RESUMEN

Excitatory and inhibitory neurons in diverse cortical regions are likely to contribute differentially to the transformation of sensory information into goal-directed motor plans. Here, we investigate the relative changes across mouse sensorimotor cortex in the activity of putative excitatory and inhibitory neurons-categorized as regular spiking (RS) or fast spiking (FS) according to their action potential (AP) waveform-comparing before and after learning of a whisker detection task with delayed licking as perceptual report. Surprisingly, we found that the whisker-evoked activity of RS versus FS neurons changed in opposite directions after learning in primary and secondary whisker motor cortices, while it changed similarly in primary and secondary orofacial motor cortices. Our results suggest that changes in the balance of excitation and inhibition in local circuits concurrent with changes in the long-range synaptic inputs in distinct cortical regions might contribute to performance of delayed sensory-to-motor transformation.

RESUMEN

Post-compression can effectively further improve the peak power of laser pulses by shortening the pulse duration. Which has been investigated in various ranges of energy and central wavelength. However, the spatial intensity profile of high-peak-power laser pulses is generally inhomogeneous due to pump lasers, imperfect optical components, and dust in the optical layout. In post-compression, the B-integral is proportional to intensity, and wavefront distortions are induced in the spectral broadening stage, leading to a decrease in focusing intensity. Moreover, the beam intensity may be strongly modulated and beam inhomogeneity will be intensified in this process, causing damage to optical components and limiting the achievement of high peak power enhancement. In this study, to address these challenges, the laser pulse is first smoothed by introducing spatial dispersion using prism pairs or asymmetric four-grating compressors, and then the smoothed pulse is used for post-compression. The simulation results indicate that this method can effectively remove hot spots from laser pulses and maintain high peak power enhancement in post-compression.

Pre-pulses caused by the post-pulses in the optical parametric chirped-pulse amplifier were comprehensively studied for the first time, including the underlying mechanism for the delay-shift of pre-pulses, the intensity variation of pre-pulses affected by the initial delay of post-pulses and the pump energy, and also the nonlinear beat noise. The simulation and measurement confirmed that the high-order dispersion of the pulse stretcher was the main cause for the delay-shift of pre-pulses, which should be similar with the chirped-pulse amplifiers. The intensity of pre-pulses would decrease significantly as the initial delay of post-pulses increased, but would increase with the growth of pump energy. Moreover, the temporal position of the nonlinear beat noise in the experiment was successfully predicted by our simulation. This work could help us better understand the pre-pulses in OPCPA and provide helpful guidance for designing high-contrast laser systems.

CONCLUSION: Upon wound formation, the wound temperature rises in the first 3-4 days until reaching its peak. It then falls at about one week after wound formation. In the second week after wound formation, the wound temperature decreases steadily to the baseline indicating a good wound condition and progression towards healing. While a continuous high temperature is often a sign of excessive inflammation or infection, which indicates urgent need of intervention and treatment.

To identify odors in complex environments accurately, insects have evolved multiple olfactory proteins. In our study, various olfactory proteins of Odontothrips loti Haliday, an oligophagous pest that primarily affects Medicago sativa (alfalfa), were explored. Specifically, 47 putative olfactory candidate genes were identified in the antennae transcriptome of O. loti, including seven odorant-binding proteins (OBPs), nine chemosensory proteins (CSPs), seven sensory neuron membrane proteins (SNMPs), eight odorant receptors (ORs), and sixteen ionotropic receptors (IRs). PCR analysis further confirmed that 43 out of 47 genes existed in O. loti adults, and O.lotOBP1, O.lotOBP4, and O.lotOBP6 were specifically expressed in the antennae with a male-biased expression pattern. In addition, both the fluorescence competitive binding assay and molecular docking showed that p-Menth-8-en-2-one, a component of the volatiles of the host, had strong binding ability to the O.lotOBP6 protein. Behavioral experiments showed that this component has a significant attraction to both female and male adults, indicating that O.lotOBP6 plays a role in host location. Furthermore, molecular docking reveals potential active sites in O.lotOBP6 that interact with most of the tested volatiles. Our results provide insights into the mechanism of O. loti odor-evoked behavior and the development of a highly specific and sustainable approach for thrip management.

In this Letter, we experimentally investigate a new kind of nanosecond pre-pulse, which originates from the bidirectional scattering of crystals in traditional Ti:sapphire multi-pass amplifiers. The experimental results demonstrate that the intensity of scattering-induced pre-pulses is very sensitive to the scattering angle, and the delay time between the pre-pulse and the main pulse is an integer multiple of the light path in each pass of the amplifier. An optimized multi-pass amplifier configuration is proposed, for what is believed to be the first time, to suppress the scattering-induced pre-pulses. The contrast ratio between pre-pulses and the main pulse is enhanced by more than two orders of magnitude, reaching a level of 10-10. This novel multi-pass amplifier configuration is very simple and economical, and provides an effective solution for the temporal contrast enhancement in the nanosecond range.

Background: Heart failure (HF) is defined as the inability of the heart's systolic and diastolic function to properly discharge blood flow from the veins to the heart. The goal of our research is to look into the possible mechanism that causes HF. Methods: The GSE5406 database was used for screening the differentially expressed genes (DEGs). Gene ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), and Protein-Protein Interaction (PPI) network were applied to analyze DEGs. Besides, cell counting Kit-8 (CCK-8) was conducted to observe the knockdown effect of hub genes on cell proliferation. Results: Finally, 377 upregulated and 461 downregulated DEGs came out, enriched in the extracellular matrix organization and gap junction. According to GSEA results, Hoft cd4 positive alpha beta memory t cell bcg vaccine age 18-45 yo id 7 dy top 100 deg ex vivo up, Sobolev t cell pandemrix age 18-64 yo 7 dy dn, and so on were significantly related to gene set GSE5406. 7 hub genes, such as COL1A1, UBB, COL3A1, HSP90AA1, MYC, STAT3 and MAPK1, were selected from PPI networks. CCK-8 indicated silencing of STAT3 promoted the proliferation of H9C2 cells and silencing of UBB inhibited the proliferation of H9C2 cells. Conclusion: Our analysis reveals that COL1A1, UBB, COL3A1, HSP90AA1, MYC, STAT3, and MAPK1 might promote the progression of HF and become the biomarkers for diagnosis and treatment of HF.

RESUMEN

The utilization of carbon framework to guide the growth of the Li dendrites is an important theme for Li metal batteries. The conductivity and electronegative sites of carbon materials will greatly affect the nucleation of Li metal. However, how much these two contributing factors affect the Li plating/stripping stability should be considered. This work presents N, O doped carbon nanofiber framework (CNF) membrane as the interlayer for protecting the Li anode. The amounts of N and O elements and their ratios, the conductivity, the thickness of CNF membrane and their effects on the Li plating/stripping process have been fully analyzed. The voltage profile and the stability of Li plating/stripping process are evaluated by symmetric and asymmetrical coin cells. The lithiophilic heteroatom doped surface mainly works as an excellent guide during the Li plating process, whereas the conductivity and mechanical stability of CNF equalize the current density and confine the volume change in during cycling. With the optimized CNF membrane as the interlayer, both Li metal and Li-S full cells exhibit good capacity properties and cyclic stability.

It is of crucial significance to investigate and suppress pre-pulses on nanosecond time scale because the intense pre-plasma generated by them may have enough time to expand and, thus, cause fatal impact on laser-matter interactions. In this research, we analyze the potential origins of pre-pulses on nanosecond time scale in a typical Ti:sapphire chirped pulse amplification laser system. Based on the analysis, the initial status of these generated pre-pulses in the SULF-1PW laser is measured and investigated. Then different measures, including fine control on the time synchronization and the replacement for the Ti:sapphire, are adopted in the SULF-1PW laser to suppress these pre-pulses with respective origins, which can promote the energy ratio between the main pulse and these pre-pulses by 2-3 orders of magnitude. This research not only improves the temporal contrast of the SULF-1PW laser on nanosecond time scale but also provides beneficial guidance for the design and construction of similar laser facilities.

In the current study, the gut microbiota of patients with and without coronary heart disease was compared and the relationship between gut microbiota distribution, intending to reveal the role of gut microbiota in the coronary atherosclerosis process, was investigated.This study included 50 patients diagnosed with coronary heart disease (CHD) who received conventional coronary angiography or computed tomography angiography and 50 patients with CHD at Changshu No. 2 People's Hospital, Suzhou, China, from May 2020 to January 2021. Trimethylamine N-oxide (TMAO) level was tested and feces were collected, the DNA of the gut microbiota was extracted, and the distribution by 16SrRNA gene sequencing was obtained from the two groups of patients.Plasma TMAO concentrations were significantly higher in patients with CHD (P < 0.001). In the CHD group, 22 patients with multivessel disease had a higher level of TMAO compared with the 28 patients who had the single-vessel disease (P < 0.001). No difference in the gut microbiota diversity was noted between the two groups (P < 0.001). Patients with CHD had a significantly lower proportion of Bacteroidetes phyla and more proportion of Epsilonbacteraeota phyla. At the genus level, patients with CHD had an increased abundance of Enterococcus, whereas healthy controls had significantly higher levels of Streptococcus. Phylogenetic Investigation of Communities by Reconstruction of Unobserved States 2 analysis found that, in the KEGG ORTHOLOGY, the level of choline trimethylamine-lyase gene expression correlated with TMAO production was higher in the fecal microbiome of the CHD group (P < 0.05).Gut microbiota and its product were expected to become a diagnostic marker and a new target for preventing CHD.

Based on the relevant data of construction waste (CW) in the People's Republic of China (PRC) from 2010 to 2018, this study applied K-means clustering algorithm and grey prediction methods to systematically investigate the spatiotemporal characteristic distribution and provincial clustering of CW in the PRC, and predicted the annual output of CW in the next five years from the scientific perspective. Results showed that the annual output of CW in the PRC displayed an overall trend of "rising first and then falling" and "being high in the middle east and low in the northwest," and the areas with obvious agglomeration gradually spread from the west to the middle and eastern regions. The law of development was consistent with the goals of the Chinese government to promulgate urban agglomeration development policies, prefabricated building encouragement policies, and CW management regulations. In the next five years, the annual output of CW in the PRC will increase by a small margin. Thus, all aspects of CW resource management should be conducted in a planned and step-by-step manner.

In this paper, we report that the conversion efficiency and spectrum of femtosecond optical parametric amplification (fs-OPA) can be significantly enhanced by employing a compact cascaded femtosecond OPA (CF-OPA) scheme with the self-compensation of the temporal walk-off between two nonlinear gain media. Correspondingly, the gain related temporal contrast can also be improved. The feasibility of the CF-OPA method using three cascaded BBO crystals is numerically and experimentally analyzed. Moreover, by replacing the conventional fs-OPA with the CF-OPA and optimizing the design, the performance of a nonlinear temporal filter combining cross-polarized wave generation and fs-OPA is comprehensively improved. The experimental results demonstrate the superiority of the CF-OPA scheme, which can generate high-performance cleaned pulses at 1 kHz repetition rate with energy of 340µJ, energy fluctuation below 0.9% (RMS), spectral width of 97 nm (FWHM), Fourier-transform-limited pulse width of 12 fs and temporal contrast better than 10-12. To the best of our knowledge, this is the first reported temporal walk-off self-compensated quasi-collinear CF-OPA geometry adopting three cascaded BBO crystals, which can be easily generalized to other wavelengths or nonlinear crystals. The above nonlinear temporal filter with a CF-OPA scheme has the rarest comprehensive parameters, which can provide excellent seed pulses for PW and 10 PW class femtosecond laser systems.

Viruses are widespread in alfalfa (Medicago sativa L.), representing a key limitation to the production of this important forage plant. Understanding the diversity of plant viruses in alfalfa and their potential vectors will play an important role in management to minimize the emergence, transmission, and impact of viruses. Next-generation sequencing (NGS) targeting the transcriptome was applied to monitor the virus communities in alfalfa and its two main pests, thrips (Odontothrips loti Haliday and Frankliniella intonsa Trybom) and aphids (Acyrthosiphon pisum Mordvilko and Therioaphis trifolii Monell). A comparison of transcriptome datasets with reference databases revealed the presence of eight candidate viruses. Five out of the eight viruses, alfalfa mosaic virus (AMV), Medicago sativa alphapartitivirus 1 (MsAPV1), Medicago sativa deltapartitivirus 1 (MsDPV1), Medicago sativa amalgavirus 1 (MsAV1), and bean yellow mosaic virus (BYMV), were confirmed by RT-PCR. We identified and determined the presence of four RNA viruses from alfalfa samples, two viruses (AMV and MsAPV1) from thrips samples, and one virus (BYMV) from T. trifolii. All sequences isolated from the insect samples were more than 95% identical to the sequences from the alfalfa samples or to sequences from the National Center for Biotechnology Information (NCBI) reference database. The RNA-seq results of this study suggest that AMV and MsAPV1 are the predominant RNA plant viruses infecting alfalfa and that they are carried by the major pests. This lays the foundation for future research on the vectors and transmission of these viruses. In addition, the sequence data have enabled the assembly of the first complete genome sequence of MsDPV1 from alfalfa.

Coherent beam combination (CBC) is a promising technology for achieving several hundred petawatts and even EW-level lasers. However, the measurement of the synchronization error and the time jitter of CBC is one of key technical issues, especially in the few-cycle PW-level laser facilities. In this paper, we demonstrate that the absolute time delay (ATD) and the relative time delay (RTD) for a tiled-aperture CBC can simultaneously be measured by using the double-humped spectral beam interferometry. The experimental study also was demonstrated. A root-mean-square deviation of approximately λ/38 (70 as) and a combining efficiency of 87.3% at 1 Hz closed feedback loop was obtained, respectively. Due to the wide adjustment range and a vast resisting beam energy disturbance capacity, this technique provide an effective and practical solution for measuring simultaneously the ATD and the RTD in the few-cycle PW-level laser pulses CBC.

A novel design of double chirped pulse amplification laser systems implementing a combination of negatively and positively chirped pulse amplification is proposed for the first time. Without utilizing any extra dispersion compensation element, this design can sufficiently cancel out the second-, third- and especially fourth-order dispersion simultaneously, just by optimizing the parameters of the stretcher and compressor in first chirped pulse amplification stage which applies negatively chirped pulse amplification. The numerical results indicate that near Fourier-transform-limited pulse duration about 20fs can be achieved in high-peak-power femtosecond laser systems up to multi-Petawatt level. This design not only provides a feasible solution for the dispersion control in high-contrast and high-peak-power femtosecond laser systems, but also can avoid the degradation of temporal contrast induced by seed energy loss in the presence of additional dispersion compensation components.

Curcumin was used to interfere with acute pancreatitis model rats to explore its possible mechanism. One hundred and twenty rats were randomly divided into blank group, model group, model+curcumin group, model+mock+curcumin group, model+antagonist+curcumin group and model+curcumin+LY294002 group, with 20 rats in each group. The wet/dry weight ratio of pancreatic tissue was measured and the pathological changes of pancreas were observed by HE staining. The apoptosis was detected by TUNEL staining; the levels of serum amylase, lipase, Bcl-2 and Bax were detected by ELISA, and the levels of PI3 K, Akt and p-Akt in pancreatic tissue were measured by Western blot. HE staining showed that curcumin could improve the pathological changes of pancreas and reduce the pathological score of pancreas, while ELISA results showed that curcumin could decrease the levels of amylase, lipase and Bax in peripheral serum and increase the concentration of Bcl-2. Western blot results showed that the expression levels of PI3 K and p-Akt in pancreatic tissue of model rats were up-regulated after the intervention of curcumin, and the apoptosis rate of pancreatic cells decreased in TUNEL staining. The above effects could be weakened by miR-198 antagonist and PI3 K-Akt signal pathway inhibitor LY294002. In conclusion, curcumin has an ideal effect on acute pancreatitis, and its mechanism may be mediated by miR-198-PI3 K-Akt axis.

We have theoretically and experimentally investigated the evolution of the temporal contrast in a 10-PW-level Ti:sapphire laser in the Shanghai Superintense Ultrafast Laser Facility (SULF). The effects induced by the grism pair, spectral shaping filter, and increase in gain on the temporal contrast were investigated. First, it was found that the energy loss of clean seed pulses in the grism pair is a major factor in contrast degradation. Because of the low transmission efficiency of the grism pair (~10%), the temporal contrast is degraded by one order of magnitude. Second, the spectral shaping filter in the regenerative amplifier degrades the temporal contrast by increasing the intracavity loss. Finally, as the amplified spontaneous emission pedestal experiences gain more than the main pulse in Ti:sapphire amplifiers, particularly during saturated amplification, the temporal contrast will further deteriorate as the gain increases in multi-stage Ti:sapphire amplifiers. In addition, the effect on the temporal contrast induced by the extraction during pumping technique in large-aperture Ti:sapphire amplifiers has been considered. According to the investigations described above, the design of the SULF can be further improved. It is predicted that a temporal contrast of over 10-11 can be achieved at a peak power of 10 PW following the improvements. The investigations conducted in this study can provide guidelines for improving the temporal contrast in ultrahigh-peak-power Ti:sapphire lasers.

A transition metal-free and external oxidant-free annulation of substrates having two nitrogen-nucleophilic sites with ClCF2COONa was demonstrated, affording a series of 1,3,5-triazines and quinazolinones in up to 96% yields. Notably, ClCF2COONa was employed as the C1 synthon for valuable heterocycles. Using this protocol, two C-N bonds were formed in one pot via the cleavage of two C-F bonds, one C-Cl bond and one C-C bond. This method avoided the use of a transition metal and an oxidant and generated low toxicity inorganic waste.

The photophysical process of localized surface plasmon resonance (LSPR) is, for the first time, exploited for broadband photon harvesting in photo-regulated controlled/living radical polymerization. Efficient macromolecular synthesis was achieved under illumination with light wavelengths extending from the visible to the near-infrared regions. Plasmonic Ag nanostructures were in situ generated on Ag3 PO4 photocatalysts in a reversible addition-fragmentation chain transfer (RAFT) system, thereby promoting polymerization of various monomers following a LSPR-mediated electron transfer mechanism. Owing to the LSPR-enhanced broadband photon harvesting, high monomer conversion (>99 %) was achieved under natural sunlight within 0.8 h. The deep penetration of NIR light enabled successful polymerization with reaction vessels screened by opaque barriers. Moreover, by trapping active oxygen species generated in the photocatalytic process, polymerization could be implemented without pre-deoxygenation.

Double deformable mirrors (DMs) with different actuator densities are cascaded to optimize the wavefront aberrations to improve the focus intensity of the Shanghai super-intense ultrafast laser facility (SULF), which plans to generate 10 PW laser pulse. The beam aberrations near the focal spot are corrected from 0.556 um to 0.112 um in RMS by a 300-mm DM with a large stroke installed after the compressor. After then, it is further optimized to 0.041 um using a 130-mm DM with a high spatial resolution working after the main amplifier. The corrected beam is focused to 2.75 × 2.87 um2 at the full width at half maximum (FWHM) with an f/2.5 off-axis parabolic mirror (OAP), which contains approximately 27.69% energy. A peak intensity of 2 × 1022 W/cm2 is achieved at the output of 5.4 PW, and it could exceed 1023 W/cm2 in the SULF 10 PW laser facility using an f/1.8 OAP.
