Every year, millions of people suffer some form of illness associated with the consumption of contaminated food. Escherichia coli (E. coli), found in the intestines of humans and other animals, is commonly associated with various diseases, due to the existence of pathogenic strains. Strict monitoring of food products for human consumption is essential to ensure public health, but traditional cell culture-based methods are associated with long waiting times and high costs. New approaches must be developed to achieve cheap, fast, and on-site monitoring. Thus, in this work, we developed optical fiber sensors based on surface plasmon resonance. Gold and cysteamine-coated fibers were functionalized with anti-E. coli antibody and tested using E. coli suspensions with concentrations ranging from 1 cell/mL to 105 cells/mL. An average logarithmic sensitivity of 0.21 ± 0.01 nm/log(cells/mL) was obtained for three independent assays. An additional assay revealed that including molybdenum disulfide resulted in an increase of approximately 50% in sensitivity. Specificity and selectivity were also evaluated, and the sensors were used to analyze contaminated water samples, which verified their promising applicability in the aquaculture field.

Capuchins are omnivorous neotropical primates that can survive in urban forests by supplementing their diet with human foods. However, few studies have analyzed the impact of these resources on their diet diversity and feeding seasonality. We aimed to assess the patterns of foraging, feeding, and diet diversity of urban capuchins (Sapajus sp.) that live in a tiny urban forest in Foz do Iguaçu, Brazil, where humans frequently feed them. We predicted that forest degradation and human foods could decrease diet diversity, though capuchins may, conversely, reduce their selectivity and expand their food repertoire. We followed the animals from dawn to dusk between May 2018 and April 2019 to list and quantify the species and items consumed. We used diversity indexes and cluster analysis to understand similarities and differences in the diet composition over the study period. We recorded 58 plant species (being 14 exotics) consumed by the monkeys, and nonconventional items, with low diet diversity overall. The diet consisted mainly of plants (69.8%), animal matter (20.6%), and processed foods (9.5%). Capuchins consumed more food from the anthropic environment (57.5%) than from the forest (42.4%), while their food from the forest included more invertebrates (47.8%) than fruits (40%). The cluster analysis showed two main groups of feeding months, in accordance with the seasons of high and low food production in local forests. Monkeys did not vary the frequencies of foraging or feeding between seasons, probably due to the omnipresent availability of human foods. Despite the high consumption of human foods, capuchins responded to the seasonality of the forest, expanding their feeding diversity in the drier period. Future studies should analyze the correspondence between food consumption and local phenology, as well as the potential role of capuchins as seed dispersers in this depauperate community.

Glutamate amidation, a secondary modification of the peptidoglycan, was first identified in Staphylococcus aureus It is catalyzed by the protein products of the murT and gatD genes, which are conserved and colocalized in the genomes of most sequenced Gram-positive bacterial species. The MurT-GatD complex is required for cell viability, full resistance to ß-lactam antibiotics, and resistance to human lysozyme and is recognized as an attractive target for new antimicrobials. Great effort has been invested in the study of this step, culminating recently in three independent reports addressing the structural elucidation of the MurT-GatD complex. In this work, we demonstrate through the use of nonstructural approaches the critical and multiple roles of the C-terminal domain of MurT, annotated as DUF1727, in the MurT-GatD enzymatic complex. This domain provides the physical link between the two enzymatic activities and is essential for the amidation reaction. Copurification of recombinant MurT and GatD proteins and bacterial two-hybrid assays support the observation that the MurT-GatD interaction occurs through this domain. Most importantly, we provide in vivo evidence of the effect of substitutions at specific residues in DUF1727 on cell wall peptidoglycan amidation and on the phenotypes of oxacillin resistance and bacterial growth.

Objectives Major and minor amputations are associated with significant rates of mortality. However, little is known about the impact of unplanned redo-amputation during the same hospitalization on outcomes. The objectives of this study were to identify the risk factors associated with in-hospital mortality after both major and minor amputations as well as the results of unplanned redo-amputation on outcome. Methods Retrospective study of 342 consecutive patients who were treated with lower extremity amputation in Brazil between January 2013 and October 2014. Results The in-hospital mortality rate was higher in major compared to minor amputation (25.6% vs. 4.1%; p < 0.0001). Whereas chronic kidney disease, chronic obstructive pulmonary disease, and planned staged amputation predicted in-hospital mortality after major amputation, age, and congestive heart failure predicted mortality after minor amputation. The white blood cell count predicted in-hospital mortality following both major and minor amputation. However, postoperative infection predicted in-hospital mortality only following major amputation. Conclusions In-hospital mortality was high after major amputations. Unplanned redo-amputation was not a predictor of in-hospital mortality after major or minor amputation. Planned staged amputation was associated with reduced survival after major but not minor amputation. Postoperative infection predicted mortality after major amputation. Systemic diseases and postoperative white blood cell were associated with in-hospital mortality. This study suggests a possible link between a pro-inflammatory state and increased in-hospital mortality following amputation.

This study aimed to identify using the infrared (IR) thermography data the injuries of guavas during cooling and storage at different temperatures. Three experiments were performed at three different temperatures with one storage time. The first experiment was done with static air in a refrigerator at 5 °C, the second experiment was conducted in a tunnel with forced air at 10 °C, and the third experiment was conducted in an air conditioned environment at 20 °C. Mechanical injuries caused by the impact of a pendulum were induced on guava surfaces. The surface temperatures were obtained for bruised and sound tissues during cooling and storage using an Infrared (IR) camera. With thermography, it was possible to distinguish the injured tissues of the fruits that were unaffected at temperatures of 5, 10 and 20 °C in first hours of cooling. The results suggest that the storage of guava fruits at 5 °C in static air resulted in cold-induced injury, while storage at 20 °C resulted in an altered activity pattern. The stored guava fruits were analyzed for mass loss, firmness, color, total sugars, total pectin and solubility. The parameters values were lower during the forced-air cooling and storage at 5 and 10 °C. When stored at 20 °C, there was fruit maturation that caused tissue softening, which makes the fruits more susceptible to deterioration and thermographic readings showed opposite trends.

Social prescribing (SP) is a promising intersectoral strategy of integrated and person-centered care that can improve individual health and well-being by going beyond medical treatment, potentially reducing overall disease burden on health system. SP addresses health and social determinants of health by linking patients to community responses, i.e. services and initiatives fostering social interaction, physical activity and creativity, among other health-promoting aspects, provided by local public, private, and non-profit entities. There is limited research on the implementation processes of SP beyond the UK, hindering improvement and scale-up. This study aims to identify enablers and barriers of implementation of an SP program conducted in Portugal. A participatory and qualitative approach was used to assess the implementation of an SP program in health units. Semi-structured interviews were conducted with family doctors, social workers and representatives of community partners who participated in SP implementation. The Consolidated Framework for Implementation Research was used to conduct thematic analysis. The main enablers of SP implementation included its recognition as an evolution towards a holistic model of health, the personal characteristics of professionals as being proactive, motivated, and concerned with social determinants of health, and the communication strategy used to engage the stakeholders. Perceived challenges included raising users' awareness of SP and ensuring intervention adherence. Lack of preparedness for intersectoral working processes, including insufficient communication channels, limited community responses and the need for a more systematic collection of data on activities adherence and progress were also highlighted as barriers to SP implementation. SP implementation seems simple, but the results show that in practice, we are facing a complex intervention with multiple stakeholders, diverse community responses and factors influencing project success. A deeper understanding of SP specificities, local context, enablers, and barriers is vital to develop strategies for improvement and successful implementation, ensuring scalability and sustainability.

AIMS: To investigate the SARS-CoV-2 Spike protein (Spk)-induced inflammatory response and its downmodulation by diminazene aceturate (DIZE). MATERIALS AND METHODS: Through inducing Spk inflammation in murine models, leukocyte migration to the peritoneum, levels of myeloperoxidase (MPO), malondialdehyde (MDA), rolling and adhesion of mesenteric leukocytes, and vascular permeability were investigated. Extracellular DNA traps (DETs) induced by Spk and the production of IL-6 and TNF-α were analyzed using human neutrophils, monocytes, and macrophages. In silico assays assessed the molecular interaction between DIZE and molecules related to leukocyte migration and DETs induction. KEY FINDINGS: Spk triggered acute inflammation, demonstrated by increasing leukocyte migration. Oxidative stress was evidenced by elevated levels of MPO and MDA in the peritoneal liquid. DIZE attenuated cell migration, rolling, and leukocyte adhesion, improved vascular barrier function, mitigated DETs, and reduced the production of Spk-induced pro-inflammatory cytokines. Computational studies supported our findings, showing the molecular interaction of DIZE with targets such as ß2 integrin, PI3K, and PAD2 due to its intermolecular coupling. SIGNIFICANCE: Our results outline a novel role of DIZE as a potential therapeutic agent for mitigating Spk-induced inflammation.

Introduction: Objective: to investigate the monocyte count and its association with nutritional status in children and adolescents with autism spectrum disorder (ASD). Methods: a cross-sectional study carried out at a Neurodevelopmental Center in the south of Brazil, with 68 ASD patients aged 3 to 18 years. The number of monocytes (per mm3) was determined in blood samples. Nutritional status was defined as BMI-for-age according to WHO standards. The Children's Eating Behaviour Questionnaire and a standard questionnaire to collect sociodemographic and clinical characteristics were administered to caregivers. Comparisons between sociodemographic, clinical, and eating behavior variables were performed with parametric tests. Linear regression was used to test the association between nutritional status and monocyte count. Results: mean age was 8.6 ± 3.3 years, 79 % were males and 66 % were overweight. In the unadjusted regression overweight was associated with higher monocyte counts compared to those non-overweight (B: 64.0; 95 % CI, 13.9 to 114.1; ß: 0.30, p = 0.01). This association remained significant after adjustment for the subscale of "emotional overeating" (B: 37.0; 95 % CI, 17.1 to 91.3; ß: 0.29; p = 0.02). The variability in monocyte count attributed to overweight was 14 %. Conclusions: overweight is associated with a higher monocyte count in children and adolescents with ASD. Nutritional intervention to control overweight is essential to mitigate the negative impact on inflammatory activity and immune dysfunction in these patients.


Introducción: Objetivo: investigar el recuento de monocitos y su asociación con el estado nutricional en niños y adolescentes con trastorno del espectro autista (TEA). Método: estudio transversal realizado en el Centro de Neurodesarrollo, en el sur de Brasil, con 68 pacientes con TEA de 3 a 18 años de edad. Se determinó el número de monocitos (por mm3) en muestras de sangre. El estado nutricional se definió como IMC para la edad según los estándares de la OMS. Se aplicó a los cuidadores el Cuestionario de Conducta Alimentaria Infantil y un cuestionario estándar para recoger características sociodemográficas y clínicas. Las comparaciones entre las variables sociodemográficas, clínicas y de conducta alimentaria se realizaron con pruebas paramétricas. Se utilizó la regresión lineal para probar la asociación entre el estado nutricional y el recuento de monocitos. Resultados: la edad media fue de 8,6 ± 3,3 años, el 79 % eran varones y el 66 % tenían sobrepeso. En la regresión no ajustada, el sobrepeso se asoció a un mayor número de monocitos en comparación con los que no tenían sobrepeso (B: 64,0; IC 95 %: 13,9 a 114,1; ß: 0,30; p = 0,01). Esta asociación siguió siendo significativa tras ajustar la subescala de "sobrealimentación emocional" (B: 37,0; IC 95 %: 17,1 a 91,3; ß: 0,29; p = 0,02). La variabilidad en el recuento de monocitos atribuida al sobrepeso fue del 14 %. Conclusiones: el sobrepeso se asocia a un mayor recuento de monocitos en niños y adolescentes con TEA. La intervención nutricional para controlar el sobrepeso es esencial para mitigar el impacto negativo sobre la actividad inflamatoria y la disfunción inmune en estos pacientes.

The incorporation of natural lignocellulosic fibers as reinforcements in polymer composites has witnessed significant growth due to their biodegradability, cost-effectiveness, and mechanical properties. This study aims to evaluate castor-oil-based polyurethane (COPU), incorporating different contents of coconut coir fibers, 5, 10, and 15 wt%. The investigation includes analysis of the physical, mechanical, and microstructural properties of these composites. Additionally, this study evaluates the influence of hydrothermal treatment on the fibers, conducted at 120 °C and 98 kPa for 30 min, on the biocomposites' properties. Both coir fibers (CFs) and hydrothermal-treated coir fibers (HTCFs) were subjected to comprehensive characterization, including lignocellulosic composition analysis, scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and thermogravimetric analysis (TGA). The biocomposites were subjected to water absorption analysis, bending tests, XRD, SEM, FTIR, and TGA. The results indicate that the 30 min hydrothermal treatment reduces the extractive content, enhancing the interfacial adhesion between the fiber and the matrix, as evidenced by SEM. Notably, the composite containing 5 wt% CF exhibits a reduced water absorption, approaching the level observed in pure COPU. The inclusion of 15 wt% HTCF results in a remarkable improvement in the composite's flexural strength (100%), elastic modulus (98%), and toughness (280%) compared to neat COPU. TGA highlights that incorporating CFs into the COPU matrix enhances the material's thermal stability, allowing it to withstand temperatures of up to 500 °C. These findings underscore the potential of CFs as a ductile, lightweight, and cost-effective reinforcement in COPU matrix biocomposites, particularly for engineering applications.

Coupling of UV-C irradiation to different peroxides (H2O2, S2O82- and HSO5-) has great potential to degrade persistent organic compounds due to the formation of HO• or SO4•- species. However, an in-depth comparison between the performance of different UV-C/peroxide processes as a function of (i) target compound degradation, (ii) generated transformation products and (iii) lethal/sub lethal toxicity effects has not yet been performed. To this end a comparison study was carried out to evaluate the effectiveness of different UV-C/peroxide processes using the herbicide tebuthiuron (100 or 500 µg L-1) as a model pollutant. TBH degradation experiments were performed at lab-scale in real municipal wastewater treatment plant effluent and distilled water. Faster degradation occurred by increasing peroxide concentration from 735 to 2206 µmol L-1 in the municipal wastewater treatment plant effluent, mainly for S2O82-. Experiments performed in the presence of peroxide trapping agents - HO• and SO4•- (methoxibenzene) or HO• (2-propanol) - revealed that oxidation in the UV-C/S2O82- system occurs mainly through SO4•-. Lower toxicity for the MWWTP effluent was obtained after oxidative treatments using hydrogen peroxide or monopersulfate as oxidants which react mainly through HO• radicals. Two mechanistic pathways were proposed for tebuthiuron degradation: (i) hydrogen abstraction by HO• (H2O2 and HSO5-) and (ii) electron transfer by SO4•- (S2O82-). In addition, one unprecedented transformation product was identified. In conclusion, results emphasize the relevance of comparing the degradation of toxic compounds in the presence of different peroxide sources and matrices and simultaneouly evaluating responses chemical and biological endpoints.

RESUMEN

Quantitative plant biology is a growing field, thanks to the substantial progress of models and artificial intelligence dealing with big data. However, collecting large enough datasets is not always straightforward. The citizen science approach can multiply the workforce, hence helping the researchers with data collection and analysis, while also facilitating the spread of scientific knowledge and methods to volunteers. The reciprocal benefits go far beyond the project community: By empowering volunteers and increasing the robustness of scientific results, the scientific method spreads to the socio-ecological scale. This review aims to demonstrate that citizen science has a huge potential (i) for science with the development of different tools to collect and analyse much larger datasets, (ii) for volunteers by increasing their involvement in the project governance and (iii) for the socio-ecological system by increasing the share of the knowledge, thanks to a cascade effect and the help of 'facilitators'.

Introduction: Objective: to investigate the speed of weight gain (WG) and the z-score (E-z) of weight in premature neonates large for gestational age (LGA) during four weeks of hospitalization. Methods: a retrospective longitudinal study with premature neonates in a neonatal intensive care unit at a university hospital. Data were obtained from January 2017 to December 2018; 115 babies with gestational age (GA) ≥ 27 and < 37 weeks, non-twin, AGA or LGA were included. The WG (g/kg/day) was obtained from the nadir weight and the E-z was calculated online based on the Intergrowth-21st curves. Repeated-measures ANOVA and multiple linear regression were used to assess the association between WP and E-z and explanatory variables; p < 5 %. Results: mean GA was 32.5 weeks, birth weight was 1910 g and weight loss was 5 % at 7 days. WG was lower in LGA babies, with GI between 32 and 37 weeks (LGA, 9.2 ± 5.6 g/kg/day vs AGA, 13.9 ± 6.0 g/kg/day). The change in WG was explained by protein supply in AGA (B = 2.5 g/kg/day; 95 % CI, 0.7 to 4.3; and ß = 0.543) and by GA in LGA (B = -0.05 g/kg/day; 95 % CI, -0.09 to -0.02; and ß = -0.574). In the 4th week of hospitalization, weight E-z decreased similarly for LGAs and AGAs, and this variation was explained by growth. Conclusions: premature LGAs had lower WG compared to AGAs during the neonatal period. The linear and brain growth explained the variation in weight E-z among these preterms.


Introducción: Objetivo: Investigar la velocidad de la ganancia de peso (GP) y la puntuación z (E-z) de peso en neonatos prematuros grandes para la edad gestacional (GEG) durante cuatro semanas de hospitalización. Métodos: estudio longitudinal retrospectivo con neonatos prematuros de una unidad de cuidados intensivos neonatales de un hospital universitario. Los datos se obtuvieron desde enero de 2017 hasta diciembre de 2018. Se incluyeron 115 bebés con edad gestacional (EG) ≥ 27 y < 37 semanas, no gemelos, AEG o GEG. El GP (g/kg/día) se obtuvo a partir del peso nadir y el E-z se calculó en línea basado en las curvas Intergrowth-21st. Se utilizaron el ANOVA de medidas repetidas y la regresión lineal múltiple para evaluar la asociación entre GP y E-z y las variables explicativas; p < 5 %. Resultados: la EG media fue de 32,5 semanas, el peso al nacer de 1910 g y la pérdida de peso del 5 % a los 7 días. El GP fue menor en los bebés GEG, con EG entre 32 y 37 semanas (GEG: 9,2 ± 5,6 g/kg/día vs. AEG: 13,9 ± 6,0 g/kg/día). El cambio en el GP se explicó por el suministro de proteínas en los AEG (B = 2,5 g/kg/día; IC 95 %: 0,7 a 4,3; y ß = 0,543) y por EG en los GEG (B = -0,05 g/kg/día; IC 95 %: -0,09 a -0,02; y ß = -0,574). En la 4ª semana de hospitalización, el peso E-z se redujo de manera similar en los GEG y los AEG, y esta variación se explicó por el crecimiento. Conclusiones: los prematuros GEG tuvieron un menor GP en comparación con los AEG durante el periodo neonatal. El crecimiento lineal y cerebral explicó la variación del peso E-z de estos prematuros.

This work presents data regarding the occurrence and treatment of Contaminants of Emerging Concern (CECs) in Brazil in the past decade. The literature review (2011-2021) revealed the detection of 87 pharmaceutical drugs and personal care products, 58 pesticides, 8 hormones, 2 illicit drugs, caffeine and bisphenol A in distinct matrices (i.e.: wastewater, groundwater, sea water, rainwater, surface water, drinking water and hospital effluent). Concentrations of CECs varied from ng-µg L-1 depending on the location, compound and matrix. The inefficiency of conventional wastewater treatment methods on the removal of CECs and lack of basic sanitation in some regions in the country aggravates contamination of Brazilian aquatic environments and poses potential environmental and health risks. Advanced oxidation processes (AOPs) are pointed out as viable and efficient alternatives to degrade CECs and prevent environmental contamination. A total of 375 studies involving the use of AOPs in Brazilian aqueous matrices were published in the last decade. Fenton and photo-Fenton processes, photo-peroxidation, ozonation, electrochemical advanced oxidation and heterogeneous photocatalysis are some of the AOPs applied by Brazilian research groups. Although many works discuss the importance of applying these technologies for CECs removal in real treatment plants, most of these studies assess the treatment of distilled water or simulated effluent. Therefore, the conduction of studies applying AOPs in real matrices are critical to drive the implementation of these processes coupled to conventional water and wastewater treatment in real plants in order to prevent the contamination of environmental matrices by CECs in Brazil.

Methicillin-resistant Staphylococcus aureus (MRSA) strains pose major treatment challenges due to their innate resistance to most ß-lactams under standard in vitro antimicrobial susceptibility testing conditions. A novel phenotype among MRSA, termed "NaHCO3 responsiveness," where certain strains display increased susceptibility to ß-lactams in the presence of NaHCO3, has been identified among a relatively large proportion of MRSA isolates. One underlying mechanism of NaHCO3 responsiveness appears to be related to decreased expression and altered functionality of several genes and proteins involved in cell wall synthesis and maturation. Here, we studied the impact of NaHCO3 on wall teichoic acid (WTA) synthesis, a process intimately linked to peptidoglycan (PG) synthesis and functionality, in NaHCO3-responsive versus -nonresponsive MRSA isolates. NaHCO3 sensitized responsive MRSA strains to cefuroxime, a specific penicillin-binding protein 2 (PBP2)-inhibitory ß-lactam known to synergize with early WTA synthesis inhibitors (e.g., ticlopidine). Combining cefuroxime with ticlopidine with or without NaHCO3 suggested that these latter two agents target the same step in WTA synthesis. Further, NaHCO3 decreased the abundance and molecular weight of WTA only in responsive strains. Additionally, NaHCO3 stimulated increased autolysis and aberrant cell division in responsive strains, two phenotypes associated with disruption of WTA synthesis. Of note, studies of key genes involved in the WTA biosynthetic pathway (e.g., tarO, tarG, dltA, and fmtA) indicated that the inhibitory impact of NaHCO3 on WTA biosynthesis in responsive strains likely occurred posttranslationally. IMPORTANCE MRSA is generally viewed as resistant to standard ß-lactam antibiotics. However, a NaHCO3-responsive phenotype is observed in a substantial proportion of clinical MRSA strains in vitro, i.e., isolates which demonstrate enhanced susceptibility to standard ß-lactam antibiotics (e.g., oxacillin) in the presence of NaHCO3. This phenotype correlates with increased MRSA clearance in vivo by standard ß-lactam antibiotics, suggesting that patients with infections caused by such MRSA strains might be amenable to treatment with ß-lactams. The mechanism(s) behind this phenotype is not fully understood but appears to involve mecA-PBP2a production and maturation axes. Our study adds significantly to this body of knowledge in terms of additional mechanistic targets of NaHCO3 in selected MRSA strains. This investigation demonstrates that NaHCO3 has direct impacts on S. aureus wall teichoic acid biosynthesis in NaHCO3-responsive MRSA. These findings provide an additional target for new agents being designed to synergistically kill MRSA using ß-lactam antibiotics.

Coronavirus disease 2019 (COVID-19) is currently a worldwide emergency caused by Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2). In observational clinical studies, statins have been identified as beneficial to hospitalized patients with COVID-19. However, experimental evidence of underlying statins protection against SARS-CoV-2 remains elusive. Here we reported for the first-time experimental evidence of the protective effects of simvastatin treatment both in vitro and in vivo. We found that treatment with simvastatin significantly reduced the viral replication and lung damage in vivo, delaying SARS-CoV-2-associated physiopathology and mortality in the K18-hACE2-transgenic mice model. Moreover, simvastatin also downregulated the inflammation triggered by SARS-CoV-2 infection in pulmonary tissue and in human neutrophils, peripheral blood monocytes, and lung epithelial Calu-3 cells in vitro, showing its potential to modulate the inflammatory response both at the site of infection and systemically. Additionally, we also observed that simvastatin affected the course of SARS-CoV-2 infection through displacing ACE2 on cell membrane lipid rafts. In conclusion, our results show that simvastatin exhibits early protective effects on SARS-CoV-2 infection by inhibiting virus cell entry and inflammatory cytokine production, through mechanisms at least in part dependent on lipid rafts disruption.

Natural lignocellulose fibers have been extensively investigated and applied as a reinforcement of polymer composites in industrial applications from food packing to automotive parts. Among the advantages of natural fibers stands their relatively low cost and sustainable characteristics. These are accentuated in the case of residual fibers such as those obtained from coffee husks, an agribusiness waste, usually burnt or disposed into the environment. As composite reinforcement, hydrophilic natural fibers display adhesion problems to the most hydrophobic polymer matrices. This adhesion might be improved with distinct types of fibers surface treatments. In the present work, the effectiveness of three surface treatments applied to coffee husk fiber wastes (CHFW) were investigated, aiming to improve the tensile performance of castor oil-based polyurethane (COPU) biocomposites. The effects of treatments associated with (i) chemical with sodium hydroxide, (ii) physical by temperature and pressure and hydrothermic treatment, and (iii) biological by fermentation with Phanerochaete Chrysosporium fungus were evaluated by means of Fourier transformed infrared spectroscopy, X-ray diffraction, thermal analyses and morphology by scanning electron microscopy for different concentration of NaOH, different hydrothermic times at 121 °C/98 kPa and exposition to P. chrysosporium. The most effective treatment was the hydrothermal one at 121 °C and 98.06 kPa for 30 min. Preliminary tensile tests were performed in COPU biocomposites reinforced with 20% CHFWs subjected to the optimized conditions for each distinct type of treatment. The results indicated that the hydrothermal treatment promoted significant enhancement in the fiber/matrix interfacial bond, increasing the tensile strength up to 60% compared to COPU reinforced with in natura CHFWs fibers. It is important to mention that these composites can be applied as plastic wood for household items' internal parts and in the automobile industry.

This is the first study to investigate ethylenediamine-N,N'-disuccinic acid (EDDS)/photo-Fenton process to polish real wastewater containing pesticides for possible water reuse. To this end, simultaneous degradation of pesticides ametrine, atrazine, imidacloprid and tebuthiuron was evaluated in distilled water (DW) and in sewage treatment plant (STP) effluent at initial pH 6.0. Several operational parameters (Fe3+-EDDS concentration, Fe3+-EDDS molar ratio, EDDS addition patterns and radiation source) were evaluated. 80-98% removal of target pesticides were obtained in DW using 30 µmol L-1 of Fe3+-EDDS with a molar ratio of 1:2 (300 µmol L-1 of H2O2). In addition, the proposed Fe3+-EDDS photo-Fenton at pH 6 was more efficient than classic photo-Fenton at pH 2.7 (30-84% removal). Experiments conducted in the presence of radical trapping agents (2-propanol or chloroform) revealed that HO• was the most active radical during treatment. Matrix composition strongly affected the degradation of target pesticides as a six-fold higher concentration of reagents (180 µmol L-1 of Fe3+-EDDS and 1800 µmol L-1 of H2O2) was needed to reach the same efficiency in STP compared to DW. Even so, first order rate constants corresponding to the degradation of pesticides in DW (k = 0.098-0.85 min-1) were nearly two-fold higher than in STP (k = 0.079-0.49 min-1) under the same radiation source (black-light or solar radiation). Finally, acute toxicity towards Vibrio fischeri and Drosophila melanogaster flies, and antibacterial activity assessed for Escherichia coli were eliminated after the application of the proposed treatment, thus indicating environmental safety for either discharge or reuse of treated wastewater for crop irrigation in agriculture.

Neutrophils release extracellular traps (NETs) after interaction with microorganisms and physiological or synthetic products. NETs consist of decondensed chromatin complexed with proteins, some of them with microbicidal properties. Because NETs can modulate the functioning of HIV-1 target cells, we aimed to verify whether they modify HIV-1 replication in macrophages. We found that exposure of HIV-1-infected macrophages to NETs resulted in significant inhibition of viral replication. The NET anti-HIV-1 action was independent of other soluble factors released by the activated neutrophils, but otherwise dependent on the molecular integrity of NETs, since NET-treatment with protease or DNase abolished this effect. NETs induced macrophage production of the anti-HIV-1 ß-chemokines Rantes and MIP-1ß, and reduced the levels of integrated HIV-1 DNA in the macrophage genome, which may explain the decreased virus production by infected macrophages. Moreover, the residual virions released by NET-treated HIV-1-infected macrophages lost infectivity. In addition, elevated levels of DNA-elastase complexes were detected in the plasma from HIV-1-infected individuals, and neutrophils from these patients released NETs, which also inhibited HIV-1 replication in in vitro infected macrophages. Our results reveal that NETs may function as an innate immunity mechanism able to restrain HIV-1 production in macrophages.
