RESUMEN

The interrogation of highly diverse repertoires of heterogeneous cell populations on a single cell basis increases the likelihood that a cell with unique characteristics will be identified. We have developed a new single cell analysis system comprising millions of bundled subnanoliter volume bioincubation chambers for the identification and recovery of target specific antibody secreting cells (ASCs). This platform integrates dual surface screening with dedicated user driven data analysis and automated cell recovery enabling multiple biophysical parameters to be tracked for millions of antibody leads in parallel. This direct clone analysis and selection technology is a clear deviation from current microfabricated well-based approaches and offers drastically enhanced screening throughput, simultaneous dual surface analysis, and rapid automated single cell recovery. The technology is also applicable to screening both bacterial and mammalian antibody secreting cells. We demonstrate the implementation and feasibility of this platform in identifying target specific antibodies from bacterial, hybridoma, and B cell libraries.

RESUMEN

A 15-year-old female Highland dancer presented to the accident and emergency department with an ankle inversion injury on a background of several weeks of pain in the right foot. A radiograph of the right foot demonstrated a stress fracture at the base of the second metatarsal. She was treated conservatively with a below knee removable supportive walking boot with a rocker bottom sole. She re-presented to the accident and emergency department 3 weeks later with pins and needles in the right foot; she was given crutches to use along side the supportive walking boot. Radiographs 12 weeks after the first presentation showed healing of the stress fracture. The patient was now asymptomatic of the injury. She was unable to fully train for 12 weeks due to the injury. Conservative management was successful in this patient.

RESUMEN

Thirteen girls with grade III-V vesicoureteric reflux were treated by endoscopic injection of Teflon paste behind the intravesical ureter. Fourteen of the 18 treated ureters showed complete absence of reflux after one injection of Teflon. Three ureters required a second injection of Teflon for successful treatment of the reflux. One ureter with grade IV reflux was converted to grade II reflux. Properly carried out, this procedure corrects reflux. It takes less than 15 minutes, may be done as a day procedure, and avoids open surgery. There have been no complications. (Reprinted with permission from Br Med J, 289: 7-9, 1984)
