ABSTRACT

In this paper, on the contrast of healthy leech, the bacterial diversities were analyzed by 16S rDNA sequence analysis of the bacteria of muscle and intestinal tract of Whitmania pigra, the environment water and sediment of cultivating the diseased Wh. pigra in high temperature by high-throughput sequencing to determine the possible pathogenic bacteria of bacterial diseases of Leech in high temperature. The results showed that the original sequence reached over 83 000, and the effective sequences accounted for more than 87%. The GC contents ranged from 52% to 54% and the bacterial diversities were abundant. Bacterial relative abundance analysis showed that the bacterial content of Proteobacteria, Bacteroidetes, and Firmicutes was the most abundant in all treatments. Compared with healthy leech muscles and intestines, the muscle and intestinal tract of pathogenic leech relative abundance of Bacteroides, Pseudomonas, and Desulfovibrio was significantly increased, and it was abundant in water and sediment of diseased leeches, Lead to the possibility that the pathogenic bacteria of this bacterial disease may be Bacteroides, Pseudomonas, Desulfovibrio.

ABSTRACT

To establish a phenylketonuria screening model by FTIR/ATR spectroscopy, and to compare the effects of different pretreatment methods, such as baseline correction, smoothing, derivation, Fourier deconvolution, on the model quality. A consensus partial least squares regression method (cPLS) was used to build the quantitative model of phenylalanine in dried blood spots. The effects of different pretreatment methods on the model performance were investigated, using the correlation coefficient (r), root mean square error of prediction (RMSEP), mean relative error (MRE) and predictive accuracy (Acc). The nine-point smoothing coupled with the first differential was found to perform the best. Compared with the model by the original spectra, its r, RMSEP, MRE and Acc were improved from 0. 822 7, 115. 8, 0. 395 and 94. 6 to 0. 889 9, 102. 2, 0. 286 and 100, respectively. With the advantages of fast speed, easy process, no reagents consumption and environmental protection, the present method is expected to become a simple and green technology for rapidly screening the neonatal phenylketonuria in a large population.

ABSTRACT

Objective: Cervical cancer is a common gynecological malignancy. In addition to the open radical hysterectomy (ORH) and laparoscopic radical hysterectomy (LRH), laparoscopic nerve-sparing radical hysterectomy (LNSRH) could be another treatment option since it could preserve urinary, colorectal, and sexual functions. However, LNSRH might result in early cancer metastasis and recurrence due to inadequate tumor resection. Additionally, whether LNSRH should be considered based on perineural invasion (PNI) status remains controversial. To assess different types of hysterectomy on the outcome of early cervical cancer with PIN. Methods: A retrospective study was performed in early cervical cancer patients who received ORH, LRH, or LNSRH between January 2012 and December 2019. Age, FIGO cancer stages, histopathological types, tumor size, histological grade, invasion depth, lymph node metastasis, lymphovascular space invasion, and PNI were documented. Disease-free survival (DFS) and overall survival (OS) were recorded. Results: A total of 174 patients were included, with 33, 69, and 72 patients received LRH, ORH, and LNSRH, respectively. Twenty-one patients (12.1%) had PNI. DFS (P = 0.000) and OS (P = 0.022) periods were shortened in positive PNI patients than in negative PNI patients (P = 0.000 and 0.022, respectively). In patients with positive PNI, lymph node metastasis, but not the surgery type, was an independent risk factor for DFS and OS (P = 0.000). Conclusion: Early cervical cancer patients with PNI had shorter postoperative DFS and OS periods. In these patients, lymph node metastasis, but not the type of hysterectomy, was independently associated with DFS and OS.

ABSTRACT

Background and objectives: The objective of this study is to investigate the outcomes of concurrent platinum-based chemoradiation therapy (CCRT), laparoscopic nerve-sparing ultra-radical hysterectomy (LNSURH), and open radical hysterectomy (ORH) on patients with locally advanced cervical carcinoma (LACC). Methods: A single-center retrospective study was conducted on LACC patients who received CCRT, ORH, or LNSURH from January 2011 to December 2019. Data on age, tumor size, overall survival (OS), disease-free survival (DFS), and early and late morbidities were collected. After 24 months of treatment, patients were asked a series of questions about their urinary, bowel, and sexual activities. Early morbidities were defined as those occurring during or within a month of treatment, whereas late morbidities and complications were defined as those occurring a month after treatment. The postoperative complications were classified with reference to the Clavien-Dindo classification (CD) system. Results: The Kaplan-Meier curves revealed no significant differences in OS and DFS among the three groups (P = 0.106 for DFS and P = 0.190 for OS). The rates of early complications in the CCRT group were comparable with those in the operated groups (P = 0.46). However, late complications were significantly lower in the ORH and LNSURH groups relative to those in the CCRT group. The scores of urinary and bowel functions were restored to the pretreatment state, although the sexual function scores were not satisfactory. Conclusions: The treatments of CCRT, ORH, and LNSURH can be considered options for patients with LACC, as their OS and DFS showed no significant difference. In addition, LNSURH exhibited a lower incidence of late complications and high sexual function scores.

ABSTRACT

BACKGROUND: A numerous studies have reported that obese patients (OP) are easily to have type 2 diabetes mellitus (T2DM). Although a variety of managements are available to treat such disorder, their efficacy is still limited. Previous studies have reported that laparoscopic sleeve gastrectomy (LSGT) can benefit OP with T2DM. However, no study specifically and systematically explores this topic. Thus, this study will assess the efficacy and complications of LSGT for the management of OP with T2DM. METHODS: The search strategy will be performed in the electronic databases from inception to the March 31, 2020 without limitations of language and publication time: PUBMED, EMBASE, Cochrane Library, Scopus, Web of Science, CINAHL, AMED, WANGFANG, VIP, and CNKI. Two authors will independently identify the articles, collect the data, and assess the risk of bias using Cochrane risk of bias tool. We will invite a third author to solve any differences between two authors. We will use RevMan 5.3 software to investigate the statistical analysis. RESULTS: This study will supply a high-quality synthesis of randomized controlled trials (RCTs) on the analysis of LSGT for the management of OP with T2DM. CONCLUSIONS: This study will help to build proposals that aim at providing high quality RCTs in the management of LSGT in OP with T2DM. SYSTEMATIC REVIEW REGISTRATION: INPLASY202040128.

ABSTRACT

Although buffer zones around aquatic areas are a useful method for controlling non-point source pollution and restoring natural ecosystem services, proper delineation methods for lakes remain poorly defined, restricting their protection and the rational utilization of resources. As the width of lake buffer zones should be set to meet the area's functional targets and requirements, this study proposes a methodology for delineating these zones that includes critical source areas for non-point source pollution and ecologically sensitive areas. The proposed method was tested on Zhushan Bay, Lake Tai, China given its poor environmental condition and a renewed focus on mitigation by the local government. Data sources and relative processing methods include vector data on land use and hydrographic networks processed by ArcGIS, digital elevation model (DEM) data with 30 m resolution, soil and socioeconomic data from local governmental statistical yearbooks, NPS pollution load into lake obtained by literatures and field survey. The results showed that a minimum buffer range could be practically determined while meeting the requirements of both environmental protection and economic needs, providing a theoretical and practical basis for the improved delineation and management of lake buffer zones in watersheds dominated by non-point source pollution.

ABSTRACT

With increasing awareness of the importance of wetlands, the number of new or restored wetlands in China is steadily growing; however, not all of them fulfill their expected ecological function. Maintaining wetlands in their optimal state is an urgent problem that requires research into the ecosystem evaluation, regulation, and biomass management of wetlands. The Ecopath with Ecosim (EWE) model, also known as the ecological channel model, is a balance model that can directly construct the ecological system structure and describe its energy flow and mass transfer through the principle of nutrition dynamics. Here, the EWE model is applied to determine the ecosystem status of a newly restored wetland, Zhushanhu wetland, in the Lake Tai buffer zone of Zhushan Bay, and evaluate the current ecological regulations and biomass control measures. Our results provide theoretical and scientific support for the management and maintenance of wetland ecological restorations.

ABSTRACT

OBJECTIVE: To evaluate the impact of closed high-pressure suction drainage on the healing of the perineal wound after abdominoperineal resection. METHODS: Patients undergoing rectal abdominoperineal resection in the Wuhan Tongji Hospital from January 2009 to January 2011 were randomized into two groups including the study group(n=61, closed high-pressure suction drainage) and the control group(n=59, presacral drainage). The drainage volume, primary healing rate, and the healing time of perineal wounds were compared. RESULTS: The total volume of the drainage in the first 3 days was (448.1±142.9) ml in the study group and (548.3±190.6) ml in the control group, the volume of the drainage on the third day was (28.1±12.7) ml and (125.9±84.3) ml respectively. The primary healing rate was 93.4%(57/61) in the study group and 74.6% (44/59) in the control group, the healing time was (13.5±3.5) days and (20.1±5.1) days respectively. CONCLUSION: Closed high-pressure suction drainage may promote perineal wound healing following rectal abdominoperineal resection.

ABSTRACT

The erythropoietin (EPO) belongs to the family of angiogenic factors, which is regulated by Hypoxia-inducible factor- 1α (HIF-1α). As known, EPO are expressed in human villi and decidua, but the function is not clear. In this study, we investigated the expression and roles of HIF-1α, EPO and its receptor (EPOR) in the biological functions of trophoblast and decidual stromal cell (DSC) in human early pregnancy. The expression of EPO, EPOR and HIF-1α was evaluated in the villi and deciduas by RT-PCR and immunohistochemistry. Thereafter, we silenced HIF-1α expression in HTR-8/SVneo cell line and decidual stromal cells (DSCs). The effects of EPO on the proliferation and apoptosis of trophoblasts and DSCs, and activation of signal molecules were investigated by BrdU proliferation assay, flow cytometry and western blot, respectively. We have observed that the HIF-1α silence results in the lower expression of EPO in trophoblasts and DSCs. The anti-EPO neutralizing antibody can inactivate the phosphorylation of STAT5 and activate p38 of these cells in a dosage-dependent manner. Furthermore, the expressions of EPO, EPOR and HIF-1α in the villi and decidua from the unexplained miscarriage were significantly lower than that of the normal early pregnancy. This study suggests that HIF-1α may regulate the expression of EPO, which plays a favorable regulatory role in the proliferation and survival of human first-trimester trophoblast cells and DSCs via inactivating p38 and activating STAT5 in an autocrine manner, while the inadequate EPO expression at maternal-fetal interface may lead to pregnancy wastage in humans.
