RESUMEN

The Orgeval watershed (104 km(2)) is a long-term experimental observatory and research site, representative of rural areas with intensive cereal farming of the temperate world. Since the past few years, we have been carrying out several studies on nitrate source, transformation and transfer of both surface and groundwaters in relation with land use and agriculture practices in order to assess nitrate (NO3(-)) leaching, contamination of aquifers, denitrification processes and associated nitrous oxide (N2O) emissions. A synthesis of these studies is presented to establish a quantitative diagnosis of nitrate contamination and N2O emissions at the watershed scale. Taking this watershed as a practical example, we compare curative management measures, such as pond introduction, and preventive measures, namely conversion to organic farming practices, using model simulations. It is concluded that only preventive measures are able to reduce the NO3(-) contamination level without further increasing N2O emissions, a result providing new insights for future management bringing together water-agro-ecosystems.

Increasing the overall use efficiency of nitrogen (N) and phosphorus (P) resources in food production while minimizing losses to the environment are required to meet the dual challenge of food security and sustainability. Yet studies quantifying the overall performance of different agro-system types and how these have changed over time remain rare, although they are essential to propose solution pathways. Here, we reconstructed fluxes of N and P within 78 watersheds of the St. Lawrence Basin (SLB) of eastern Canada between 1901 and 2011, using the Generalized Representation of Agro-Food System model (GRAFS). This analysis allowed us to classify different agro-food system types and to evaluate how agricultural specialization influenced nutrient efficiencies and potential losses to the environment over time. Using a cluster analysis, we identified four agro-food system types with different overall outcomes in efficiencies and losses. We show that agricultural practices in the SLB were similar until the 1950's and deemed unsustainable in several watersheds by depleting agricultural soils of their nutrients (particularly N). With the advent of manufactured fertilizers and the intensification of livestock farming, the SLB then rapidly shifted through the 1970s and 1980s to more intensified and highly unsustainable agro-food system types, where, in 2011, ~77 % of N and ~ 94 % of P inputs were lost to the environment. We also show that nutrient pollution continued to increase despite gains in the nutrient use efficiency of animal farming due to higher nutrient throughput from intensive production. The increased proportion of confined animals, disconnected from croplands, indeed resulted in inefficient nutrient recycling. While nutrient use efficiency may mitigate nutrient pollution, reducing the absolute nutrient flux through agro-food systems should be a priority, likely through a reconnection of crop and animal farming and an overall reduction of meat production, specifically from concentrated, intensive livestock systems.

Marine eutrophication in the North-East Atlantic (NEA) strongly relies on nutrient enrichment at the river outlets, which is linked to human activities and land use in the watersheds. The question is whether human society can reduce its nutrient emissions by changing land use without compromising food security. A new version of Riverstrahler model (pyNuts-Riverstrahler) was designed to estimate the point and diffuse nutrient emissions (N, P, Si) to the rivers depending on land use in the watersheds across a large domain (Western Europe agro-food systems, waste water treatment). The loads from the river model have been used as inputs to three marine ecological models (PCOMS, ECO-MARS3D, MIRO&CO) covering together a large part of the NEA from the Iberian shelf to the Southern North Sea. The modelling of the land-ocean continuum allowed quantifying the impact of changes in land use on marine eutrophication. Pristine conditions were tested to scale the current eutrophication with respect to a "natural background" (sensu WFD), i.e. forested watersheds without any anthropogenic impact. Three scenarios representing potential management options were also tested to propose future perspectives in mitigating eutrophication. This study shows that a significant decrease in nitrogen fluxes from land to sea is possible by adapting human activities in the watersheds, preventing part of the eutrophication symptoms in the NEA rivers and adjacent coastal zones. It is also shown that any significant achievement in that direction would very likely require paradigmatic changes at social, economic and agricultural levels. This requires reshaping the connections between crop production and livestock farming, and between agriculture and local human food consumption. It also involves cultural changes such as less waste production and a shift towards lower-impact and healthier diets where half of the animal products consumption is replaced by vegetal proteins consumption, known as a demitarian diet (http://www.nine-esf.org/node/281/index.html).

A model (the Riverstrahler model) is used to describe nutrient transfer and transformation at the scale of the whole drainage network of the Seine based on information concerning the basic mechanisms governing N, P and Si inputs to the drainage network and in-stream transformation and retention. It was used to calculate the budget of these nutrients through the whole river continuum from land to sea. With the help of historical documents, the constraints used as forcing function in this model were reconstructed to express the changing conditions of land-use and urban population over the last five hundred years. The corresponding scenarios were run for different hydrological regimes including dry, mean and wet conditions. The results were validated on the long-term series of nutrient measurements spanning more than a century available at some stations on the Seine, upstream and downstream of the city of Paris. The model was also used to explore past and future trends in nutrient loading, retention and delivery to the coastal zone, in response to human management of the terrestrial watershed. Beside the initial pristine state, used as an idealized reference state (with N, P and Si delivered fluxes of about 45-110 kg N km(-2) yr(-1), 2-5 kg P km(-2) yr(-1), 510-1325 kg Si km(-2) yr(-1)), four periods were distinguished. The first one is that of the traditional cottage economy which prevailed, with quite a constant total population, until the end of the 18th century. N, P and Si fluxes were about 235-750 kg N km(-2) yr(-1), 15-60 kg P km(-2) yr(-1) and 425-1280 kg Si km(-2) yr(-1), depending on hydrological conditions. The second period, from the beginning of the 19th century to about the 1950's, corresponded to rapid increase in the total and urban population with a corresponding increase of point sources of N and P. From 1950 onwards, modern farming practices resulted in a dramatic increase in diffuse sources of nitrogen and to a lesser extent phosphorus: riverine N and P export reached 1320-2800 kg N km(-2) yr(-1), and 310-340 kg P km(-2) yr(-1): silica export remained fairly constant at around 410-1260 kg Si km(-2) yr(-1) depending on the hydrological conditions. In the 1990's, the fourth period is represented by a stabilized population and improved wastewater treatment, when the export of phosphorus is reduced to values as low as 40-60 kg P km(-2) yr(-1), but without as effective a reduction of nitrogen export. This represents an unprecedented situation for the marine coastal system, i.e. a shift from nitrogen to phosphorus limitation, as nitrogen is still delivered far in excess of the amount of silica available for diatom blooms.

To explore the evolution of a human impacted river, the Seine (France), over the 21st century, three driving factors were examined: climate, agriculture, and point source inputs of domestic and industrial origin. Three future scenarios were constructed, by modification of a baseline representative of recent conditions. A climate change scenario, based on simulations by a general circulation model driven by the SRES-A2 scenario of radiative forcing, accounts for an average warming of +3.3 degrees C over the watershed and marked winter increase and summer decrease in precipitation. To illustrate a possible reduction in nitrate pollution from agricultural origin, a scenario of good agricultural practices was considered, introducing catch crops and a 20% decrease in nitrogen fertilisation. Future point source pollution was estimated following the assumptions embedded in scenario SRES-A2 regarding demographic, economic and technologic changes, leading to reductions of 30 to 75% compared to 2000, depending on the pollutants. Four models, addressing separate components of the river system (agronomical model, hydrogeological model, land surface model and water quality model), were used to analyse the relative impact of these scenarios on water quality, in light of their impact on hydrology and crop production. The first-order driving factor of water quality over the 21st century is the projected reduction of point source pollution, inducing a noticeable decrease in eutrophication and oxygen deficits downstream from Paris. The impact of climate change on these terms is driven by the warming of the water column. It enhances algal growth in spring and the loss factors responsible for phytoplankton mortality in late summer (grazers and viruses). In contrast, increased seasonal contrasts in river discharge have a negligible impact on river water quality, as do the changes in riverine nitrate concentration, which never gets limiting. The latter changes have a similar magnitude under the three scenarios. Under climate change, riverine and groundwater nitrate concentrations increase and crop production is advantaged with reduced growing cycles and increased yields. In contrast, nitrate concentrations decrease under the good agricultural practices scenario, with a limited decrease in crop production. When these two scenarios are combined, the changes in nitrate concentrations balance each other and crop yields increase. The results of this numerical exercise indicate that the potential changes to the Seine River system during the 21st century will not lead to severely degraded water quality.

A set of 90 novel 2-(arylsulfonyl)-1,2,3, 4-tetrahydroisoquinoline-3-carboxylates and -hydroxamates as inhibitors of the matrix metalloproteinase human neutrophil collagenase (MMP-8) was designed, synthesized, and investigated by 3D-QSAR techniques (CoMFA, CoMSIA) and X-ray structure analysis. Docking studies of a reference compound are based on crystal structures of MMP-8 complexed with peptidic inhibitors to propose a model of its bioactive conformation. This model was validated by a 1. 7 A X-ray structure of the catalytic domain of MMP-8. The 3D-QSAR models based on a superposition rule derived from these docking studies were validated using conventional and cross-validated r2 values using the leave-one-out method, repeated analyses using two randomly chosen cross-validation groups plus randomization of biological activities. This led to consistent and highly predictive 3D-QSAR models with good correlation coefficients for both CoMFA and CoMSIA, which were found to correspond to experimentally determined MMP-8 catalytic site topology in terms of steric, electrostatic, and hydrophobic complementarity. Subsets selected as smaller training sets using 2D fingerprints and maximum dissimilarity methods resulted in 3D-QSAR models with remarkable correlation coefficients and a high predictive power. This allowed to compensate the weaker zinc binding properties of carboxylates by introducing optimal fitting P1' residues. The final QSAR information agrees with all experimental data for the binding topology and thus provides clear guidelines and accurate activity predictions for novel MMP-8 inhibitors.

Although coastal eutrophication is generally recognised as a recent phenomenon related to the well-documented increase in riverine nutrient delivery during the last 30 or 40 years, a few historical records paradoxically show that, in some places like the Southern Bight of the North Sea, or the Northern Adriatic, algal proliferation as intense as presently observed was already regularly occurring at the end of the 19th century. Estimated riverine nutrient loads from diffuse sources or from domestic point sources of waste water at that time are too low to account for these observations. We attempted a retrospective evaluation of the possible contribution of industrial activity to nutrient river loading. The figures indicate that, by the end of the last century, large scale use of traditional processes in textile and paper industries, in tanneries, candles factories and others was responsible for a dominant part of the nutrient load carried by rivers in Western Europe and could have caused nutrient inputs to coastal zones similar to the present ones.

We propose a method for determining the mortality rate for allochthonous bacteria released in aquatic environments without interference due to the loss of culturability in specific culture media. This method consists of following the disappearance of radioactivity from the trichloroacetic acid-insoluble fraction in water samples to which [3H]thymidine-prelabeled allochthonous bacteria have been added. In coastal seawater, we found that the actual rate of disappearance of fecal bacteria was 1 order of magnitude lower than the rate of loss of culturability on specific media. Minor adaptation of the procedure may facilitate assessment of the effect of protozoan grazing and bacteriophage lysis on the overall bacterial mortality rate.

A method is proposed which provides a minimum estimate of the rate of bacterial mortality in growing natural populations of planktonic bacteria. This estimate is given by the rate of decrease of radioactivity from the DNA of a [H]thymidine-labeled natural assemblage of bacteria after all added thymidine has been exhausted from the medium. Results obtained from river water, estuarine water, and seawater show overall bacterial mortality rates in the range 0.010 to 0.030 h, in good agreement with the range of growth rates measured in the same environments. Use of selective filtration through Nuclepore filters (pore size, 2 mum) allowed us to determine the contribution of microzooplankton grazing to overall bacterial mortality. Grazing rates estimated by this method ranged from 0 to 0.02 h.

Determination of [H] thymidine incorporation into bacterial DNA versus other macromolecules is usually achieved by NaOH and hot trichloroacetic acid hydrolysis. This procedure was found not to be specific enough. An alternative method founded on DNase treatment is proposed. Under the new method, the fraction of thymidine incorporated into DNA ranged from 10 to 83%.

Grazing rates of mixed cultures of freshwater, heterotrophic nanoflagellates on two populations of bacterial prey present together at varying concentrations were measured by using fluorescently labeled bacteria. The effect of one population on the ingestion kinetics of the other was consistent with a theory based on competitive inhibition of enzymatic reactions. However, allochthonous bacteria, when present in low concentrations within a much larger population of small autochthonous bacteria, may be preferentially grazed, which is due to their large size.
