NPs to tumor sites through the EPR effect, effectively suppressing tumor growth and metastasis. By combining CaP NPs and liposomes, this study not only resolves the toxicity of CaP but also enhances the stability of liposomes. The CaP@Lip NPs developed in this study have significant implications for biomedical applications and inspire the development of intelligent and smart drug nanocarriers and release systems for clinical use.

BACKGROUND: Simethicone (SIM) is a commonly used antifoaming agent in the clinic. However, it has not been clarified whether SIM can improve the quality of intestinal preparation and the detection rates of adenomas (ADR) and polyps (PDR). This systematic review and meta-analysis were carried out to mainly evaluate the effect of SIM in bowel preparation for colonoscopy. MATERIALS AND METHODS: An electronic and a manual search of the literature for studies was conducted in PubMed, EMBASE, and Web of Science in all published data before February 1, 2020. The primary outcomes were the quality of bowel preparation and the ADR and PDR. All the data were calculated using a pooled estimate of risk ratio with 95% confidence intervals, and a random-effect model was used for the calculation. RESULTS: Eighteen randomized controlled trials with 7187 patients were included in this meta-analysis. Polyethylene glycol (PEG) with SIM improved colon cleansing (P<0.00001), PDR (P=0.006) and the detection rate of lesions in the right colon (P<0.00001) when compared with PEG alone. There was no difference in the ADR (P=0.68), withdrawal time (P=0.06), cecal intubation rate (P=0.98), and cecal intubation time (P=0.65) between 2 groups. The rate of abdominal bloating rate was higher in the PEG group, but there was no significant difference in vomiting (P=0.65), and abdominal pain (P=0.25). CONCLUSIONS: SIM improves the quality of bowel cleanliness and PDR but not ADR. Besides, SIM improves the detection rate of lesions in the right colon and decreased abdominal bloating, but do not affect vomiting and abdominal pain or cramping.

Cytochrome P450s (CYPs) are widespread proteins that interact with exogenous chemicals from the diet or the environment. CYP9A subfamily genes are important in the silkworm Bombyx mori. We previously reported transcriptional levels of two CYP9A genes in different tissues and their responses to sodium fluoride (NaF). In this study, promoter truncation analysis using a dual-luciferase reporter assay in B. mori ovary cells (BmN) showed that the regions -1,496 to -1,102 bp for CYP9A19, and -1,630 to -1,210 bp for CYP9A22 were essential for basal transcriptional activity. Sequence analysis of these regions revealed several transcriptional regulatory elements but no typical promoter elements. Promoter activities were regulated after NaF induction and with an obvious dose effect. Although the dual-luciferase assay has been widely used to determine the activity of a given promoter in cell lines, problems with it still exist. Our results indicate that both plasmid size and construct protocols affect the experimental results.

Intrinsically stretchable gas sensors possess outstanding advantages in seamless conformability and high-comfort wearability for real-time detection toward skin/respiration gases, making them promising candidates for health monitoring and non-invasive disease diagnosis and therapy. However, the strain-induced deformation of the sensitive semiconductor layers possibly causes the sensing signal drift, resulting in failure in achievement of the reliable gas detection. Herein, a surprising result that the stretchable organic polymers present a universal strain-insensitive gas sensing property is shown. All the stretchable polymers with different degrees of crystallinity, including indacenodithiophene-benzothiadiazole (PIDTBT), diketo-pyrrolo-pyrrole bithiophene thienothiophene (DPPT-TT) and poly[4-(4,4-dihexadecyl-4H-cyclopenta[1,2-b:5,4-b']dithiophen-2-yl)-alt-[1,2,5]thiad-iazolo [3,4-c] pyridine] (PCDTPT), show almost unchanged gas response signals in the different stretching states. This outstanding advantage enables the intrinsically stretchable devices to imperceptibly adhere on human skin and well conform to the versatile deformations such as bending, twisting, and stretching, with the highly strain-stable gas sensing property. The intrinsically stretchable PIDTBT sensor also demonstrates the excellent selectivity toward the skin-emitted trimethylamine (TMA) gas, with a theoretical limit of detection as low as 0.3 ppb. The work provides new insights into the preparation of the reliable skin-like gas sensors and highlights the potential applications in the real-time detection of skin gas and respiration gas for non-invasive medical treatment and disease diagnosis.

Glutathione S-transferases (GSTs) are a multifunctional super gene family, some of which play an important role in insecticide resistance. In this research, we used a real-time quantitative RT-PCR method, and a novel strategy, to measure the transcriptional level per gene copy using an exogenous RNA reference and DNA reference. The transcription levels of six BmGST genes in different tissues of fifth instar Bombyx mori larvae and their responses to insecticide and fluoride were investigated. The results show different levels and patterns of expression of the different BmGSTs in the various tissues observed. The BmGSTs can be induced by insecticide and fluoride, but their responses to each are different. The results of this research are helpful in studying the tissue-specific expression of BmGSTs in Bombyx mori, and in developing new pesticide resistant silkworm varieties.

Fabricating a high-performance adsorbent as a desirable candidate for removing Pb2+ from aqueous water remains a challenge. Aramid nanofibers (ANFs) are promising building blocks that have realized multifunctional applications due to their intrinsic mechanical and chemical stability. Herein, an in situ loading strategy for preparing nanofiber composite aerogel was proposed by assembling ANFs into a 3D aerogel and applying it as host media for the in situ polymerization of pyrrole followed by facile redox reaction between the polypyrrole (PPy) and MnO4-1 to load manganese dioxide (MnO2). The idea was to fully exploit the structural advantages of ultra-low bulk density, large specific surface area, and high porosity of ANFs, and the possible chemical adsorption characteristics of MnO2 on the basis of ion exchange reaction. The adsorption capacity of 3D ANF/MnO2 composite aerogel was as large as 554.36 mg/g for Pb2+. The adsorption mechanism based on an exchange reaction between Pb2+ and protons on the surface of MnO2 was also investigated. The desorption results showed that the adsorption performance could remain up to 90% after five times of usage. In conclusion, this research provides promising insights into the preparation of high-performance lead adsorbent for water treatment.

BACKGROUND: Chitosan-coated liposome containing both coenzyme Q10 and alpha-lipoic acid (CCAL) was a novel multifunctional nano delivery system exhibited long-term stability at room temperature and high encapsulation efficiency. Which containing two kinds of antioxidants (coenzyme Q10 and alpha-lipoic acid) and two antibacterial agents (chitosan and alpha-lipoic acid). However, the further biocompatibility, such as cytotoxicity and antioxidant activity, and antimicrobial activity of CCAL, has not been systematically evaluated. OBJECTIVES: This study aims to evaluate the cytotoxicity, antioxidant activity and antimicrobial activity of CCAL. METHODS: This article evaluated the in vitro cytotoxicity and antioxidant activity of CCAL by CCK8 assay, and antimicrobial activity on Staphylococcus aureus and Escherichia coli was investigated. RESULTS: For antioxidant activity study, CCAL displayed significant antioxidant effect when compared with control group (P<.01). Finally, CCAL also exhibited antimicrobial activity, and CCAL showed stronger bactericidal effect with Gram-positive bacteria than Gram-negative bacteria. CONCLUSIONS: CCAL could be a potential antioxidant delivery system for cosmetics or pharmaceuticals without cytotoxicity and artificial preservatives free.
