RESUMEN

In the spinal cord, attenuation of the inhibitory action of glycine is related to an increase in both inflammatory and diabetic neuropathic pain; however, the glycine receptor involvement in diabetic neuropathy has not been reported. We determined the expression of the glycine receptor subunits (α1-α3 and ß) in streptozotocin-induced diabetic Long-Evans rats by qPCR and Western blot. The total mRNA and protein expression (whole spinal cord homogenate) of the α1, α3, and ß subunits did not change during diabetes; however, the α2 subunit mRNA, but not the protein, was overexpressed 45 days after diabetes induction. By contrast, the synaptic expression of the α1 and α2 subunits decreased in all the studied stages of diabetes, but that of the α3 subunit increased on day 45 after diabetes induction. Intradermal capsaicin produced higher paw-licking behavior in the streptozotocin-induced diabetic rats than in the control animals. In addition, the nocifensive response was higher at 45 days than at 20 days. During diabetes, the expression of the glycine receptor was altered in the spinal cord, which strongly suggests its involvement in diabetic neuropathy.

RESUMEN

The human genome is highly dynamic and only a small fraction of it codes for proteins, but most of the genome is transcribed, highlighting the importance of non-coding RNAs on cellular functions. In addition, it is now known the generation of non-coding RNA fragments under particular cellular conditions and their functions have revealed unexpected mechanisms of action, converging, in some cases, with the biogenic pathways and action machineries of microRNAs or Piwi-interacting RNAs. This led us to the question why the cell produces so many apparently redundant molecules to exert similar functions and regulate apparently convergent processes? However, non-coding RNAs fragments can also function similarly to aptamers, with secondary and tertiary conformations determining their functions. In the present work, it was reviewed and analyzed the current information about the non-coding RNAs fragments, describing their structure and biogenic pathways, with special emphasis on their cellular functions.

This study investigated the application of adsorption with activated carbons (ACs) and photodegradation to reduce the concentration of triclosan (TCS) in aqueous solutions. Concerning adsorption, ACs (Darco, Norit, and F400) were characterised and batch experiments were performed to elucidate the effect of pH on equilibrium. The results showed that at pH = 7, the maximum adsorption capacity of TCS onto the ACs was 18.5 mg g-1 for Darco, 16.0 mg g-1 for Norit, and 15.5 mg g-1 for F400. The diffusional kinetic model allowed an adequate interpretation of the experimental data. The effective diffusivity varied and increased with the amount of TCS adsorbed, from 1.06 to 1.68 × 10-8 cm2 s-1. In the case of photodegradation, it was possible to ensure that the triclosan molecule was sensitive to UV light of 254 nm because the removal was over 80 % using UV light. The removal of TCS increased in the presence of sulfate radicals. It was possible to identify 2,4-dichlorophenol as one of the photolytic degradation products of triclosan, which does not represent an environmental hazard at low concentrations of triclosan in water. These results confirm that the use of AC Darco, Norit, and F400 and that photodegradation processes with UV light and persulfate radicals are effective in removing TCS from water, reaching concentration levels that do not constitute a risk to human health or environmental hazard. Both methods effectively eliminate pollutants with relatively easy techniques to implement.

Plant-animal mutualisms such as seed dispersal are key interactions for sustaining plant range shifts. It remains elusive whether the organization of interactions with seed dispersers is reconfigured along the expansion landscape template and, if so, whether its effects accelerate or slow colonization. Here we analyse plant-frugivore interactions in a scenario of rapid population expansion of a Mediterranean juniper. We combined network analyses with field surveys, sampling interactions between individual plants and frugivores by DNA-barcoding and phototrapping over two seasons. We assess the role of intrinsic and extrinsic intraspecific variability in shaping interactions and we estimate the individual plant contributions to the seed rain. The whole interaction network was highly structured, with a distinct set of modules including individual plants and frugivore species arranged concordantly along the expansion gradient. The modular configuration was partially shaped by individual neighbourhood context (density and fecundity) and phenotypic traits (cone size). Interaction reconfiguration resulted in a higher and more uneven propagule contribution, with most effective dispersers having a prominent role at the colonization front stand, where a distinct subset of early arriving plants dominated the seed rain. Our study offers new insights into the key role of mutualistic interactions in colonization scenarios by promoting fast plant expansion processes.

Internet of Things (IoT) has made significant strides in energy management systems recently. Due to the continually increasing cost of energy, supply-demand disparities, and rising carbon footprints, the need for smart homes for monitoring, managing, and conserving energy has increased. In IoT-based systems, device data are delivered to the network edge before being stored in the fog or cloud for further transactions. This raises worries about the data's security, privacy, and veracity. It is vital to monitor who accesses and updates this information to protect IoT end-users linked to IoT devices. Smart meters are installed in smart homes and are susceptible to numerous cyber attacks. Access to IoT devices and related data must be secured to prevent misuse and protect IoT users' privacy. The purpose of this research was to design a blockchain-based edge computing method for securing the smart home system, in conjunction with machine learning techniques, in order to construct a secure smart home system with energy usage prediction and user profiling. The research proposes a blockchain-based smart home system that can continuously monitor IoT-enabled smart home appliances such as smart microwaves, dishwashers, furnaces, and refrigerators, among others. An approach based on machine learning was utilized to train the auto-regressive integrated moving average (ARIMA) model for energy usage prediction, which is provided in the user's wallet, to estimate energy consumption and maintain user profiles. The model was tested using the moving average statistical model, the ARIMA model, and the deep-learning-based long short-term memory (LSTM) model on a dataset of smart-home-based energy usage under changing weather conditions. The findings of the analysis reveal that the LSTM model accurately forecasts the energy usage of smart homes.

RESUMEN

The Internet of Things (IoT) is an innovative technology that presents effective and attractive solutions to revolutionize various domains. Numerous solutions based on the IoT have been designed to automate industries, manufacturing units, and production houses to mitigate human involvement in hazardous operations. Owing to the large number of publications in the IoT paradigm, in particular those focusing on industrial IoT (IIoT), a comprehensive survey is significantly important to provide insights into recent developments. This survey presents the workings of the IoT-based smart industry and its major components and proposes the state-of-the-art network infrastructure, including structured layers of IIoT architecture, IIoT network topologies, protocols, and devices. Furthermore, the relationship between IoT-based industries and key technologies is analyzed, including big data storage, cloud computing, and data analytics. A detailed discussion of IIoT-based application domains, smartphone application solutions, and sensor- and device-based IIoT applications developed for the management of the smart industry is also presented. Consequently, IIoT-based security attacks and their relevant countermeasures are highlighted. By analyzing the essential components, their security risks, and available solutions, future research directions regarding the implementation of IIoT are outlined. Finally, a comprehensive discussion of open research challenges and issues related to the smart industry is also presented.

Adaptive equalization is crucial in mitigating distortions and compensating for frequency response variations in communication systems. It aims to enhance signal quality by adjusting the characteristics of the received signal. Particle swarm optimization (PSO) algorithms have shown promise in optimizing the tap weights of the equalizer. However, there is a need to enhance the optimization capabilities of PSO further to improve the equalization performance. This paper provides a comprehensive study of the issues and challenges of adaptive filtering by comparing different variants of PSO and analyzing the performance by combining PSO with other optimization algorithms to achieve better convergence, accuracy, and adaptability. Traditional PSO algorithms often suffer from high computational complexity and slow convergence rates, limiting their effectiveness in solving complex optimization problems. To address these limitations, this paper proposes a set of techniques aimed at reducing the complexity and accelerating the convergence of PSO.

Breast cancer, due to its high incidence and mortality, is a public health problem worldwide. Current chemotherapy uses non-specific cytotoxic drugs, which inhibit tumor growth but cause significant adverse effects. (-)-Epicatechin (EC) is part of a large family of biomolecules called flavonoids. It is widely distributed in the plant kingdom; it can be found in green tea, grapes, and cocoa. Several studies in animals and humans have shown that EC induces beneficial effects in the skeletal muscle and the cardiovascular system, reducing risk factors such as arterial hypertension, endothelial dysfunction, damage to skeletal muscle structure, and mitochondrial malfunction by promoting mitochondrial biogenesis, with no adverse effects reported. Recently, we reported that EC had an antitumor effect in a murine triple-negative mammary gland tumor model, decreasing tumoral size and volume and increasing survival by 44%. This work aimed to characterize the effects of flavanol EC on proliferation, migration, and metastasis markers of triple-negative murine breast (4T1) cancer cells in culture. We found proliferation diminished and Bax/Bcl2 ratio increased. When the migration of culture cells was evaluated, we observed a significant reduction in migration. Also, the relative expression of the genes associated with metastasis, Cdh1, Mtss1, Pten, Bmrs, Fat1, and Smad4, was increased. In conclusion, these results contribute to understanding molecular mechanisms activated by EC that can inhibit metastatic-associated proliferation, migration, and invasion of murine breast cancer cells.

(-)-Epicatechin (EC) is part of a large family of biomolecules called flavonoids and is widely distributed in the plant kingdom. Several studies have shown the beneficial effects of EC consumption. Many of these reported effects are exerted by activating the signaling pathways associated with the activation of two specific receptors: the G protein-coupled estrogen receptor (GPER), a transmembrane receptor, and the pregnane X receptor (PXR), which is a nuclear receptor. However, the effects of EC are so diverse that these two receptors cannot describe the complete phenomenon. The apelin receptor or APLNR is classified within the G protein-coupled receptor (GPCR) family, and is capable of activating the G protein canonical pathways and the ß-arrestin transducer, which participates in the phenomenon of receptor desensitization and internalization. ß-arrestin gained interest in selective pharmacology and mediators of the so-called "biased agonism". With molecular dynamics (MD) and in vitro assays, we demonstrate how EC can recruit the ß-arrestin in the active conformation of the APLN receptor acting as a biased agonist.

This work characterizes two alternative materials to substitute the most expensive microbial fuel cells (MFCs) components: proton exchange membrane (PEM) and cathode. Crude glycerol biodegradation was studied in MFCs using a clay cup as a PEM and activated carbon and camphor carbon mixture (CAC) as a cathode. The cathode performance was compared with Platinum on carbon cloth. Two clay cup single-chamber MFCs were operated with each cathode and fed with 2000 mg/L of crude glycerol. Electrochemical properties were characterized by linear sweep voltammetry, electrochemical impedance spectroscopy, and chronoamperometry. Biodegradation efficiencies were estimated with the chemical oxygen demand (COD) removal percentage. MFCs with CAC showed a maximum power density of 100 mW/m2. This result was a 43.47% power response regarding MFCs with Platinum. COD removal efficiencies of 94% were achieved in 37 days for both cells. The Columbic efficiencies were 24.04% and 22.78% for the MFCs with Platinum and CAC. The economic analysis showed a cost of USD 9.97 for MFCs with CAC. This cost is five times lower than when using Platinum. MFCs utilizing clay cups and CAC showed an acceptable performance for the bioenergy production from crude glycerol biodegradation above all economic advantage in the cell cost.

A risk assessment model for a smart home Internet of Things (IoT) network is implemented using a Bayesian network. The directed acyclic graph of the Bayesian network is constructed from an attack graph that details the paths through which different attacks can occur in the IoT network. The parameters of the Bayesian network are estimated with the maximum likelihood method applied to a data set obtained from the simulation of attacks, in five simulation scenarios. For the risk assessment, inferences in the Bayesian network and the impact of the attacks are considered, focusing on DoS attacks, MitM attacks and both at the same time to the devices that allow the automation of the smart home and that are generally the ones that individually have lower levels of security.

Ibuprofen degradation and energy generation in a single-chamber Microbial Fuel Cell (MFC) were evaluated using a bioanode fabricated from devil fish bone char (BCA) synthesized by calcination in air atmosphere. Its performance was compared with conventional carbon felt (CF). Bone char textural properties were determined by nitrogen adsorption. Before and after, the bacterial colonization on the materials was analyzed by environmental scanning electron microscopy. Energy generation was evaluated by electrochemical techniques as open-circuit potential, linear sweep voltammetry, and electrochemical impedance spectroscopy. Ibuprofen degradation was analyzed by High-Performance Liquid Chromatography-Ultraviolet, and the chemical oxygen demand (COD) removal was measured. Results showed a specific area of 136 m2/g for BCA, having enough space to immobilize microorganisms. The micrographs confirmed the biofilm formation on the electrode materials. Over the 14 days, MFC with BCA reached a maximum power density of 4.26 mW/m2, 175% higher than CF, and an electron transfer resistance 2.1 times lower than it. This coincides with the COD removal and ibuprofen degradation efficiencies, which were 43.6% and 34% for BCA and 31.8% and 27% for CF. Hence, these findings confirmed that BCA in MFC could provide an alternative electrode material for ibuprofen degradation and energy generation.

Non-medulloblastoma CNS embryonal tumors (former PNET/Pineoblastomas) are aggressive malignancies with poor outcome that have been historically treated with medulloblastoma protocols. The purpose of this study is to present a tumor-specific, real-world data cohort of patients with CNS-PNET/PB to analyze quality indicators that can be implemented to improve the outcome of these patients. Patients 0-21 years with CNS-PNET treated in eight large institutions were included. Baseline characteristics, treatment and outcome [progression-free and overall survival (PFS and OS respectively)] were analyzed. From 2005 to 2014, 43 patients fulfilled entry criteria. Median age at diagnosis was 3.6 years (range 0.0-14.7). Histology was pineoblastoma (9%), ependymoblastoma (5%), ETANTR (7%) and PNET (77%). Median duration of the main symptom was 2 weeks (range 0-12). At diagnosis, 28% presented with metastatic disease. Seventeen different protocols were used on frontline treatment; 44% had gross total resection, 42% craniospinal radiotherapy, 86% chemotherapy, and 33% autologous hematopoietic stem cell transplantation (aHSCT). Median follow-up for survivors was 3.5 years (range 1.7-9.3). 3-year PFS was 31.9% (95% CI 17-47%) and OS 35.1% (95% CI 20-50%). Age, extent of resection and radiotherapy were prognostic of PFS and OS in univariate analysis (p < 0.05). Our series shows a dismal outcome for CNS-PNET, especially when compared to patients included in clinical trials. Establishing a common national strategy, implementing referral circuits and collaboration networks, and incorporating new molecular knowledge into routine clinical practice are accessible measures that can improve the outcome of these patients.

Retinal detachment with subsequent silicone oil retinopexy is not uncommon. A known complication of silicone retinopexy is intraventricular migration of the intraocular silicone oil. While the oil itself does not result in direct pathology, misdiagnosis may lead to an unnecessary diagnostic workup and possibly predispose the patient to surgery intervention. Silicone oil typically appears hyperdense on computer tomography (CT) and hyperintense on T1-weighted magnetic resonance (MR). These imaging findings may mimic a mass or blood products. However, MR imaging of silicone results in chemical shift artifact which should help narrow the imaging differential. We present a patient with incidental CT and MRI findings which resulted in a prolonged hospital course following misidentification of intraventricular silicone oil. Although the imaging differential for an intraventricular lesion may include metastasis, lymphoma, hemorrhage, choroid plexus papilloma/carcinoma, meningioma, subependymoma, and ependymoma, secondary imaging findings should be noted to ensure an accurate diagnosis. In patients with evidence of prior silicone retinopexy, visualization of an intraventricular lesion with associated chemical shift artifact should raise the possibility of intraventricular silicone oil migration.

Purpose To review a single-center experience with the cortical tangential approach during computed tomography (CT)-guided native medical renal biopsy and to evaluate its efficacy and safety compared with those of a non-cortical tangential approach. Materials and Methods This retrospective study received institutional review board approval, with a waiver of the HIPAA requirement for informed consent. The number of cores, glomeruli, and complications were reviewed in 431 CT-guided medical renal biopsies performed between July 2007 and September 2015. A biopsy followed a cortical tangential approach if the needle path was parallel to the renal cortical surface, at a depth closer to the renal capsule than the renal pelvic fat. A sample was considered adequate if the biopsy yielded at least 10 glomeruli at light microscopy, one glomerulus at immunofluorescence microscopy, and one glomerulus at electron microscopy. The χ2 test, the t test, the Mann-Whitney test, and logistic regression modeling of sample adequacy were performed. Results One hundred fifty-six (36%) of 431 biopsies were performed with the cortical tangential approach. More cores were obtained for the cortical tangential group (2.6 vs 2.4, P = .001); biopsy needle gauge was not significantly different (P = .076). More adequate samples were obtained in the cortical tangential group (66.7% vs 49.8%, P = .001), with more glomeruli (23 vs 16, P = .014). Results were significant after controlling for needle gauge and number of cores (P = .008). The cortical tangential group had fewer complications (1.9% vs 7.3%, P = .018). Conclusion The cortical tangential approach, when applied to CT-guided native medical renal biopsies, results in higher rates of sample adequacy and lower rates of postprocedural complications. © RSNA, 2016.

B-cell lymphoma 2 (BCL2)-interacting killer (apoptosis inducing) (BIK) has been proposed as a tumor suppressor in diverse types of cancers. However, BIK's overexpression in breast cancer (BC) and in non-small lung cancer cells (NSCLCs), associated with a poor prognosis, suggests its participation in tumor progression. In this study, we evaluated the global expression pattern of microRNAs (miRNAs), messenger RNA (mRNA) expression changes in autophagy, and autophagic flux after BIK interference. BIK gene expression was silenced by small interfering RNA (siRNA) in BC cell MDA-MB-231, and BIK interference efficiency was tested by real-time PCR and by Western blotting. BIK expression levels decreased by 75 ± 18 % in the presence of 600 nM siRNA, resulting in the abolishment of BIK expression by 94 ± 30 %. BIK interference resulted in the overexpression of 17 miRNAs that, according to the DIANA-miRPath v3.0 database, are mainly implied in the control of cell signaling, gene expression, and autophagy. The autophagy array revealed downregulation of transcripts which participate in autophagy, and their interactome revealed a complex network, where hepatocyte growth factor-regulated tyrosine kinase substrate (HGS), α-synuclein (SNCA), unc-51-like autophagy activating kinase 1/2 (ULK1/2), and mitogen-activated protein kinase 3 (MAPK3) were shown to be signaling hubs. LC3-II expression-an autophagy marker-was increased by 169 ± 25 % after BIK interference, which indicates the involvement of BIK in autophagy. Altogether, our results indicate-for the first time-that BIK controls the expression of miRNAs, as well as the autophagic flux in MDA-MB-231 cells.

Majority of women with estrogen receptor (ER)-positive breast cancers initially respond to hormone therapies such as tamoxifen (TAM; antagonist of estrogen). However, many tumors eventually become resistant to TAM. Therefore, understanding the various cellular components involved in causing resistance to TAM is of paramount importance in designing novel entities for efficacious hormone therapy. Previously, we found that suppression of BIK gene expression induced TAM resistance in MCF-7 breast cancer cells. In order to understand the response of these cells to TAM and its association with resistance, a microarray analysis of gene expression was performed in the BIK-suppressed MCF-7 cells and compared it to the TAM-only-treated cells (controls). Several genes participating in various cellular pathways were identified. Molecules identified in the drug resistance pathway were 14-3-3z or YWHAZ, WEE1, PRKACA, NADK, and HSP90AA 1. Further, genes involved in cell cycle control, apoptosis, and cell proliferation were also found differentially expressed in these cells. Transcriptional and translational analysis of key molecules such as STAT2, AKT 3, and 14-3-3z revealed similar changes at the messenger RNA (mRNA) as well as at the protein level. Importantly, there was no cytotoxic effect of TAM on BIK-suppressed MCF-7 cells. Further, these cells were not arrested at the G0-G1 phase of the cell cycle although 30 % of BIK-suppressed cells were arrested at the G2 phase of the cycle on TAM treatment. Furthermore, we found a relevant interaction between 14-3-3z and WEE1, suggesting that the cytotoxic effect of TAM was prevented in BIK-suppressed cells because this interaction leads to transitory arrest in the G2 phase leading to the repair of damaged DNA and allowing the cells to proliferate.

We report on a 16-year-old girl with a complex phenotype, including intellectual disability, facial dysmorphisms, and obesity. During her infancy, she presented with weak sucking, global developmental delay, and later with excessive eating with central obesity. The girl was clinically diagnosed with probable Prader-Willi syndrome. Chromosomal analysis showed a de novo deletion 46,XX,del(15)(q21q22). However, the use of the Affymetrix CytoScan HD Array defined the exact breakpoints of the deleted 15q21q22 region. The imbalance, about 10.5 Mb in size, is to date the second largest deletion ever described in this chromosomal region. In addition, our patient carries a microdeletion in the 1q44 region and a gain in 9p24. The array result was arr[hg19] 9p24.1(6,619,823-6,749,335)×3, 1q44(248,688,586-248,795,277)×1, 15q21.2 q22.2(50,848,301-61,298,006)×1. Although our patient presents additional chromosomal alterations, we provide a correlation between the clinical findings and the phenotype of the 15q21 deletion syndrome.

The aim of this work is to evaluate the antimicrobial and cytotoxic activity of a formulation containing silver nanoparticles and chitosan, provisionally called nano silver fluoride (NSF), against Streptococcus mutans in comparison to chlorhexidine and silver diamine fluoride (SDF). The product was characterised by transmission electron microscopy and UV-Vis absorption spectroscopy. The minimum inhibition concentration (MIC) was evaluated by the spectrophotometric microdilution method and turbidity. The minimum bactericide concentration (MBC) was evaluated in brain heart infusion plates, and cytotoxicity was evaluated by haemolytic activity. The MIC and MBC for NSF were, respectively, 33.54 ± 14.52 and 50.32 µg/mL; for SDF were 33.33 ± 14.43 and 50.0 µg/mL, respectively; and for CHX were 3.3 ± 0.5 and 6 µg/mL, respectively. An ANOVA for MIC gave P = 0.032, and for MBC P = 0.035. The cytotoxic effect of NSF compared to SDF demonstrated a statistically significant difference in the MIC value (t test P < 0.05). The NSF formulation may be effective against S. mutans with much lower doses, may have lower toxicity than SDF, and may not stain teeth.
