ABSTRACT

The purpose of this study was to evaluate the dose- and time-dependent effect of caffeine treatment on the motility and viability of stallion spermatozoa at different temperatures. Six dose groups (A to F) were established with changing caffeine concentrations (from 0.625 to 10 mg/mL). The control samples were prepared by diluting the ejaculate only with physiological salt solution. The samples were examined after 0, 1, 2 and 3 h of incubation at 5 °C and 37 °C. The motility parameters were evaluated by Computer Assisted Semen Analyzer (CASA) system, and the viability was assessed by the mitochondrial toxicity test at the end of the incubation. A positive effect of the lowest tested caffeine concentration on the motility parameters was observed throughout the incubation period at 5 °C. At the end of the 3h incubation, the viability in every sample in these groups, treated with any caffeine concentration, showed lower values compared to the control. At the higher incubation temperature (37 °C), caffeine positively affected the motility in samples B (P < 0.05) and D, E, F (P < 0.001) after 3 h of incubation; however, the viability showed a slightly decreasing tendency. Our results suggest that caffeine, in an optimal concentration, may be used as a component of stallion semen extenders.

ABSTRACT

Currently, in animal nutrition, the replacement of synthetic substances with natural ones was expected to improve animal health. The aim of the present study was to evaluate the effects of a dietary brown seaweed and plant polyphenol mixture in adult male rabbits on the haematological profile and antioxidant markers. Twenty-four adult male rabbits were divided into three experimental groups receiving a control diet (C) or diets supplemented with 0.3% (T1) and 0.6% (T2) of a feed additive containing brown seaweed (Laminaria spp.) and plant extracts of seaweed origin. The trial lasted for 90 days. A lower potassium concentration was observed at 30 days in the T2 group, compared with the T1 and C groups. An increase in the antioxidant status was observed (P < 0.05) from day 60 of the trial in the rabbits fed diets with an algae-polyphenolic supplement (T1 and T2 groups). Concluding, the diet supplementation of brown seaweed and polyphenol stimulates the antioxidant status of the blood, however, it does not affect the haematological profile.

ABSTRACT

Aminoglycoside antibiotics have been used for treating serious but also routine infections in veterinary and human medicine for many years. The basic aim of this work is to evaluate the cytotoxicity of dihydrostreptomycin and neomycin in vitro on three cell cultures - BHK-21 (Syrian golden hamster kidney fibroblast), VERO (African green monkey kidney fibroblast) and FEA (feline embryonic fibroblast) cells. The morphological changes were examined by Giemsa staining. Cells were dried and visualized under fluorescence microscope. After the exposure to different experimental doses of dihydrostreptomycin (812.5-20000 µg/mL) and neomycin (1000-20000 µg/mL) during 24 h, the viability of BHK-21, FEA and VERO cell lines were evaluated by MTT assay. Viability of BHK-21 cells significantly (P < 0.001) decreased after treatment with 3500; 5500 and 7500 µg/mL of dihydrostreptomycin and 9000; 10000 and 20000 µg/mL of neomycin. The FEA cell viability decreased significantly (P < 0.001; P < 0.01) at 2500 and 3000 µg/mL dihydrostreptomycin and at 3000 µg/mL of neomycin treatment. Only the highest concentration of dihydrostreptomycin (20000 µg/mL) reduced VERO cell viability significantly (P < 0.01). Based on or results we can assume the effect of different antibiotics in different concentrations on cell lines is various. Detection of antibiotic toxicity to animal cells is very important because of the increasing resistance of bacteria. One of the solutions is drug dose increasing, but only to a certain concentration, since the toxic effect over the therapeutic one will prevail, which we have also shown in this work.

ABSTRACT

The purpose of the present study was to evaluate the effects of aflatoxin B1 (AFB1) and benzo[a]pyrene (BaP) on the heart muscle of chicken embryos of the broiler line Ross 308. The benzo[a]pyrene in the organic oil solution was injected in ovo on the 6th day of the incubation in doses of: 0.1, 0.5, and 1 mg/kg weight of eggs; the aflatoxin B1 in the organic oil solution was injected in ovo on the 6th day of the incubation into the yolk in doses of 80, 120 and 240 ng/kg weight of eggs. Multiple biochemical and hepatic parameters have been observed, including sodium, potassium, chloride, cholesterol, uric acid, total proteins, aminotransferase aspartate, and aminotransferase alanine. A low dose of AFB1 and BaP administered in ovo during early embryonic development had a significant impact on chicken embryonic development, as demonstrated by alterations in biochemical, mineral, and hepatic parameters.

ABSTRACT

Copper is an environmental risk factor, which has various effects on reproductive endocrinology. In this study human adrenocortical carcinoma (NCI-H295R) cell line was used as an in vitro biological model to study the effect of copper sulfate (CuSO4.5H2O) on steroidogenesis and cytotoxicity. The cell cultures were exposed to different concentrations (3.90, 62.50, 250, 500, 1000 µM) of CuSO4.5H2O and compared to control group (medium without CuSO4.5H2O). Cell viability was measured by the metabolic activity assay. Quantification of sexual steroid production directly from the medium was performed by ELISA assay. Following 48 h culture of NCI-H295R cell line in the presence of CuSO4.5H2O a dose-dependent depletion of progesterone release was observed even at the lower concentrations of CuSO4.5H2O. The lowest levels of progesterone were detected in groups with the higher doses (≥ 250 µM) of CuSO4.5H2O, which elicited significant cytotoxic action. Testosterone production decreased significantly, and this decline was more prominent in comparison to that of progesterone. The lowest release of testosterone was recorded at 1000 µM of CuSO4.5H2O. The cytotoxic effect of CuSO4.5H2O was evident at all concentrations used in the study. The presented data suggest that copper has detrimental effects on sexual steroid hormones and consecutively on reproductive physiology.

ABSTRACT

The use of artificial insemination in cattle breeding has evolved to global extent, and insemination doses are often shipped via air transport which requires strict radiation-based examinations. For the determination of effect of non-ionizing radiation (NIR), to which are beings frequently exposed due to protection of airport or cultural event security, freshly ejaculated and cryopreserved bovine spermatozoa were used as experimental model. Following radiation with hand-held metal detector in various exposition times (0, 10 s, 15, 30 and 60 min-groups FR, FR10, FR15, FR30 and FR60) the spermatozoa underwent motility and DNA fragmentation analyses. Study on cryoconserved semen treated with NIR was performed in time intervals 0, 10 s, 1 and 5 min (insemination doses radiated before cryoconservation-CB, CB10, CB1, CB5; samples radiated after freezing-CA, CA10, CA1 and CA5). Fresh semen and insemination doses radiated after cryoconservation showed significantly lower total and progressive motility. No effect on motility parameters was detected in semen extended with cryopreservative medium and radiated prior to freezing. Surprisingly, NIR showed a potential to stimulate spermatozoa velocity; however, the effect was modulated throughout the post-thawing incubation. Based on the DNA fragmentation assay, sperm DNA stayed intact. Present study underlines the potential harm of NIR, which is frequently used in everyday life, with overall adverse impact on human and animal reproduction. Current study also points out on interesting short-term spermatozoa stimulation induced by NIR.

ABSTRACT

The goal of this study was to determined polychlorinated biphenyls (PCBs) and organochlorine pesticides in the depot fat of roe deer (Capreolus capreolus) coming from south-western Slovakia. The mutual correlations of the organic pollutants were analyzed. The study included dichlorodiphenyltrichloroethane (DDT), hexachlorobenzen (HCB), alpha-hexachlorocyclohexane and beta-hexachlorocyclohexane (α + ß-HCH), gamma-hexachlorocyclohexane (γ-HCH), and polychlorinated biphenyls (PCB-delor). The gas chromatograph with an electron capture detector ECD was used for analysis. The accumulations of organic pollutant in depot fat of roe deer were in following order: DDT > PCB-delor > α + ß-HCH > HCB > γ-HCH. Among all pollutants, DDT was accumulated significantly in the highest level in the samples. The significantly higher content of DDT, HCB, α + ß-HCH, and γ-HCH was detected in the adult animals when compared to the juveniles. Some strong positive correlations among pollutants, between HCB and DDT, α + ß-HCH and HCB, α + ß-HCH and HCB, between γ-HCH and other pollutants, and between PCB-delor and γ-HCH were found. Game animals are a part of human food chain and monitoring of the environment pollution by PCBs and other organic pollutants are worthy to study.

ABSTRACT

The goal of this study was to monitor the accumulation of aflatoxin B1 in the liver and kidney of brown hares (Lepus europaeus Pall) in the region of south-western Slovakia. A total of 65 samples were involved for analysis by RIA method. Brown hares were divided into the groups according to age, sex and season (month). The sex was determined visually after shooting, and the age was assigned from dried eye lens. The average concentration of AFB1 in the liver of hares was 0.54 ± 0.053 µg/kg, and lower values were measured in the kidney (0.41 ± 0.038 µg/kg). The significantly (P < 0.05) higher values were found in winter months when compared to summer months. According to the age, juvenile animals showed significant higher accumulation of B1 in both organs than adults (P < 0.05). Wild animals can serve as a good model of real environmental contamination. Thus, monitoring of risk factors such as mycotoxins in the environment is important with regard to public health, as game animals constitute an important part of food chain for humans.

ABSTRACT

This study investigated the role of exogenous α-lipoic acid (ALA) in the inflammation caused by zymosan application. Seventy-two adult male white mice were divided into twelve groups: three control groups, three Zymosan groups, three ALA groups and three groups being the combination of Zymosan and ALA. In the experimental groups, the animals were decapitated after 3, 6 and 24 hours after the injection. The activity of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) were determined in the brain, liver and kidneys of the mice. After the injection of Zymosan, it was found that the activity of SOD, CAT and GSH-Px in the brain, liver and kidneys of mice was significantly lower in all time periods. The administration of ALA resulted in an opposite effect, namely, it increased the activity of the enzymes studied in the selected organs of mice. The Zymosan and ALA combination significantly inhibited the decrease in the activity of the enzymes compared with the values obtained in the groups of animals which received Zymosan only. The results of our study, using the Zymosan-induced inflammation, clearly indicate that ALA is an anti-inflammatory agent.

ABSTRACT

Bee drone brood is a beehive by-product with high hormonal activity used in natural medicine to treat male infertility. The aim of the study was to assess the effect of drone brood on stallion spermatozoa during a short-term incubation for its potential use in the equine semen extenders. Three different forms of fixed drone brood (frozen (FR), freeze-dried (FD), and dried extract (DE)) were used. Solutions of drone brood were compared in terms of testosterone, protein, total phenolic content, and antioxidant activity. The stallion semen was diluted with prepared drone brood solutions. The computer-assisted semen analysis (CASA) method was employed to evaluate the movement characteristics of the diluted ejaculate. To determine spermatozoa viability, the mitochondrial toxicity test (MTT) and Alamar Blue test were performed. In terms of testosterone content and antioxidant activity, a close likeness between FR and FD was found whereas DE's composition differed notably. FR had a positive effect mainly on progressive motility, but also on sperm distance and speed parameters after 2 and 3 h of incubation. On the contrary, FD and DE acted negatively, depending on increasing dose and time. For the first time, a positive dose-dependent effect of fixed drone brood on spermatozoa survival in vitro was demonstrated.

BACKGROUND: The main objective of the study was to analyze the potential ability of vitamins E, C, and D, used as nutritional supplements, in averting inflammation and oxidative stress in the course of diabetes mellitus. METHODS: Male mice were divided into eight groups. Diabetes was induced (groups II, VI, VII, and VIII) by an intraperitoneal injection of streptozotocin (STZ). The third and sixth groups were given vitamin C (50 mg/kg) 3 times per week, the fourth and seventh groups were given vitamin E (300 mg/kg) 3 times per week, and the fifth and eight groups were given vitamin D (2000 IU/day). Interleukin-6 levels were measured in serum. Glutathione (GSH) levels and glutathione peroxidase (GPx), superoxide dismutase (SOD), and catalase (CAT) activity were measured in the liver tissue. RESULTS: STZ resulted in a significant decrease in all tested enzymes and glutathione levels, and an increase in IL-6 level in comparison to control animals (p < 0.05). Mice treated with vitamins had a significantly (p < 0.05) higher content of enzymes and glutathione in liver than diabetic mice, however IL-6 concentration showed a significant decrease. Concurrent administration of STZ and vitamins caused a significant increase (compared to the diabetes group) in SOD, CAT, GPx, GSH content, and a decrease in IL-6 levels (p < 0.05). CONCLUSION: These results indicate the preventive role of vitamin C, E, and D against STZ-induced diabetic oxidative stress and inflammation. Hence these vitamins could be used as an adjuvant therapy for the prevention and/or management of diabetes.

ABSTRACT

The aim of this study was to investigate the effects of lead (Pb) and cadmium (Cd) content on basic motility characteristics (motility-MOT, progressive motility-PROG) as well as selected markers of the prooxidant-antioxidant balance (catalase-CAT, glutathione-GSH, malondialdehyde-MDA) in bovine seminal plasma and spermatozoa. Twenty five semen samples were collected from breeding bulls and used in the study. Motility analysis was carried out using the Computer Assisted Sperm Analysis (CASA) system. The samples were centrifuged, fractions of seminal plasma and spermatozoa were separated, lysates were prepared from the sperm cell fractions. Pb and Cd concentrations were determined by the voltametric method (ASV), antioxidants and MDA were analyzed by UV/Vis spectrophotometry. The analysis showed that the average concentration of Pb in the seminal plasma was 0.23 ± 0.02 µg/mL, while its amount in the sperm cells was significantly higher (0.41 ± 0.07 µg/mL; P < 0.05). The average Cd content in bovine seminal fractions was similar and non-significant: 0.09 ± 0.01 µg/mL in the seminal plasma and 0.11 ± 0.01 µg/mL in spermatozoa (P > 0.05). The correlation analysis revealed that both heavy metals were significantly negatively correlated with MOT and PROG (P < 0.05; P < 0.01; P < 0.001), CAT (P < 0.05; P < 0.01) as well as GSH (P < 0.05; P < 0.01) but significantly positively associated with MDA as the marker of lipid peroxidation (P < 0.05; P < 0.01). Moreover the samples were categorized in three quality groups (Excellent, Good, Moderate) according to their motility values. The lowest Pb, Cd and MDA concentrations but the best antioxidant characteristics were found in samples of the best quality, moderate quality samples exhibited the highest Pb, Cd and MDA content together with the worst antioxidant capacity. This study demonstrates that Pb and Cd are serious toxic elements, which are able to increase the risk of seminal oxidative stress development and a subsequent decrease of male fertility.

ABSTRACT

The performance of sport horses is conditioned not only by the quality of its gene pool, but also by a large number of external factors. The most dominant being nutrition, quality of breeding, level of zootechnical care and the quality of the sports rider and coach. Important factor is the process of individuals' adaptation to the training load occurring during the training itself. This study was focused on the analysis of salivary cortisol levels as well as hematological and biochemical blood parameters in relation to load to which the tested horses were subjected. In the study 14 horses of sport breeds were analyzed a all tested horses were in the same (medium) level of training load. Tested horses underwent following stages of workload - transportation, jumping training, parkour competition, treadmill training, riding training, shoeing and lunging of various intensity. Saliva samples were obtained using a tampon on a string which was inserted into horse's oral cavity, chewed by the horse and placed in a sterile tube with a closable lid. Afterwards, the samples were then stored in deep-freezing boxes at temperature of -80 °C. The EIA cortisol kit was used in this study. The absorbance was read at the wavelength of 450 nm against a reference wavelength of 620-630 nm or a blank sample. Blood samples were obtained at the beginning of the experiment, after half a year of running the experiment and at the end of the experiment from v. jugularis. Hematological analysis were carried out using automatic hematologic analyser and multiple parameters were observed. Analysis of biochemical parameters in blood serum were realized using commercial DiaSys kits and semiautomatic biochemical spectrophotometer. Sodium, potassium and chlorides were measured using automatic analyzer EasyLytePlus. In all monitored forms of exercise (transportation, jumping training, parkour competition, treadmill training, riding training, horse shoeing, lunging), an increase in cortisol concentrations immediately after the exercise was recorded, but only spotted statistically significant differences were found during the transportation of monitored horses. The levels of blood parameters were within the reference range during the experiment period. From a comprehensive evaluation of the results, it can be stated that there were no visible health changes to the horses that underwent the experimental load and that manipulation with horses is an important factor that has effect on horses' stress response. In general, the results of this study show no visible impact of training and/or load on the health status of horses over entire 12-month duration of the experiment.

ABSTRACT

This study was aimed to determine the impact of different taurine and caffeine combinations on the motility, viability, and oxidative markers of chilled stallion spermatozoa. Each stallion semen sample was diluted in a ratio of 1:2, with various taurine and caffeine concentrations (2.5-7.5 mg/mL taurine + 0.625-1.25 mg/mL caffeine) dissolved in a conventional extender. The control samples (CON) were prepared by diluting ejaculate only using the conventional extender. The motility was analyzed using a CASA system at different time intervals (0, 6, 12, 24, and 30 h) and the viability was evaluated using a mitochondrial toxicity test (MTT) performed at the end of the incubation at 5 °C. The liquid part of experimental samples was separated by centrifugation after 30 h of incubation and underwent the evaluation of oxidative stress via the quantification of markers ferric reducing ability of plasma (FRAP) and total oxidant status (TOS). The samples that were treated with a combination of taurine and caffeine significantly improved the motility parameters, mainly after 12, 24, and 30 h of incubation. Samples extended with combination of taurine and caffeine neither compromise viability nor alterations of redox status. The results of this study describe the combination of taurine and caffeine as an optimal supplement for improving the quality of stallion semen during chilled storage.

BACKGROUND: Conservation of species diversity is the need of the hour for preserving life forms on Earth. Extinction of any part of the ecosystem has negative impacts on many processes and systems. The objective of this work was to analyze some biochemical and molecular indicators and their correlations to biogenic elements and heavy metals in Testudo hermanni (n = 16). METHODS: Biochemical parameters were analyzed using the commercial kit DiaSys and biochemical analyzer Randox RX Monza. Sodium, potassium, and chlorides were measured using the EasyLite analyzer. Oxidative stress was evaluated using colorimetric and luminometric methods. Quantification of chemical elements in the blood was carried out using inductively coupled plasma mass spectrometry (ICPS). RESULTS: Biochemical values of analyzed samples from Hermann's tortoises were almost the same as referential values described by multiple authors, with minor aberrations in the total protein parameter. Values of arsenic (As) and nickel (Ni) showed correlation with biochemical parameters and the parameters of oxidative stress. Cadmium (Cd) exhibited correlation with aspartate aminotransferase (AST). CONCLUSIONS: This study reports correlations among four heavy metals, and their levels were again correlated with biochemical and molecular parameters in Hermann's tortoises.

The aim of the study was to delineate the protective effect of ascorbic acid with plausible mechanism after single and repetitive cadmium administration to Swiss mice. The effects of single high dose administration of CdCl(2) (6 mg/kg) or ascorbic acid (AsA) (50 mg/kg) and chronic (three times) administration of Cd at low dose (2 mg/kg) or AsA at same dose (50 mg/kg) were compared in Swiss albino mice. Changes of lipid peroxidation [determined by the malonyldialdehyde (MDA) concentration] were taken as a measure of the oxidative stress intensity. Lipid fatty acid's unsaturation related to the permeability of cell membranes was also examined. Mobilization of the immune system was determined by analyzing changes in antioxidant concentrations of AsA and glutathione (GSH), and by measuring the activation of antioxidant enzymes SOD, GPx and CAT. In addition, the level of free polyamines and variation in their proportions were examined. In conclusion, exposure to higher levels of cadmium will have more deleterious effects on the body rather than chronic exposure at lower levels with this toxic metal, while this study clearly demonstrated the protective effects of AsA in a mouse model.

ABSTRACT

Health state of animals undergoing experimental procedures is an important topic nowadays, as even the small changes can influence the outcome of entire outcomes. Main aim of the study was to evaluate the influence of horse training on variety of blood parameters including mineral profile, energy profile, hepatic profile and haematology. In the experiment, the studied group of horses underwent training programme which consisted of transportation, lounging, riding, jumping, racing, treadmill training and shoeing. Blood samples were collected and later evaluated at the beginning, in the middle and at the end of 1-year lasting process. Our results show multiple significant changes in blood parameters, including changes to multiple minerals, such as Ca, K, Na as well as significant changes of total proteins, urea and certain hepatic profile parameters. Haematology results have also been affected in individual sample collections. Based on results of our study we can state that there have been changes to the internal milieu of the horse but also that there have not been any visible changes of the health status of the animals over the duration of the experiment.

ABSTRACT

This study monitored the chemical and biochemical composition of bovine seminal plasma (SP). Freshly ejaculated semen (n = 20) was aliquoted into two parts. The first aliquot was immediately assessed to determine the sperm motion parameters. Another motility measurement was performed following an hour-long co-incubation of spermatozoa with SP at 6 °C. The other aliquot was processed to obtain the SP. Seminal plasma underwent the analyses of chemical composition and quantification of selected proteins, lipids and RedOx markers. Determined concentrations of observed parameters served as input data to correlation analyses where associations between micro and macro elements and RedOx markers were observed. Significant correlations of total oxidant status were found with the content of Cu and Mg. Further significant correlations of glutathione peroxidase were detected in relation to Fe and Hg. Furthermore, associations of chemical elements and RedOx markers and spermatozoa quality parameters were monitored. The most notable correlations indicate beneficial effects of seminal Fe on motility and Mg on velocity and viability of spermatozoa. On the contrary, negative correlations were registered between Zn and sperm velocity and seminal cholesterol content and motility. Our findings imply that seminal plasma has a prospective to be developed as the potential biomarker of bull reproductive health.

Zinc plays a very important role in various biological activities of the body. Multifaceted role of zinc is also known in testes development, spermatogenesis, capacitation and has effect on spermatozoa motility. On the other hand, the growing industry of nanotechnology has created reasonable interest of the risk assessment for nanoparticles. The aim of this study was to evaluate in vitro effect of zinc oxide (ZnO) nanoparticles on rabbit spermatozoa. Fresh semen was collected from sexually mature New Zealand rabbits. Experimental groups were prepared by diluting semen with ZnO nanoparticles in seven different concentrations (6-391 mg/mL). The experimental groups were compared with control group. Semen was assessed using computer assisted semen analysis (CASA) at intervals of 0, 1, 2 and 3 h of incubation. The mitochondrial toxicity assay (MTT) assay was used to determine cell viability. The results of monitored motility parameters in experimental groups showed a decreasing trend during whole experiment. Significant decrease (P < 0.001) of motility and progressive motility was observed after 3 h of incubation in samples cultured with higher ZnO nanoparticles in comparison to the control group. After 3 h of incubation, viability of rabbit spermatozoa showed slightly increased values in group with the lowest concentration of ZnO nanoparticles, but in other groups viability showed non-significant decrease compared to control. Similar tendency was detected for spermatozoa membrane integrity. These original data show the negative dose-dependent effect of ZnO nanoparticles on spermatozoa motility and viability parameters.

The aim of present study was to verify antagonistic effect of acrylamide (AA) and ethanol (Et) on bone quality parameters. Adult mice (n = 20) were segregated into four groups following 2 weeks administration of toxins: group E1, which received AA (20 mg/kg body weight daily); group E2, which received 15% Et (1.7 g 100% Et/kg body weight daily); group E12, which received simultaneously both toxins; and a control group. An insignificant impact of individual applications of AA, Et or their simultaneous supplementation on the total body weight of mice and the length and weight of their femoral bones was identified. In group E1, higher levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), triglyceride (TG), a decreased level of glutathione (GSH) and elevated endocortical bone remodelling were determined. A significantly lower relative volume of cortical bone, bone mineral density (BMD), elevated endocortical bone remodelling and cortical porosity, higher levels of ALT, AST, lower values for total proteins (TP), GSH, alkaline phosphatase (ALP), calcium, and phosphorus were recorded in group E2. In the mice from group E12, the highest endocortical bone remodelling, decreased values for BMD, TP, GSH and ALP and increased levels of ALT and AST were found. Our findings confirmed the antagonistic impact of AA and Et at doses used in this study on biochemical and morphological parameters consistent with bone health in an animal model.
