RESUMEN

It is established that histamine in the concentration of 10(-4)-10(-5) M possesses an inhibitory effect in the lymphocyte adherence inhibition test in patients with breast cancer. Using specific antagonists (H1-blocker dimedrol and H2-blocker cimetidine) it is shown that the revealed histaminergic suppression of the lymphocyte adherence inhibition is mediated through the stimulating influence of histamine on H2-receptors.

RESUMEN

Alteration of the cholinoreceptor sensitivity of peripheral blood lymphocytes in patients with breast cancer was studied by the lymphocyte adhesion inhibition test (LAI-test). It has been shown that acetylcholine stimulates the LAI-reaction in optimal concentrations of 10(-7)-10(-8) M. Specific antagonists (m-cholinoblocker of atropin and n-cholinoblocker of hexonium) being used, it is demonstrated that the revealed cholinergic enhancement of the lymphocyte adherence inhibition is mediated through the stimulating influence of acetylcholine on m-cholinoreceptors. It is established that the lymphocyte cholinoreceptor sensitivity of the patients with breast cancer at the advanced stages is significantly lower than in patients with breast cancer at early stages.

RESUMEN

A comparative study of immunological reactivity of healthy volunteers and cancer patients with different stages of the disease was carried out. The study of 8 types of suppressor cells, chemoreceptor sensitivity and blast transformation of lymphocytes, natural killer cells, the level of immunoglobulins, regulatory subpopulation of lymphocytes, etc., has revealed several alterations in regulatory and effector links of immunity in the patients with late stages of the disease. The detailed immunological monitoring in the course of varying schemes of treatment is emphasized.

RESUMEN

Sensitivity of blood lymphocytes to the stimulating effect of met-enkephalin and dopamine was shown to decrease at late stages of the gastric cancer (IIIB-IV) or in the case of metastases to other organs, while to the inhibitory effect of serotonin it increased as compared to early stages (I-IIA). Using specific blockers of opioid, dopamine and serotonin receptors it was shown that these effects were mediated through stimulation of respective type receptors.

RESUMEN

The simplified micromodification of the leukocyte adhesion inhibition (LAI) test has shown that the LAI tests both for lymphocytes and for leukocytes may be equally used as additional methods of diagnostics of the early stages of breast cancer. The detailed study of the kinetics and cellular mechanisms of the method has shown that the LAI reaction belongs to the earliest reactions of T-cellular immunity and is mediated by T-cell lymphokine. The study of sensitivity and specificity of the method revealed that the suggested micromodification of the LAI test may be used in experimental and clinical research for the study of the subpopulation of T-cells-producers of the LAI factor as well as for monitoring of the T-cell immunity in the cancer patients during treatment.

RESUMEN

The inhibitory action of serotonin (10(-6) M and higher) in an in vitro lymphocyte adherence inhibition test appeared to be levelled off by methysergide, suggesting the involvement of serotonin receptors in the effect. A comparative evaluation of serotonin action in patients with stage I-IIA (group 1), stage IIIB-IV (group 2) rectal cancer and recurrent disease (group 3) showed lymphocyte sensitivity to serotonin in vitro to be significantly higher in groups 2 and 3, as compared to group 1.

RESUMEN

A simplified micromodification of the leukocyte adherence inhibition (LAI) test using both leukocytes and lymphocytes from peripheral blood of cancer patients is discussed. Data on oncoimmunologic specificity and cellular mechanisms underlying the proposed procedure as well as the results of a comparative study of LAI factor-producing T-cell and immunoregulatory (Tmu and T gamma) lymphocyte subpopulations in breast cancer patients are presented. The LAI-test micromodification may be adjunct in breast cancer diagnosis. It may be employed for monitoring T-cell specific immunity in cancer patients and further study of LAI factor-producing T-cells as a component of the structure of T-cell subpopulations.

RESUMEN

With the help of spontaneous lymphocyte adhesion test the influence of dopamine and histamine on the adhesion of peripheral blood lymphocytes has been studied in healthy donors. It was shown that dopamine enhanced spontaneous lymphocyte adhesion, with the optimal concentrations of the substance for the realization of a stimulating effect being 10(-4)-10(-6) M. The effect of the enhancement was pharmacologically specific, as it was completely blocked by dopamine receptor-blocker--haloperidol. It was demonstrated that low concentrations of histamine (10(-7)-10(-8) M) enhanced, while higher concentrations (10(-6)-10(-3) M) inhibited lymphocyte adhesion. Dimedrol, but not cimetidine abolished the enhancement of spontaneous lymphocyte adhesion, while the adhesion inhibition was blocked by cimetidine, but not dimedrol. Thus, the revealed histaminergic enhancement and inhibition of spontaneous lymphocyte adhesion is mediated through the influence of histamine on H1- and H2-receptors, respectively.

RESUMEN

With the help of spontaneous lymphocyte adhesion test it was demonstrated that acetylcholine and carbacholine enhanced the adhesion of lymphocytes. The enhancement revealed was completely abolished by a specific M-cholinergic blocker--atropine, but not N-cholinergic blocker--hexonium. Thus, the enhancement of spontaneous human lymphocyte adhesion by cholinergic stimulation is mediated by M-cholinoreceptors. It is suggested that the phenomenon revealed is mediated by the enhancement of cGMP intracellular levels after cholinergic stimulation.

The effect of dopamine in the test of lymphocyte adhesion inhibition in vitro was studied in patients with early (I-IIA) and advanced (IIIB-IV) stages of breast cancer (BC). It was shown that the stimulating effect of dopamine revealed at concentrations of 10(-7)-10(-5) M was blocked by haloperidol (10(-6) M). During a comparative study of dopamine effect at the early and advanced stages of BC it was found that sensitivity of lymphocytes to the stimulating effect of dopamine at the advanced stages of BS was significantly lower as compared to that at the early stages.

The influence of adrenergic agents: alpha, beta-agonists adrenalin and noradrenaline, alpha-agonist mesaton (phenylephrine) and beta-agonist isadrin on spontaneous adhesion of lymphocytes of healthy donors in vitro was studied. It was established, that the influence on lymphocyte alpha-adrenoreceptors caused stimulation, and on beta-adrenoreceptors--inhibition of the lymphocyte spontaneous adhesion. With the help of alpha-adrenoblocking agent phentolamine and beta-adrenoblocking agent propranolol it was demonstrated, that the adrenergic effects revealed were pharmacologically specific.

RESUMEN

With the aid of lymphocytes' spontaneous adhesion test and the reaction of blast transformation with phytohemagglutinin (PHA) the influence of met-enkephaline upon the functional activity of lymphocytes of healthy donors was studied in vitro. It was shown, that met-enkephaline stimulate lymphocyte spontaneous adhesion and the lymphocyte PHA-induced proliferative activity. The stimulating effects of met-enkephalin revealed in vitro were blocked by the opioid receptor blocker naloxone.

It was found that the stimulating effect of alpha-adrenomimetic mesatone in the test of lymphocyte adhesion inhibition in patients with rectum cancer in vitro was eliminated by beta-adrenergic blocker phentolamine and the inhibitory effect of beta-adrenergic antagonist propranolol. It was shown in vitro that lymphocyte chemoreceptor sensitivity to the stimulating effect of mesatone decreased and sensitivity to the inhibitory effect of isadrine increased in patients with advanced stages of rectum cancer.

The reaction of MIF-producing T cells was studied on the 1st, 6th and 13th days after intravenous immunization in vivo by irradiated spleen cells, immunized and immunizing strains of mice differing in: (a) H-2 antigens; (b) H-2 and minor histocompatibility antigens (non-H-2); (c) non-H-2 antigens, Mls identical; (d) non-H-2 antigens, Mls different. It was demonstrated that in combinations of mice differing in H-2 antigens as well as in H-2 and non-H-2 antigens, the reaction of MIF-producing cells was revealed already on the 1st day after immunization and maintained also on the 6th and 13th days. After intravenous immunization against non-H-2 antigens (Mls identical combination of mice) MIF-production was revealed on the 1st day, whereas in combination of mice differing in non-H-2 antigens and Mls locus on the 1st and 6th days after immunization. The data obtained show that in-vivo immunization by the minor transplantation antigens effectively induces MIF-producing T lymphocytes, and first of all, a special category of these cells--"early" MIF-producers.

It has been demonstrated with the help of intravenous in-vivo immunization with irradiated splenocytes of A.AL anti-A.TL (difference at the K-region of H-2), A. TH anti-A. TL and A. TL anti-A. TH (difference at the I-region of H-2), C57BL/10Sn anti-B10.D2(R107) and B10.D2 (R107) anti-C57BL/10Sn (difference at the D-region of H-2) mice that T cells--MIF producers, induced at different times after induction of the alloimmune response, react to the products of the K- and I- but not to those of the D-region of the H-2 complex.

The macrophage migration stimulation factor (MSF) is produced during the first hours of mixed lymphocyte culture (MLC) in the H-2 system. The activity of MSF is characterized by sharp peaks of migration stimulation in the culture fluids of allogeneic MLC. The peaks appeared every 5 hours. After 12--16 hours in the culture fluids of allogeneic MLC, there has been demonstrated macrophage migration inhibition factor whose activity rose by 48 hours. Optimal concentrations were revealed for stimulation and inhibition of macrophage migrations on the dilution of 1--2-day MLC culture fluids. The existence of the biorhythmical mechanism that controls the macrophage mobility through the balance of soluble mediators with alternative activity is suggested.

C57BL/6 (H-2b) mice were immunized once intravenously by irradiated (1500 rad) spleen cells of mutant strains B6.C-H (z1)Y (H- 2bm1 ) and BB. M505Y (H- 2bm3 ), by the cells of allogeneic strain B10.D2 (H-2d) (in positive control) and syngeneic strain C57BL/6 (in negative control). It was demonstrated that MIF-producing T cells can react to mutant H-2 antigens and that this reaction is revealed on the 1st day after intravenous immunization in vivo. It is suggested that MIF-producing T cells can participate in the immunological surveillance of slight changes in the major histocompatibility complex antigens through mobilization of macrophage effector functions.

The ability of T-cells producing the migration inhibition factor (MIF) to recognize point mutations of the K-end of the H-2 complex was revealed in a primary mixed lymphocyte culture. Nonimmune C57BL/6Y (H-2bm) lymphocytes produce MIF in response to the H-2 antigens of B6..C-G (zI)/Y (H-2bm1) and B6.M505/Y (H-2bm3) mutant strains. Similarly lymphocytes of the mutant strains react to the H-2b wild-type and reciprocally to each other. T-cell dependence of these reactions was shown by their abolition after pretreatment of responder cells with Thy-1.2 antiserum in the presence of complement. The ability of MIF-producing cells to recognize serologically unrecognizable H-2bm1 and H-2bm3 mutant antigens demonstrates high immunologic specificity and sensitivity of the MIF-reaction.

Lymphocytes B10D2 anti-B10 (anti-KbIbDb) produce MIF when incubated with spleen cells of the recombinant KbIb-bearing R107 and Db-bearing R101 lines, possessing either public or private H-2 specificities of the immunizing complex. Cross reactivity revealed in the reaction with "unrelated" lines one week after immunization by allogeneic tumour disappeared during the second week; simultaneously the intensity of the reaction to KbIb and Db region products changes. The cross reaction of anti-Kb MIF producers obtained one week after immunization with cells of Kba and Kbd mutants disappeared two weeks after immunization: the migration inhibition which occurred one week after immunization turns to stimulation during the second week, while the degree of inhibition in the reaction with B10 cells remains unchanged.

The influence of monoclonal antibodies (Mabs) ICO-11 on the ability of lymphocytes from breast cancer patients to react in lymphocyte adherence inhibition (LAI) test has been studied in vitro. It has been demonstrated that Mabs ICO-11 in the dilutions 1/4 and higher blocked the reaction to tumor extracts in LAI-test, without affecting the reaction to the extracts of normal tissues and a spontaneous adhesion of lymphocytes of healthy donors as well. The addition of the control supernatants of myeloma cells X63.Ag8. 653 to the test-system in the same dilutions caused no influence upon LAI-reaction and spontaneous adhesion of lymphocytes from healthy donors. A possible participation of alpha-chains of function-associated antigens in the binding of tumor-associated antigens to T-cells in the inductive phase of the reaction of lymphocyte adherence inhibition in vitro has been suggested.
