ABSTRACT

World mushroom production in 2018 was approximately 8.99 million tonnes. The most commonly cultivated species in the Czech Republic are Agaricus spp., which are sold fresh or canned. In 2017, 2018 mushroom production in the Czech Republic was approximately 540 tonnes. Vermicomposting is an easy and less ecologically harmful way to process the spent mushroom substrate. Earthworms, which are referred to as the engine of the process of vermicomposting, and microorganisms, help convert organic waste into fertilizer. This study is concerned with laboratory vermicomposting in a system of continuous feeding of earthworms Eisenia andrei. It compares the differences between variants with and without earthworms. The dry matter percentage was approximately 20% or more in both variants. The variant with earthworms showed a significant decrease in electrical conductivity. The C/N ratio was very low in both variants. The highest total P was recorded in the variant with earthworms, but the highest values of K and Mg were found in the control. Both variants recorded higher content of bacteria than fungi. All values of microorganism contents were higher in the vermicomposter without earthworms, but the bacterial/fungal ratio was higher in the variant with earthworms. The highest content in both variants shows the bacteria especially G-bacteria, on the other side, the lowest content shows the actinobacteria. The highest activity of ß-D-glucosidase and acid phosphatase was measured in the vermicomposter with earthworms, but the activity of other enzymes was higher in the control. In both vermicomposters laccase activity was below the detection limit. The method of classical vermicomposting can be used for processing the spent mushroom substrate. However, in terms of higher content of total and available nutrients, there seems to be a better method of processing the substrate without earthworms.

ABSTRACT

Cadmium (Cd) and its mobility and toxicity in the food chain receives considerable public and scientific attention. Levels in soil have been increasing, partly caused by mineral fertiliser use. In many areas in the EU, intake of Cd is not far from the maximum tolerable level. Now, urban and more circular approaches to food production are being promoted, in which waste materials substitute growing media and fertilizers. Can this make the situation worse, or is it, on the contrary, possible to extract Cd from the cycle using practical microbial methods? Cd concentration, availability and accumulation in materials and crops in a closed greenhouse system was investigated first. Secondly, we tested if fungal remediation was able to remove a significant amount of Cd. A sequential extraction analysis showed how anaerobic digestion and composting changed chemical Cd availability. Cd levels in the system were comparable to agricultural soils. The vegetable crops did not accumulate Cd. However, the culture of the edible mushroom Agaricus subrufescens in the greenhouse had a strong effect on Cd mobility: It extracted 80% of the Cd from the growing medium into the mushrooms, mostly at the start of fruiting. These early Cd enriched mushrooms can be treated as special waste, while the later harvest complied with EU Cd limits for foodstuffs and can potentially be consumed. Thus, growing this mushroom species in an integrated waste-to-crop system can be a cost-efficient way to remediate Cd from the food chain.

ABSTRACT

The main by-product of wine-making is grape marc. With proper treatment, grape marc may return to the vineyard as a fertiliser. This study deals with the vermicomposting of grape marc in a continuous feeding system in outdoor conditions for more than 12 months. The N-NH4+, dissolved organic carbon (DOC), and N-NH4+/N-NO3- contents were greater in the top layers. The pH value was about 8 in all the layers. The electrical conductivity was the greatest in the bottom layer. The ion-exchange capacity did not modify significantly during vermicomposting. The microbial biomass was the greatest in the upper layer, as well as the number and the biomass of the earthworms. The process of vermicomposting seems to be an ideal way of processing residues from the winemaking industry. This vermicompost has very good properties for use as a fertiliser, and for returning the nutrients and organic matter to the soil, for example, in a vineyard.

ABSTRACT

Processing sewage sludge can be problematic due to its potential environmental toxicity. It may contain high concentrations of pharmaceuticals, polycyclic aromatic hydrocarbons, and heavy metals, as well as pathogenic microorganisms. However, it is a good source of organic matter and rich in microbial communities and enzymatic activity. This study deals with composting and vermicomposting of pre-composted mixtures of two different kinds of sewage sludge blended with moulded pulp in an operating composting plant. Of the total number and concentration of pollutants detected in individual piles, a large percentage of them were reduced by the composting process. The composting 2 process resulted in the greatest reduction in contaminating substances--a total of 19 substances by 4.39-90.4%. Some pharmaceuticals accumulated in earthworm bodies during vermicomposting; a total of 11 substances were detected. Atorvastatin showed the highest percentage reduction in compost 2 (90.4%), vermicompost 1 (65.2%) and vermicompost 2 (97.3%). Both composting and vermicomposting appeared to be effective for removal of heavy metals. A higher content of microbial phospholipid fatty acids (PLFAs) was found in composts than vermicomposts. There was a significant reduction in the content of pathogenic microorganisms in both processes, but the reduction in enterococci was not significant.

ABSTRACT

Environmental pollution by heavy metals affects both urban and non-urban areas of Europe and the world. The use of bioindicator plants for the detection of these pollutants is a common practice. An important property of potential bioindicators is their easy availability and wide distribution range, which means that they can be practically used over a wide area. Therefore, common and widely distributed weeds: Trifolium pratense L., Rumex acetosa L., Amaranthus retroflexus L., Plantago lanceolata L., ornamental species Alcea rosea L., and Lolium multiflorum L. var. Ponto were selected as a potential bioindicators of heavy metals (Cd, Pb, Cu, Zn). Plants were exposed in the same soil conditions in three sample sites in the Poznan city. It was found that all species had heavy metal accumulation potential, especially A. rosea, P. lanceolata and L. multiflorum for Zn (BCF = 6.62; 5.17; 4.70) and A. rosea, P. lanceolata for Cd (BCF = 8.51; 6.94). Translocation of Cu and Zn was the most effective in T. pratense (TFCu = 2.55; TFZn = 2.67) and in A. retroflexus (TFCu = 1.50; TFZn = 2.23). Cd translocation was the most efficient in T. pratense (TFCd = 1.97), but PB was the most effective translocated in A. retroflexus (TFPb = 3.09).. Based on physiological response to stress, it was detected an increasing level of hydrogen peroxide (H2O2) in roots and leaves of all samples, with the highest in all organs of A. rosea. Enzymatic activity levels of CAT, APOX, and also the marker of polyunsaturated fatty acid peroxidation MDA, were higher after 6 weeks of exposure in comparison to control samples and varied in time of exposure and between species and exposure. After the experiment, in almost all samples we detected a reduction of chlorophyll content and relative water content, but in efficiency of photosynthesis parameters: net photosynthesis rate, intercellular CO2 concentration and stomatal conductance, we noted increased values, which proved the relatively good condition of the plants. The examined weeds are good bioindicators of heavy metal contamination, and their combined use makes it possible to comprehensively detection of environmental threats.

ABSTRACT

The study was aimed to evaluate the influence of the vascular disease, atherosclerotic obliterans (AO), on the location and concentration of elements in the arterial wall and serum. Use of a modern method for studying element's concentration and distribution in samples of clinical material, i.e. laser ablation inductively coupled plasma mass spectrometry, is presented. Elements are not equally distributed between the inner (intima) and the outer (media + adventitia) layer of the arterial wall. Among the studied elements, calcium was found to have an unquestionable role in the calcification of the wall. Increased concentration of calcium found in the inner part of the atherosclerotic arterial wall and in the plaque, as compared to the control arterial wall samples, demonstrates the unquestionable role of this element in the calcification of the wall observed in AO. Applied chemometric methods were useful for demonstrating the differences in the element's concentration in blood serum and the arterial wall samples between AO and the control group.

ABSTRACT

Pleurotus ostreatus is one of the most cultivated mushrooms in the Czech Republic. The production of 1 kg of mushrooms generates about 5 kg of spent mushroom substrate. A gentle and fast method for using this substrate is vermicomposting. Vermicomposting of spent mushroom substrate using Eisenia andrei was conducted for seven months. For control purposes, a treatment without earthworms was also prepared. The vermicomposting process used vertical continuous feeding vermicomposters. The agrochemical and biological parameters were analysed. Values of electrical conductivity were very high in both vermicomposters (higher than 2000 µS/cm). During the vermicomposting process the C/N ratio decreased. The number and biomass of earthworms decreased with the age of the layers. The values of total P, K and Mg were higher in the vermicomposter without earthworms. There were also lower microbial phospholipid fatty acids content - than in the vermicomposter with earthworms. However, the fungal phospholipid fatty acids content were two times higher in the vermicomposter without earthworms. The highest hydrolytic enzyme activity was found in lipase, acid phosphatase and ß-D-glucosidase. Most hydrolytic enzymes were more active in the vermicomposter without earthworms, with the exception of arylsulphatase. Mn-peroxidase activity was higher in the vermicomposter without earthworms and laccase activity was below the detection limit.

ABSTRACT

Amniotic fluid is the substantial factor in the development of an embryo and fetus due to the fact that water and solutes contained in it penetrate the fetal membranes in an hydrostatic and osmotic way as well as being swallowed by the fetus. Elemental composition of amniotic fluid influences the growth and health of the fetus, therefore, an analysis of amniotic fluid is important because the results would indicate abnormal levels of minerals or toxic elements. Inductively coupled plasma mass spectroscopy (ICP-MS) is often used for determination of trace and ultra-trace level elements in a wide range of matrices including biological samples because of its unique analytical capabilities. In the case of trace and ultra-trace level analysis detailed characteristics of analytical procedure as well as properties of the analytical result are particularly important. The purpose of this study was to develop a new analytical procedure for multielemental analysis of 18 elements (Al, As, Ba, Ca, Cd, Co, Cr, Cu, Mg, Mn, Ni, Pb, Sb, Se, Sr, U, V and Zn) in amniotic fluid samples using ICP-MS. Dynamic reaction cell (DRC) with two reaction gases, ammonia and oxygen, was involved in the experiment to eliminate spectral interferences. Detailed validation was conducted using 3 certified reference mterials (CRMs) and real amniotic fluid samples collected from patients. Repeatability for all analyzed analytes was found to range from 0.70% to 8.0% and for intermediate precision results varied from 1.3% to 15%. Trueness expressed as recovery ranged from 80% to 125%. Traceability was assured through the analyses of CRMs. Uncertainty of the results was also evaluated using single-laboratory validation approach. The obtained expanded uncertainty (U) results for CRMs, expressed as a percentage of the concentration of an analyte, were found to be between 8.3% for V and 45% for Cd. Standard uncertainty of the precision was found to have a greater influence on the combined standard uncertainty than on trueness factor.

ABSTRACT

The effect of soil ozonation on Pb and Zn extraction with EDTA, bioavailability (Ruby's Physiologically Based Extraction Test, PBET) and mobility (Toxicity Characteristic Leaching Procedure, TCLP) of Pb was studied on contaminated soils taken from 7 different locations in the Mezica Valley, Slovenia. EDTA extraction (40 mmol kg(-1)) removed from 27.4+/-1.5% to 64.8+/-1.5% of Pb, and from 1.9+/-0.2% to 22.4+/-2.0% of Zn from tested soils, and significantly reduced soil Pb bioavailability (PBET) and mobility (TCLP). Pretreatment of tested soils with ozone before EDTA extraction enhanced EDTA extractability of Pb for 11.0 to 28.9%, but had no effect on the extractability of Zn. In most of the soils, ozonation had no statistically significant effect on bioavailability and mobility of Pb, residual after EDTA extraction. Using linear regression analysis we found a significant increase (p<0.01) in EDTA extractability of Pb after soil ozonation in soils with a higher initial Pb content. EDTA extractability of Pb after soil ozonation was also significantly higher for soils with a lower Pb extractability when treated with EDTA alone. We found no correlation between soil organic matter content and the percentage of the Pb fraction bound to soil organic matter (where from 25.6+/-1.3% to 73.2+/-0.6% of Pb reside in tested soils) and Pb extractability with EDTA after soil ozonation.
