ABSTRACT

Objective To investigate the modeling elements of various types of animal models for acute liver injury,and to provide references and suggestions to establish and evaluate animal models of acute liver injury(ALI).Methods The animal experimental literature of ALI from 2002 to 2022 was searched in the databases of the China Knowledge Network,WanFang,Chongqing Vip(VIP),Chinese Medical Journal Full Text Data(Yiigle),and PubMed.The animal species,positive control drugs,modeling method,modeling drugs,and drug administration of the animal models of ALI in the literature were summarized.The result were analyzed using Excel,SPSS Modeler 18.0,and Cytoscape 3.8.2.Results A total of 896 articles were included in the databases.The most used animal models for ALI were male KM mice.The modeling method were mainly chemical liver injury,alcoholic liver injury,drug-related liver injury,and immune liver injury.①The corresponding main modeling method were intraperitoneal injection of 10 mL/kg of 0.1%CC14 in vegetable oil at 24 h before experiments,②gavage of 12.0 mL/kg of 50.0%~56.0%ethanol at 16 h before experiments,③intraperitoneal injection of 300 mg/kg APAP at 24 h before experiments,④tail vein injection of 20 mg/kg Con A at 8 h before experiments.Evaluation of the models was based on liver pathological indexes as the gold standard combined with biochemical indexes of serum ALT,AST,and SOD and MDA contents and activities in liver tissue homogenate as direct indicators.Conclusions Because the causes of ALI vary in clinical practice,the preparation of animal models of ALI should be based on the specific study content and characteristics,and the corresponding modeling method should be selected.

ABSTRACT

Objective:To optimize the ethanol reflux extraction process of total saponins in total saponins of Trillium tschonoskii Rhizome. Methods:On the basis of single factor tests, making the total extraction rate of three main compounds [paris saponin Ⅵ, paris saponin Ⅶ and pennogenin-3- O-α-L-rhamnopyranosyl-(1→4)-[O-α-L-rhamnopyranosyl (1→2)]- O-β-D-glucopyranoside (PRRG)] as the indicator, the optimal extraction parameter was selected with the main influencing factors: the ethanol concentration, solid-liquid ratio, and extraction time by the central composite design-response surface method.Results:The optimal extraction parameter for the ethanol extract of total saponins of Trillium tschonoskii Rhizome was as follows: ethanol concentration 69%, extraction time 1.9 h, and solid liquid ratio 1∶9.7. The binomial fitting complex correlation coefficient r = 0.966 1, and the deviation between the extracted predicted value and the actual value is 4.68%. Conclusion:The central composite design-response surface method is simple and reliable for the optimization of extraction process of total saponins of Trillium tschonoskii Rhizome.

ABSTRACT

Coumarins is a kind of important components in traditional Chinese medicine. Due to their special structure, they have significant pathological connections with many diseases. In this research, we established a rapid method to analyse the coumarins from Fructus citri Sarcodactylis by using the technique of ultra-performance liquid chromatography with quadruple time-of-flight mass spectrometry. Twenty-one coumarins were identified from Fructus citri Sarcodactylis, 10 were discovered for the first time. At the same time, a rapid sensitive method for identifying coumarins from Fructus citri Sarcodactylis was established.

Objective Summarize the anatomical features of the aortic dissection of right axillary and femoral artery cannulation in Sun's surgery.Exploring whether right axillary and femoral artery cannulation can reduce the incidence of organ malperfusion and improve patient prognosis.Methods From January 2015 to December 2017,181 cases with aortic dissection were treated in Beijing Anzhen Hospital affiliated to Capital Medical University,Beijing Great Blood Vessel Research and Treatment Center,122 patients were enrolled in the right axillary and femoral artery cannulation group,and 59 patients in control group were enrolled.Retrospective analysis the data of two groups,intraoperative,and postoperative univariate were compared between the two groups.Results A total of 17 patients died in the postoperative group,with 9 (7.38%) in the combined perfusion group and 8 (13.56%) in the control group,P =0.t81.Among the complications of the two groups,18 patients (14.75%) in combined perfusion group had renal insufficiency and 17 patients (28.81%) in the control group,P =0.025.0 case had stransient spinal cord injury in combined perfusion group,3 cases(5.08%) in control group,P =0.033.Conclusion Right axillary and femoral artery cannulation can reduce the incidence of complications of postoperative organ dysfunction and improve patient prognosis.

ABSTRACT

This study reported the analysis of plasma phospholipid metabolism of the rats and the pathological biomarkers between the type 2 diabetes model control group (MC) and the normal control group (NC). SD rats were randomly divided into 2 groups: NC and MC. To investigate state of plasma metabolite profiling in normal body, type 2 diabetes mellitus (T2DM) model group using UPLC-Q-TOF/MS which was used as analysis tool in this research. The compounds were identified by UPLC-Q-TOF/MS based on MS/MS fragment ions information, element composition in MassLynx 4.1 and the Lipid Maps database. The sign of two groups of samples in specific markers for screening was through a software package in R software (BioMark software). The results show that the pathological markers were mainly phosphatidylcholine (PC) and triglycerides (TG); the 2-acyl PC in the MC group was less more obviously than that in the NC group; high carbon number and high degree of unsaturation of the TG was reduced under the condition of type 2 diabetes. In the state of type 2 diabetes, metabolic changes occurred in rat plasma phospholipids obviously, which had a close relationship with the occurrence and development of T2DM.

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the styryllactone components in Goniothalamus cheliensis Hu (Annonaceae).</p><p><b>METHOD</b>UPLC-Q-TOF-MS was used to identify the main styryllactone components in G. cheliensis. The chromatographic separation was performed on ACQU ITY UPLC BEH C18 column and eluted by actonitrile and 0.1% acetic acid in water gradiently. The mass spectrometer equipped with electrospray ionization souce was used as detector under the positive ion modes.</p><p><b>RESULT</b>Twelve styryllactons were identified based on their MS data and published literatures, and the MS fragmentation regularity of the styryllactones was also proposed.</p><p><b>CONCLUSION</b>It is an accurate and effective method to obtain the structural information of styryllactones.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the flavonoids in Rhododendron mariae.</p><p><b>METHOD</b>The constituents in R. mariae were determined by UPLC/Q-TOF-MS. The chromatographic separation was performed on a C18 column (2.1 mm x 50 mm, 1.7 microm) with a gradient elufion of methanol-water containing 0.1% formic acid. The mass specfrometer eqaipped with elecfrospay ionizafion source was usedas defecfor and operated in data was collected under the positive and negative ion modes.</p><p><b>RESULT</b>Seven constituents were identified as myricetin-3-O-beta-D-gluconside (1), myricetin-3'-O-beta-D-xylopyraoside (2), hyperoside (3), isoquercitrin (4), avicularin (5), quercitroside (6) and quercetin (7).</p><p><b>CONCLUSION</b>In this study, the main flavonoids in R. mariae were separated by UPLC, and identified through the information of positive ion and negative ion and relative molecular mass which were determined by Q-TOF-MS. It is an accurate and effective method which can be applied for the constituent identification of R. mariae.</p>

ABSTRACT

AIM: To establish the quality standard for Xiangpu Ganmao Capsule (Herba Moslae, Cortex Magnoliale Officinalis, Radix Scutellariae). METHODS: Herba Moslae, Cortex Magnoliae Officinalis and Radix Scutellariale of Xiangpu Ganmao Capsule were identified by TLC. Thymol, magnolol and honokiol of capsule were determined by HPLC together. RESULTS: The characteristic identification by TLC was distict and highly specific. The quantitative evaluation of thymol had the linear range of 0.32-3.2 ?g, The average recovery was 99.44% and RSD was 1.3. The quantitative evaluation of magnolol had the linear range of 0.20-2.0 ?g. The average recovery was 99.78% and RSD was 1.58. The quantitative evaluation of honokiol had the linear range of 0.49-(4.89 ?g.) The average recovery was 99.41% and RSD was 0.51. CONCLUSION: The method is reliable, accurate and specific. It can be used for quality control of Xiangpu Ganmao Capsule.

AIM: To establish the quality standard for Tianqi Tongjing Capsule(Radix et Rhizoma Notoginseng, Rhizoma Corydalis, Fructus Foeniculi, etc.). METHODS: Rhizoma corydalis, Rhizoma Chuanxiong, Radix Aucklandiae Pollen Typhae, Faeces Trogopteror in Tianqi Tongjing Capsule were identified by TLC. Ginsenoside Rg1 wsa determined by HPLC. RESULTS: The average recovery was 98.66% and RSD was 0.56% (n=5), respectively. CONCLUSION: The method is reliable, accurate and specific. It can be used for quality control of Tianqi Tongjing Capsule.

ABSTRACT

AIM: To establish a quantitative method of determining thymol and carvacrol in Shushi Ganmao Ling(dripping pill)(Herba Moslae,Herba Artemisiae Annuae,Radix Scutellariae,etc.). METHODS: The GC method was used with HP-17 quartz capillary column(HP 50% PhMe Silicone,10 m?0.25 mm?0.25 ?m).The initial temperature of chromatographic column was at 130 ℃ for 14 min,then elevated to 220 ℃ with the rate of 20 ℃/min,and retention for 36 min. RESULTS: The linearities of thymol and carvacrol were respective in the range of 12.51 ?g/mL-125.1 ?g/mL(r=0.999 8) and 21.52 ?g/mL~215.2 ?g/mL(r=0.999 7).The average recoveries were 97.9%(RSD was 1.69%) and 100.6%(RSD was 2.27%).RSD of repeatability were 1.88% and 1.60%,respectively. CONCLUSION: The method is reliable,accurate and specific.It can be used for quality control of Shushi Ganmao Ling(dripping Pill).

AIM: To establish the quality standard for Tongfeng Huadu Tincture (Radix Gentianae Macrophyllae, Radix Angelicae Pubescentis, Rhizoma Chuanxiong, Herba Asari, etc.). METHODS: The Radix Gentianae Macrophyllae, Radix Angelicae Pubescentis, Rhizoma Chuanxiong, Herba Asari of Tongfeng Huadu Tincture were identified by TLC. Strychnine of Tincture was determined by HPLC. RESULTS: The average recovery was 97.07% and RSD and 2.1% (n=5). CONCLUSION: The method is reliable, accurate and specific. It can be used for quality control of Tongfeng Huadu Tincture.

ABSTRACT

[Objective] Molecular distillation technique was applied for the separation and purification of Atractylodes Lancea oil (ALO) to increase the yield of atractylodin. [Methods] Atractylodes Lancea oil prepared with supercritical extraction was refined by molecular distillation technique. The optimum refining conditions were selected by uniform design containing two factors of temperature and vacuum and five levels. The content of atractylodin from refined Atractylodes Lancea oil was measured and the remainder after isolation was detected by high performance liquid chromatography (HPLC). [Results] A yield rate of atraotylodin from Atractylodes Lancea oil was 52.17% at the temperature of 105℃ and under the vacuum of 100 Pa. [Conclusion] The application of molecular distillation technique makes it easy to collect the volatile atractylodin from Atractylodes Lancea oil at lower temperature and under higher vacuum; this method ensures the yield rate of atractylodin over 50% and has no problem of environmental pollution.
