RESUMEN

RTL1/PEG11, which has been associated with anxiety disorders, is a retrotransposon-derived imprinted gene in the placenta. However, imprinting patterns and functions of RTL1 in the brain have not been well-investigated. We found Rtl1 was paternally, but not maternally, expressed in brain stem, thalamus, and hypothalamus of mice, and imprinting status of RTL1 was maintained in human brain. Paternal Rtl1 knockout (Rtl1m+/p-) mice had higher neonatal death rates due to impaired suckling, and low body weights beginning on embryonic day 16.5. High paternal expression of Rtl1 was detected in the locus coeruleus (LC) and Rtl1m+/p- mice showed an increased delay in time of onset for action potentials and inward currents with decreased neuronal excitability of LC neurons. Importantly, Rtl1m+/p- mice exhibited behaviors associated with anxiety, depression, fear-related learning and memory, social dominance, and low locomotor activity. Taken together, our findings demonstrate RTL1 is imprinted in brain, mediates emotional and social behaviors, and regulates excitability in LC neurons.

BACKGROUND: Metagenomic next-generation sequencing (mNGS) has been increasingly applied in sepsis. We aimed to evaluate the diagnostic and therapeutic utility of mNGS of paired plasma and peritoneal drainage (PD) fluid samples in comparison to culture-based microbiological tests (CMTs) among critically ill patients with suspected acute intra-abdominal infections (IAIs). METHODS: We conducted a prospective study from October 2021 to December 2022 enrolling septic patients with suspected IAIs (n = 111). Pairwise CMTs and mNGS of plasma and PD fluid were sent for pathogen detection. The mNGS group underwent therapeutic regimen adjustment based on mNGS results for better treatment. The microbial community structure, clinical features, antibiotic use and prognoses of the patients were analyzed. RESULTS: Higher positivity rates were observed with mNGS versus CMTs for both PD fluid (90.0% vs. 48.3%, p < 0.005) and plasma (76.7% vs. 1.6%, p < 0.005). 90% of enrolled patients had clues of suspected pathogens combining mNGS and CMT methods. Gram-negative pathogens consist of most intra-abdominal pathogens, including a great variety of anaerobes represented by Bacteroides and Clostridium. Patients with matched plasma- and PD-mNGS results had higher mortality and sepsis severity. Reduced usage of carbapenem (30.0% vs. 49.4%, p < 0.05) and duration of anti-MRSA treatment (5.1 ± 3.3 vs. 7.0 ± 8.4 days, p < 0.05) was shown in the mNGS group in our study. CONCLUSIONS: Pairwise plasma and PD fluid mNGS improves microbiological diagnosis compared to CMTs for acute IAI. Combining plasma and PD mNGS could predict poor prognosis. mNGS may enable optimize empirical antibiotic use.

Dry eye disease (DED) is a prevalent ocular disorder with a multifactorial etiology. The pre-angiogenic and pre-inflammatory milieu of the ocular surface plays a critical role in its pathogenesis. DZ2002 is a reversible type III S-adenosyl-L-homocysteine hydrolase (SAHH) inhibitor, which has shown excellent anti-inflammatory and immunosuppressive activities in vivo and in vitro. In this study, we evaluated the therapeutic potential of DZ2002 in rodent models of DED. SCOP-induced dry eye models were established in female rats and mice, while BAC-induced dry eye model was established in female rats. DZ2002 was administered as eye drops (0.25%, 1%) four times daily (20 µL per eye) for 7 or 14 consecutive days. We showed that topical application of DZ2002 concentration-dependently reduced corneal neovascularization and corneal opacity, as well as alleviated conjunctival irritation in both DED models. Furthermore, we observed that DZ2002 treatment decreased the expression of genes associated with angiogenesis and the levels of inflammation in the cornea and conjunctiva. Moreover, DZ2002 treatment in the BAC-induced DED model abolished the activation of the STAT3-PI3K-Akt-NF-κB pathways in corneal tissues. We also found that DZ2002 significantly inhibited the proliferation, migration, and tube formation of human umbilical endothelial cells (HUVECs) while downregulating the activation of the STAT3-PI3K-Akt-NF-κB pathway. These results suggest that DZ2002 exerts a therapeutic effect on corneal angiogenesis in DED, potentially by preventing the upregulation of the STAT3-PI3K-Akt-NF-κB pathways. Collectively, DZ2002 is a promising candidate for ophthalmic therapy, particularly in treating DED.

The biology and diversity of glomerular parietal epithelial cells (PECs) are important for understanding podocyte regeneration and crescent formation. Although protein markers have revealed the morphological heterogeneity of PECs, the molecular characteristics of PEC subpopulations remain largely unknown. Here, we performed a comprehensive analysis of PECs using single-cell RNA sequencing (scRNA-seq) data. Our analysis identified five distinct PEC subpopulations: PEC-A1, PEC-A2, PEC-A3, PEC-A4 and PEC-B. Among these subpopulations, PEC- A1 and PEC-A2 were characterized as podocyte progenitors while PEC-A4 represented tubular progenitors. Further dynamic signaling network analysis indicated that activation of PEC-A4 and the proliferation of PEC-A3 played pivotal roles in crescent formation. Analyses suggested that upstream signals released by podocytes, immune cells, endothelial cells and mesangial cells serve as pathogenic signals and may be promising intervention targets in crescentic glomerulonephritis. Pharmacological blockade of two such pathogenic signaling targets, proteins Mif and Csf1r, reduced hyperplasia of the PECs and crescent formation in anti-glomerular basement membrane glomerulonephritis murine models. Thus, our study demonstrates that scRNA-seq-based analysis provided valuable insights into the pathology and therapeutic strategies for crescentic glomerulonephritis.

BACKGROUND: Extravillous trophoblasts (EVTs) are essential cells during the formation of the placenta, with the major function of invading the maternal decidua, anchoring the developing placenta to the uterus, remodeling uterine arteries, and regulating immune responses to prevent rejection. During early pregnancy, the decidua undergoes a hypoxic and acidic microenvironment, which has been shown to participate in tumor cell migration, invasion, growth, and angiogenesis. Nevertheless, the mechanisms by which EVTs sense and respond to the acidic microenvironment, thereby executing their functions, remain poorly understood. METHODS: The effects of G protein-coupled receptor 65 (GPR65) on cell adhesion and other cellular functions were tested using JAR spheroids, mouse blastocysts, and HTR-8/SVneo cells. Specifically, we employed HTR-8/SVneo cells for gene overexpression and silencing to investigate the underlying mechanism of GPR65's impact on trophoblast cell function under acidic conditions. Additionally, villus tissue samples obtained from early pregnancy loss patients were utilized to explore the potential association between GPR65 and its related signaling pathway molecules with the disease. RESULTS: This study identified GPR65 expression widely in trophoblasts, with the highest level in EVTs. Importantly, optimal GPR65 levels are required for maintaining normal adhesion, migration, and invasion, whereas overexpression of GPR65 inhibits these functions by activating the cAMP-ERK signaling pathway, upregulating myosin light chain kinase (MYLK) and MYLK3 expression, and subsequently downregulating fibronectin. Consistently, elevated expression of GPR65, MYLK, and MYLK3 is observed in patients suffering from early pregnancy loss. CONCLUSIONS: This work offers insights into the suppressive effects of GPR65 on EVT function under acidic conditions and highlights a putative target for therapeutic intervention in early pregnancy complications. In order to develop efficient protocols for CO2 reduction with less expensive and more convenient hydrogen sources, the catalytic reactivities of group 10 metal hydride complexes supported by a PNCNP pincer ligand, [2,6-(tBu2PNH)2C6H3]MH (M = Ni, 1a; Pd, 1b; Pt, 1c), against the hydroboration of CO2 with NH3·BH3 and NaBH4 have been explored. Both 1a and 1b readily react with CO2 at room temperature to form the corresponding formato complexes, [2,6-(tBu2PNH)2C6H3]MOC(O)H (M = Ni, 2a; Pd, 2b), in nearly quantitative yields. Treatment of NH3·BH3 with CO2 (1 atm) in 1,4-dioxane or THF at room temperature in the presence of 0.05-1.0 mol % of 1b followed by hydrolysis of the resulting mixtures produces formic acid in 105-186% yields, and initial turnover frequencies of up to 2000 h-1 are observed. In the presence of 1.0 mol % of 1b, NaBH4 reacts with CO2 (1 atm) in THF at room temperature to form NaB[OC(O)H]4 (3) in 87% isolated yield. In situ NMR spectroscopy indicates that the reactions proceed through the insertion of the C═O bond in CO2 into the Pd-H bond in 1b to form 2b, which sequentially reacts with the hydrides in NH3·BH3 or NaBH4 to produce boron formato species and regenerate 1b. This work represents one of the rare examples of catalytic transfer hydrogenation of CO2 with NH3·BH3 to the formic acid level under very mild conditions without any additives and also the first example of 4 equiv of CO2 uptake by NaBH4 in a reaction.

BACKGROUND: Acute Myeloid Leukemia (AML) is a hematological cancer characterized by heterogeneous hematopoietic cells. Through the use of multidimensional sequencing technologies, we previously identified a distinct myeloblast population, CD34+CD117dim, the proportion of which was strongly associated with the clinical outcome in t (8;21) AML. In this study, we explored the potential value of the CD34+CD117dim population signature (117DPS) in AML stratification. METHODS: Based on the CD34+CD117dim gene signature, the least absolute shrinkage and selection operator (LASSO) Cox regression analysis was performed to construct the 117DPS model using the gene expression data from Gene Expression Omnibus (GEO) database (GSE37642-GPL96 was used as training cohort; GSE37642-GPL570, GSE12417-GPL96, GSE12417-GPL570 and GSE106291 were used as validation cohorts). In addition, the RNA-seq data from The Cancer Genome Atlas (TCGA)-LAML and Beat AML projects of de-novo AML patients were also analyzed as validation cohorts. The differences of clinical features and tumor-infiltrating lymphocytes were further explored between the high-risk score group and low-risk score group. RESULTS: The high-risk group of the 117DPS model exhibited worse overall survival than the low-risk group in both training and validation cohorts. Immune signaling pathways were significantly activated in the high-risk group. Patients with high-risk score had a distinct pattern of infiltrating immune cells, which were closely related to clinical outcome. CONCLUSION: The 117DPS model established in our study may serve as a potentially valuable tool for predicting clinical outcome of patients with AML.

Chikungunya fever is an acute infectious disease caused by the chikungunya virus (CHIKV) that is characterized by fever, rash, and joint pain. CHIKV has infected millions of people in Africa, Asia, America, and Europe since it re-emerged in the Indian Ocean region in 2004. Here, we report an outbreak of Chikungunya fever that occurred in Ruili of Yunnan Province, a city located on the border between China and Myanmar, in September 2019. The outbreak lasted for three months from September to December. Overall, 112 cases were confirmed by a real-time reverse-transcription polymerase chain reaction in the Ruili People's Hospital, and they showed apparent temporal, spatial, and population aggregation. Among them, 91 were local cases distributed in 19 communities of Ruili City, and 21 were imported cases. The number of female patients was higher than that of male patients, and most patients were between 20 and 60 years old. The main clinical manifestations included joint pain (91.96%), fever (86.61%), fatigue (58.04%), chills (57.14%), rash (48.21%), headache (39.29%), and so forth. Biochemical indexes revealed increased C-reactive protein (63.39%), lymphopenia (57.17%), increased hemoglobin (33.04%), neutrophilia (28.57%), and thrombocytopenia (16.07%). Phylogenetic analysis of the complete sequences indicated that the CHIKV strains in this outbreak belonged to the Indian Ocean clade of the East/Central/South African genotype. We speculated that this chikungunya outbreak might be caused by CHIKV-infected persons returning from Myanmar, and provided a reference for the formulation of effective treatment and prevention measures.

The preparation of primary amines from nitriles has been a subject of continuing interest, and many different methods have been reported for this process. We report in this paper an alternative method for transforming nitriles into primary amines. In this work, a wide range of nitriles were reduced to primary amines by 1.2 equiv of ammonia borane under thermal decomposition conditions without any catalyst and the corresponding primary amines were isolated in good to excellent yields. The reactions are environmentally benign with H2 and NH3 generated as byproducts. The reactions are also tolerant of many functional groups. Nitriles are likely reduced by the in situ-generated aminodiborane, the application of which in organic synthesis has never been reported before. By using our protocol, primary amines containing multifluorinated aromatic rings, which are greatly important in pharmaceutical synthesis and have rarely been prepared via catalytic processes, were successfully prepared.

This study investigated the effect of salidroside on phenotypic transformation of rat pulmonary artery smooth muscle cells(PASMCs) induced by hypoxia. Rat pulmonary arteries were isolated by tissue digestion and PASMCs were cultured. The OD values of cells treated with salidroside at different concentrations for 48 hours were measured by cell counting kit-8(CCK-8) to determine the appropriate concentration range of salidroside. The cells were divided into a normal(normoxia) group, a model(hypoxia) group, and three hypoxia + salidroside groups(40, 60, and 80 µg·mL~(-1)). Quantitative real-time PCR(qRT-PCR) was used to detect the mRNA expression of cell contractile markers in each group, such as α-smooth muscle actin(α-SMA), smooth muscle 22(SM22), and calcium-binding protein(calponin), and synthetic marker vimentin. The expression levels of cell phenotypic markers and proliferating cell nuclear antigen(PCNA) were detected by Western blot. The proliferation of cells in each group was detected by the 5-ethynyl-2'-deoxyuridine(EdU) assay. Cell migration was measured by Transwell assay. As revealed by results, compared with the normal group, the model group showed decreased mRNA and protein expression of contractile phenotypic markers of PASMCs and increased mRNA and protein expression of synthetic markers. Compared with the conditions in the model group, salidroside could down-regulate the mRNA and protein expression of synthetic markers in PASMCs and up-regulated the mRNA and protein expression of contractile phenotypic markers. Compared with the normal group, the model group showed potentiated proliferation and migration. Compared with the model group, the hypoxia + salidroside groups showed blunted proliferation and migration of cells after phenotypic transformation. The results suggest that salidroside can inhibit the expression of synthetic markers in PASMCs and promote the expression of contractile markers to inhibit the hypoxia-induced phenotypic transformation of PASMCs. The mechanism of salidroside in inhibiting the proliferation and migration of PASMCs is related to the inhibition of the phenotypic transformation of PASMCs.

BACKGROUND: Mitochondrial DNA (mtDNA) is a critical activator of inflammation. Circulating mtDNA released causes lung injury in experimental models. We hypothesized that elevated plasma mtDNA levels are associated with acute lung injury (ALI) in septic patients. METHODS: We enrolled 66 patients with sepsis admitted to the Department of Critical Care Medicine of Peking Union Medical College Hospital between January 2019 and October 2019. Respiratory, hemodynamic and bedside echocardiographic parameters were recorded. Plasma mtDNA, procalcitonin, interleukin 6, and interleukin 8 levels were examined. RESULTS: Plasma mtDNA levels within 24 h after admission were significantly increased in the group of septic patients with ALI [5.01 (3.38-6.64) vs 4.13 (3.20-5.07) log copies/µL, p 0.0172]. mtDNA levels were independently associated with mortality (hazard ratio, 3.2052; 95% CI 1.1608-8.8500; p 0.0253) and ALI risk (odds ratio 2.7506; 95% CI 1.1647-6.4959; p 0.0210). Patients with high mtDNA levels had worse outcomes, and post hoc tests showed significant differences in 28-day survival rates. Increased mtDNA levels were seen in patients with abdominal infection. CONCLUSIONS: Increased plasma mtDNA levels within 24 h after admission were significantly associated with ALI incidence and mortality in septic patients.

Under the background of global warming, the summer temperature of the North and Northeast China (NNEC) has significantly increased since 2017, which was accompanied by the aggravated ozone (O3) pollution. In 2018, the NNEC experienced a record-breaking summer of the past 40 years. Influenced by the abnormal high temperatures, a regional ozone event occurred on 2-3 August, over 63% of 79 selected cities in the NNEC were exposed to O3 pollution, and the maximum value of MDA8 O3 reached 268 µg/m3. Observations indicated that ozone concentrations agree well with the maximum temperature at 2 meters (MT2M) over NNEC with a correlation coefficient of 0.69. During the pollution episode, strong downdraft in the local high (35°N-42.5°N, 112.5°E-132.5°E; LH) over the NNEC created the favourable meteorological conditions for O3 formation. By analyzing the horizontal wind and wave activity fluxes (WAFs) at 200 hPa, we found that the LH formation was resulted from the Rossby wave propagation from upstream along the mid-latitude Asian jet. The split polar vortex intrusion further strengthened the amplitude of the Rossby wave and reinforced the LH. Moreover, a secondary circulation between Typhoon Jongdari and the LH contributed to the enhanced LH with strong subsidence. On the other hand, the stratospheric intrusions under the deep subsidence also contributed to the enhanced surface O3. In this study, the deep-seated meteorological dynamical mechanisms contributing to the abnormal high temperatures were investigated, which can lead to a better understanding of the regional O3 pollution over NNEC under the global-warming background.

An intensive and persistent regional ozone pollution event occurred over eastern China from 25 June to 5 July 2017. 73 out of 96 selected cities, most located in the Beijing-Tianjin-Hebei and the surrounding area (BTHS), suffered severe ozone pollution. A north-south contrast ozone distribution, with higher ozone (199 ± 33 µg/m3) in the BTHS and lower ozone (118 ± 25 µg/m3) in the Yangtze River Delta (YRD), was found to be dominated by the position of the West Pacific Subtropical High (WPSH) and mid-high latitude wave activities. In the BTHS, the positive anomalies of geopotential height at 500 hPa and temperature at the surface indicated favorable meteorological conditions for local ozone formation. Prevailing northwesterly winds in the mid-high troposphere and warm advection induced by weak southerly winds in the low troposphere resulted in low-moderate relative humidity (RH), less total cloud cover (TCC), strong solar radiation and high temperatures. Moreover, southerly winds prevailing over the BTHS aggravated the pollution due to regional transportation of O3 and its precursors. On one hand, the deep sinking motion and inversion layer suppressed the dispersion of pollutants. On the other hand, O3-rich air in the upper layer was maintained at night due to temperature inversion, which facilitated O3 vertical transport to the surface in the next-day morning due to elevated convection. Generally, temperature, UV radiation, and RH showed good correlations with O3 in the BTHS, with rates of 8.51 (µg/m3)/°C (within the temperature range of 20-38°C), 59.54 (µg/m3)/(MJ/m2) and -1.93 (µg/m3)/%, respectively.

RESUMEN

The historical preparation methods of Glycyrrhizae Radix et Rhizoma were summarized and analyzed by consulting relevant literatures of herbal medicines and medical classics. This study also reviewed the records of Glycyrrhizae Radix et Rhizoma processing methods in previous editions of the Chinese Pharmacopoeia and the regulations on processing technology of Glycyrrhizae Radix et Rhizoma decoction pieces in China. This paper summarized the processing history of Glycyrrhizae Radix et Rhizoma and defined the development process of Glycyrrhizae Radix et Rhizoma processing. According to textual research from ancient times to today, there are many ways to process Glycyrrhizae Radix et Rhizoma. The processing methods without auxiliary materials include braising, frying, cooking, simmering and adding such auxiliary materials as wine, vinegar, salt, oil, ginger, honey, water and bile. There are 9 editions of the published Chinese Pharmacopoeia that document the processing of Glycyrrhizae Radix et Rhizoma, and 24 provinces and cities nationwide record the processing of Glycyrrhizae Radix et Rhizoma. At present, the 2015 edition of the Chinese Pharmacopoeia only records the processing technology of Glycyrrhizae Radix et Rhizoma honey, and the honey processing method is still widely usedtoday. Whether or not Zhigancao should be used uniformly for honey-processed Zhigancao today should be based on the processing methods of Chinese herbal medicine and its clinical use in previous ancient medical books. This paper provides a reference and historical basis for subsequent studies on other processing techniques of Glycyrrhizae Radix et Rhizoma, the rational selection of Glycyrrhizae Radix et Rhizoma varieties and the further development and utilization of corresponding medicinal materials.

A catalytic asymmetric [3 + 2] cycloaddition of 2-indolylmethanols with α,ß-unsaturated aldehydes was developed for the first time. This transformation was achieved by a synergistic catalytic system consisting of a palladium complex, a Brønsted acid, and a chiral secondary amine to synthesize biologically active cyclopenta[b]indole derivatives with excellent diastereo- and enantioselectivities (up to >20:1 dr, up to 99% ee).

A novel magnetic MIPs (DUMIPs) was prepared by surface molecular imprinting method using superparamagnetic core-shell nanoparticle (Fe3O4@SiO2) as the sacrificial support matrix, herbicide diuron as template, α-methacrylic acid as the functional monomer, trimethylolpropane trimethacrylate as the crosslinker, azobisisobutyronitrile as the initiator, and acetonitrile as the porogen. Highly cross-linked porous surface and excellent magnetic property were characterized by Fourier-transform infrared spectroscopy, transmission electron microscopy, and vibrating sample magnetometer, respectively. The adsorption capacity of DUMIPs was 8.1 mg g-1, 2.6-fold over its corresponding non-imprinted polymers (DUNIPs). The adsorption in DUMIPs was considered as multilayer adsorption and posed high affinity to diuron, due to the better fitting to Freundilich isotherm. Competitive recognition study demonstrated DUMIPs had highly selective binding diuron. DUMIPs, as an influential sorbent has been used for selective extraction of diuron from environmental samples (paddy field water, paddy soil and grain seedlings) and the elution was determined by high efficiency liquid chromatography (HPLC). In this analytical method, various factors affecting the extraction efficiency such as pH, sorbent dosage, utilization efficiency and volumes of eluent were simultaneously investigated. Under the optimal conditions, the linearity of the method obtained is in the range of 0.02-10.0 mg L-1. The limit of detection is 0.012 mg L-1. In four spiked levels (0.04, 0.2, 1.0, and 4.0 mg kg-1), the recoveries of diuron in real samples are in the range of 83.56%-116.10% with relative standard deviations in the range of 1.21-6.81%. Importantly, compared to C18-SPE column, the MMIPs exhibited convenient separation by external magnetic field, strong clean-up capacity, and selective enrichment for diuron. Thus, the DUMIPs-based method is great potential for efficient sample preparation in the determination of trace amounts of diuron residues in complex matrices.

Nanocellulose has been subjected to a wide range of chemical modifications towards increasing its potential in certain fields of interest. These modifications either modulated the chemistry of the nanocellulose itself or introduced certain functional groups onto its surface, which varied from simple molecules to polymers. Among many, aliphatic and aromatic mono- and di-isocyanates are a group of chemicals that have been used for a century to modify cellulose. Despite only being used recently with nanocellulose, they have shown great potential as surface modifiers and chemical linkers to graft certain functional chemicals and polymers onto the nanocellulose surface. This review discusses the modification of cellulose and nanocellulose using isocyanates including phenyl isocyanate (PI), octadecyl isocyanate (OI), toluene diisocyanate (TDI), diphenylmethane diisocyanate (MDI), hexamethylene diisocyanate (HMDI), and their derivatives and polymers. It also presents the most commonly used nanocellulose modification strategies including their advantages and disadvantages. It finally discusses the challenges of using isocyanates, in general, for nanocellulose modification.

In order to establish a more realistic radiation model of the sea surface, the effects of solar radiation, sky radiation, and atmospheric thermal radiation on sea surface radiation are taken into consideration, on the basis of which the infrared radiative transfer equation of the sea surface is deduced in this paper. A method for calculating the bidirectional reflection characteristics of the sea surface based on measured data is proposed according to the projection imaging of beam propagation. Based on the measurements of sea surface temperature, incident sky radiation, incident solar radiation, and radiance of sea crests at different times, the radiative transfer equation is used to retrieve the bidirectional reflectance of a midwave infrared sea surface. Meanwhile, the results of the method mentioned above are compared with the calculated results of Cox-Munk, Mermelstein, Wu, and Beckmann bidirectional reflection characteristics models. Research shows that the bidirectional reflectance at the wave crest of a sea surface increases gradually, when the solar incident zenith angle changes from 56.39° to 76.02° as well as the direction of observation remaining constant (θr=80.0°; ϕr=73.0°). The reflection ability at the wave crest of the sea surface is strongest when the incident direction of the sun is close to the observation direction, which is in accordance with the law of reflection. The Cox-Munk model and Wu model are closer to our values when the solar incidence zenith angle is small (θi≤65.93°). On the other hand, the calculated values of the Mermelstein and Wu models are closer to the values in this paper when the solar incidence zenith angle is large (θi≤65.93°). In general, the error of the Beckmann model is a little greater than that of the other three models.

In this study, a new method for the simultaneous quantitative determination of triclopyr and aminopyralid in forage grass, hay, and soil was developed and validated using gas chromatography coupled with electron capture detector (GC-ECD). In this method, a simple and maneuverable esterification reaction was applied to convert the two acidic herbicides into their ester form with methanol. The target compounds were extracted with 1% hydrochloric acid-acetonitrile, esterified, purified by florisil solid-phase extraction cartridge, and detected in a single run by the GC-ECD. The average recoveries using this method, at different fortified levels, ranged from 80% to 104% with intra-day and inter-day RSDs in the range of 1.2-10.8% and 3.3-10.3% for both the herbicides, respectively. The LODs were below 0.02 mg/kg while the LOQs were below 0.05 mg/kg, both of which were much lower than the maximum residue limits (MRLs) of 25-700 mg/kg in pastures, as established by the USA (the code of federal regulations). The open field dissipation and residual analysis in pastures and soil were conducted with the commercial formulation at two locations. With time, both triclopyr and aminopyralid dissipated via first-order kinetics. In forage grass, both compounds degraded rapidly over the first 14- or 21-d period and at a slow rate over the remainder of experimental days. In soil, they degraded at a relatively slow rate, and dissipated steadily to below or close to the LOQ by 60-d post application. The half-lives of triclopyr were 1.4-1.8 d and 6.2-9.0 d and aminopyralid were 1.7-2.1 d and 8.2-10.6 d in terms of forage grass and soil, respectively. The terminal residue results indicated that on 7 d after the treatment, the residues of aminopyralid and triclopyr in forage grass and hay were lower than the MRLs set by the USA. This work can provide guidance on the reasonable use of these herbicides and also provide an analytical method for the determination of triclopyr and aminopyralid in pasture and soil.

OBJECTIVE: To evaluate the clinical efficacy and safety of Qilin Pills in the treatment of oligoasthenospermia in infertile men. METHODS: This multi-centered randomized double-blind controlled clinical trial included 216 infertile males with oligoasthenospermia, 108 in the trial group and the other 108 in the control, the former treated with Qilin Pills at the dose of 6 g tid while the latter with Wuziyanzong Pills at 6 g bid, both for 12 weeks. We examined the total sperm count, sperm motility and the count of progressively motile sperm of the patients before and at 4, 8 and 12 weeks after medication and evaluated the safety of the drug based on the adverse events and the laboratory results of blood and urine routine examinations and liver and kidney function tests. RESULTS: Compared with the baseline, the patients in the trial group showed a significant time-dependent improvement after 4, 8 and 12 weeks of medication in sperm motility (21.75% vs 27.54%, 29.04% and 32.95%, P <0.05), total sperm count (156.27 ×106 vs 177.33, 188.18 and 205.44 ×106, P <0.05), and the count of progressively motile sperm (32.08 ×106/ml vs 46.33, 50.98 and 61.10 ×106/ml, P <0.05). The three parameters above were also improved in the controls, but more significantly in the trial group (P <0.05). CONCLUSIONS: Qilin Pills can evidently improve the semen quality of oligoasthenospermia patients with no obvious adverse events.
