RESUMEN

The fibroblast growth factors (FGFs) are important for embryo development, wound healing, hematopoiesis, and angiogenesis. FGF-1, a member of FGF family, is involved in both receptor-dependent pathways and an intracrine pathway. Studies have recently shown that FGF-1 is overexpressed in the early stages of several kinds of cancer. Thus, FGF-1 is a candidate for cancer immunotargeting. To study the potential use of therapeutic antibodies against FGF-1, a monoclonal hybridoma 1C9 secreting monoclonal antibody specific for FGF-1 was developed. Then, a single-chain variable fragment (scFv) antibody was genetically engineered from hybridama 1C9. The binding of the scFv1C9 to the antigen FGF-1 was demonstrated by ELISA and immunoprecipitation assays. Functional analysis showed that the overexpressed scFv1C9 in MCF-7 cells targeted endogenous FGF-1 and prevented the translocation of FGF-1 into the nucleus, resulting in the blockade of the intracrine pathway of FGF-1, which caused the G1 arrest by p21 up-regulation. These results suggest that the generated scFv1C9 is an effective inhibitor of the intracrine pathway of FGF-1 and has a potential application as anti-tumoral agent in breast cancer.

RESUMEN

Free vibration of summation resonance of suspended-cable-stayed beam is investigated in the article. A 3-DOF model of the coupled structure is built, with the main cable and sling (vertical cable) considered to be geometrically nonlinear, and the beam is taken as linear Euler beam. Hamilton's principle is used to derive the dynamic equilibrium equations of the coupled structure. Then, the dynamic equilibrium equations are solved by means of multiple scales method, the second order approximation solutions of single-modal motion of the coupled structure are obtained. Numerical examples are presented to discuss time history of free vibration of the summation resonance, with and without damping. Additionally, fourth-order Runge-Kutta method is directly used for the dynamic equilibrium equations to complement and verify the analytical solutions. The results show that the coupled structure performs strongly nonlinear and coupled characteristics, which is useful for engineering design.

RESUMEN

Complete mitochondrial genome of Scapharca subcrenata was determined in this report. It is 48,161 bp in length, being the largest mitochondrial genome among reported shellfish at present. The entire mitochondrial genome consists of 57 genes including 12 protein-coding genes, 2 ribosomal RNAs and 41 transfer RNAs.

RESUMEN

Objective To compare the clinical feasibility of excising epiglottis cyst by CO2 laser and high frequency under laryngomicroscope. Methods 44 cases with epiglottic cysts were divided into two groups, with 22 cases in observation group (OG) and also 22 cases in control group (CG). All patients were operated by a laryngomicroscopic surgery under a general anesthesia, while cases in OG under CO2 laser and patients in CG under high frequency electric knife to remove the cyst. Then, analyse and compare the clinical feasibility. Results The average operating time is significantly shorter , and the VAS of sore throat is obviously lower in OG than that in CG. Thanks to the accurate and precise operation by CO2 laser, the epiglottis cysts are able to be removed intact, the surrounding tissues and epiglottic cartilage are better protected. There is no recurrence in both groups. Conclusion The operation performed with CO2 laser under micro-laryngoscope holds such advantages as accurate and easy to operate, better protect cyst wall, and fewer side hurting.
